従って、本分野では、より有効的な神経変性疾患の治療薬の開発が切望されている。
M1-L-M2 式I
(式中において、
M1はヒトコロニー刺激因子G-CSFの第一の単量体で、
M2はヒトコロニー刺激因子G-CSFの第二の単量体で、
Lは前述の第一の単量体と第二の単量体の間に位置し、前述第一の単量体と第二の単量体を連結するリンカーである。)
ここで、前述G-CSFの二量体は、G-CSFの単量体の生物活性を保持し、且つ前述第一の単量体および前述第二の単量体のいずれかの2倍以上の血清半減期を有する。
(i) 3〜50個のアミノ酸からなる短鎖ペプチドと、
(ii) 式IIで示されるポリペプチドと、
-Z-Y-Z- 式II
(式中において、
Yは担体タンパク質で、
Zはなし、または1〜30個のアミノ酸の短鎖ペプチドで、

(a)G-CSF-Fc複合体をコードする、SEQ ID NO:10またはSEQ ID NO:9で示されるようなDNA配列を含有する発現ベクターで哺乳動物細胞をトランスフォームする工程、
(b)前述哺乳動物細胞を培養し、G-CSF-Fc複合体とG-CSF二量体を発現させる工程、および
(c)前述G-CSF二量体を分離して精製する工程。
ここで、前述G-CSF二量体は、2つのG-CSF-Fc複合体を含み、前述G-CSF-Fc複合体は、アミノ酸配列がSEQ ID NO:2〜7で示される。
前述の医薬品組成物は、固体製剤または液体製剤である。
前述の医薬品組成物は、0.01-99wt%のコロニー刺激因子G-CSFの二量体と、薬理学的に許容される賦形剤または担体を含む。
前述の賦形剤または担体は、セルロースおよびその誘導体、ゼラチン、タルク、固体潤滑剤、硫酸カルシウム、植物油、多価アルコール、乳化剤、湿潤剤、着色剤、調味剤、安定剤、酸化防止剤、防腐剤、発熱性物質除去蒸留水からなる群から選ばれる。

(a) 疎水性アミノ酸であるGlyとProからなる、3〜15個のアミノ酸の配列、例えばGly-Pro-Gly-Pro-Gly-Pro、または、GlyとSerからなる、3〜20個のアミノ酸の配列、例えばGSGG。
(b) 多回クローニングサイトでコードされるアミノ酸配列。このような配列は、通常、5〜20個、好ましくは10〜20個のアミノ酸で、その例として、TGLQPTRGIDDITSPVDを含むが、これらに限定されない。
(c) G-CSF単量体以外のタンパク質由来のアミノ酸配列、例えばIgGまたはアルブミン由来のアミノ酸配列。
(d) (a)、(b)および(c)の組合せで形成されるアミノ酸配列。
好ましい連結ぺプチドの一つは、ASTKGP(SEQ ID NO:4における175-180個目)である。
SEQ ID NO:1は、G-CSF二量体の配列の一つで、構造が図1に示され、ここで、1-174個目はG-CSF単量体で、175-190個目は連結ぺプチドで、191-364個目はもう一つのG-CSF単量体である。
SEQ ID NO:2は、G-CSF二量体のFc断片のついたG-CSF単量体の配列で、ここで、1-174個目はG-CSF単量体で、175-190個目は連結ぺプチドで、191-418個目はヒトIgG2のFc断片である。図2に示すように、二つのFc断片のついたG-CSF単量体は、Fcのジスルフィド結合のペアリング作用で二量体を構成する。
SEQ ID NO:3は、G-CSF二量体のFc断片のついたG-CSF単量体の配列で、ここで、245-418個目はG-CSF単量体で、229-244個目は連結ぺプチドで、1-228個目はヒトIgG2のFc断片である。二つのFc断片を含むG-CSFの単量体は、Fcのジスルフィド結合のペアリング作用で二量体を構成する。
SEQ ID NO:4は、G-CSF二量体のFc断片のついたG-CSF単量体の配列で、ここで、1-174個目はG-CSF単量体で、175-180個目は連結ぺプチドで、181-403個目はヒトIgG2のFc断片である。二つのFc断片を含むG-CSFの単量体は、Fcのジスルフィド結合のペアリング作用で二量体を構成する。
SEQ ID NO:5は、G-CSF二量体のFc断片のついたG-CSF単量体の配列で、ここで、230-403個目はG-CSFで、224-229個目は連結ぺプチドで、1-223個目はヒトIgG2のFc断片である。二つのFc断片を含むG-CSFの単量体は、Fcのジスルフィド結合のペアリング作用で二量体を構成する。
SEQ ID NO:6は、G-CSF二量体のFc断片のついたG-CSF単量体の配列で、ここで、1-174個目はG-CSFで、175-190個目は連結ぺプチドで、191-413個目はヒトIgG2のFc断片である。二つのFc断片を含むG-CSFの単量体は、Fcのジスルフィド結合のペアリング作用で二量体を構成する。
SEQ ID NO:7は、G-CSF二量体のFc断片のついたG-CSF単量体の配列で、ここで、240-413個目はG-CSFで、224-239個目は連結ぺプチドで、1-223個目はヒトIgG2のFc断片である。二つのFc断片を含むG-CSFの単量体は、Fcのジスルフィド結合のペアリング作用で二量体を構成する。
SEQ ID NO:9は、SEQ ID NO:2のcDNA配列である。
SEQ ID NO:10は、SEQ ID NO:6のcDNA配列である。
本発明のG-CSF二量体または融合タンパク質をコードするDNA配列は、全部人工合成としてもよい。PCR増幅または合成の方法でG-CSFの第一の単量体及び/又はG-CSFの第二の単量体のコードDNA配列を得た後、これらを連結し、本発明の融合タンパク質をコードするDNA配列を形成してもよい。
ここで用いられるように、用語の「ベクター」は、プラスミド、コスミド、発現ベクター、クローンベクター、ウイルスベクターなどを含む。
(a)G-CSF-Fc複合体をコードする、SEQ ID NO:10またはSEQ ID NO:9で示されるようなDNA配列を含有する発現ベクターで哺乳動物の細胞をトランスフォームする工程、
(b)前述哺乳動物細胞を培養し、G-CSF-Fc複合体とG-CSF二量体を発現させる工程、および
(c) 前述G-CSF二量体を分離して精製する工程。
を含む。
ここで、前述G-CSF二量体は、2つのG-CSF-Fc複合体を含み、前述G-CSF-Fc複合体は、アミノ酸配列がSEQ ID NO:2〜7で示される。
本発明のG-CSF二量体は、優れた血清半減期を有するため、本発明のG-CSF二量体および主な活性成分として本発明のG-CSF二量体を含む医薬品組成物は、神経損傷疾患の治療、およびニューロンの保護に有用である。ここで、前述の神経損傷疾患は、パーキンソン病(PD)、アルツハイマー病(AD)、ハンチントン病(HD)、筋萎縮性側索硬化症(ALS)、脊髄性筋萎縮症(SMA)、原発性側索硬化症(PLS)、脊髓小脳失調症(SCA)からなる群から選ばれる。
これらの不活性希釈剤の他、組成物は助剤、例えば、湿潤剤、乳化剤、懸濁剤、甘味料、矯味剤や香料を含んでもよい。
活性化合物の他、懸濁液は、懸濁剤、例えば、エトキシ化イソオクタデカノール、ポリオキシエチレンソルビトールやソルビタンエステル、微晶質セルロース、メトキシアルミニウムや寒天またはこれらの物質の混合物などを含んでもよい。
1.より長い体内における生物半減期。
2.PDモデル動物の線条体におけるドーパミンの濃度を顕著に向上させ、PDモデル動物の線条体におけるドーパミン作動性神経線維の欠失および黒質におけるドーパミン作動性ニューロンの欠失を顕著に抑制し、ドーパミン作動性ニューロンの機能を上げる。
3.海馬区域のニューロン細胞のアポトーシスを顕著に減少し、ADモデル動物の学習記憶能力を改善する。
4.神経変性疾患に顕著な神経保護作用を有し、有効的に神経変性疾患を治療する。
本発明のG-CSF二量体は、SEQ ID NO:1で構成し、またはSEQ ID NO:2-7から選ばれるG-CSF-Fc複合体から図1-3の二量体を形成する。製造方法は以下の通りである。
全遺伝子で合成されたG-CSF-Fc複合体のcDNA配列(SEQ ID NO:10またはSEQ ID NO:9に示すように)を使用し、ヒトG-CSF単量体の遺伝子とIgG2のFc遺伝子断片を連結し、5’にHindIII部位および哺乳動物細胞の発現に必要なエレメント、例えばKozak配列やシグナルペプチドを、3’にEcoRI部位を導入し、pUC19プラスミドにクローンし、pG-CSF-Fcと名づけ、E.coli TG1を形質転換した。pUC19プラスミドをHind IIIとEcoRIの酵素で切断し、約1400bpのG-CSF-IgG2Fc断片を回収し、Hind IIIとEcoRIの酵素で切断したpcDNA3(Invitrogen)発現プラスミドと連結し、発現プラスミドpEX-G-CSF-Fcを構築した。発現プラスミドpEX-G-CSF-Fcを直鎖化し、精製し、エレクトロポレーションでCHO細胞を形質転換し、スクリーニングし、ELISA法で発現量を検出し、蛋白質生産量の高い細胞株を選択し、細胞を冷凍保存し、細胞ライブラリーを調製した。
細胞ライブラリーから一種の細胞(〜 1×107 細胞/ml)を取り、10cmのシャーレで戻させ、10mLの基礎培地で培養し、37℃、5% CO2で24時間培養した。
種の増幅:10mLの培養物を30〜40mLの培地に継代し、細胞の濃度が1.0〜1.5×106細胞/mLで、且つ生存率≧90%の時点で、段階的に300〜400mLの培地に拡大し、振とうフラスコに置いて37℃、5% CO2で培養し、回転数が120rpmであった。
細胞タンクにおける増幅(3L-10L):細胞の濃度が1.0〜3.0×106細胞/mLで、且つ生存率≧90%の時点で、300〜400mLの培養物を3-10Lの細胞タンクに接種して培養し、培地のpHを6.8にし、溶解酸素が約50%で、回転数が65-100rpmであった。
細胞タンクによる生産(30L-100L):3-10Lの細胞タンクにおける細胞の濃度が1.0〜3.0×106細胞/mLで、且つ生存率≧90%の時点で、3-10Lの細胞タンクにおける培養物を30-100Lの細胞タンクに接種して培養し、培地のpHを6.8にし、溶解酸素が約50%で、回転数が65-100rpmで、12-48時間培養してから培地におけるグルコース濃度(<1g/L)をコントロールし、補充培養を行った。
細胞タンクにおける培養が終わった後、細胞上清(G-CSF-Fc複合体、G-CSF二量体、G-CSF-Fc多量体および代謝物を含む。)を収集し、収集後細胞上清をろ過、多段ゲルクロマトグラフィーで精製した。例えば、rProtein A Sepharose FF (GE Healthcare、cat#17-1279-04)で捕獲し、50 mM クエン酸/クエン酸ナトリウム、0.2M NaCl、pH 3.7-3.8の緩衝液で溶離し、純度>90%のG-CSF二量体を得た後、Capto Adhereクロマトグラフィーを使用し、50 mM NaAc/HAC、0.2 M NaCl、pH 4.5-5.0の緩衝液で溶離し、さらにSP Sepharose FF (GE Heathcare Cat #17-0729-04)を使用し、10mM PB (pH 6.0±0.1)緩衝液で平衡を取り、10mM PB、0.2M NaCl (pH 7.2±0.1)で溶離し、溶離液が低pHによるウイルス除去、ろ過などを経て、最終的にG-CSF二量体を得た。
ラットに1回100 μg/kgでG-CSF二量体(二量体が2つのSEQ ID NO:3に示すようなG-CSF-Fc複合体からなる)を注射し、薬物動態学のパラメーターは下述表1に示す(n=6)。単量体G-CSFは、ラットにおける半減期が約2時間である。
24人の健康の被験者を無作為にそれぞれ30、60、120、240ug/kg投与量群の4群に分け、G-CSF二量体(二量体が2つのSEQ ID NO:6に示すようなG-CSF-Fc複合体からなる)の投与量がそれぞれ30、60、120、240ug/kgで、1回で投与し、投与後0.5、1、2、4、8、16、24、36、48、72、96時間、6(120時間)、7、9、11、13および15日目で、採血して血清を分離して-70℃の冷蔵庫に冷凍保存した。ELISA法で血中薬物濃度(ELISA, Quantikine human G-CSF ELISA kit, R&D System, Inc. Minneapolis, Min, Cat:PDCS50)を検出した。検出結果は、ノンコンパートメントモデルで薬物動態学のパラメーター(ソフトウェアWinNonlin v 5.2、Pharsight Corporation、USA)を使用し、結果を下述表2に示す。
PC12細胞は、ラットのクロム親和性細胞腫の細胞系で、体外培養でドーパミンを合成・代謝・伝達することができ、体外モデルとして活性を有する化合物を選択することが可能である。
妊娠14日のSDラットの胎児の全脳を取って冷却しておいたD-Hanks液に入れ、解剖顕微鏡で黒質を取り、1mm3の大きさに切り、10mLの0.125%トリプシンを入れ、且つ37℃のインキュベーターで15min消化し、さらに組織を冷却しておいたDMEM+10%FBSの入った遠心管に移して最終消化を行い、ピペットで数回吹き、静置後上清を取ってもう一つの遠心管内に入れた。上述工程を2〜3回繰り返した。無血清ニューロン基礎培地neurobasal(invitrogen、製品番号:21103049)+無血清添加剤B27(invitrogen、製品番号:17504044)で8日培養し、2日おきに液を換えた。8日目に培養し、それぞれ異なる濃度のG-CSF二量体(G-CSF-D)とG-CSF単量体(G-CSF二量体の最終濃度:1、10、100ng/mL、G-CSFの最終濃度:0.4、4、40ng/mL)でニューロンを15分間処理した(Schneider Aら.J Clin Invest 2005, 115(8):2083-2098)。培地を完全に吸い出し、細胞をPBSで2回洗浄し、細胞溶解液のマニュアルで細胞を溶解した。細胞溶解液(Cell Signaling Technology、製品番号:9803、主要成分:20 mM Tris-HCl (pH 7.5)、150 mM NaCl、1 mM Na2EDTA、1 mM EGTA、1% Triton、2.5 mM sodium pyrophosphate、1 mM beta-glycerophosphate、1 mM Na3VO4、1 μg/ml leupeptin、1 mM PMSF)を氷上で細胞を20分間溶解し、細胞をかき取った。細胞溶解液を収集し、12000rpm、4℃で10分間遠心した。上清を吸い取り、蛋白質の濃度を測定した。別に100μlの上清を取ってSTAT3 [pY705] ELISAキット(invitrogen、製品番号:KH00481)でSTATのリン酸化レベルの変化を検出した。
1-メチル-4-フェニル-1,2,3,6-エトラヒドロピリジン(MPTP)は、ドーパミン作動性ニューロンを特異に損傷し、黒質におけるドーパミン作動性ニューロンを大量に欠失させ、パーキンソン病に似た症状を出すことができる。
50匹のマウスを無作為にそれぞれ10匹ずつ溶媒対照群、MPTPモデル群、MPTP+G-CSF 40μg/kg群、MPTP+GCSF-D 40μg/kg群、MPTP+GCSF-D 100μg/kg群の5群に分け、ここで、GCSF-DはSEQ ID NO:2-7に示すような配列から選ばれるG-CSF-Fc複合体からなるG-CSF二量体である。
MPTPは30mg/kgの投与量で腹腔注射し、5日間連続投与し、1日回復した後(7日目から)、MPTP+G-CSF 40μg/kg群は、G-CSFを40μg/kgで皮下注射し、G-CSFを毎日1回、5日間連続、即ち、7〜11日目で、MPTP+GCSF-D 40μg/kg群は、40μg/kgで皮下注射し、7、9日目にそれぞれGCSF-Dを1回、MPTP+GCSF-D 100μg/kg群は、100μg/kgで皮下注射し、7、9日目にそれぞれGCSF-Dを1回、溶媒対照群は、等体積の生理食塩水を投与した。
12日目に、動物に以下の評価を行った。
本実験では、MPTPの最後の投与後10日目に行動学測定を行った。方法:ポール試験(pole test)でPDにおける典型的な行動学症状である動作緩慢(Matsuuraら, 1997; Arakiら, 2001; Katoら, 2004)を測定した。
マウスを頭が上のように軽く粗いポールのトップに置いた(直径8mm、高さ55cm)。マウスが頭が上の状態から完全下に向いたまでの時間をT-turn(time to turn)と記録し、マウスが下への運動から四肢がすべてポールの底についたまでの時間をT-LA(locomotion activity time)と記録し、30sを超えた場合30sと記録した。すべてのマウスに対して5回測定を繰り返して平均値を取った。
方法:マウスを断頭し、線条体を取って重量を量った後、1.5mlの遠心管に入れ、すぐ砕氷に置いた。サンプル10mgあたりに300 μlの氷水浴中のサンプル処理液を入れた(0.2M過塩素酸、0.2mM二亜硫酸ナトリウム、0.01%EDTA-2Na、同時に内部標準として0.3μM DHBAを含有する)。以上の混合液を超音儀で超音波粉砕し、さらにそれを4℃、10000gで20 min遠心した。その上清液を取って0.22μMの水相ろ膜でろ過し、高速液相クロマトグラフィーで線条体におけるドーパミンの濃度を検出した。
方法:10%抱水クロラールで麻酔し、4%パラホルムアルデヒドで灌流した後、脳を取った。4%パラホルムアルデヒドで24時間固定し、さらにサンプルを10%、20%、30%のショ糖溶液に移してサンプルが底に沈殿するまで勾配脱水し、-20℃で冷凍スライサーで中脳と線条体の部位の冠状切片を作り、マウスの切片の厚さが20μmであった。THはドーパミン作動性ニューロンの特異的標識である。第一抗体は、モノクローナルマウス抗TH(1:1000, CHEMICON)で、線条体と中脳の部位の脳切片と共に4℃で一晩インキュベートし、PBSで3回洗浄した後、ビオチン化第二抗体で室温で1時間インキュベートした。SABC複合体を室温で1時間インキュベートした。DAB呈色を行い、エタノールで勾配脱水し、キシレンで透明にし、中性樹脂で封じた。黒質TH陽性細胞を計数し、線条体TH陽性染色の光学密度をスキャンした。
妊娠17日のSDラットの胎児の全脳を取って冷却しておいたD-Hanks液に入れ、解剖顕微鏡で海馬を取った。1mm3の大きさに切り、10mLの0.125%トリプシンを入れ、且つ37℃のインキュベーターで15min消化し、さらに組織を冷却しておいたDMEM+10%FBSの入った遠心管に移して最終消化を行い、ピペットで数回吹き、静置後上清を取ってもう一つの遠心管内に入れた。上述工程を2〜3回繰り返した。無血清ニューロン基礎培地neurobasal(invitrogen、製品番号:21103049)+無血清添加剤B27(invitrogen、製品番号:17504044)で8日培養し、2日おきに液を換えた。8日目に培養し、それぞれ異なる濃度のG-CSF二量体(SEQ ID NO:2-7に示すような配列から選ばれるG-CSF-Fc複合体からなる)とG-CSF(G-CSF二量体の最終濃度:1、10、100ng/mL、G-CSF単量体の最終濃度:0.4、4、40ng/mL)でニューロンを15分間処理した(Schneider Aら.J Clin Invest 2005, 115(8):2083-2098)。培地を完全に吸い出し、細胞をPBSで2回洗浄し、細胞溶解液のマニュアルで細胞を溶解した。細胞溶解液(Cell Signaling Technology、製品番号:9803、主要成分:20 mM Tris-HCl (pH 7.5)、150 mM NaCl、1 mM Na2EDTA、1 mM EGTA、1% Triton、2.5 mM sodium pyrophosphate、1 mM beta-glycerophosphate、1 mM Na3VO4、1 μg/ml leupeptin、1 mM PMSF)を氷上で細胞を20分間溶解し、細胞をかき取った。細胞溶解液を収集し、12000rpm、4℃で10分間遠心した。上清を吸い取り、蛋白質の濃度を測定した。別に100μlの上清を取ってSTAT3 [pY705] ELISAキット(invitrogen、製品番号:KH00481)でSTATのリン酸化レベルの変化を検出した。
PC12細胞は、神経因子(NGF)で誘導された後、突出が現れ、神経細胞の特性を有し、Aβ(βアミロイド)によるPC12細胞のアポトーシスは体外でADモデルを模擬することができる。
オス、180〜220gのSDラットを室温24±2℃の環境で飼い、明-暗12時間循環交代を維持し、充分な飼料を与え、自由に水を飲ませた。
50匹のマウスを無作為にそれぞれ10匹ずつ溶媒対照群、Aβモデル群、Aβ+G-CSF 40μg/kg群、Aβ+GCSF-D 40μg/kg群、Aβ+GCSF-D 100μg/kg群の6群に分けた。ここで、GCSF-DはSEQ ID NO:2-7に示すような配列から選ばれるG-CSF-Fc複合体からなるものである。
実験ラットは、1%ペントバルビタールトンナトリウム(40mg/kg)で麻酔した後、頭部を固定し、皮膚を消毒し、頭蓋の中線に沿って2cm切り口を入れ、骨膜を分離して頭骨を露出させ、大泉門から後ろに3.0mm、中線の左右でそれぞれ2.2mm開き、歯科ドリルで頭蓋骨に穴を開け、垂直に微量サンプラー針2.8mmを入れ、モデル群と各薬物処理群では5μl
Aβ1-40(βアミロイド)(2μg/μl)溶液を注入し、偽手術群では5ulの生理食塩水を注射した。
動物モデルを構築した後3日目に投与を開始し、Aβ+G-CSF 40μg/kg群は、G-CSFを40μg/kgで皮下注射し、G-CSFを毎日1回、5日間連続で、Aβ+GCSF-D 40μg/kg群は、40μg/kgで皮下注射し、3、5日目にそれぞれGCSF-Dを1回、Aβ+GCSF-D 100μg/kg群は、100μg/kgで皮下注射し、3、5日目にそれぞれGCSF-Dを1回、溶媒対照群は、等体積の生理食塩水を投与した。
10日目にMorris水迷宮で行動学測定を行った。
1-メチル-4-フェニル-1,2,3,6-エトラヒドロピリジン(MPTP)は、ドーパミン作動性ニューロンを特異に損傷し、黒質におけるドーパミン作動性ニューロンを大量に欠失させることで、パーキンソン病(PD)に似た症状が現れる。チロシンヒドロキシラーゼ(TH)は、ドーパミン作動性ニューロンの特異的標識で、黒質におけるドーパミン作動性ニューロンの数を定量に検出することができる。
MPTP+GCSF-D 30μg/kg群:30mg/kgの投与量でMPTPを腹腔注射し、5日間連続投与し、1日回復した後(即ち7日目から)30μg/kgでG-CSF-Dを皮下注射し、7、9、11日目にそれぞれG-CSF-Dを1回投与した。
MPTP+GCSF-D 100μg/kg群:30mg/kgの投与量でMPTPを腹腔注射し、5日間連続投与し、1日回復した後(即ち7日目から)100μg/kgでG-CSF-Dを皮下注射し、7、9、11日目にそれぞれG-CSF-Dを1回投与した。
MPTPモデル群:30mg/kgの投与量でMPTPを腹腔注射し、5日間連続投与し、動物を1日回復させた後、7日目から等体積の溶媒(0.5%マウス血清/PBS)を投与した。
正常対照群:等体積の生理食塩水を5日間連続投与し、動物を1日回復させた後、7日目から等体積の溶媒(0.5%マウス血清/PBS)を投与した。
前述G-CSF二量体(G-CSF-D)は2つのSEQ ID NO:6に示すような配列のG-CSF-Fcからなる。
12日目に、動物を殺処分し、線条体におけるドーパミン濃度を検出し、線条体におけるドーパミン作動性神経線維と黒質におけるドーパミン作動性ニューロンの様子を観察した。
方法:マウスを断頭し、線条体を取って重量を量った後、1.5mlの遠心管に入れ、すぐ砕氷に置いた。サンプル10mgあたりに300 μlの氷水浴中のサンプル処理液を入れた(0.2M過塩素酸、0.2mM二亜硫酸ナトリウム、0.01%EDTA-2Na、同時に内部標準として0.3μM DHBAを含有する)。以上の混合液を超音儀で超音波粉砕し、さらにそれを4℃、10000gで20 min遠心した。その上清液を取って0.22μMの水相ろ膜でろ過し、高速液相クロマトグラフィーで線条体におけるドーパミンの濃度を検出した。
方法:10%抱水クロラールで麻酔し、4%パラホルムアルデヒドで灌流した後、脳を取った。4%パラホルムアルデヒドで24時間固定し、さらにサンプルを10%、20%、30%のショ糖溶液に移してサンプルが底に沈殿するまで勾配脱水し、-20℃で冷凍スライサーで中脳と線条体の部位の冠状切片を作り、マウスの切片の厚さが20μmで、TH免疫組織化学分析を行った。第一抗体は、モノクローナルマウス抗TH抗体(1:1000、Sigma)で、線条体と中脳の部位の脳切片と共に4℃で一晩インキュベートし、PBSで3回洗浄した後、ビオチン化第二抗体(ヒツジ抗マウス)で室温で1時間インキュベートした。SABC複合体を室温で1時間インキュベートした。DAB呈色を行い、エタノールで勾配脱水し、キシレンで透明にし、中性樹脂で封じた。線条体TH陽性染色の光学密度のスキャン、黒質緻密部TH陽性細胞の計数などの測定を行った。
結果は、図5A、図5B、図6Aおよび図6Bに示された通りである。
実験方法(US7723302を参照):マウスに30mg/kgでMPTPを腹腔注射し、連続5日で投与し、PDマウスモデルを得た。動物を1日回復させた後、250 μg/kg G-CSF(Neupogen、Amgen)を投与し、連続7日で投与し、最後の投与後(即ち投与終了後)の異なる時刻で黒質緻密部TH陽性ドーパミン作動性ニューロンの数を観察した。
モデル群:1回目の投与前の黒質緻密部TH陽性ドーパミン作動性ニューロンの数。
正常群:MPTPを注射しなかったマウス。
投与終了後1、7、14日目に黒質緻密部TH陽性ドーパミン作動性ニューロンは大体それぞれ正常群の70%、80%、77%に回復した。
実施例10に記載されるように、本発明のG-CSF二量体は、毎回30 μg/kgまたは100 μg/kgの投与量で、間欠的に3回投与した後、投与終了後の1日目に、30 μg/kg群、100 μg/kg群では、黒質におけるTH陽性ドーパミン作動性ニューロンは大体それぞれ正常対照群の86%、99%に回復した。特に、100 μg/kg群では、投与終了後の1日目に、TH陽性ドーパミン作動性ニューロンはほとんど正常対照群のレベルに回復した。
比較例1では、250 μg/kgの投与量のG-CSFで連続7日投与しても、投与終了後の1日目に、TH陽性ドーパミン作動性ニューロンは正常群の70%にしか回復しなかった。
合計投与量から見ると、G-CSF二量体100 μg/kg群の合計投与量が300μgで、比較例1では、G-CSF単量体の合計投与量が1750μgである。
G-CSF単量体のモル分子濃度から見ると、G-CSF単量体とG-CSF二量体の分子量比が約1:5であるため、本発明のG-CSF二量体100 μg/kg群では、G-CSF単量体のモル分子濃度は比較例1の1/15だけである。
Claims (9)
- 神経変性疾患を治療・予防する組成物の製造におけるコロニー刺激因子G-CSF二量体の使用であって、
前記コロニー刺激因子G-CSF二量体は2つのG-CSF-Fc複合体を含み、
前記G-CSF-Fc複合体は、いずれも、SEQ ID NO:2〜7からなるグループから選択されるアミノ酸配列を含む、使用。 - 前述神経変性疾患は、アルツハイマー病、ハンチントン病、筋萎縮性側索硬化症、脊髄性筋萎縮症、原発性側索硬化症、および脊髓小脳失調症のうちのいずれかであることを特徴とする請求項1に記載の使用。
- 前述コロニー刺激因子G-CSFの二量体は、ヒトコロニー刺激因子G-CSF二量体であることを特徴とする請求項1または2に記載の使用。
- 前述コロニー刺激因子G-CSFの二量体は、構造が式Iで示され、

ことを特徴とする請求項1または2に記載の使用。 - 活性成分としてコロニー刺激因子G-CSFの二量体を含む、ことを特徴とする神経変性疾患を治療する医薬品であって、
前記コロニー刺激因子G-CSF二量体は2つのG-CSF-Fc複合体を含み、
前記G-CSF-Fc複合体は、いずれも、SEQ ID NO:2〜7からなるグループから選択されるアミノ酸配列を含む、医薬品。 - 前述神経変性疾患は、パーキンソン病、アルツハイマー病、ハンチントン病、筋萎縮性側索硬化症、脊髄性筋萎縮症、原発性側索硬化症、脊髓小脳失調症からなる群から選ばれることを特徴とする請求項5に記載の医薬品。
- 前述コロニー刺激因子G-CSFの二量体は、ヒトコロニー刺激因子G-CSF二量体であることを特徴とする請求項5または6に記載の医薬品。
- 前述コロニー刺激因子G-CSFの二量体は、構造が式Iで示され、
