Description
濾液の限外濾過は、50kDaのカットオフレベルを有する膜を用いて行われる。
適切な大きさのCellufine C−500カルボキシメチルゲル(Amicon)のカラムを用いて作製されたカチオン交換クロマトグラフィーを使用した。
分析的等電点電気泳動は、PhastシステムにおけるpH3から9用にPharmaciaにより製造された既製ゲルを用いて行われる。顕色は、クマシーブルーを用いて行われる。
アンソン(Anson)アッセイ
このアッセイにより、特異性なしでタンパク質分解活性を測定できる。該アッセイは、放出されたオリゴペプチドに含有されたチロシンの分光光度測定による、変性ヘモグロビンの加水分解を測定して行われる。
このアッセイは、オレンジ色素により染色されたカゼインの加水分解を440nmの分光光度測定により決められる。このアッセイは特異的でない。酵素とのインキュベーション後、残りのアゾカゼインはトリクロロ酢酸を用いて沈殿する。光学密度を測定することにより、放出されたペプチドの存在を確認でき、したがって酵素活性を確認できる。
エラスチンコンゴレッドアッセイ
このアッセイは、赤色色素により染色されたエラスチンの加水分解により放出された染色ペプチドを495nmで分光光度的に測定することに存する。
(N−Bloc−L−アラニネート−パラ−ニトロフェニルエステル) このアッセイは、放出されたパラニトロフェノールを判定して、エラスターゼに存在するエステラーゼ活性を347.5nmで分光光度的に測定して行われる(Vissier L.ら Biochim、Biophys Acta 268(1972)207.260)。
アゾコルアッセイ
このアッセイは、アゾコル(ボルドーレッド色素で染色されたコラーゲン粉末(ground collagen))の加水分解により放出されたペプチドを520nmで分光光度的に測定することに存する(Chavira、Analytical Biochemistry 136(1984) 446−450頁)。
組成物は、酵素複合体を添加する10kgのバッチに対して調製される:
小麦粉 2.5kg
ラクトース 4.5kg
ふすま 3kg
本発明による酵素複合体 0.2g/kg
このような組成物が、家禽用飼料1トン当たり0.5kgの割合で組み込まれる。
組成物は、酵素複合体が添加された20.1kgのバッチに対して調製され、次いで家禽用飼料に組み込まれる:
小麦粉 1.5kg
エンバク粉 4.5kg
セルロース 14kg
コロイダルシリカ 0.100kg
本発明による酵素複合体 0.1g/kg
組成物は、酵素複合体が添加された10kgのバッチに対して調製され、次いでブタ用飼料に組み込まれる:
小麦粉 4kg
ふすま 2kg
カゼイン 3kg
ジャガイモデンプン 1kg
本発明による酵素複合体 0.2g/kg
組成物は、
コメデンプン 0.5kg
ジャガイモデンプン 2.5kg
本発明による酵素複合体 0.2g/kg
を用いて同じように調製される。
組成物は、酵素複合体が添加される10kgのバッチに対して調製される:
トウモロコシ 5.65kg
大豆ミール 3.23kg
綿実粕 0.3kg
小麦胚芽粉 0.4kg
チョーク 0.13kg
リン酸二カルシウム 0.16kg
NaCl 0.03kg
アミノ酸複合体 0.1kg
本発明による酵素複合体 0.2g/kg
本発明による酵素複合体はまた、これらの調製物を補うために市販の家禽用飼料組成物に添加することもできる。
以下の組成物は、慣例に従って魚用飼料に調製され、組み込まれる。
魚粉 50.8%
動物性脂肪および油 28%
大豆ミール 9.9%
小麦粉 10%
ビタミン複合体(A、D、E、C、...) 1%
本発明による酵素複合体 0.3%
Claims (10)
- 家畜飼料を補うための、ストレプトマイセス・フラジエ(Streptomyces fradiae)菌株を培養して得られたプロテアーゼの混合物を含む酵素複合体の使用であって、プロテアーゼのこの混合物のうち、プロテアーゼの1つが7.0付近の等電点を有し、もう1つが8.0付近の等電点を有することを特徴とし、前記酵素複合体は、前記2つのタイプのプロテアーゼを優勢的に含み、前記2つのプロテアーゼの比率は前記混合物の活性の80%を超え、前記酵素複合体は、農場動物の全身状態を改善するための、飼料組成物との混合に適した粉末又は液体の飼料サプリメントとしての酵素複合体の使用。
- 等電点が7.0付近であるプロテアーゼが、タンパク質1mg当たり約150,000アンソン単位の比活性を有することを特徴とする、請求項1に記載の使用。
- 等電点が8.0付近であるプロテアーゼが、タンパク質1mg当たり約38,000アンソン単位の比活性を有することを特徴とする、請求項1に記載の使用。
- 粉末形態の酵素複合体が、均一化されるまで回転ドラム中で飼料組成物と乾燥混合されることを特徴とする、請求項2又は3に記載の使用。
- 液体形態の酵素複合体が、流動床中で飼料組成物に噴霧され、次いで顆粒化されることを特徴とする、請求項2又は3に記載の使用。
- 家禽用飼料における、請求項1〜5のいずれか一項に記載の使用。
- ブタ用飼料における、請求項1〜5のいずれか一項に記載の使用。
- 魚用飼料における、請求項1〜5のいずれか一項に記載の使用。
- 請求項1〜5のいずれか一項に記載の酵素複合体を含む、家畜用飼料組成物。
- 請求項1〜9のいずれか一項に記載の酵素複合体の製造方法であって、ストレプトマイセス・フラジエ菌株を培養すること、発酵ブロスを濾過すること、この後に酵素複合体を、限外濾過およびイオン交換クロマトグラフィーとこれに続く等電点電気泳動により抽出すること、最後に、こうして得られた複合体を凍結乾燥することを特徴とする、方法。
