Description
〔1〕 多能性幹細胞または神経前駆細胞をN−アセチル−D−マンノサミンの存在下で培養する工程を含む、オレキシンニューロンの製造方法。
〔2〕 多能性幹細胞が胚性幹細胞、体性幹細胞または人工多能性幹細胞である〔1〕に記載の製造方法。
〔3〕 神経前駆細胞が胎児由来神経塊である〔1〕に記載の製造方法。
〔4〕 〔1〕〜〔3〕いずれかに記載の製造方法により得られうるオレキシンニューロン。
〔5〕 〔4〕に記載のオレキシンニューロンを用いることを特徴とする、覚醒-睡眠の調節に作用する薬物のスクリーニング方法。
〔6〕 薬物がナルコレプシーの治療薬である、〔5〕に記載のスクリーニング方法。
〔7〕 N−アセチル−D−マンノサミンの有効量をそれを必要とする対象に投与または摂取させる工程を含む、オレキシンニューロンの誘導方法。
〔8〕 N−アセチル−D−マンノサミンを有効成分として含有してなる、神経前駆細胞または神経細胞からオレキシンニューロンの誘導剤。
〔9〕 多能性幹細胞または神経前駆細胞をN−アセチル−D−マンノサミンならびに、サーチュイン(Sirt)阻害剤およびO-結合型β-N-アセチルグルコサミントランスフェラーゼ(OGT)阻害剤からなる群より選ばれる少なくとも1種の阻害剤の存在下で培養する工程を含む、オレキシンニューロンの製造方法。
〔10〕 多能性幹細胞が胚性幹細胞、体性幹細胞または人工多能性幹細胞である〔9〕に記載の製造方法。
〔11〕 神経前駆細胞が胎児由来神経塊である〔9〕に記載の製造方法。

〔15〕 N−アセチル−D−マンノサミンならびに、サーチュイン1(Sirt1)阻害剤およびO-結合型β-N-アセチルグルコサミントランスフェラーゼ(OGT)阻害剤からなる群より選ばれる少なくとも1種の阻害剤を有効成分として含有してなる、神経前駆細胞または神経細胞からオレキシンニューロンの誘導剤。
オレキシンニューロンの減少または欠損による症状を呈する病気に対して、ManNAc(その誘導体、前駆体もしくはプロドラッグを含む)、またはManNAcとSirt阻害剤および/またはOgt阻害剤との組合せは有効である。ManNAc(その誘導体、前駆体もしくはプロドラッグを含む)、またはManNAcとSirt阻害剤および/またはOgt阻害剤との組合せを用いて製造したオレキシンニューロンにより、新たな薬物療法や薬物スクリーニングを提供することができる。本発明により、オレキシンニューロン回復を目指した再生医療にも貢献することができる。
N−アセチル−D−マンノサミンは、例えば、下記式(II)で示される化合物であってもよい。
置換基としてはF、Cl、Brを用いることができる。
本発明は、多能性幹細胞または神経前駆細胞をN−アセチル−D−マンノサミンの存在下で培養する工程を含む、オレキシンニューロンの製造方法を提供する。
また、本発明は、多能性幹細胞または神経前駆細胞をN−アセチル−D−マンノサミンならびに、Sirt阻害剤およびOgt阻害剤からなる群より選ばれる少なくとも1種の阻害剤の存在下で培養する工程を含む、オレキシンニューロンの製造方法を提供する。
本発明は、本発明の製造方法により得られうるオレキシンニューロンを用いて、覚醒−睡眠の調節に作用する薬物のスクリーニング方法を提供する。
本発明は、N−アセチル−D−マンノサミンを有効成分として含有してなる、神経前駆細胞または神経細胞からオレキシンニューロンの誘導剤を提供する。
また、本発明は、(1)N−アセチル−D−マンノサミン、ならびに
(2)サーチュイン1阻害剤およびO-結合型β-N-アセチルグルコサミントランスフェラーゼ阻害剤からなる群より選ばれる少なくとも1種の阻害剤
を有効成分として含有する試薬を別々の容器に収容してなる、神経前駆細胞または神経細胞からオレキシンニューロンを誘導するためのキットを提供する。
Stromal cell-derived inducing activity (SDIA)分化培養系を用いた解析
分化培養
ES細胞用培地[15% FBS (Biowest)、1 mM sodium pyruvate (Invitrogen)、100 mM β-mercaptoethanol (Invitrogen)、2 mM L-glutamine (Invitrogen)、1 mM non essential amino acid (Invitrogen)、50 U/ml penicillin/50 μg/ml streptomycin (Invitrogen)、2000 U leukemia inhibitory factor (Chemicon、CA) をDMEM (Wako) に添加]で維持したマウスES細胞(J1株)を、10cm培養皿1枚当たり1x105個になるように、予めPA6間質細胞がまかれた培養皿に播種し、神経分化用培地 [10% KSR (Invitrogen)、100 mM β-mercaptoethanol (Invitrogen)、1 mM non essential amino acid (Invitrogen)、50 U/ml penicillin/50μg/ml streptomycin (Invitrogen)をGlasgow MEM (Invitrogen) に添加]で10日間分化誘導を行った。分化培養4および7日目に培地の交換を行った。また、分化培養4日目から5 nM recombinant human BMP4(Wako)を添加した(図1)。分化培養10日目に細胞を回収し、遠心操作により細胞をペレット状にし、培養液を除去した。その後、直ちに液体窒素中で凍結させ、使用するまで-80℃で保存した。

回収した細胞からRNeasy plus Mini Kit (QIAGEN)を用いてtotal RNAを抽出した。次いでSuperScript III First Strand Synthesis System (Invitrogen) を用いて、total RNA 1 μgおよびOligo(dT)を含む反応液中で逆転写反応を行いcDNAを合成した。
PCR反応は10 μlスケールで行い、0.5 μlのcDNA、5 μlの2xGC Buffer I、200 μM各dNTP、1.5 mM MgCl2、終濃度0.2 μM各プライマーに1 UのLA-Taq DNA Polymerase (Takara) を加え、95℃ 3分の熱変性後、(95℃ 30秒、55℃ 30秒、72℃ 30秒)x35サイクル (Actbの場合は20サイクル) の条件で行った。用いたプライマーは以下に示す。

各PCR産物は2% アガロースゲルで電気泳動し、エチジウムブロマイド染色後、UV照射によりバンドの観察を行った。
(実験1、図2)
分化培養開始時からGlcNAc(N-アセチル-D-グルコサミン)、ManNAcおよびNeu5Ac(N-アセチルノイラミン酸)を0.2、1.0mMになるように神経分化培地中に添加し、分化培養10日目に細胞を回収してRT-PCRに供した。オレキシン遺伝子(Hcrt)の発現解析を行った結果、ManNAc添加区のみでHcrtの発現が認められた。なお、ヒト由来の多能性幹細胞でも同様の効果が得られた。
次に、GlcNAc、ManNAcおよびNeu5Acの添加する時期を変えて1.0 mMの濃度で添加し神経分化培養を行った。RT-PCRの解析の結果、いずれの添加時期においてもManNAcを添加した区でのみHcrtの発現が観察された。また、1.0 mM ManNAc添加区の中でも、分化7日目以降に培地にManNAcが添加されている区 (4-10日、7-10日および0-10日目に添加)はその他の2区 (0-4日および0-7日に添加)に比べて発現が高かった。

分化培養
ES細胞用培地で維持したマウスES細胞(J1株)を、1ウェル当たり10,000個になるように96ウェル低接着丸底培養皿(Nunc)に播種し、gfCDM [5 mg/ml BSA (SIGMA)、450μM Monothioglycerol (Wako)、1x chemically defined lipid concentrate (Invitrogen)、50 U/ml penicillin/50μg/ml streptomycinをIMDM/F12(Invitrogen)に添加]で7日間培養した。1ウェル当たり150 μlの培地で培養した。分化培養7日目にDFK培地 [7 g/l glucose、10% KSR、50 U/ml penicillin/50μg/ml streptomycinをDMEM/F12(Invitrogen)に添加]に交換し、10日目で半量の培地を10 ng/ml CNTFを含むDFNB培地 [7 g/l glucose、1x N2 (Wako)、1xB27 (Invitrogen)]と交換した。培養13日目に細胞を0.05% トリプシン/EDTAで剥離し、10 cm培養皿1枚当たり4.8x106個になるようにポリ-D-リシン/ラニミンで処理した培養皿(Falcon)に播種し、10% FBS、10 ng/ml CNTF、50 ng/ml BDNFおよび50 ng/ml NT3を含むDFNB培地で培養した。培地交換は3日毎に行った。分化培養25日目に細胞を回収し、遠心操作により細胞をペレット状にし、培養液を除去した。その後、直ちに液体窒素中で凍結させ、使用するまで-80℃で保存した。

分化培養7日目からGlcNac、ManNAcおよびNeu5Acを1.0 mMの濃度で培地に添加し、神経分化誘導を行った。その結果、ManNAcおよびNeu5Ac添加区でHcrtの発現が認められた。また、Hcrtの発現はNeu5Acに比べてManNAc添加区の方が高かった。
終脳由来神経塊(ニューロスフィア、NSph)の培養およびニューロンへの分化誘導
Ciccolini and Svendsen(1998, J Neurosci. 18(19):7869-80)の方法に基づいて、培養および分化誘導実験を行った。概略を図6に示す。氷冷PBS/0.6%グルコース中で、胎齢14.5日のC57BL/6Nマウス胎仔より終脳を摘出した。ピペッティングにより単一の細胞に分散し、NSph用培地[1xB27 (Invitrogen)、20 ng/ml EGF (Sigma)、20 ng/ml FGF-basic (PEPROTECH)、5 μg/ml heparin (Sigma)を含むDMEM/F12 (Wako)]に懸濁して2x106cells/10 mlの量で10 cmペトリディッシュに播種した。培養開始3日後に培地を半量交換し、さらに3日間の培養を行った(合計6日間)。
培養開始6日後にNSphを回収し、PBS/0.6%グルコース中でのピペッティングにより細胞を分散した。遠心分離により細胞を集め、分化誘導用培地(NSph用培地のEGF、FGF-basicおよびheparinを含まないもの)に細胞を懸濁した。4.5x105cells/7.5 mlの量で、予めpoly-L-lysine/laminin (ともにSigma)コート処理を施した6 cmディッシュに細胞を播種し分化誘導を行った。分化誘導開始5日後に培地を半量交換し、さらに5日間の培養を行った(合計10日間)。PBSで細胞を洗浄した後、0.5 mlのTRIzol Reagent (Invitrogen)で細胞を溶解して1.5 mlマイクロチューブに回収し、使用するまで-80℃で保存した。
TRIzol Reagentで溶解した細胞からtotal RNAを抽出した。SuperScript III First Strand Synthesis System (Invitrogen)を用い、90 ngのtotal RNAおよびOligo(dT)を含む10 μlの反応液中で逆転写反応を行いcDNAを合成した。反応液を10 mM Tris-HCl (pH 8.0)/0.1 mM EDTAで5倍希釈しPCRに用いた。
1 x ImmoBuffer、1.5 mM MgCl2、0.2 mM each dNTP、0.2 μM forward primer、0.2 μM reverse primer、1 U BIOTAQ HS Polymerase (BIOLINE)および2 μlのcDNAを含む20 μlの反応液を調整し、95℃で7分を1サイクルの後、94℃で30秒、62℃で30秒、72℃で30秒を24 (Actb)/32 (Prkcz)/36 (Hcrt)サイクルの条件でPCRを行った。用いたプライマーの塩基配列を以下に示す。

Prkcz reverse; 5’- ATATCCTTTCGCTGCACTGG -3’(配列番号:8)
Hcrt forward; 5’- CTCCAGGCACCATGAACTTT -3’(配列番号:1)
Hcrt reverse; 5’- AGTTCGTAGAGACGGCAGGA -3’(配列番号:2)
PCR産物を2%アガロースゲルで電気泳動しエチジウムブロマイドで染色後、UV照射によりバンドの観察を行った。
NSphの培養開始時にGlcNAc、ManNAcあるいはNeuAcを1 mMとなるように添加して6日間の培養を行った。添加された条件で引き続き10日間の分化誘導を行った後細胞を回収した(図6)。
Hcrtの発現をRT-PCRにより解析した結果、1.0 mM ManNAc添加区のみでバンドが検出された(図7)。
分化培養
実施例1に記載の方法と同様にして、マウスES細胞(J1株)を10日間、神経細胞への分化誘導のために培養した。神経細胞への分化誘導プロトコルを図11の上段に示す。分化培養開始時(0日)からManNAcを1.0mMになるように神経分化培地中に添加し、分化培養4および7日目に培地の交換を行った。また、分化培養4日目から5 nM recombinant human BMP4(Wako)を添加した。分化培養7日目にEX-527(SIGMA-ALDRICH)を50nMになるように神経分化培地に添加し、あるいはBADGP(SIGMA-ALDRICH)を1または5mMになるように神経分化培地に添加し、分化培養10日目に細胞を回収してRT-PCRに供した。RT-PCRの条件は、実施例1と同様である。
(実験3、図8)
分化培養開始時からManNAcを1.0mMになるように神経分化培地中に添加し、分化培養7日目にEX-527(SIGMA-ALDRICH)を50nMになるように神経分化培地に添加し、分化培養10日目に細胞を回収してRT-PCRに供した。オレキシン遺伝子(Hcrt)の発現解析を行った結果、EX-527添加区でHcrtの発現が認められたが、ManNAcとEX-527とを併用することにより、Hcrtの発現が増強することがわかった。
次に、分化培養開始時からManNAcを1.0mMになるように神経分化培地中に添加し、分化培養7日目にBADGP(SIGMA-ALDRICH)を1mMまたは5mMになるように神経分化培地に添加し、分化培養10日目に細胞を回収してRT-PCRに供した。オレキシン遺伝子(Hcrt)の発現解析を行った結果、ManNAc添加区で認められたHcrtの発現が、ManNAcとBADGPとを併用することにより増強されることがわかった。Hcrtの発現は、BADGPの濃度に依存して増大される傾向が観察された。
分化培養
ヒトiPS細胞株201B7(HSP0001)は、ナショナルバイオリソースプロジェクト(MEXT、日本)を介して、理研バイオリソースセンターより入手した。ヒトiPS細胞は、5 ng/mLのrecombinant human bFGF(Wako)を補足した霊長類ES培地(ReproCELL)中、マイトマイシンC処理STO/Neo resistant/LIF(SNL)フィーダー細胞のフィーダー層上で維持した。SIDAとBMP4系による神経への分化誘導のため、ヒトiPS細胞をPA6とともに20日間共培養した。5nM BMP4を、7日目から補足した。詳細な神経細胞への分化誘導プロトコルを図11の下段に示す。分化培養開始時(0日)からManNAcを1.0mMになるように神経分化培地中に添加し、分化培養7、10および14日目に培地の交換を行った。また、分化培養7日目から5 nM recombinant human BMP4(Wako)を添加した。分化培養14日目にEX-527(SIGMA-ALDRICH)を50nMになるように神経分化培地に添加し、あるいはBADGP(SIGMA-ALDRICH)を5mMになるように神経分化培地に添加し、分化培養20日目に細胞を回収してRT-PCRに供した。RT-PCRの条件は、実施例1と同様である。
RT-PCRによるオレキシン遺伝子(Hcrt)の発現解析を行った結果、ManNAc添加区およびEX-527添加区で同程度に認められたHcrtの発現が、ManNAcとEX-527、ManNAcとBADGP、ManNAcとEX-527とBADGPとを併用することにより増強されることがわかった。Hcrtの発現は、ManNAcとEX-527とBADGPの3種併用によって最も増大された。
実施例5に記載された方法で4ウェル培養プレートで分化誘導させた細胞を、4%パラホルマリン溶液で固定した後、0.2% Triton-X100溶液で細胞膜透過性処理を行った。5%牛血清アルブミン溶液で処理した後、一次抗体(抗ヒトオレキシン抗体、抗ヒトチューブリンIII抗体)で4℃、一夜処理をした。二次標識抗体(Alex-Fluor 488標識ロバ抗ヤギ抗体、Alexa-Fluor 594標識ウサギ抗マウス抗体)と反応させた後、DAPIで核DNAを染色した。蛍光は蛍光顕微鏡で観察し、オレキシン抗体との反応が認められた神経分化コロニー中のオレキシン陽性細胞の割合を計測した。
Claims (13)
- 多能性幹細胞または神経前駆細胞をN−アセチル−D−マンノサミンの存在下で培養する工程を含む、オレキシンニューロンの製造方法。
- 多能性幹細胞が胚性幹細胞、体性幹細胞または人工多能性幹細胞である請求項1に記載の製造方法。
- 神経前駆細胞が胎児由来神経塊である請求項1に記載の製造方法。
- 請求項1〜3のいずれか1項に記載の方法により、オレキシンニューロンを製造する工程、並びに
製造されたオレキシンニューロンを被験物質の存在下で培養し、Hcrt遺伝子の発現又は培地中に放出されたオレキシン若しくは細胞内に蓄積されたオレキシンを検出又は測定する工程、
を含む、覚醒−睡眠の調節に作用する薬物のスクリーニング方法。 - 薬物がナルコレプシーの治療薬である、請求項4に記載のスクリーニング方法。
- N−アセチル−D−マンノサミンの有効量をそれを必要とするヒトを除く対象に投与する、または摂取させる工程を含む、オレキシンニューロンの誘導方法。
- N−アセチル−D−マンノサミンを有効成分として含有してなる、神経前駆細胞または神経細胞からオレキシンニューロンの誘導剤。
- 多能性幹細胞または神経前駆細胞をN−アセチル−D−マンノサミンならびに、サーチュイン阻害剤およびO-結合型β-N-アセチルグルコサミントランスフェラーゼ阻害剤からなる群より選ばれる少なくとも1種の阻害剤の存在下で培養する工程を含む、オレキシンニューロンの製造方法。
- 多能性幹細胞が胚性幹細胞、体性幹細胞または人工多能性幹細胞である請求項8に記載の製造方法。
- 神経前駆細胞が胎児由来神経塊である請求項8に記載の製造方法。
- 請求項8〜10のいずれか1項に記載の方法により、オレキシンニューロンを製造する工程、並びに
製造されたオレキシンニューロンを被験物質の存在下で培養し、Hcrt遺伝子の発現又は培地中に放出されたオレキシン若しくは細胞内に蓄積されたオレキシンを検出又は測定する工程、
を含む、覚醒−睡眠の調節に作用する薬物のスクリーニング方法。 - 薬物がナルコレプシーの治療薬である、請求項11に記載のスクリーニング方法。
- (1)N−アセチル−D−マンノサミン、ならびに
(2)サーチュイン阻害剤およびO-結合型β-N-アセチルグルコサミントランスフェラーゼ阻害剤からなる群より選ばれる少なくとも1種の阻害剤を有効成分として別々の容器に収容してなる、神経前駆細胞または神経細胞からオレキシンニューロンを誘導するためのキット。
