Description
本出願は、2008年11月6日提出の米国仮特許出願第61/112,018号の利益を主張し、これは参照によりその全体が本明細書に組み込まれる。

本明細書に開示の態様および実施形態は、造血幹細胞および前駆細胞の生着を強化するための材料および方法に関する。
材料
マウス C57B1/6マウスは、Jackson Laboratories(Bar Harbor、ME、USA)から購入した。B6.SJL−PtrcAPep3B/BoyJ(BOYJ)およびF1 C57B1/6/BOYJハイブリッドを、研究所内で繁殖させた。すべての動物を、食物および酸性化水を連続提供されるマイクロアイソレーターケージにおいて飼育した。移植研究に使用するマウスは、放射時、および移植後30日の間、ドキシサイクリンの供給を受けた。The Animal Care and Use Committee of Indiana University School of Medicineは、すべての動物プロトコルを承認した。
限界希釈競合的および非競合的移植
WBM細胞(CD45.2)を、1マイクロモル濃度のdmPGE2(Cayman Chemical、Ann Arbor、MI)/1×106細胞または滅菌、非発熱性PBS中0.01%のETOHのどちらかを用いて、氷上において2時間処置した。インキュベーション後、細胞を2回洗浄し、2×105のコンジェニックCD45.1競合骨髄細胞と、0.075:1、0.25:1、1:1および2.5:1の割合で混合し、致死的に放射線(1100−cGy分割線量)を浴びたCD45.1マウスに、尾静脈注射により移植した(5マウス/希釈)。PB中のCD45.1およびCD45.2細胞を、フローサイトメトリによって、月1回決定した。1対1競合移植のために、CD45.1マウスおよびCD45.2マウス由来のWBMを、ビヒクルまたはdmPGE2を用いて処置しCD45.1/CD45.2マウス由来の2×105の競合骨髄細胞と、0.075:1、0.25:1、1:1および2.5:1の割合で混合し、致死的に放射線を浴びたCD45.1/CD45.2マウスに移植した。PB中のCD45.1、CD45.2およびCD45.1/CD45.2細胞の一部(proportion)を、月1回決定した。HSPCの頻度を、L−CALCソフトウェア(Stem Cell Technologies、Vancouver BC、Canada)を使用して、Poisson統計によって定量化し、キメラ現象に対する寄与は<5%であり、陰性レシピエントと考えられる。競合的再構築単位(CRU)を、記載のように(Harrison、1980)計算した。二次移植のために、移植後20週に、すでに移植を受けた1:1比のF1ハイブリッドマウス由来の2×106のWBMを、致死的に放射線を浴びたF1ハイブリッドマウスに非競合的様式で注射し、PBのキメラ現象および三血球系の再構成を月1回評価した。
CD45.1/CD45.2ハイブリッドマウスに移植されたCD45.2およびCD45.1のコンジェニック移植体を利用する限界希釈競合移植モデルの使用により、HSPCをPGE2に短期曝露することが、HSPCおよび競合的再構築単位(CRU)の頻度の長期強化を生み出すことが実証された。ここで図1Aを参照すると、CD45.1またはCD45.2マウス由来の骨髄は、それぞれ、ビヒクルまたはdmPGE2を用いて処置された。CD45.1/CD45.2ハイブリッド骨髄細胞を競合相手として使用した。限界希釈を、致死的に放射線(1100cGys、分割線量)を浴びたCD45.1/CD45.2ハイブリッドマウスに移植し、PB中のキメラ現象を20週間にわたって分析した。個々の細胞集団を検出する代表的フロープロットを示す(下段パネル)。
伝えられるところでは、PGE2は、4種の特異的な、高度に保存されたGタンパク質共役受容体;EP1〜EP4と相互作用する(SugimotoおよびNarumiya、2007年;Tsuboiら、2002年)。EP受容体のレパートリーは、多重の、時としてPGE2に起因する逆の応答の原因となる(Breyerら、2001年)。HSPCにおけるPGE2受容体サブタイプの発現は今まで知られていない。ここで図2Aを参照すると、造血前駆細胞(HPC)に富んだc−kit+Linneg(KL)細胞およびHSPCに富んだSca−1+c−kit+Linneg(SKL)細胞におけるEP受容体の分析は、4種すべてのEP受容体(EP3+、EP2、EP1およびEP4)が発現されたことを示した。さらにここで図2A(右パネル)を参照すると、QRT−PCRによりFACSにより選別されたKLおよびSKL細胞中に、4種すべてのEP受容体に関するmRNAが検出されていた。ここで図2A(中央パネル)を参照すると、EP3に関する解離曲線はいくつかのピークを示し、EP3の公知の多重スプライス変異体と一致する(Nambaら、1993年)。KLまたはSKL細胞に関して、任意のEP受容体サブタイプの表面発現またはmRNAレベルの間に、有意な定量的差異は見られなかった。ここで図2Bを参照すると、ヒトCD34+UCB細胞の表面において、マウス細胞の類似体(右パネル)は4種すべての受容体サブタイプを発現し、QRT−PCR分析により、4種すべてのEP受容体のmRNAが検出されていた(図2B)。
PGE2に対するパルス曝露において観察されるHSPCの生着の強化は、HSPCの数および/または細胞の促進をもたらす、または宿主の骨髄におけるHSPCのホーミングもしくは増殖をもたらす細胞周期状態に起因すると思われる。その原因と無関係に、骨髄ニッチはHSPCにとって自己再生および増殖に必要であり、これらのニッチへのHSPCのホーミングだけが長期再構築を提供し得る可能性が非常に高い。さらにここで図3Aを参照すると、HSPCのホーミングを評価するために、CFSE標識された全骨髄(WBM)CD45.2細胞は、dmPGE2またはビヒクルを氷上で2時間パルスされ、洗浄され、致死的に放射線を浴びたCD45.2宿主にIV注射された。16時間後、骨髄にホーミングするCFSE+細胞の合計およびKLおよびSKL細胞集団内のホーミング事象の数を定量した。総WBM細胞を評価した場合、骨髄へのCFSE+細胞のホーミングのパーセントにおいて、dmPGE2処置細胞およびビヒクル処置細胞の間に差異は観察されなかったが、対照よりも有意に多くのSKL細胞が骨髄にホーミングした。ここで図3Bを参照すると、ビヒクル処置細胞または未操作細胞と比較して、コンジェニックモデルにおいて、dmPGE2処置細胞に関してSKL細胞の有意により大きなパーセントが観察された。未処置細胞およびビヒクル−処置細胞の間にホーミング効率の差は見られなかった。
ここで図4Aを参照すると、ストロマ細胞由来因子−1アルファ(SDF−1a)/CXCR4軸は、HSPCの輸送およびホーミングと関係している。本研究は、dmPGE2処置HSPCのホーミングの改善は、SDF−1a/CXCR4のシグナル伝達の増加の結果であったかどうかを評価した。Linneg細胞のdmPGE2に対するパルス曝露は、KLおよびSKL細胞におけるCXCR4の発現を増加させ(図4A);同様に、dmPGE2のパルス曝露は、予想通りCD34+UCB細胞におけるCXCR4の発現を増加させた。QRT−PCRは、ビヒクルと比較してdmPGE2処置細胞においてCXCR4 mRNAレベルが上昇し、最大の上昇は6時間目に観察されたことを実証した(データ非掲載)。
PGE2処置は、HSPCの頻度およびCRUを4倍増加させる(図1)が、ホーミングはわずか2倍強化され(図3)、他の事象が生着の強化に関与していることを示唆する。アポトーシスは、正常な造血または悪性の造血において重要な制御過程であり、PGE2は抗アポトーシスシグナル伝達に関係している。さらに、EP受容体の下流シグナル伝達分子であるcAMPの活性化は、CD34+細胞においてアポトーシスを阻害する。これらの結果に一致する1つの仮説は、dmPGE2処置が、HSPCの生存および/または増殖に影響を与え、生着の強化に寄与するというものである。HSPCの生存に対するdmPGE2の効果を評価するために、Linneg細胞に、0.1ナノモル濃度〜1マイクロモル濃度のdmPGE2またはビヒクルをパルスし、成長因子を含まない低血清培養培地において培養した。dmPGE2に対するパルス曝露は、用量依存性様式でSKL細胞においてアポトーシスを低下させ(図5A)1マイクロモル濃度で約65%の阻害に達した。
サバイビンは、HSPCが細胞周期に進入し、進行することを制御する。さらに、HSPCの増殖および自己再生に関係するβ−カテニンは、EP受容体経路の下流に位置する。これらの細胞周期制御因子を調節するPGE2の能力は、HSPCの自己再生および増殖における増加が、dmPGE2をパルスされた細胞の生着の強化に寄与しうることを示唆している。この仮説を試験するために、in vitroでdmPGE2またはビヒクルをパルスされたSKL細胞の細胞周期状態を分析した。ここで図6Aを参照すると、dmPGE2に対するパルス曝露が、細胞循環の増加の指標である、SKL細胞中のDNA含有量を増加させた(左パネル、右上の象限)。3つの実験において、対照と比較してdmPGE2処置後に60%を超えるSKL細胞が細胞周期のS+G2/M期にあった(図6A、右パネル)。KLまたはLinneg細胞の細胞周期速度に関して有意な効果は見られず(非掲載)、dmPGE2が初期HSPCのサイクル状態を選択的に増加させることを示唆している。
PGE2は、CXCR4受容体の発現を増加させるので、骨髄において、SDF−1/CXCR4のシグナル伝達はHSPCの輸送および保有にとって重要である。これらの結果と一致する1つの仮説は、COX1/COX2二重阻害剤のインドメタシンによるPGE2の内因性生合成の阻害もまたHSPCを動員することである。ここで図7Aおよび7Bを参照すると、図7Aおよび7Bは、150μg/kgのインドメタシンまたは150ug/kgのバイカレインを単独で(図7A)またはG−CSFと一緒に(図7B)4日間、毎日SC投与することによる、CFU−GMの動員に関する効果を示す。ここで図7Aを参照すると、150μg/kgのインドメタシンをSCで1日1回4日間の投与することにより、動員される前駆細胞の数が4倍増加した。ここで図7Bを参照すると、インドメタシンとG−CSFとを同時投与することにより、末梢血幹細胞の動員において高度に相乗的な増加が起こった。リポキシゲナーゼの阻害剤であるバイカレインは、ベースラインまたはG−CSF誘導性CFU−GM動員に関して効果を有さず、観察された効果が、シクロオキシゲナーゼ経路の阻害に特異的であったことを示唆する。データは、それぞれ個別にアッセイしたN=3マウスに関する、動員されたCFU−GM/血液mlの平均±SEMを表す。
CXCR4を評価するために、Lineagenegマウスの骨髄細胞またはCD34+UCBを、1マイクロモル濃度のdmPGE2またはビヒクル対照のどちらかを用いて2時間氷上で処置し、洗浄し、次いでRPMI−1640/10%HI−FBS中で37℃において24時間培養し、SKL(マウス細胞)またはCD34(ヒト)およびCXCR4に関して染色し、FACSにより分析した。ここで図4Aを参照すると、dmPGE2を用いて処置後24時間のKLおよびSKL細胞ならびにヒトCD34+UCB細胞におけるCXCR4の発現である。データは、dmPGE2またはビヒクルを用いた処置による、CXCR4の平均蛍光強度(MFI)の変化の平均±SEMで表す(n=3)。QRT−PCRによる分析で、CXCR4mRNAの2.65倍の増加が実証される。
新たに単離されたLineagenegマウス骨髄細胞に、dmPGE2またはビヒクルを2時間パルスし、洗浄し、10%HI−FCSを含む培地に再懸濁し、37℃において16時間培養した。インキュベーション後、細胞を洗浄し、RPMI/0.5%BSAに再懸濁し、トランスウェルにおいてrmSDF−1αに4時間遊走させた。細胞遊走の合計を、フローサイトメトリにより測定した。ここで図9を参照すると、SKL細胞遊走の合計は、dmPGE2をパルスした細胞がより高かった。データは、3つの実験に関する遊走パーセントの平均±SEMである。ビヒクルを用いて処置した細胞と比較したdmPGE2処置細胞に関して、†P<0.05。
新たに単離されたUCB CD34+細胞に、dmPGE2またはビヒクルを2時間パルスし、洗浄し、10%HI−FCSを含む培地に再懸濁し、37℃において16時間培養した。インキュベーション後、細胞を洗浄し、RPMI/0.5%BSAに再懸濁し、rhSDF−1への遊走をフローサイトメトリにより測定した。遊走アッセイの前に、CXCR4受容体をブロックするために、複製細胞を、5マイクログラム/mlのAMD3100と共に30分間インキュベートした。ここで図10を参照すると、データは、3つの実験に関する遊走パーセントの平均±SEMである。
ホーミングにおけるCXCR4の役割を評価するために、LineagenegCD45.2細胞を、ビヒクルまたは1マイクロモル濃度のdmPGE2+10マイクロモル濃度のAMD3100を用いて処置し、2×106の処置細胞を、致死的に放射線を浴びたCD45.1マウスに注射し、移植後16時間でホーミングしたSKL細胞を回収しFACSにより分析した。ここで図11を参照すると、10マイクロモル濃度のAMD3100不在下または存在下の、ビヒクルおよびdmPGE2処置細胞の骨髄へのホーミング効率である。ホーミングアッセイの前に、細胞を、AMD3100と共に30分間インキュベートした。
Lineageneg細胞を、ビヒクルまたは1マイクロモル濃度のdmPGE2どちらかを用いて2時間処置し、洗浄し、rmSCF、rhFlt3およびrhTpoを含む培地において培養した。20時間後、細胞を、SKLおよびHoechst−33342およびPyronin−Yに対して染色した。細胞周期におけるSKL細胞の割合を、FACSにより測定した。ここで図12を参照すると、ゲーティングされたSKL細胞の細胞周期分布を示す代表的フロープロットであり、それぞれ個別にアッセイした平均±SEM、n=9マウスの3つの実験による、ビヒクル対照と比較した、dmPGE2処置細胞に関する細胞周期における倍数増加を組み合わせたデータである。細胞周期におけるSKL細胞の割合を、FACSにより測定した。ゲーティングされたSKL細胞の細胞周期分布を示す代表的フロープロットであり、それぞれ個別にアッセイした平均±SEM、n=9マウスの3つの実験による、ビヒクル対照と比較した、dmPGE2処置細胞に関する細胞周期における倍数増加を組み合わせたデータである。
ここで図8を参照すると、ビヒクルまたは1マイクロモル濃度のdmPGE2どちらかを用いて2時間処置し、成長因子(50ng/mlのrmSCF、それぞれ100ng/mlのrhFlt−3およびrhTPO)の存在下で20時間培養し、SLAM SKL、Hoechst−33342およびPyronin−Yに対して染色し、細胞周期のG0、G1、SおよびG2/M期にあるSLAM SKL細胞の割合を、FACSによるDNAおよびRNA含有量の定量化によって決定したLinneg骨髄細胞を使用して収集したデータを要約した表である。データは、それぞれ個別にアッセイしたn=9マウスに関する平均±SEMである。(b)G1+S+G2Mにある細胞のパーセント;n=9マウスに関する組み合わせデータ。(*)ビヒクル対照と比較してP<0.05。
CD45.1Lineageneg骨髄細胞を、dmPGE2またはビヒクルを用いて処置し、致死的に放射線を浴びたCD45.2マウスに移植した。移植直後、BrdUを飲料水中に提供し、IP注射によって投与した。16時間後骨髄を分析し、BrdU+であったCD45.1+、SKL細胞の割合を、FACS分析により分析した。ここで図13を参照すると、CD45.1Linneg骨髄細胞をdmPGE2またはビヒクルを用いて処置し、致死的に放射線を浴びたCD45.2マウスに移植した。移植直後、BrdUを飲料水中に提供し、IP注射によって投与した。16時間後骨髄を分析し、BrdU+であったCD45.1+、SKL細胞の割合を、FACS分析により分析した。PGE2を用いて処置したSKL細胞がより高い割合で骨髄にホーミングした。データは、平均±SEM、n=5/マウス/群であり、それぞれ個別にアッセイした。
1対1競合分析のために、CD45.1およびCD45.2マウス由来のWBMを、ビヒクルまたはdmPGE2を用いて処置し、様々な割合で2×105のCD45.1/CD45.2マウス由来の競合骨髄細胞と混合し、致死的に放射線を浴びたCD45.1/CD45.2マウスに移植した。PB中のCD45.1、CD45.2およびCD45.1/CD45.2細胞の割合を、月1回決定した。二次、三次および四次移植のために、1:1比ですでに移植されたCD45.1/CD45.2マウス由来の2×106のWBMを、致死的に放射線を浴びたCD45.1/CD45.2マウスに非競合的様式で注射した。PB中のCD45.1、CD45.2およびCD45.1/CD45.2細胞の割合を、月1回決定した。ここで図14を参照すると、一次移植の20週目(二次移植の時)およびサブコホートにおける32週目(二次移植の12週目分析の時)、二次移植の12週および24週目、三次および四次移植も同様にそれぞれ12週目に関する、dmPGE2処置細胞対ビヒクルのキメラ現象の増加である。20週の一次移植に関するデータは、それぞれ個別にアッセイした、n=5マウス/群/実験の2つのプールした実験からである。二次、三次および四次移植に関するデータは、それぞれ個別にアッセイした、n=5マウス/群からである。
マウスに、ゼラチン中の150マイクログラム/kgのインドメタシンまたは150マイクログラム/kgのバイカレイン(リポキシゲナーゼ阻害剤)を、G−CSFと共に、または無しで48時間ごとに4日間のSC処置を与えた。CFU−GMの動員を、すでに記載のように決定した(Pelusら、Experimental Hematology33巻(2005年)295〜307頁)。ここで図7Aおよび7Bを参照すると、シクロオキシゲナーゼ二重阻害剤のインドメタシンおよびG−CSFの併用は、マウスHSPCを相乗的に動員している。150μg/kgインドメタシンまたは150ug/kgのバイカレイン(リポキシゲナーゼ阻害剤)を単独で(図7A)、またはG−CSFと一緒に(図7B)4日間、毎日SC投与することによるCFU−GMの動員に関する効果である。データは、それぞれアッセイした、N=3マウスの動員されたCFU−GM/血液mlの平均±SEMとして表す。
マウスに、ビヒクルまたはインドメタシン(50マイクログラム/マウス)を、4日間毎日、1日2回(bid)SC注射した。5日目に、マウスに、ビヒクルまたはAMD3100(5mg/kg)のどちらかを与えた。1時間後、マウスを犠牲にし、CFU−GMの動員を、先に記載(Pelusら、Experimental Hematology 33巻(2005年)295〜307頁)のように決定した。ここで図18を参照すると、ビヒクルまたはインドメタシン処置単独によるCFU−GMの動員である(左パネル)。AMD3100またはインドメタシン処置+AMD3100の単回投与によるCFU−GMの動員のグラフである(右パネル)。データは、それぞれ個別にアッセイされたN=5マウス/群の平均±SEMとして表す。
マウスを、ビヒクル、インドメタシン(50マイクログラム/マウス、1日2回SC、4日間)、AMD3100(5mg/kg、5日目)、G−CSF(1マイクログラム/マウス、1日2回SC、4日間)、AMD3100+GROβ(それぞれ、5ミリグラム/kgおよび20ミリグラム/kg、5日目)、AMD3100+インドメタシン(インドメタシン50マイクログラム/マウス、1日2回4日間;AMD3100 5ミリグラム/kg 5日目)またはG−CSF+インドメタシン(それぞれ、1マイクログラムおよび50マイクログラム、1日2回SC、4日間)により処置した。CFU−GMの動員を、先に記載(Pelusら、Experimental Hematology 33巻(2005年)295〜307頁)のように決定した。ここで図19を参照すると、上に要約した様々な処置レジメンに関してプロットしたCFU−GM/末梢血mLである。
CD45.1マウスを、G−CSF(1マイクログラム/マウス、1日2回、SC、4日間)またはG−CSF+インドメタシン(50マイクログラム/マウス、1日2回、SC、4日間)を用いて動員し、末梢血単核細胞(PBMC)を5日目に採取した。PBMCを、様々な比率でCD45.2骨髄と混合し、致死的に放射線を浴びた(1100cGy、分割線量)CD45.2マウスに移植した。ここで図21を参照すると、移植後12週目の競合的再構築単位を示す(左パネル)。データは、それぞれ個別にアッセイされたN=5マウス/群/実験の2つの実験から、平均±SEMとして表す。G−CSF単独と比較して、インドメタシンとG−CSFとの同時投与により動員されたPBMCの生着に改善がなかったので、ホーミングの欠損は、CXCR4受容体の発現の減少の結果として起こると思われ、これは、インドメタシン処置とG−CSF処置とに時間差を与えることによって緩和できるという仮説を立てた。CD45.2マウスを、G−CSF(1マイクログラム/マウス、1日2回、SC、4日間)、時間差無しのG−CSF+インドメタシン(50マイクログラム/マウス、1日2回、SC、4日間)、1日の時間差のG−CSF+インドメタシン(インドメタシンを1日目に開始し4日間与え、G−CSFをインドメタシン処置の2日目から開始して4日間与え、PBMSの採取前に、G−CSFを与えるが、インドメタシンは与えない日を1日作る)または2日の時間差のG−CSF+インドメタシン(インドメタシンを1日目に開始し4日間与え、G−CSFをインドメタシン処置の3日目から開始して4日間与え、PBMSの採取前に、G−CSFを与えるが、インドメタシンは与えない日を2日作る)により動員した。ここで図21を参照すると(右パネル)、SKL細胞におけるCXCR4の発現を示す。データは、それぞれ個別にアッセイされた、N=5マウス/群の平均±SEMとして表す。
CD45.1マウスを、G−CSFまたはG−CSF+インドメタシン(1日ずらして)を用いて動員し、PBMCを、CD45.2競合骨髄細胞と共に致死的に放射線を浴びたCD45.2マウスに移植した。ここで図22を参照すると、移植後12週目における、複数ドナーのキメラ現象:競合相手の比(左パネル)および競合的再構築単位(右パネル)を示す。データは、それぞれ個別にアッセイされた、n=5マウス/群の平均±SEMとして表す。
マウスを、G−CSFまたはG−CSF+メロキシカム(1日ずらして)を用いて動員し、2×106のPBMCを、致死的に放射線を浴びたレシピエントに移植した。血中好中球を、Hemavet950FS(Drew Scientific)により、(対照のサブセットと比較して)完全に回復するまで1日おきに数えた。血中血小板を、Hemavet950FS(Drew Scientific)により、(対照のサブセットと比較して)完全に回復するまで1日おきに数えた。ここで図23を参照すると、末梢血(PB)中の好中球は、(対照のサブセットと比較して)完全に回復するまで1日おきに数えられた。これらのデータは、それぞれ個別にアッセイされた、n=10マウス/群の平均±SEMとして表す。ここで図24を参照すると、末梢血(PB)中の血小板は(対照のサブセットと比較して)完全に回復するまで1日おきに数えられた。これらのデータは、それぞれ個別にアッセイされた、n=10マウス/群の平均±SEMとして表す。
ここで図25を参照すると、ヒヒの第1群を、以下の10ug/体重kgのG−CSFの投与レジメン処置を用いて動員し、次いで2週間の洗い流し期間後に、動物を、10ug/kgのG−CSF+0.1mg/kgのメロキシカムを用いて処置した。ヒヒの第2群を、以下の10ug/kgのG−CSF+0.1mg/kgのメロキシカムのレジメンを用いて動員し、2週間の洗い流し期間後、10ug/kgのG−CSFを用いる期間であった。ここで図26を参照すると、PB中のCD34+細胞がFACS分析により決定され、先に記載のようにCFU−GM/血液mlを決定した。G−CSFとメロキシカムとの同時投与は、CD34+細胞の動員を増加させ(左パネル)、取り出した血液単位当たりのCFU−GMを測定した。
マウスを、G−CSF用いて動員し、PB中のCFU−GMを、G−CSFおよび様々なNSAIDS(アスピリン[COX−1およびCOX−2];リコフェロン[COX−2および5−LOX];SC−560[COX1];サリチル酸バレリル[COX−1];バルデコキシブ[COX−2];NS−398[COX−2])との併用による動員レジメンと比較した。ここで図27を参照すると、これら試験の結果が要約されており、これらのデータは、それぞれ個別にアッセイされた、n=4マウス/群の平均±SEMとして表される。COX1および2の両方を阻害することが公知である化合物は、2種のアイソザイムの1種だけに対して高度に選択性であると思われる化合物より、コロニー形成細胞の動員においてより優れていた。2種の一般的に使用されるCOX阻害剤の効率を試験するために、マウスを、G−CSFまたはG−CSFと、アスピリンまたはイブプロフェンとの併用により動員した(PO、1日2回、4日間)。CFU−GMを先に記載のように決定した。ここで図28を参照すると、G−CSF+アスピリンおよびG−CSF+イブプロフェンによる、CFU−GMの末梢血への動員の用量反応分析は、G−CSFだけで処置したマウスの対照群;G−CSF+10、20または40ミリグラム/kgのアスピリンを用いて処置したマウス;およびG−CSF+10、20または40ミリグラム/kgのイブプロフェンを用いて処置したマウスに関して棒グラフの形態で提示される。データは、それぞれ個別にアッセイされた、n=4マウス/群の平均±SEMとして表す。
マウスを、G−CSFおよび以下のメロキシカム用量、メロキシカムに対して0.0(対照)、0.02、0.2、0.5、1.5および3ミリグラム/体重kgを用いて動員し(1日2回、SC、4日間)、CFU−GMを先に記載のように決定した。ここで図29を参照するとHSPCに対するメロキシカムの用量反応は、動物の末梢血、または動物の骨髄において測定された(図30)。これらのデータは、それぞれ個別にアッセイされた、n=3マウス/群の平均±SEMとして表す。
本発明は以下の態様を含み得る。

非ステロイド系抗炎症性化合物が、アスピリン、セレコキシブ、ロフェコキシブ、エトリコキシブ、バルデコキシブ、イブプロフェン、ナプロキセン、ジクロフェナク、エトドラク、ケトロラクおよびリコフェロンからなる群から選択される、請求項2に記載の方法。

非ステロイド系抗炎症性化合物が少なくとも3日間患者に投与される、請求項2に記載

少なくとも1種のPGE 2 受容体のアンタゴニストである化合物を提供するステップ、ならびに
ドナーから造血幹細胞および前駆細胞を採取する前に、有効量の前記化合物を造血幹細胞および/または前駆細胞ドナーに投与するステップ、
を含む、ドナー由来の造血幹細胞および/または前駆細胞の動員を強化する方法。

少なくとも1種のPGE 2 受容体のアンタゴニストが、N−[[4’−[[3−ブチル−1,5−ジヒドロ−5−オキソ−1−[2−(トリフルオロメチル)フェニル]−4H−1,2,4−トリアゾール−4−イル]メチル][1,1’−ビフェニル]−2−イル]スルホニル]−3−メチル−2−チオフェンカルボキシアミドおよび4−(4,9−ジエトキシ−1,3−ジヒドロ−1−オキソ−2H−ベンズ[f]イソインドール−2−イル)−N−(フェニルスルホニル)−ベンゼンアセトアミドからなる群から選択される、請求項11に記載の方法。

供給源においてPGE 2 活性を減少させる少なくとも1種の化合物を用いて処置された、当該供給源由来の造血幹細胞および/または前駆細胞を含む細胞の群を採取するステップ、

を含む、レシピエントにおいて造血幹細胞および/または前駆細胞の生着を強化する方法。

Claims (5)
- 造血幹細胞および/または前駆細胞の供給源においてPGE2の生合成および/またはPGE2の活性を前記細胞がその有効量と接触するときに減少させる化合物を含む、造血幹細胞および/または前駆細胞の動員強化剤であって、
ここで、前記PGE 2 活性を減少させる化合物が非ステロイド系抗炎症性化合物であり、前記非ステロイド系抗炎症性化合物が、シクロオキシゲナーゼ−1およびシクロオキシゲナーゼ−2の両方に作用する剤。 - 造血幹細胞および/または前駆細胞の供給源においてPGE 2 の生合成および/またはPGE 2 の活性を前記細胞がその有効量と接触するときに減少させる化合物を含む、造血幹細胞および/または前駆細胞の動員強化剤であって、
ここで、前記PGE 2 活性を減少させる化合物が非ステロイド系抗炎症性化合物であり、前記非ステロイド系抗炎症性化合物が、シクロオキシゲナーゼ−2に作用する剤。 - 造血幹細胞および/または前駆細胞の供給源においてPGE 2 の生合成および/またはPGE 2 の活性を前記細胞がその有効量と接触するときに減少させる化合物を含む、造血幹細胞および/または前駆細胞の動員強化剤であって、
