Description
〔1〕式(1):
〔2〕前記式(1)及び前記式(2)で示される2種のレスベラトロール誘導体の重量比が1:9〜9:1である前記〔1〕記載の代謝促進剤組成物、
〔3〕脂肪組織の代謝を促進する前記〔1〕又は〔2〕記載の代謝促進剤組成物、
〔4〕前記〔1〕〜〔3〕いずれか1項に記載の代謝促進剤組成物を含有することを特徴とするPGC−1遺伝子発現促進剤、
〔5〕前記〔1〕〜〔3〕いずれか1項に記載の代謝促進剤組成物を含有することを特徴とするミトコンドリア脱共役タンパク質遺伝子発現促進剤、
〔6〕前記〔1〕〜〔3〕のいずれか1項に記載の代謝促進剤組成物を含有することを特徴とする概日リズム調整剤、
〔7〕前記〔1〕〜〔3〕いずれか記載の代謝促進剤組成物を含有する食品、医薬品又は医薬部外品
に関する。
また、本発明の代謝促進剤組成物は、PGC−1遺伝子発現促進剤、ミトコンドリア脱共役タンパク質遺伝子発現促進剤の有効成分としても有用である。
また、本発明の代謝促進剤組成物は、PGC−1遺伝子発現促進作用を有することから、新規な概日リズム調整剤の有効成分としても有用である。
また、本発明の代謝促進剤組成物を、代謝促進、メタボリックシンドロームの改善、概日リズムの調整などを目的として、食品、医薬品、医薬部外品に添加することだできる。
前記脂肪細胞における各遺伝子の発現については、具体的には、後述の実施例に記載の方法により測定することができる。
前記レスベラトロール含有原料としては、ブドウ果皮、ピーナッツ果皮、イタドリ等の原料からの抽出物、該抽出物の凍結乾燥品等を使用してもよい。
また、レスベラトロールは、レスベラトロール含有溶液中においてレスベラトロール誘導体(1)及び(2)の生成反応前に完全に溶解していなくともよい。
なお、レスベラトロール含有溶液中の金属塩の含有量は特に限定されず、レスベラトロール誘導体(1)及び(2)を生成可能な量であればよい。
また、本発明の代謝促進剤組成物は、PGC−1遺伝子発現促進剤及びミトコンドリア脱共役タンパク質遺伝子発現促進剤の有効成分として有用である。また、本発明の代謝促進剤組成物は、新規な概日リズム調整剤の有効成分としても有用である。
以下においては、代謝促進剤組成物、PGC−1遺伝子発現促進剤、ミトコンドリア脱共役タンパク質遺伝子発現促進剤及び概日リズム調整剤をまとめて本発明の薬剤と総称することがある。
結合剤としては、例えば、単シロップ、ブドウ糖液、デンプン液、ゼラチン溶液、ポリビニルアルコール、ポリビニルエーテル、ポリビニルピロリドン、カルボキシメチルセルロース、セラック、メチルセルロース、エチルセルロース、水、エタノール、リン酸カリウム及びこれらの混合物等が挙げられる。
安定化剤としては、例えば、ピロ亜硫酸ナトリウム、エチレンジアミン四酢酸、チオグリコール酸、チオ乳酸及びこれらの混合物等が挙げられる。
pH調整剤及び緩衝剤としては、例えば、クエン酸ナトリウム、クエン酸、酢酸ナトリウム、リン酸ナトリウム及びこれらの混合物等が挙げられる。
トランス−レスベラトロール(東京化成工業(株)製)700mgをエタノール14mLに溶解し、2.5%NaHCO3水溶液を14mL加えて、レスベラトロール含有溶液(pH9.9)を得た。このレスベラトロール含有溶液をオートクレーブ(商品名:SANYO LABO AUTOCLAVE、三洋電機(株)製、以下同じ)にて130℃で20分間加熱した。得られた反応液に、エタノール14mL及び5.0%NaHCO3水溶液14mLを加え、再度、オートクレーブにて130℃で20分間加熱した。得られた反応液のうち1mLをメタノールにて全量50mLにメスアップし、このうちの10μLをHPLCにより分析した。その結果を図1に示す。

UHA4002及びUHA4003含有組成物によるPGC−1α遺伝子の発現量を、3T3―L1細胞(マウス由来脂肪前駆細胞)を用いて評価した。
このようにUHA4002及びUHA4003含有組成物は、PGC−1α発現促進作用を有することから、脂肪細胞の代謝を亢進する作用、具体的には、PGC−1遺伝子発現促進作用に基づく抗肥満作用及びメタボリックシンドローム予防・治療作用を有していることが明らかである。
また、式(1)で示されるUHA4002及び式(2)で示されるUHA4003の2種を有効成分として含む本発明の薬剤は、PGC−1遺伝子発現促進剤としても使用でき、さらにはPGC−1α発現促進作用を有することから概日リズム調整剤としても使用できることがわかる。
UHA4002及びUHA4003含有組成物によるUCP−2遺伝子の発現量を、3T3―L1細胞(マウス由来脂肪前駆細胞)を用いて評価した。
このようにUHA4002及びUHA4003含有組成物は、UCP−2発現促進作用を有することから、脂肪細胞の代謝を亢進する作用、具体的には、UCP−2発現促進作用に基づく抗肥満作用及びメタボリックシンドローム予防・治療作用も有していることが明らかである。
また、UHA4002及びUHA4003含有組成物を有効成分として含む本発明の薬剤は、ミトコンドリア脱共役タンパク質遺伝子発現促進剤としても使用できることがわかる。
実施例2と同様にして得られたUHA4002及びUHA4003含有組成物の1gを、エタノール100mLに溶解し、得られた溶液に砂糖500g及び水飴400gを混合溶解し、更に生クリーム100g、バター20g、練乳70g及び乳化剤1.0gを混合した。得られた混合物を、真空釜にて−550mmHg減圧下及び115℃の温度下で濃縮し、水分値3.0重量%のミルクハードキャンディを得た。
実施例2と同様にして得られたUHA4002及びUHA4003含有組成物の1gをエタノールに溶解し、得られた溶液を微結晶セルロースに吸着させて、減圧乾燥した。このUHA4002及びUHA4003吸着体を用い、下記の配合で打錠品を得た。

実施例2と同様にして得られたUHA4002及びUHA4003含有組成物の1.2gの10mLエタノール溶液、タウリン20g、ビタミンB1硝酸塩0.12g、安息香酸ナトリウム0.6g、クエン酸4g、砂糖60g及びポリビニルピロリドン10gを精製水に溶解し、全量を1000mLにメスアップした。なお、pHは、希塩酸を用いて3.2に調整した。得られた溶液1000mLのうち50mLをガラス瓶に充填し、80℃で30分間滅菌して、医薬部外品であるドリンク剤を得た。

- 前記式(1)及び前記式(2)で示される2種のレスベラトロール誘導体の重量比が1:9〜9:1である請求項1に記載の代謝促進剤組成物。
- 脂肪組織の代謝を促進する請求項1又は2記載の代謝促進剤組成物。
- 請求項1〜3いずれか1項に記載の代謝促進剤組成物を含有することを特徴とするPGC−1遺伝子発現促進剤。
- 請求項1〜3いずれか1項に記載の代謝促進剤組成物を含有することを特徴とするミトコンドリア脱共役タンパク質遺伝子発現促進剤。
- 請求項1〜3のいずれか1項に記載の代謝促進剤組成物を含有することを特徴とする概日リズム調整剤。
- 請求項1〜3のいずれか1項に記載の代謝促進剤組成物を含有する食品、医薬品又は医薬部外品。
