Description
細胞接着性人工ペプチド(P)が、遺伝子組換微生物によって合成され、細胞接着性最小アミノ酸配列(X)を1分子中に少なくとも1個有するポリペプチド(P1)であって、
ポリペプチド(P1)が、さらにGAGAGS(21)で表されるアミノ酸配列を1分子中に30〜1,000個有してなる生体組織用細胞接着性材料である。
アミノ基含有モノマーの重合体の重量平均分子量は、500〜100万が好ましく、さらに好ましくは1,000〜80万、特に好ましくは2,000〜50万である。なお、重量平均分子量は、ゲルパーミエーションクロマトグラフィー(GPC)で測定することができる{基準物質:分子量420〜20,600,000のポリスチレンスタンダード(東ソー製)等}。
これらのうち、結合強度の観点等から、(1)の化学結合させる方法が好ましく、さらに好ましくは共有結合させる方法である。
RGD配列(1)の13個と(GAGAGS)9配列(22)の12個とを有し、これらが交互に位置する構造を有する数平均分子量(Mn)約11万のペプチド(23){「プロネクチンF」、プロネクチンは三洋化成工業(株)の登録商標(日本及び米国)である。三洋化成工業(株)製<以下同じ>};
特表平3−502935号公報(対応するPCT出願;WO90/05177号パンフレット;これに開示された開示内容を参照により本出願に取り込む。)中の実施例記載の方法に準じて、RGD配列(1)の13個と(GAGAGS)9配列(22)の12個とを有し、これらが交互に位置する構造を有する数平均分子量(Mn)約11万のペプチド(23){細胞接着性人工ペプチド(P1−0)}を遺伝子組換え大腸菌により製造した。
細胞接着性人工ペプチド(P1−1)を細胞接着性人工ペプチド(P1−0)に変更したこと以外、実施例1と同様にして、本発明の生体組織用細胞接着性材料(A2)を得た。この生体組織用細胞接着性材料(A2)に結合された細胞接着性人工ペプチド(P1−0)の製造直後の結合量(J)は、70ng/mm2であった。
塩酸N,N−ジメチルアミノエチルクロリド(特級試薬)をクロロ酢酸(特級試薬)に変更したこと以外、実施例1と同様にして、細胞接着性人工ペプチド(P1−2)を得た。細胞接着性人工ペプチド(P1−2)に導入されたクロロ酢酸の数は、1分子中に12個であった。
細胞接着性人工ペプチド(P1−1)をRGD配列の5個と(GAGAGS)3配列(24)の5個とを有しこれらが交互に位置する構造を有する数平均分子量(Mn)約2万のペプチド(P2−0)(特表平3−502935号公報中の実施例記載の方法に準じて調製した)に変更したこと以外、実施例1と同様にして、参考用の生体組織用細胞接着性材料(A4)を得た。この生体組織用細胞接着性材料(A4)に結合された細胞接着性人工ペプチド(P2−0)の製造直後の結合量(J)は、60ng/mm2であった。
細胞接着性人工ペプチド(P1−0)を細胞接着性人工ペプチド(P2−0)に変更したこと以外、実施例1と同様にして、細胞接着性人工ペプチド(P2−1)を得た。細胞接着性人工ペプチド(P2−1)に導入された塩酸N,N−ジメチルアミノエチルクロリドの数は、1分子中に6個であった。
細胞接着性人工ペプチド(P1−1)をYIGSR配列(4)の5個と(GAGAGS)3配列(24)の5個とを有しこれらが交互に位置する構造を有する数平均分子量(Mn)約2万のペプチド(P3−0)(特表平3−502935号公報中の実施例記載の方法に準じて調製した)に変更したこと以外、実施例1と同様にして、参考用の生体組織用細胞接着性材料(A6)を得た。この生体組織用細胞接着性材料(A6)に結合された細胞接着性人工ペプチド(P3−0)の製造直後の結合量(J)は、60ng/mm2であった。
細胞接着性人工ペプチド(P1−1)をIKVAV配列(9)の5個と(GAGAGS)3配列(24)の5個とを有しこれらが交互に位置する構造を有する数平均分子量(Mn)約2万のペプチド(P4−0)(特表平3−502935号公報中の実施例記載の方法に準じて調製した)に変更したこと以外、実施例1と同様にして、参考用の生体組織用細胞接着性材料(A7)を得た。この生体組織用細胞接着性材料(A7)に結合された細胞接着性人工ペプチド(P4−0)の製造直後の結合量(J)は、60ng/mm2であった。
「両極間に3.0Vの電圧を加えて10分間通電し、電気化学反応を行った」を実施しなかったこと以外、実施例1と同様にして、比較用の生体組織用細胞接着性材料(B1)を得た。この生体組織用細胞接着性材料(B1)に結合された細胞接着性人工ペプチド(P1−1)の製造直後の結合量(J)は、60ng/mm2であった。
細胞接着性人工ペプチド(P1−1)と3−グリシドキシプロピルトリエトキシシラン各々200μg/gの濃度で脱イオン水に溶解した溶液を作製し、これらを等量混合した溶液200mL中に生体組織用材料(チタンプレート、100mm×100mm×0.1mmの直方体)を浸漬し、20〜30℃で2時間放置した。その後、チタンプレートを取り出し、脱イオン水200mLに浸漬し直ぐに取り出す洗浄操作を3回行い、次いで、60℃の循風乾燥機の中で2時間乾燥して、比較用の生体組織用細胞接着性材料(B2)を得た。この生体組織用細胞接着性材料(B2)に結合された細胞接着性人工ペプチド(P1−1)の製造直後の結合量(J)は、90ng/mm2であった。
「両極間に3.0Vの電圧を加えて10分間通電し、電気化学反応を行った」を実施しなかったこと、及び細胞接着性人工ペプチド(P1−1)を細胞接着性人工ペプチド(P1−0)に変更したこと以外、実施例1と同様にして、比較用の生体組織用細胞接着性材料(B3)得た。この生体組織用細胞接着性材料(B3)に結合された細胞接着性人工ペプチド(P1−1)の製造直後の結合量(J)は、60ng/mm2であった。
細胞接着性人工ペプチド(P1−1)をウシ血清アルブミン(BSA)に変更したこと以外、実施例1と同様にして、比較用の生体組織用細胞接着性材料(B4)得た。この生体組織用細胞接着性材料(B4)に結合されたウシ血清アルブミン(BSA)の製造直後の結合量(J)は、50ng/mm2であった。
(チタンプレートと細胞接着性人工ペプチドの結合強度評価:超音波洗浄後の結合量)
試験片{生体組織用細胞接着性材料(A1)〜(A7)及び(B1)〜(B4)}を0.5重量%ドデシル硫酸ナトリウム(特級試薬)水溶液500mLに浸漬し、超音波洗浄器(テックジャム社製WS−600−28S、28kHz)内で10分間超音波を加えた後、試験片を取り出し、脱イオン水500mLに浸漬し直ぐに取り出す洗浄操作を3回行い、次いで、60℃の循風乾燥機の中で2時間乾燥した。試験片に結合している細胞接着性人工ペプチド又はウシ血清アルブミン(BSA)の結合量を実施例1と同様にしてトリニトロベンゼンスルホン酸(TNBS)法により定量した。これらの結果を表1に示す。
(歯肉上皮増殖試験評価)
試験片{生体組織用細胞接着性材料(A1)〜(A7)及び(B1)〜(B4)を6×6mm×0.1mmに切断した直方体}を用いて、以下のようにして歯肉上皮増殖率(%)を求めた。
歯肉上皮増殖率(%)=〔1−(面積H/面積G)〕×100
(チタンプレートとウサギ上顎頬側歯肉との接着強度評価)
試験片{生体組織用細胞接着性材料(A1)〜(A7)及び(B1)〜(B4)を4×8mm×0.1mmに切断した直方体}を用いて、以下のようにして接着強度を求めた。
(チタンプレートとラット上顎頬側歯肉との接着強度評価)
試験片{生体組織用細胞接着性材料(A1)〜(A7)及び(B1)〜(B4)を4×0.1×0.1mmに切断した立方体}を用いて以下のようにして接着強度を求めた。
Claims (6)
- 細胞接着性人工ペプチド(P)が生体組織用材料の表面に電気化学反応により固定された生体組織用細胞接着性材料であって、
細胞接着性人工ペプチド(P)が、遺伝子組換微生物によって合成され、細胞接着性最小アミノ酸配列(X)を1分子中に少なくとも1個有するポリペプチド(P1)であって、
ポリペプチド(P1)が、さらにGAGAGS(21)で表されるアミノ酸配列を1分子中に30〜1,000個有してなる生体組織用細胞接着性材料。 - ポリペプチド(P1)1分子中の細胞接着性最小アミノ酸配列(X)の数が3〜50個である請求項1に記載の生体組織用細胞接着性材料。
- 細胞接着性最小アミノ酸配列(X)が、アミノ酸一文字表記で現わされる、RGD配列(1)、LDV配列(2)、REDV配列(3)、YIGSR配列(4)、PDSGR配列(5)、RYVVLPR配列(6)、LGTIPG配列(7)、RNIAEIIKDI配列(8)、IKVAV配列(9)、LRE配列(10)、DGEA配列(11)及びHAV配列(12)からなる群より選ばれる少なくとも1種の配列である請求項1又は2に記載の生体組織用細胞接着性材料。
- 血管内皮細胞、平滑筋細胞、線維芽細胞、筋肉細胞、脂肪細胞、神経細胞、肝実質細胞、膵ラ島細胞、腎上皮細胞、近位尿細管上皮細胞、メサンギウム細胞、上皮細胞、視細胞、角膜上皮細胞、角膜内皮細胞、間質細胞、骨芽細胞、骨細胞、破骨細胞、軟骨芽細胞、軟骨細胞、歯根膜細胞及びこれらの幹細胞からなる群より選ばれる少なくとも1種の細胞を接着させるために用いられる請求項1〜3のいずれかに記載の生体組織用細胞接着性材料。
- 人工臓器用基材、歯科用基材、整形外科用基材又は眼科用基材に用いる請求項1〜4のいずれかに記載の生体組織用細胞接着性材料。
- 請求項1〜5のいずれかに記載された生体組織用細胞接着性材料から構成される医療用器具。
