Abstract
본 발명은 암세포 표면 항원에 대한 항체의 N-말단 아미노 그룹에 위치 특이적으로 싸이토톡신이 결합된 항체-싸이토톡신 결합체 및 이의 제조방법에 관한 것이다. 또한 관련된 병리적 질환을 시험관내, 계내 및 생체 내에서 진단 또는 치료하기 위해 상기 항체-싸이토톡신 결합체를 사용한 암 진단용 또는 치료용 조성물에 관한 것이다. 본 발명의 항체-싸이토톡신 결합체를 포함하는 암치료용 조성물을 이용하여, 종양 세포 특이적 표적화를 높이고 강력한 세포독성을 통해, 항체 또는 싸이토톡신 단독 치료요법에 비해 증가된 항체의존성 세포독성 및 부작용 완화의 효과를 나타낼 수 있다. 또한, 결합체가 동질성을 유지하기 때문에 기존 항체-싸이토톡신 결합체의 이질성 문제를 극복할 수 있으며, 유전자 조작되지 않은 항체를 사용하여 항체의 N-말단, α-아민에 싸이토톡신을 결합시키기 때문에 보다 효과적인 항체-싸이토톡신 결합체 개발에 활용될 수 있을 것이다.
Description
본 발명은 암세포 표면 항원에 대한 항체의 N-말단 아미노 그룹에 위치 특이적으로 싸이토톡신이 결합된 항체-싸이토톡신 결합체 및 이의 제조방법에 관한 것이다. 또한 관련된 병리적 질환을 시험관내 및 생체 내에서 진단 또는 치료하기 위해 상기 항체-싸이토톡신 결합체를 사용한 암 진단용 또는 치료용 조성물에 관한 것이다.
종래에 암을 치료하고자 할 때, 종양 세포를 억제시키거나 사멸시키기 위하여 세포독성제 또는 세포증식 억제제 등의 역할을 하는 싸이토톡신(cytotoxin)을 단독으로 사용하였다. 하지만, 메이탄시노이드와 오리스타틴 등으로 대표되는 싸이토톡신은 세포독성(cytotoxicity)은 강하지만 종양에 대한 선택성이 낮아 종양 세포뿐만 아니라 정상 세포에 대해서도 허용 가능하지 않은 수준의 독성을 야기시킨다는 문제점을 가지고 있었다. 이러한 문제점 때문에 싸이토톡신을 암 치료 임상에 사용하는 것이 극도로 제한되어 왔으며, 실제로 중추 신경계와 위장계에 대한 심각한 부작용으로 인해 임상 시도가 중단되기도 하였다.
싸이토톡신의 독성으로 인한 부작용을 최소화하고 치료 효과를 증대시키고자, 종양 세포에 대한 싸이토톡신의 특이적 표적화 기술이 연구되었다. 싸이토톡신의 표적화 수치를 높이기 위해 종양 세포 특이적 항원과 결합하는 항체에 싸이토톡신을 접합시키는 실험이 시도되었다. 인간 결장직장암에 대해 단일클론 항체 C242에 연결된 메이탄시노이드를 포함하는 면역결합체를 처리한 경우에 배양된 결장암 세포에 대해 강력한 세포독성을 나타내었으며, 생체 내 종양 성장 검정에서 항종양 활성을 나타내었다(Liu et al., Proc. Natl. Acad. Sci. USA 93:3618-8632 (1996)). 또한, 메이탄시노이드를 디설파이드 링커를 통하여, 인간 결장 암 세포주 상의 항원과 결합하는 쥐의 항체 A7과 결합시키거나 또는 HER-2/neu 종양 유전자와 결합하는 또다른 쥐의 단일클론 항체 TA.1과 결합시킨 면역결합체를 제조하려는 시도도 있었다(Chari et al., Cancer Research 52:127-131 (1992)). CD30에 특이적인 cAC10 항체에 연결된 오리스타틴E를 포함하는 면역결합체가 보고되었고(Klussman et al., Bioconjugate chemistry 15(4):765-773 (2004)), 리툭산과 같은 항-CD20항체에 연결된 오리스타틴E를 포함하는 면역결합체에 대하여 보고된 바 있다(WO 04/032828).
종양 세포 항원과 특이적으로 결합하는 항체에 싸이토톡신을 접합한 결합체(antibody-drug conjugates, ADC)의 제조 시, 통상적으로 공유 결합을 통해 접합되며, 일반적으로 싸이토톡신이 항체에 존재하는 여러 부위에 부착되어 이질성 혼합물을 생성시킨다. 예를 들어, 제넨텍사(Genentech)에서 개발중인 T-DM1의 경우, 싸이토톡신이 항체에 존재하는 다수의 라이신 잔기에 접합하게 되는데, 반응조건에 따라 싸이토톡신이 0개부터 8개까지 접합된 이질성 결합체를 생성하며, 또한 싸이토톡신이 접합되는 라이신 잔기의 위치에 있어서도 이질성을 가지게 된다(WO 01/00244).
항체에 디티오트레이톨(DTT), 트리카르보닐에틸포스핀(TCEP) 등의 환원제를 사용하여 인위적으로 유리 티올을 생성시킨 후 싸이토톡신을 접합하는 씨애틀제네틱스사(Seattle genetics)의 면역결합체 제조 방법은 제넨텍사의 제조방법과 차이가 있지만, 0개부터 8개까지 접합된 싸이토톡신의 개수에 따른 이질성 결합체를 생성하며, 접합되는 유리 티올의 위치에 있어서도 이질성을 나타낸다(US7659241). 또한, 인위적으로 유리 티올을 유도하는 결합체 제조방법은 항체의 3차 구조를 손상시켜 항체의 항원 결합 특이성을 약화시키거나 상실시킬 수 있다(Zhang et al., Anal. Biochem. 311:1-9 (2002)).
종양 세포 표적화를 위한 항체와 싸이토톡신의 결합체 제조 시, 동질성을 높인 면역 결합체를 제조하기 위하여 항체의 중쇄에 인위적으로 시스테인을 삽입하고, 삽입된 시스테인의 티올 잔기에 선택적으로 싸이토톡신을 접합시키는 면역 결합체 제조 방법을 제넨텍사에서 개발하였다(US7521541). 2개의 시스테인이 삽입된 항체를 이용하여 2개의 싸이토톡신을 원하는 위치에 결합하여 동질성을 가진 면역 결합체를 제조하였지만, 인위적으로 삽입한 시스테인의 티올 잔기가 반응성을 가지도록 활성화시키기 위하여 항체의 환원과정과 산화과정을 거쳐야 하는 다단계 접합 공정을 따르고 있으며, 시스테인을 삽입한 항체이기 때문에 배양, 정제 과정 및 티올 활성화 과정에서 단백질 이량체 또는 다량체를 형성할 수 있는 문제점을 가지고 있다. 또한, 인위적으로 유전자 조작된 항체를 사용하여 면역결합체를 제조해야 하므로, 인체 내에서 면역원성(immunogenicity)을 가질 위험성을 가지며 항체의 생산성 또한 유전자 조작되지 않은 항체에 비해 현저히 낮아질 수 있는 단점을 가지고 있다.
이러한 배경 하에, 본 발명자들은 동질성을 갖는 동시에 면역원성이 적도록 항체와 싸이토톡신을 결합시키기 위해 예의 노력한 결과, 링커를 갖는 싸이토톡신과 유전자 조작되지 않은 단일 클론 항체를 사용하여 싸이토톡신의 항체 접합 개수와 위치가 동질성을 가지는 항체-싸이토톡신 결합체를 제조하여 본 발명을 완성하였다.
본 발명의 하나의 목적은 암세포 표면 항원에 대한 항체의 N-말단 아미노 그룹에 위치 특이적으로 싸이토톡신이 결합된 항체-싸이토톡신 결합체를 제공하는 것이다.
본 발명의 다른 목적은 상기 항체-싸이토톡신 결합체를 제조하는 방법을 제공하는 것이다.
본 발명의 또 다른 목적은 상기 항체-싸이토톡신 결합체와 약제학적으로 허용가능한 담체를 포함하는 암치료용 조성물을 제공하는 것이다.
본 발명의 또 다른 목적은 상기 항체-싸이토톡신 결합체 또는 상기 조성물을 개체에 투여하는 단계를 포함하는 암치료 방법을 제공하는 것이다.
상기 목적을 달성하기 위한 하나의 양태로서, 본 발명은 암세포 표면 항원에 대한 항체의 N-말단 아미노 그룹에 위치 특이적으로 싸이토톡신이 결합된 항체-싸이토톡신 결합체를 제공한다.
본 발명에서 용어, “항체-싸이토톡신 결합체”란 항체와 싸이토톡신의 생물학적 활성을 저하시키지 않으면서 싸이토톡신에 연결된 링커를 통하여 항체와 싸이토톡신을 화학적으로 연결한 형태를 지칭한다. “링커”는 항체를 싸이토톡신에 공유 결합시키는 원자쇄를 포함하는 화학적 부분을 의미한다. 링커는 싸이토톡신과 연결된 형태로 제조되며, 링커 말단에는 항체와 연결시킬 수 있는 반응기를 갖는다.
본 발명에서 용어, "항체"란 면역학적으로 특정 항원과 반응성을 갖는 면역글로불린 분자를 포함하며, 다클론 항체 및 단일클론항체를 모두 포함한다. “다클론 항체”란 상이한 결정기(에피토프)에 대항하여 발생된 상이한 항체를 지칭하며, "단일클론 항체"란 단일 항원성 부위에 대하여 발생된 항체로, 실질적으로 동질적 항체 집단, 즉 집단을 차지하고 있는 개개의 항체가, 미량으로 존재할 수 있는 천연 발생적 돌연변이를 제외하고는 동일한 집단으로부터 수득한 항체를 지칭한다.
본 발명에서 항체는 IgA, IgD, IgE, IgM 및 IgG를 포함하며, IgG는 아형(subtype)으로, IgG1, IgG2, IgG3 및 IgG4를 포함한다. 또한, 항체 단편을 포함할 수 있다. 항체 단편은 항원 결합 기능을 보유하고 있는 단편을 의미하며, Fc 단편, Fab, Fab', F(ab')2 및 Fv 등을 포함한다. 상기 Fc 단편은 Fc 수용체와 같은 세포 표면 수용체와 결합할 수 있는 항체의 말단 부위를 의미하며, 항체의 두 개의 중쇄의 2번 또는 3번째 보존 도메인(constant domain)으로 구성된다. 상기 Fab는 경쇄 및 중쇄의 가변영역과 경쇄의 불변 영역 및 중쇄의 첫 번째 불변 영역(CH1)을 가지는 구조로 1개의 항원 결합 부위를 가진다. Fab'는 중쇄 CH1 도메인의 C 말단에 하나 이상의 시스테인 잔기를 포함하는 힌지 영역(hinge region)을 가진다는 점에서 Fab와 차이가 있다. F(ab')2 항체는 Fab'의 힌지 영역의 시스테인 잔기가 디설파이드 결합을 이루면서 생성된다. Fv(variable fragment)는 중쇄 가변부위 및 경쇄 가변부위만을 가지고 있는 최소의 항체조각을 의미한다. 이중쇄 Fv(dsFv, disulfide-stabilized variable frament)는 디설파이드 결합으로 중쇄 가변부위와 경쇄 가변부위가 연결되어 있고 단쇄 Fv(scFv, single chain variable frament)는 일반적으로 펩타이드 링커를 통하여 중쇄의 가변 영역(VH)과 경쇄의 가변 영역(VL)이 공유 결합으로 연결되어 있다. 본 발명에서 상기 scFv는 완전한 항원-인식 및 항원-결합 부위를 갖는 최소 항체 단편을 의미하며, 항체의 VH 및 VL 도메인을 포함하고, 여기서 이 도메인은 단일 폴리펩타이드쇄에 존재한다. 상기와 같은 항체 단편은 단백질 가수분해 효소를 이용해서 얻을 수 있다(예를 들어, 전체 항체를 파파인으로 제한 절단하며 Fab를 얻을 수 있고 펩신으로 절단하면 F(ab')2 단편을 얻을 수 있다).
바람직하게는 천연형 항체일 수 있다. 천연형 항체란 유전자 조작을 가하지 않은 항체를 뜻하며, 생체 내에서 유전자 조작된 항체가 지닐 수 있는 면역원성(immunogenicity)의 위험성이 현저히 낮다. 본 발명의 목적상 유전자 조작이 되지 않은 천연형 항체로 상기 항체-싸이토톡신 결합체를 제조하는 것이 바람직하며, 이와 같은 천연형 항체 사용으로 동질성 및 생체 내에서 면역원성의 위험성이 낮아져, 암조직만 특이적으로 사멸시키고 그 이외에 불필요한 면역반응을 일으키지 않을 수 있다.
본 발명에서 용어, "유전자 조작"이란 아미노산 서열을 변화시키는 행위로서, 해당 아미노산 서열을 코딩하는 본래 서열 폴리펩타이드와 어느 정도 상이한 아미노산 서열을 갖는 폴리펩타이드에 대한 조작을 포함한다. 아미노산 서열 변이체는 본래 아미노산 서열 내의 특정 위치에서 아미노산이 치환, 결실되거나 외부 삽입된 아미노산 서열을 함유하게 된다.
본 발명에서 용어, "암세포 표면 항원"이란 정상세포에는 생성되지 않거나 생성되더라도 세포 표면에 노출되어 있지 않은 물질이 암세포에서만 특이적으로 세포 표면에 노출되어 있는 물질을 말한다. 해당 물질이 항체에 의해 인식되는 경우 이를 항원이라고 표현한다.
바람직하게는, 본 발명의 암세포 표면 항원은 본 발명의 항체가 특이적으로 인식할 수 있는 암세포 표면 물질은 제한없이 포함되나, 그 예로 CD19, CD20, Her2, IGF-1R, EpCAM (Epithelial cell adhesion molecule) 및 EGFR (epidermal growth factor receptor)로 이루어진 군에서 선택된 것일 수 있으며, 보다 바람직하게는 Her2, IGF-1R, 및 EpCAM로 이루어진 군에서 선택된 것일 수 있다. 그러나, 이에 제한되는 것은 아니다. 본 발명의 일실시예에서는 Her2, IGF-1R, 및 EpCAM를 인식하는 anti-HER2 항체, anti-IGF-1R 항체, 그리고 anti-EpCAM 항체를 모델항체로 사용하였다(실시예 1).
본 발명에서 용어, "위치 특이적 결합"은 결합되는 두 물질의 결합을 이루는 반응기 위치가 정해져 있는 것을 의미한다. 곧, 단백질의 경우에는 특정 아미노산의 특정 반응기만이 결합에 참여하는 경우를 말하며, 화합물의 경우에도 비슷한 여러 반응기 중에서도 3차원적으로 특정 위치에 존재하는 반응기만이 결합에 참여하는 경우를 말한다. 본 발명의 항체-싸이토톡신 결합체는 항체의 N-말단에 존재하는 아미노기가 싸이토톡신과 결합하는바 위치 특이적 결합이 일어난다. 특히, 아미노기에 존재하는 α-아민이 싸이토톡신에 연결된 링커에 존재하는 알데히드기와 위치 특이적으로 결합을 일으킬 수 있다.
본 발명에서 용어, "동질성"은 두 물질이 결합되어 형성되는 결합체에 있어서 두 물질의 결합 비율 및 결합 위치가 균질한 경우를 말한다. 결합체가 동질성을 갖게 되면 전체적으로 균질하게 되고 투여량에 따른 효능을 정확하게 측정할 수 있어 투여량, 투여 횟수 등을 표준화 할 수 있게 된다. 본 발명에서는 항체-싸이토톡신 결합체를 제조함에 있어 항체에 결합하는 싸이토톡신의 개수가 일정하고, 결합이 일어나는 위치가 일정하게 유지되는 성질을 의미한다. 바람직하게는 항체의 N-말단 아미노 그룹 위치에 싸이토톡신에 결합된 링커의 알데히드기가 결합하며 각 항체마다 4개의 싸이토톡신이 결합할 수 있다.
본 발명에서 용어, “싸이토톡신”이란 세포독성 또는 세포증식 억제성 효과를 갖는 임의의 화합물을 지칭한다. “세포독성”이란 세포의 기능을 억제하거나 저하시켜 세포의 파괴를 야기시키는 효과를 의미하며, “세포증식 억제성”이란 세포성장 또는 세포 증식의 제한 등과 같이 세포 생장 기능을 제한하는 효과를 의미한다.
본 발명에서 싸이토톡신은 마이크로튜불린 억제제, 유사분열 억제제, 토포이소머라아제 억제제 및 DNA 인터컬레이터(intercalators) 등의 기능을 할 수 있는 화학요법제, 효소적으로 기능할 수 있는 단백질 독소 및 방사선 동위원소를 포함한다. 바람직하게는, 메이탄시노이드, 오리스타틴, 돌라스타틴, 트리코테센(trichothecene), CC1065, 칼리케아미신 및 다른 에네디인 항생제, 탁산, 안트라시클린 및 그의 입체이성질체, 유도체가 될 수 있다. 보다 바람직하게는, 도 1에서 개시하고 있는 Monomethyl Auristatin F (MMAF)일 수 있다. 다만, 이에 제한되는 것은 아니다.
본 발명에서 상기 싸이토톡신은 링커를 포함할 수 있다. 본 발명에서 용어, “링커”는 항체에 싸이토톡신을 공유 결합시키는 원자쇄를 포함하는 화학적 부분을 의미한다. 링커는 싸이토톡신과 연결된 형태로 제조되며, 링커 말단에는 항체와 연결시킬 수 있는 반응기를 갖는다. 바람직하게는, 링커는 알데히드기를 반응기로 가지며, 항체의 N-말단 아미노 그룹의 α-아민에 위치 특이적으로 결합하는 특성을 갖는 링커일 수 있다.
바람직하게는, 본 발명의 항체-싸이토톡신 결합체는 종양 세포 항원과 특이적으로 결합하는 항체의 N-말단의 아미노기에, 알데히드 반응기를 가지는 링커와 연결된 싸이토톡신을 위치 특이적 및 개수 특이적으로 결합한 면역결합체이다.
알데히드 반응기는 비특이적 반응을 최소화하여 항체의 N-말단의 아미노기, 특히, α-아민에 싸이토톡신을 결합시키기에 효과적이다. 알데히드 결합에 의한 환원성 알킬화로 생성된 최종 산물은 아미드 결합으로 연결된 것보다 훨씬 안정적이다. 알데히드는 낮은 pH에서 N-말단 α-아민과 선택적으로 반응하는 특성을 가진다. 이에 따라, 본 발명자들은 항체의 α-아민에 위치 특이적으로 싸이토톡신을 연결하기 위하여 결합 반응 시 pH6.0 이하로 반응시킬 경우, 싸이토톡신이 라이신 잔기에 존재하는 ε-아민과 결합하는 것을 최소화할 수 있음을 알아냈다. 이를 통해, 상기 결합체는 항체의 N-말단 α-아민에 위치 특이적으로 싸이토톡신이 연결된 동질성을 가지게 된다. 따라서, 기존 항체와 싸이토톡신의 결합체의 결합의 개수 및 위치의 이질성 때문에 생기는 일률적인 약효 및 질을 담보할 수 없다는 문제점을 극복하였다.
또 하나의 양태로서, 본 발명은 (1) 알데히드 반응기를 갖는 링커와 연결된 싸이토톡신과 항체를 반응시켜서 싸이토톡신과 항체의 N-말단의 아미노 그룹과 연결시키는 단계; 및 (2) 상기 (1)단계의 반응 혼합물로부터 결합체를 형성하지 않은 항체 및 싸이토톡신을 제거하여 항체-싸이토톡신 결합체를 분리 및 정제하는 단계를 포함하는, 상기 항체-싸이토톡신 결합체를 제조하는 방법을 제공한다.
항체, 싸이토톡신, 및 항체-싸이토톡신 결합체는 상기에서 설명한 바와 같다.
바람직하게는 (1)의 항체와 싸이토톡신-링커는 pH4.0 이상 pH6.5 이하에서 연결되며, 더욱 바람직하게는 pH5.5 이상 pH6.5 이하에서 연결되며, 가장 바람직하게는 pH6.0에서 연결된다. 상기에서 서술한 바와 같이 낮은 pH에 의해서 링커에 존재하는 알데히드기와 항체의 N-말단 α-아민 결합이 특이적으로 일어나게 된다.
또 하나의 양태로서, 본 발명은 상기 항체-싸이토톡신 결합체와 약제학적으로 허용가능한 담체를 포함하는 암치료용 약학적 조성물을 제공한다.
본 발명에서 용어, "암"은 암의 종류를 제한 없이 포함하나, 그 예로 식도암, 위암, 대장암, 직장암, 구강암, 인두암, 후두암, 폐암, 결장암, 유방암, 자궁 경부암, 자궁 내막체암, 난소암, 전립선암, 고환암, 방광암, 신장암, 간암, 췌장암, 골암, 결합 조직암, 피부암, 뇌암, 갑상선암, 백혈병, 호지킨(Hodgkin) 질환, 림프종, 또는 다발성 골수종혈액암일 수 있다. 상기 암세포 표면 항원에 특이적인 항체의 종류에 따라 치료할 수 있는 암을 선택할 수 있다.
본 발명에서 용어, "약학적으로 허용가능한 담체"란 생물체를 자극하지 않고 투여 화합물의 생물학적 활성 및 특성을 저해하지 않는 담체 또는 희석제를 말한다. 액상 용액으로 제제화되는 조성물에 있어서 허용되는 약학적 담체로는, 멸균 및 생체에 적합한 것으로서, 식염수, 멸균수, 링거액, 완충 식염수, 알부민 주사용액, 덱스트로즈 용액, 말토 덱스트린 용액, 글리세롤, 에탄올 및 이들 성분 중 1 성분 이상을 혼합하여 사용할 수 있으며, 필요에 따라 항산화제, 완충액, 정균제 등 다른 통상의 첨가제를 첨가할 수 있다.
상기 담체는 특별히 이에 제한되지는 않으나, 경구투여시에는 결합제, 활택제, 붕해제, 부형제, 가용화제, 분산제, 안정화제, 현탁화제, 색소, 향료 등을 사용할 수 있고, 주사제의 경우에는 완충제, 보존제, 무통화제, 가용화제, 등장화제, 안정화제 등을 혼합하여 사용할 수 있으며, 국소투여용의 경우에는 기제, 부형제, 윤활제, 보존제 등을 사용할 수 있다.
본 발명의 조성물의 제형은 상술한 바와 같은 약제학적으로 허용되는 담체와 혼합하여 다양하게 제조될 수 있다. 예를 들어, 경구 투여시에는 정제, 트로키, 캡슐, 엘릭서, 서스펜션, 시럽, 웨이퍼 등의 형태로 제조할 수 있으며, 주사제의 경우에는 단위 투약 앰플 또는 다수회 투약 형태로 제조할 수 있다. 기타, 용액, 현탁액, 정제, 환약, 캡슐, 서방형 제제 등으로 제형화할 수 있다.
한편, 제제화에 적합한 담체, 부형제 및 희석제의 예로는 락토즈, 덱스트로즈, 수크로즈, 솔비톨, 만니톨, 자일리톨, 에리스리톨, 말티톨, 전분, 아카시아, 알지네이트, 젤라틴, 칼슘 포스페이트, 칼슘 실리케이트, 셀룰로즈, 메틸 셀룰로즈, 미정질 셀룰로즈, 폴리비닐피롤리돈, 물, 메틸하이드록시벤조에이트, 프로필하이드록시벤조에이트, 탈크, 마그네슘 스테아레이트 또는 광물유 등이 사용될 수 있다. 또한, 충진제, 항응집제, 윤활제, 습윤제, 향료, 방부제 등을 추가로 포함할 수 있다.
또한, 본 발명의 약학적 조성물은 각각 통상의 방법에 따라 정제, 환제, 산제, 과립제, 캡슐제, 현탁제, 내용액제, 유제, 시럽제, 멸균된 수용액, 비수성용제, 현탁제, 유제, 동결건조제제 및 좌제로 이루어진 군으로부터 선택되는 어느 하나의 제형을 가질 수 있다.
또한, 상기 조성물은 약학적 분야에서 통상의 방법에 따라 환자의 신체 내 투여에 적합한 단위투여형의 제제, 바람직하게는 펩타이드 의약품의 투여에 유용한 제제 형태로 제형화시켜 당업계에서 통상적으로 사용하는 투여 방법을 이용하여 경구, 또는 피부, 정맥내, 근육내, 동맥내, 골수내, 수막강내, 심실내, 폐, 경피, 피하, 복내내, 비강내, 소화관내, 국소, 설하, 질내 또는 직장 경로를 포함하는 비경구투여 경로에 의하여 투여될 수 있으나, 이들에 한정되는 것은 아니다.
또한, 상기 결합체는 생리식염수 또는 유기용매와 같이 약제로 허용된 여러 전달체(carrier)와 혼합하여 사용될 수 있고, 안정성이나 흡수성을 증가시키기 위하여 글루코스, 수크로스 또는 덱스트란과 같은 카보하이드레이트, 아스코르브 산 (ascorbic acid) 또는 글루타치온과 같은 항산화제 (antioxidants), 킬레이팅 물질 (chelating agents), 저분자 단백질 또는 다른 안정화제 (stabilizers)들이 약제로 사용될 수 있다.
또 하나의 양태로서, 본 발명은 상기 항체-싸이토톡신 결합체 또는 상기 조성물을 이용하여 암을 치료하는 방법을 제공한다.
상기 방법은 치료를 필요로 하는 개체에 상기 약학적 조성물을 투여하는 단계를 포함하는 암을 치료하는 방법일 수 있으며, 이때, 사용되는 항체-싸이토톡신 결합체, 담체 및 암의 종류는 상기에서 설명한 바와 동일하다.
상기 조성물은 약학적으로 유효한 양으로 단일 또는 다중 투여될 수 있다. 이때, 조성물은 액제, 산제, 에어로졸, 캡슐제, 장용피 정제 또는 캡슐제 또는 좌제의 형태로 투여할 수 있다. 투여 경로는 복강내 투여, 정맥내 투여, 근육내 투여, 피하내 투여, 내피 투여, 경구 투여, 국소 투여, 비내 투여, 폐내 투여, 직장내 투여 등을 포함하지만, 이에 제한되지는 않는다. 그러나 경구 투여시, 단백질인 항체는 소화가 되기 때문에 경구용 조성물은 활성 약제를 코팅하거나 위에서의 분해로부터 보호되도록 제형화 되어야 한다. 또한, 제약 조성물은 활성 물질이 표적 세포로 이동할 수 있는 임의의 장치에 의해 투여될 수 있다. 또한, 본 발명의 항암 치료용 조성물은 개별 치료제로 투여하거나 다른 치료제와 병용하여 투여될 수 있고 종래의 치료제와는 순차적 또는 동시에 투여될 수 있다.
본 발명의 상기 항체-싸이토톡신 결합체를 포함하는 조성물은 약제학적으로 유효한 양으로 투여한다. "약제학적으로 유효한 양"이란 의학적 치료 또는 예방에 적용 가능한 합리적인 수혜/위험 비율로 질환을 치료 또는 예방하기에 충분한 양을 의미하며, 유효 용량 수준은 질환의 중증도, 약물의 활성, 환자의 연령, 체중, 건강, 성별, 환자의 약물에 대한 민감도, 사용된 본 발명 조성물의 투여 시간, 투여 경로 및 배출 비율 치료기간, 사용된 본 발명의 조성물과 배합 또는 동시 사용되는 약물을 포함한 요소 및 기타 의학 분야에 잘 알려진 요소에 따라 결정될 수 있다.
또 하나의 양태로서, 본 발명은 상기 항체-싸이토톡신 결합체로 제조하기에 적합한 항체를 선별하는 방법을 제공한다. 항체에 유전자조작을 가하지 않은 상태로 항체의 N-말단, α-아민에 싸이토톡신을 결합시키기 때문에 효과적으로 항체-싸이토톡신 결합체로 제조하기에 적합한 항체를 검색하여 선별할 수 있다.
본 발명의 항체-싸이토톡신 결합체를 포함하는 암치료용 조성물을 이용하여, 종양 세포 특이적 표적화를 높이고 강력한 세포독성을 통해, 항체 또는 싸이토톡신 단독 치료요법에 비해 증가된 항체의존성 세포독성 및 부작용 완화의 효과를 나타낼 수 있다. 또한, 결합체가 동질성을 유지하기 때문에 기존 항체-싸이토톡신 결합체의 이질성 문제를 극복할 수 있으며, 유전자 조작되지 않은 항체를 사용하여 항체의 N-말단, α-아민에 싸이토톡신을 결합시키기 때문에 보다 효과적인 항체-싸이토톡신 결합체 개발에 활용될 수 있을 것이다.

이하, 하기 실시예를 통하여 본 발명을 더욱 상세하게 설명하기로 한다. 이들 실시예는 단지 본 발명을 예시하기 위한 것으로, 본 발명의 범위가 이들 실시예에 의해 제한되는 것으로 해석되지는 않는다.
실시예
1: 단일클론 항체의 제조
본 발명의 항체-싸이토톡신 결합체 제조에 있어서, 링커가 존재하는 싸이토톡신이 위치 특이적으로 항체에 결합하는지 확인하기 위해 anti-HER2 항체, anti-IGF-1R 항체, 그리고 anti-EpCAM 항체를 모델 항체로 사용하였다.

3: 단일클론 항체-
싸이토톡신
결합체 제조
20mM 인산칼륨 pH6.0 완충 용액에 항체 5mg을 넣어 최종부피가 1ml이 되도록 준비한다. 이 후 알데히드 반응기를 가지는 링커와 연결되어 있는 Monomethyl Auristatin F (MMAF)(레고켐바이오, 한국)를 10% DMSO 용매에 녹여 0.5mg/ml이 되도록 한다. 그리고 나서 최종 10mM 인산칼륨 pH6.0, 항체농도 2.5mg/ml, 2.5% DMSO, MMAF 농도 0.125mg/ml, 항체의 α-아민과 MMAF의 몰비가 약 1:2가 되도록 준비된 항체 용액과 MMAF 용액을 혼합하였다. 상기 반응 용액에 NaCNBH3(Sigma사, 미국)를 최종 20mM이 되도록 첨가한 후, 4에서 20시간동안 서서히 교반시키면서 반응시켰다. 반응하지 않은 항체 및 링커와 연결된 MMAF를 분리하기 위하여 세파덱스 G-25컬럼(GE헬스케어, 미국) 또는 리소스페닐컬럼(Resource Phe, GE헬스케어, 미국)을 사용하였다. 항체 한 분자에 4개의 Monomethyl Auristatin F (MMAF) 독소가 아미노 말단에 선택적으로 결합된 형태를 정제하였다(도 2).

4: 항원 결합력 확인
싸이토톡신 결합 후에도 항원결합력이 유지되는지를 확인하기 위하여, BIAcore 방법으로 독소가 결합된 항체의 항원 결합력을 측정하였다. 대조항체는 천연형 항체를 사용하였다.

제조한 항체-싸이토톡신 결합체의 in vitro 효능을 확인하기 위하여 BT474, SKBR3, LS174T, MCF7, JIMT-1, HT-80 세포주를 이용하여 anti-proliferation assay를 수행하였다. 각 세포를 배양하여 1*105 cells/ml로 서스펜션하여 96 well 플레이트에 100 ㎕씩 로딩한다. 세포 배양기에서 3시간 배양 후, well당 다양한 농도구간의 항체-싸이토톡신 결합체를 각각 100 ㎕씩 넣고 세포 배양기에서 5일간 배양한다. 알라바마 블루(인비트로젠, 미국)를 1:10으로 희석하여 각 well에 처리 후, 호일에 싸서 세포 배양기에 6시간 처리 한다. 150rpm으로 15분간 교반 후, 스펙트라맥스 제미닉스를 이용하여 530nm에서 형광세기를 측정한다.
실시예
6: 항암모델동물에서 항암효과 확인실험
SCID/NOD 마우스와 허셉틴 저항성 세포주인 JIMT-1을 사용하여 일반적인 xenograft 실험을 진행하였다. 약물투여 후 암세포 크기를 측정하여 항암효과를 확인하였다.
Claims (13)
- 암세포 표면 항원에 대한 항체의 N-말단 아미노 그룹에 위치 특이적으로 싸이토톡신이 결합된 항체-싸이토톡신 결합체.
- 제1항에 있어서,
상기 싸이토톡신은 링커가 연결된 것으로, 링커에 의해 상기 항체의 N-말단 아미노 그룹에 위치 특이적으로 결합된 것인, 항체-싸이토톡신 결합체.
- 제2항에 있어서,
상기 링커는 알데히드기를 반응기로 가지는 것인, 항체-싸이토톡신 결합체.
- 제3항에 있어서,
상기 알데히드기를 반응기로 가지는 링커와 연결된 싸이토톡신을 항체의 N-말단 아미노 그룹의 α-아민에 위치 특이적으로 결합된 것인, 항체-싸이토톡신 결합체.
- 제1항에 있어서,
상기 암세포 표면 항원과 결합하는 항체는 IgA, IgD, IgE, IgM, IgG, Fc 단편, Fab, Fab', F(ab')₂및 Fv로 이루어진 군에서 선택된 것인 항체-싸이토톡신 결합체.
- 제1항에 있어서,
상기 항체는 천연형인 것인, 항체-싸이토톡신 결합체.
- 제1항에 있어서,
상기 암세포 표면 항원은 CD19, CD20, Her2, IGF-1R, EpCAM (Epithelial cell adhesion molecule) 및 EGFR (epidermal growth factor receptor)로 이루어진 군에서 선택된 것인 항체-싸이토톡신 결합체.
- 제1항에 있어서,
상기 싸이토톡신은 마이크로튜불린 억제제, 유사분열 억제제, 토포이소머라아제 억제제 및 DNA 인터컬레이터(intercalators)로부터 선택되는 것인, 항체-싸이토톡신 결합체.
- 제1항에 있어서,
상기 싸이토톡신은 메이탄시노이드, 오리스타틴, 돌라스타틴, 칼리케아미신 으로부터 선택되는 것인, 항체-싸이토톡신 결합체.
- (1) 알데히드 반응기를 갖는 링커와 연결된 싸이토톡신과 항체를 반응시켜서 싸이토톡신과 항체의 N-말단의 아미노 그룹과 연결시키는 단계; 및
(2) 상기 (1)단계의 반응 혼합물로부터 결합체를 형성하지 않은 항체 및 싸이토톡신을 제거하여 항체-싸이토톡신 결합체를 분리 및 정제하는 단계를 포함하는, 제1항 내지 제9항 중 어느 한 항의 항체-싸이토톡신 결합체 제조방법.
- 제10항에 있어서,
상기 (1)단계의 알데히드 반응기를 갖는 링커와 연결된 싸이토톡신을 항체의 N-말단 아미노 그룹의 α-아민과 연결시키는 것인 방법.
- 제1항 내지 제9항 중 어느 한 항의 항체-싸이토톡신 결합체와 약제학적으로 허용가능한 담체를 포함하는 암치료용 약학적 조성물.
- 제12항에 있어서,
상기 암은 식도암, 위암, 대장암, 직장암, 구강암, 인두암, 후두암, 폐암, 결장암, 유방암, 자궁 경부암, 자궁 내막체암, 난소암, 전립선암, 고환암, 방광암, 신장암, 간암, 췌장암, 골암, 결합 조직암, 피부암, 뇌암, 갑상선암, 백혈병, 호지킨(Hodgkin) 질환, 림프종, 및 다발성 골수종혈액암으로 이루어진 군에서 선택된 것인 암치료용 약학적 조성물.
