Description
本発明の他の目的は、前記抗体−薬物結合体の製造方法を提供することである。
本発明のさらに他の目的は、前記抗体−薬物結合体を含む組成物を提供することである。
本発明のさらに他の目的は、前記抗体−薬物結合体を癌の疑いがある個体に投与する段階を含む、癌の治療方法を提供することである。
本発明のさらに他の目的は、前記抗体−薬物結合体を自己免疫疾患の疑いがある個体に投与する段階を含む、自己免疫疾患の治療方法を提供することである。
本発明のさらに他の目的は、前記抗体−薬物結合体で製造するのに適合した抗体を選別する方法を提供することである。
前記抗体−薬物結合体およびこれの構成要素に対しては上述したとおりである。
具体的に、前記製造方法は、α−アミン基と反応して架橋できる反応基を含む薬物と抗体を反応させて、抗体の重鎖または軽鎖のN−末端のα−アミン基に薬物を結合させる段階を含む。
具体的に前記製造方法で抗体と薬物は、pH4.0以上pH6.5以下、より具体的にはpH5.5以上pH6.5以下で連結され、より具体的にはpH6.0で連結される。上述したとおり、低いpHによって薬物、あるいはこれのリンカーに存在するアルデヒド基と抗体のN−末端α−アミンとの間の結合が特異的に起きる利点を持つ。
前記組成物は、前記抗体−薬物結合体を含む癌または自己免疫疾患治療用薬学的組成物の形態を含む。この場合、前記抗体は、特に癌細胞表面抗原または自己免疫疾患が発生した組織表面抗原に特異的に結合するものであってもよい。本発明の薬学的組成物は、薬学的に許容可能な担体をさらに含んでもよい。
前記方法は治療を必要とする個体に前記薬学的組成物を投与する段階を含む癌または自己免疫疾患を治療する方法であってもよく、この時、使用される抗体−薬物結合体および担体の種類は前記説明したものと同様である。
本発明の抗体−薬物結合体に対して代表的に抗体−サイトトキシン結合体を製造するために、リンカーが存在するサイトトキシンが位置選択的(Site selective)に抗体に結合するのか確認するために、抗−HER2抗体であるトラスツズマブ(Trastuzumab)、抗−CD56抗体であるロボツズマブ(Lorvotuzumab)、抗−CD30抗体であるブレンツキシマブ(Brentuximab)、および抗−GPNMB(glycoprotein NMB)抗体であるグレムバツムマブ(Glembatumumab)をモデル抗体として使用した。
前記抗体は、公知のアミノ酸配列情報を利用して発現ベクターを構築してCHO細胞株で安定発現株(stable cell line)構築、あるいは臨時発現を介して発現させて培養、精製した。
アルデヒドリンカーが末端に連結されたモノメチルオリスタチンF(Monomethyl Auristatin F、MMAF)毒素を合成した(レゴケムバイオサイエンスまたはXcessBioscience)(図1)。
また、MMAF外に他の種類の毒素にも本発明に係わるN−末端コンジュゲーション方法の適用の可能性を確認するために、モノメチルオリスタチンE(Monomethyl Auristatin E、MMAE)を合成して比較した(XcessBioscience、米国)。
3−1.本発明に係わるモノクローナル抗体−薬物結合体の製造
抗体を100mMリン酸カリウムpH5.49緩衝溶液に交換して、約7.1mg/mLに濃縮した。以後、アルデヒド反応基を持つリンカーと連結されているMMAF(レゴケムバイオサイエンス、韓国)を50%DMSO(Dimethyl sulfoxide)溶媒に溶かして2.5mg/mLになるようにした。それから最終70mMリン酸カリウムpH6.0、抗体濃度5.0mg/mL、14%DMSO、MMAF濃度0.3mg/mL、抗体のα−アミンとMMAFのモル比が約1:2.3(または、抗体とMMAFのモル比が1:9)になるように用意された抗体溶液とMMAF溶液を混合した。前記反応溶液にNaCNBH3(Sigma社、米国)を最終20mMになるべく添加した後、4℃で12時間徐々に撹はんさせながら反応させた。反応しなかった抗体およびリンカーと連結されたMMAFを分離するために、セファデックスG−25カラム(GEヘルスケア、米国)またはリソスフェニルカラム(Resource Phe、GEヘルスケア、米国)を使用した。このような過程で抗体一分子に約三つのMMAF毒素が抗体のアミノ末端に選択的に結合された形態を製造した(図2)。
既存技術で製造した対照群抗体−薬物結合体は、システイン結合(Cys conjugation,Thiomab(HC−A114C)+Mal−C6−MMAF)、チオール結合(Mal−C6−MMAF)、およびリシン結合(SMCCリンカー、SH−C6−MMAF)を介して実現した。
チオール結合抗体は、pH8.0でTCEPで抗体を還元させた後、Mal−C4−MMAFを入れて0℃で3時間反応させた。反応後システインを入れて追加反応させた後、反応を終結させて、G25 Desaltingカラム(GEヘルスケア、米国)を使用して1X PBSで置換して反応を終了した。
システイン結合抗体は、精製された抗体のシステインを活性化させた後、Mal−C6−MMAFを入れてチオール結合抗体と類似した過程を行ってコンジュゲーションを行った。
リシン結合抗体の結合体は、Immunogen社特許(WO2005037992)を参考に製造した。まず抗体とSMCCリンカーを反応させて、反応しなかったSMCCをバッファー交換によって除去した。抗体−SMCC結合体は、さらにチオール基を含むSH−C4−MMAF(Concortis bioscience、米国)と反応させて抗体−SMCC−MMAF結合体を製作した。
本発明に該当するα−アミン結合によって製造された抗体−サイトトキシン結合体を含んでまとめると、下記表1のとおりである。
4−1.分子量分析
抗体−薬物結合体(T−N−MMAF)の分子量をLC−MS分析によって確認した。使用した薬物(MMAF)の結合時分子量の理論値は、824.54Daで、トラスツズマブの分子量は145kDaである。したがって、質量分析によって薬物の抗体結合の有無と抗体一分子に結合した薬物の個数も同時に確認することができた。
実施例3により製造したT−N−MMAFに対してDARを決めるためにLC/MSで分子量を分析した。製造された試料は、PNGaseFで糖鎖を除去した後、ACQUITY UPLC BEH 200 SECカラムを介して分離後、Waters Synapt G2−Sシステムに試料を注入して質量を分析して、その結果を図3に示した。
その結果、図3に示した通り、薬物が一つも結合しなかった化学種(D0)から最高7個がついた化学種(D7)まで検出されて、付着した薬物の個数は各ピーク間分子量差が薬物の分子量と一致するか類似することに基づいて判断して、その相対含有量は下記の表2に示したとおりである。DARは、各化学種の加重平均で求めて、その値はDAR=3.2であった。
製造されたT−N_MMAF結合体で薬物が結合された位置は、ペプチドマッピングによって確認した。実施例3で製造したDAR3.2のT−N−MMAF ADCをRapigest(Waters)で変成させた後、トリプシン(Roche)で反応させて切片を作った。反応物は、ACQUITY UPLC PST(BEH) C18カラムで分離して分離した各ピークはWaters Synapt G2−S Q/TOFシステムを介して質量分析してどの配列に該当するのか決めて、その結果を図4に示した。
その結果、図4に示した通り、コンジュゲーション(conjugation)されなかった母抗体では発見されないピークがクロマトグラムで検出された。質量分析結果、該当切片は重鎖N−末端、軽鎖N−末端、重鎖一部分、そしてその他の小さい切片と確認されてその比率は下の表3に示したとおりである。したがって、全薬物のうち75%がN−末端に結合されていて、92%が明確に定義できる三地点であるN−末端と重鎖−CH2領域に選択的に結合されていると言える。
製造したT−N−MMAF結合体の凝集体(aggregate)含有量を確認するために、SE−HPLCおよびSDS−PAGE分析により純度分析を進行した。TSK−Gel3000SWXLカラムにPBSを移動相にしてサイズ排除クロマトグラフィーを行ってNovex NuPAGE 4−12%ゲルを利用してSDS−PAGEを行った。クロマトグラフィー結果について図5に示した。
その結果、図5に示したとおり、単量体の純度は98.8%であって効力試験に適した純度を確認して、フラグメンテーションや交差結合(cross−linking)等は観察されなかった。
薬物結合後にも抗原結合力が維持されるかを確認するために、ピアコアTM(BiacoreTM)を利用した表面プラスモン共鳴(surface plasmon resonance)を測定する方法で薬物が結合された抗体の抗原結合力を測定した。対照抗体としては、天然型抗体を使用した。抗原(ErbB2)結合力はピアコアT200を利用して分析してCM5センサーチップ(GEヘルスケア、米国)に各抗体をアミンカップリングキットを利用して固定化させた後、ErbB2(R&D systems)を50、16.67、5.56、1.85、0.62、0.21nMの濃度で30μL/min速度で注入してon/off速度を測定、分析してkD(M)を求めた。
製造した抗体−サイトトキシン結合体のin vitro効能を確認するために、HER2発現腫瘍細胞株であるBT474、HCC1954、SKOV−3、JIMT−1細胞株を利用して抗−増殖アッセイを行った。各細胞を培養して1*105細胞/mLでサスペンションして96ウェルプレートに100μLずつローディングした。細胞培養器で3時間培養後、ウェル当り様々な濃度区間の抗体−サイトトキシン結合体を各々100μLずつ入れて細胞培養器で4日間培養する。CCK−8(Dojindo)を1:10で希釈して各ウェルに処理後、ホイルで包んで細胞培養器に2〜5時間反応させた後SpectraMax190マイクロプレートリーダー(Molecular Device)を利用して450nmで吸光度を測定した。
6−1.In vitroヒト血清安定性
実施例3で製造したT−N−MMAFと天然型抗体、T−C−MMAF、Thiomab−MMAFなどの対照区を使用してIn vitroヒト血清安定性試験を行った。抗体−サイトトキシン結合体を1xPBSで緩衝液交換して3.33mg/mLに濃縮した後、抗体−サイトトキシン結合体とヒト血清(Sigma,米国)の比率(v/v)を1:9で混合して37℃で7日間静置した。7日後、保管試料はMabSelectSure(GEヘルスケア、米国)を使用して試料に含まれた抗体−サイトトキシン結合体以外のたんぱく質を除去して、LC/MS分析時干渉を最小化した。試験管条件でヒト血清での安定性はLC/MSで分析して結果を下の表6に示した。
ラットPK実験を介して生体内安定性を比較および確認した。3種ADCs(T−K−MMAF、T−C−MMAF、T−N−MMAF)とトラスツズマブを2.5mg/kg用量で雌Sprague−Dawleyラットに1回静脈注射した。物質投与後、0.05時間、0.5時間、1時間、6時間、24時間、72時間、168時間、240時間、336時間後に採血した。血液内ErbB2に結合する全ての抗体を分析する全抗体(Total antibody)分析法と薬物結合が維持されている抗体を分析する結合抗体(Conjugated antibody)分析法をELISA方法で実施した。
全抗体の含有量は、下記のようなELISA法で分析した:
96ウェルマイクロプレートをErbB2(R&D systems)でコーティングした後、試料をプレートに入れて37℃の温度で1時間反応した。PBSTを使用して固定されない全ての残余物を洗浄した後に、HRP結合された抗−ヒトカッパ軽鎖抗体(HRP−conjugated anti−Human kappa light chain antibody)と3,3’,5,5’−テトラメチルベンジジン(TMB,Sigma,T0440)を使用して450nmで吸光度を測定する方法で試料内全抗体量を決めた。
結合抗体分析法は、前述した方法と類似する方法で進行して抗−MMAF抗体(YOUNGINフロンティア)で96−ウェルマイクロプレートをコーティングした後、試料をプレートに入れて37℃の温度で1時間反応した。以後パイオチニル化されたErbB2 (ACROBIOSYSTEMS、米国)、ストレプトアビジン−HRP、TMBを順に入れて発色後450nmで吸光度を測定して結合抗体の濃度を決めて、その結果を図6〜8および下記の表7に示した。
互いに異なる技術で製作された3種のADCの効力と薬物−抗体比率(DAR)による効果の差を比較するために、ヌードラットを利用した乳癌(HCC 1954)異種移植モデルでin vivo効能テストを行った。
4種のADC、すなわちT−N−M(DAR約1.6、3.2)、T−C−M(DAR約3.7)、T−K−M(DAR約3.9)をHCC1954細胞が移植されたラットに各々1mg/kgの用量で単回静脈投与した後、各試験群間の移植腫瘍成長抑制程度を比較して、その結果を下記の図9および10に示した。
その結果、図9および10に示した通り、本発明に係わる抗体は、対照群および比較抗体結果と比較して抗癌効果が優れている。
ADCの製造機法によって変わる安定性が毒性に影響を与えるのかSDラットを利用した単回投与毒性試験を行った。3種類のADCを各々高用量である200mpkで静脈を介して単回投与した。比較群として単独抗体とMMAFも200mpkの用量の露出に合わせて投与した。体重は、試験物質投与時点から実験終了時(12日)まで毎日測定した。血液、生化学分析は、投与後5日に行われた。測定項目は、肝毒性と代表的な血液学的毒性確認のためにASTとALT、好中球(Neutrophil)、血小板(Platelet)である。
体重変化に対する結果を図11に示した。その結果、図11に示した通り、T−C−MMAF、T−K−MMAF群は、T−N−MMAF群および残りの群に比べて明確な体重減少を見せた。特にT−C−MMAF投与群は、8日目以後一匹を除いたすべての個体が死んだ。
肝毒性誘発の有無を比較してみるために、投与後5日目に採取した血液に対して血液生化学分析を行った。Au480(Beckman coulter、米国)化学分析器を利用して行ってそのうちの肝毒性の指標になるAST(Aspartate Aminotransferase)、ALT(Alanine Aminotransferase)の役価を図12に表示した。
その結果、図12に示した通り、本発明に係わるT−N−MMAF群は、PBSを含んだ他の対照群と大きい違いが生じない変化を見せて急激かつ深刻な肝毒性を誘発していないことを観察することができた。しかし、T−C−MMAF、T−K−MMAF群では相当な増加が観察されて薬物投与による肝毒性の発生を観察することができた。
現在許可を受けたADCの主な臨床的毒性が血液学的特性で現れるので、投与後5日目に採取した血液に対してHemavet 950 FS血液学分析器(Drew Scientific Inc.,米国)を利用して血液学的分析を行って、その結果を図13に表示した。
その結果、好中球(Neutrophil)の数字の場合、T−N−MMAF投与群はPBSを含んだ対照群に比べて著しい変化を見せなく急激で深刻な血液学的毒性は誘発されなかったと見られた。しかし、T−C−MMAF群では顕著な減少を見せてT−K−MMAF群の場合は顕著に増加した数値を見せたが、投与直後減少して再び上昇したと推定された。したがって、この二つの群に対しては薬物投与によって急激な血液学的毒性が誘発されたと結論を出すことができた。
血小板数値の場合、T−N−MMAF投与群は、PBSを含んだ他の対照群に比べて差を確認できる程度で小幅に減少した。しかし、T−C−MMAF、T−K−MMAF群の場合は、顕著な減少を見せて投与物質によって急激な毒性が発生したことを見せた。
本発明の抗体−薬物結合体製造方法が、様々な抗体−薬物結合体に使用できるか可能性を確認した。これのために、様々な薬物または様々な抗体および様々な抗体形態に適用して機能を確認した。
様々な薬物に対して前記本発明の抗体−薬物結合体製造方法が使用できるかを確認するために、代表的にモデル抗体としてトラスツズマブ抗体を利用して、様々な薬物でN末端結合を進行した。具体的にはMMAFとMMAEの2種の薬物を使用してMMAFを使用して得た結果は、先立って、いくつかの実施例に記述した。実施例3で記述した方法に従って抗体−薬物結合体を製造して各々製造した抗体−薬物結合体を収得してDAR分析およびin vitro安定性、Rat PKを前述した実施例の方法により実施した。
実施例3の方法に従って、MMAE(XcessBioscience、米国)と抗体との間の結合体を作って、DARを決めるためにLC/MSで分子量を分析した結果を図14および下記の表8に示した。
実施例6の方法に従って、T−N−MMAE ADCの血清内安定性を評価した。各試料でのADCの濃度は、ELISAを利用した全抗体分析法により測定してDARの変化はLC/MSを利用して測定した。
製造されたMMAE結合体の体内安定性を評価するために、SDラットを利用したPK測定試験を前述した実施例6と類似するように行った。それをまとめると、2.5mg/pkのADCを雌SDラットに投与して12分、30分、1時間、6時間、24時間、3日、7日、10日、14日、17日、21日後に採血してその濃度をELISA手法を使用して全抗体および結合抗体に分けて前述した方法で測定した。
製作されたMMAEの生物学的活性を確認するために、4種の腫瘍細胞株を使用してその活性を測定して、その結果を下記の表11に示した。使用した方法は実施例5と類似している。
様々な抗体に対して前記本発明の抗体−薬物結合体製造方法が使用できるか確認するために、3種の抗癌抗体(Brentuximab、Lorvotuzumab、Glembatumumab)にN末端結合を進行して、DAR分析およびin vitro安定性を測定した。
9−2−1−1.ブレンツキシマブ−N−MMAFの製造
CHO細胞株から発現させたブレンツキシマブを使用して、実施例3の方法によりブレンツキシマブ−N−MMAF(B−N−MMAF)を製造した。製造されたADCは、図16および下記の表12のようなLC/MSプロファイルを示して、D0−D6までの化学種が検出されて、DARは2.90と計算された。
コンジュゲーション(Conjugation)により抗体の特性が変わるかを判断するために、抗原に対する結合力をELISA方法で測定、比較した。抗原であるCD30(R&D systems)を96ウェルマイクロプレートに100μgコーティングした後、1%BSAで37℃、1時間ブロッキングした。ブロッキング溶液を除去した後、試料をプレートに入れて37℃、1時間反応した。PBST(PBS+0.05%ツイン20)で5回洗浄後、HRPコンジュゲーションされた抗−ヒトカッパ(kappa)軽鎖抗体を1000倍希釈してプレートに入れて37℃、1時間反応した。PBSTで5回洗浄した後、TMB(Sigma)を入れて10分発色した。1N H2SO4を入れて反応を停止させた後、450nmで吸光度を測定して、その結果を図17に示した。図17で、円(○)で表示した線はコンジュゲーションを行わなかったブレンツキシマブに対する結果、四角形(◇)で表示した線はDAR2.90のB−N−MMAFに対する結果、そして三角形(△)で表示した線はDAR4.22のB−N−MMAFに対する結果を示したものである。結果からコンジュゲーション実行後にもDAR値に関係なく抗原に対する結合力は変わらないことが分かった。
製造した抗体−サイトトキシン結合体のin vitro効能を確認するために、CD30発現細胞株であるKarpas−299、L−540細胞株を利用して抗−増殖アッセイを行った。
具体的に、各細胞を培養して1*105細胞/mLでサスペンションして96ウェルプレートに100μLずつローディングした。細胞培養器で3時間培養後、ウェル当り様々な濃度区間の抗体−サイトトキシン結合体を各々100μLずつ入れて細胞培養器で4日間培養した。CCK−8(Dojindo)を1:10で希釈して各ウェルに処理後、アルミホイルで包んで細胞培養器に2〜5時間反応した。SpectraMax190マイクロプレートリーダーを利用して450nmで吸光度を測定して、その結果を下記の表13に示した。
9−2−2−1.ロボツズマブ−N−MMAFの製造
CHO細胞から臨時発現したロボツズマブを使用して、前述した実施例3の方法によりロボツズマブ−N−MMAF(L−N−MMAF)を製造した。その結果、製造されたADCは図18および下記表14のようなコンジュゲーションプロファイルを見せて、DARは3.33と決められた。
コンジュゲーションによる抗体の特性変化があるのか調べるために、コンジュゲーション前後抗原に対する結合力が変わるかELISA方法を利用してテストした。抗原であるCD56(R&D systems、2408−NC−050)を96ウェルマイクロプレートに1μg/mLの濃度でコードした後、1%BSAで37℃、1時間ブロッキングした。ブロッキング溶液を除去した後、試料をプレートに入れて37℃の温度で1時間反応した。PBST(PBS+0.05%tween20)で5回洗浄した後、HRPコンジュゲーションされた抗−ヒトカッパ軽鎖抗体を1000倍希釈してプレートに入れて37℃、1時間反応した。PBSTで5回洗浄した後、TMB(Sigma)を入れて10分発色した。1N H2SO4を入れて反応を停止させた後、450nmで吸光度を測定して、その結果を図19に示した。図19で円(○)で表示した線は薬物とコンジュゲーションしていない抗体に対する結果を、三角形(△)で表示した線はDAR2.5のL−N−MMAFに対する結果を、四角形(◇)で表示した線はDAR3.3のL−N−MMAFに対する結果を示したものである。その結果、DAR程度に関係なくその結合力が維持されていることが分かった。
製造した抗体−サイトトキシン結合体のin vitro効能を確認するために、OPM−2細胞株を利用して抗−増殖アッセイを行った。各細胞を培養して1*105細胞/mLでサスペンションして96ウェルプレートに100μLずつローディングした。細胞培養器で3時間培養後、ウェル当り様々な濃度区間の抗体−サイトトキシン結合体を各々100μLずつ入れて細胞培養器で4日間培養した。CCK−8(Dojindo)を1:10で希釈して各ウェルに処理後、ホイルで包んで細胞培養器に2〜5時間反応した。SpectraMax190マイクロプレートリーダー機を利用して450nmで吸光度を測定して、その結果を表15に示した。
9−2−3−1.In vitro細胞毒性
製造した抗体−サイトトキシン結合体のin vitro効能を確認するために、皮膚癌細胞株であるSK−MEL−2細胞株を利用して抗−増殖アッセイを行った。各細胞を培養して1*105細胞/mLでサスペンションして96ウェルプレートに100μLずつローディングした。細胞培養器で3時間培養後、ウェル当り様々な濃度区間の抗体−サイトトキシン結合体を各々100μLずつ入れて細胞培養器で4日間培養した。CCK−8(Dojindo)を1:10で希釈して各ウェルに処理後、ホイルで包んで細胞培養器に2〜5時間反応した。SpectraMax190マイクロプレートリーダー機を利用して450nmで吸光度を測定して、その結果を表16に示した。
Claims (16)
- 抗体の重鎖または軽鎖N−末端アミノ酸残基に細胞毒性薬物が結合された抗体−薬物結合体(ADC)であって、前記細胞毒性薬物は、前記抗体の重鎖または軽鎖のN−末端のα−アミン基に結合され、前記細胞毒性薬物は、アルデヒド反応基を持つリンカーを介して前記抗体に結合される、前記抗体−薬物結合体。
- 前記抗体は、全長抗体または抗原結合ドメインを含む抗体断片を含むことを特徴とする請求項1に記載の抗体−薬物結合体。
- 前記抗体は、IgG、scFv、Fv、Fab、Fab’およびF(ab’)2からなる群から選択された形態を持つことを特徴とする請求項2に記載の抗体−薬物結合体。
- 前記細胞毒性薬物は、マイクロチューブリン構造形成抑制剤、有糸分裂抑制剤、RNA重合酵素抑制剤、トポイソメラーゼ抑制剤、DNAインターカレーター、DNAアルキレーター、リボソーム抑制剤、放射線同位元素および毒素からなる群から選択されることを特徴とする請求項1に記載の抗体−薬物結合体。
- 前記細胞毒性薬物は、メイタンシノイド、オリスタチン、トラスタチン、チュブリシン、カリケアミシン、ピロロベンゾジアゼピン、ドキソルビシン、テュオカルマイシン、カルボプラチン(パラプラチン)、シスプラチン、シクロホスファミド、イホスファミド、ニドラン、窒素マスタード(メクロレタミン塩酸塩)、ブレオマイシン、ミトマイシンC、シタラビン、フルオロウラシル、ゲムシタビン、トリメトキサート、メトトレキサート、エトポシド、ビンブラスチン、ピノレルビン、アリムタ、アルトレタミン、プロカルバジン、タキソール、タキソテレ、トポテカン、イリノテカン、トリコテセン、CC1065、α−アマニチン、他のエンジイン抗生物質、外毒素および植物毒素からなる群から選択されることを特徴とする請求項4に記載の抗体−薬物結合体。
- 前記オリスタチンは、モノメチルオリスタチンEまたはモノメチルオリスタチンFであることを特徴とする請求項5に記載の抗体−薬物結合体。
- 前記抗体は、癌細胞表面抗原に特異的に結合することを特徴とする請求項1に記載の抗体−薬物結合体。
- 前記癌細胞表面抗原は、CD19、CD20、CD30、CD33、CD37、CD22、CD56、CD70、CD74、CD138、Muc−16、mesothelin、HER2、HER3、GPNMB、IGF−1R、BCMA、PSMA、EpCAMおよびEGFRからなる群から選択されることを特徴とする請求項7に記載の抗体−薬物結合体。
- 前記抗体は、抗−HER2抗体、抗−CD30抗体、抗−CD56抗体、抗−GPNMB抗体からなる群から選択されることを特徴とする請求項1に記載の抗体−薬物結合体。
- 前記抗体は、トラスツズマブ、ロボツズマブ、ブレンツキシマブ、およびグレムバツムマブからなる群から選択されることを特徴とする請求項9に記載の抗体−薬物結合体。
- 抗体の重鎖または軽鎖N−末端アミノ酸残基に免疫抑制剤が結合された抗体−薬物結合体(ADC)であって、前記免疫抑制剤は、前記抗体の重鎖または軽鎖のN−末端のα−アミン基に結合され、前記免疫抑制剤は、アルデヒド反応基を持つリンカーを介して前記抗体に結合される、前記抗体−薬物結合体。
- α−アミン基と反応して架橋できる反応基を含む細胞毒性薬物または免疫抑制剤と抗体を反応させて、抗体の重鎖または軽鎖のN−末端のα−アミン基に細胞毒性薬物または免疫抑制剤を結合させる段階を含むことを特徴とする請求項1〜11のいずれか一項に記載の抗体−薬物結合体の製造方法。
- 前記製造方法は、結合体を形成しなかった抗体および細胞毒性薬物または免疫抑制剤を含む反応産物から抗体−薬物結合体を分離する段階をさらに含むことを特徴とする請求項12に記載の製造方法。
- 前記α−アミン基と反応して架橋できる反応基は、イソチオシアネート、イソシアネート、アシルアジド、NHSエステル、スルホニルクロリド、アルデヒド、グリオキサール、エポキシド、オキシラン、カルボネート、アリールハライド、イミドエステル、カルボジイミド、アンハイドライドおよびフルオロフェニルエステルからなる群から選択されることを特徴とする請求項12に記載の製造方法。
- 請求項1〜10のいずれか一項に記載の抗体−薬物結合体を含む、癌治療用薬学的組成物。
- 請求項11に記載の抗体−薬物結合体を含む、自己免疫疾患の治療用薬学的組成物。
