Description

騒音曝露は、世界中の難聴の主な原因である。騒音曝露後には、通常蝸牛中の有毛細胞とシナプス接合を形成するらせん神経節神経線維などの蝸牛中の多様な構造に損傷が認められる。これらのシナプスはらせん神経節が音響情報を蝸牛から脳幹中の高次構造に伝達することを可能にする。騒音曝露後、有毛細胞は、神経突起における興奮毒性損傷につながる神経伝達物質を放出し、シナプス分裂や神経突起退縮をもたらす(Kujawa, S.G. et al., J. 本実施例は、内因性NAD+生合成経路の遺伝子による能力増強が騒音誘発難聴を回復させることを示す。ウォラー変性スロー(WldS)マウスは、NAD生合成酵素ニコチンアミドモノヌクレオチドアデニリルトランスフェラーゼ1およびUbe4aの遺伝子融合の3つの繰り返しを発現する(Coleman, M.P, et al., Proc. Natl. Acad. Sci. USA, 95, 9985-9990 (1998))。WldSは、ニューロン中に高く発現され、その発現は、軸索トランザクションの後の末端軸索フラグメントの変性を著しく遅らせる(Araki, T. et al., Science, 305, 1010-1013 (2004))。WldS発現は、種々の神経変性疾患にかかりやすいマウスの疾病表現型も緩和する(Samsam, M., et al., J. Claims (7)
- ニコチンアミドリボシドを含む、哺乳動物の難聴を緩和又は治療するための医薬製剤であって、難聴が騒音曝露によるものであり、難聴を起こす騒音曝露の前又は騒音曝露中に投与するための医薬製剤。
- ニコチンアミドリボシドが細胞中の酸化的損傷を抑制する請求項1に記載の医薬製剤。
- ニコチンアミドリボシドがSIRT3を活性化する請求項1又は2に記載の医薬製剤。
- 経口投与のための請求項1〜3のいずれか1項に記載の医薬製剤。
- ニコチンアミドリボシドが、らせん神経節神経細胞、内有毛細胞および外有毛細胞、支持細胞ならびにシュワン細胞からなる群より選ばれる1以上の細胞中の細胞内NAD+を増加させる請求項1に記載の医薬製剤。
- 注射による投与のための請求項1〜3のいずれか1項に記載の医薬製剤。
- 中耳洞への鼓室内注射による投与のための請求項1〜3のいずれか1項に記載の医薬製剤。
