Description

・ 少なくとも第一相が、約60‐160μmの範囲の平均直径を有する空間(void)を含み;そして
・ 少なくとも第二相が、約170‐850μmの範囲の平均直径を有する空間を含んでいる

・ 少なくとも第一相が、約35‐55%の範囲の細孔容積(pore volume)(多孔度)を有する細孔を含み;そして
・ 少なくとも第二相が、約56‐95%の範囲の細孔容積(多孔度)を有する細孔を含んでいる
を含む。
・ 少なくとも第一相が、平均直径、細孔容積またはそれらの組み合わせを有する空間を含み、そしてそれは、前記固形形態が分離された天然のサンゴと一致し;そして
・ 少なくとも第二相が、平均直径、細孔容積またはそれらの組み合わせを有する空間を含み、そしてその平均空間直径、細孔容積またはそれらの組み合わせは、前記第一相のそれよりも約15‐100%より大きい
を含む。
・ アラゴナイトまたはカルサイトの固形形態の一部と、カルシウムキレート剤および酸を接触させて、前記固形形態の少なくとも一部に拡大された空間を含む固形形態を生成させ;および
・ 適用された陰圧下、前記固形形態を洗浄および乾燥すること
を含む。
・ 所望のサイズおよび形の固形アラゴナイトを、酸化剤を含む溶液と接触させ;そして
・ 前記固形アラゴナイトを洗浄および乾燥すること
を含み、それによって、前記ステップの一つまたはそれぞれは適用される陰圧下、実施される。
本発明は、とりわけ、対象の軟骨および/または骨組織の修復および/または形成のための足場、ツール、およびそれらの使用方法を提供する。本発明は、さらに対象の軟骨および/または骨組織の修復および/または形成のためのキットを提供する。
・ 前記2相の第一相が、ヒアルロン酸を含んでいる固形サンゴまたはバイオラティスを含み、そして前記第一相はさらに前記第一相において縦軸に沿って一連の中空を含み;そして

・ 少なくとも第二相で、平均直径、細孔容積、またはそれらの組み合わせを有する空間を含み、そしてそれらの平均空間直径、細孔容積またはそれらの組み合わせは、前記第一相のそれよりも約15‐100%大きい
を含む。
・ 少なくとも第一態様で、平均直径、細孔容積またはそれらの組み合わせを有する空間を含み、そしてそれは、天然のサンゴから分離した前記固体形態に相当する;そして
・ 少なくとも第二態様で、平均直径、細孔容積、またはそれらの組み合わせを有する空間を含み、そしてそれらの平均空間直径、細孔容積またはそれらの組み合わせは、前記第一態様のそれよりも約15‐900%大きい
・ アラゴナイトまたはカルサイトの固形形態の一部のみと、カルシウムキレート剤および酸を接触させて、前記固形形態の少なくとも一部に拡大した空間を含む固形形態を生成させ;および
・ 適用された陰圧下で、前記固相を洗浄しおよび乾燥すること
を含む。
本発明のサンゴに基づく足場は、それを必要とする対象について、軟骨、骨またはそれらの組み合わせの中に挿入されてもよい。
骨軟骨欠損の復元は、成熟したヤギについて、直径6mmおよび長さ8mmの丸みを帯びたインプラントを使用することで行った。軟骨および骨組織の5.5×8mmコアは、それぞれのヤギの大腿骨内側顆の外に穴を開け(図2A)、そしてインプラントを軟骨および骨修復の部位の中に押し込んだ(図2Bおよび2C)。
それぞれの相の細孔容積(多孔度)に関して変化し、および/またはそれぞれの相に存在する空間の直径に関して変化する多相性の足場を作るために、直径5.2mmおよび長さ7.5mmのプラグを、シリコンホルダー内に配置し、それによってただ1mmのプラグが露出し、そしてプラグを有するホルダーを、反転された位置に配置し、そして反応混合物の中に浸漬し、プラグの上部1mmのみが、混合物と直接接触するようにした。
インプラントの軟骨相の軟骨形成の可能性を評価するため、アラゴナイトに基づく足場をヒアルロン酸を浸透させたアラゴナイトの足場と比較した。インビトロアッセイは、マウス間葉系幹細胞(MSCs)ATCC/CRL‐12424を使用して行い、そしてそれらの軟骨形成系列に向かった分化を評価した。MSCsを、ヒアルロン酸(NaHA 1%)を含むまたは含まないサンゴ(コブハマサンゴ類)に基づくアラゴナイトの1mgの小さい粒子(〜1mmのサイズ)の上に5000のMSCsを播種した。任意のインプラントも含まない1/3の群を、対象として利用した。足場浸透の方法は、本明細書に記載した。使用したヒアルロン酸(HA)は、Bio‐Technology General(Israel)LTD.Lot:RDO131Bのアースレース(商標)として市場に出される1%ヒアルロン酸ナトリウムの注射用ゲルである。
インプラントは、例えば、膝関節修復のための、任意の適当な配置であってもよく、インプラントは、大腿骨内側顆(MFC)、大腿骨外側顆(LFC)、膝蓋骨、滑車切痕(Trochlear Groove(TG))、および脛骨(Tibia)内であってもよい。
アラゴナイトコア足場の調製:100‐150μmの平均細孔サイズを有するコブハマサンゴ(hydrocoral Porites Lutea)由来のサンゴを、採取し、その外観、密度、多孔度について視覚的に評価し、そしてFTIR分析を行った。アミノ酸定量を、また決定した。サンゴは、その時、外部の有機組織の除去のため5%次亜塩素酸ナトリウム中に浸漬した。
Claims (13)
- 組織修復足場であって、前記足場が2相からなり、
ここで、前記2相の第一相が、固形のサンゴ及び生体適合性ポリマーを含み、ここで前記生体適合性ポリマーが、ヒアルロン酸、ヒアルロン酸ナトリウム、架橋ヒアルロン酸、又はそれらの組合せを含むグリコサミノグリカンを含み、そして前記第一相がさらに、前記第一相において縦軸(longitudinal axis)に沿って一連の中空(hollow)を含み、そしてここで前記生体適合性ポリマーが、前記中空内に満たされ;及び
前記2相の第二相が、固形のサンゴを含み、そして前記第二相において縦軸に沿って一連の中空をさらに含まない、足場。 - 前記生体適合性ポリマーが、他のグリコサミノグリカン、コラーゲン、エラスチン、シルク、キトサン、アルギン酸塩、又はそれらの組み合わせをさらに含む、請求項1に記載の足場。
- 前記グリコサミノグリカンが、ヒアルロン酸又はヒアルロン酸ナトリウムである、請求項2に記載の足場。
- 前記生体適合性ポリマーが、アルファヒドロキシ酸を重合することによって得られる生分解性合成ポリマーをさらに含む、請求項1に記載の足場。
- 前記足場がさらに、サイトカイン、キレート剤、細胞集団、抗炎症性化合物、抗感染性化合物、成長因子、抗生物質、血管新生促進因子、治療上の化合物、又はそれらの組み合わせを含む、請求項1に記載の足場。
- 前記細胞集団が、間葉系幹細胞、骨芽細胞、骨細胞、破骨細胞、軟骨芽細胞、軟骨細胞、線維芽細胞、又はそれらの組み合わせ、又は軟骨若しくは骨修復に関わる他の細胞を含む、請求項5に記載の足場。
- 前記成長因子が、上皮成長因子(EGF)、β型トランスフォーミング増殖因子(TGF‐β)、ヒト内皮細胞増殖因子(ECGF)、顆粒球マクロファージコロニー刺激因子(GM‐CSF)、骨形成タンパク質(BMP)、神経成長因子(NGF)、血管内皮増殖因子(VEGF)、線維芽細胞成長因子(FGF)、インスリン様成長因子(IGF)、軟骨由来形成タンパク質(CDMP)、血小板由来成長因子(PDGF)、又はそれらの組み合わせである、請求項5に記載の足場。
- 前記足場が、円柱、円錐、スクリュー、長方形棒(rectangular bar)、プレート、ディスク、ピラミッド、顆粒(granule)、ボール、又はキューブの形に近似する、請求項1に記載の足場。
- 前記足場が、膝若しくは肘に関する半月板(meniscus);関節;骨の関節面、胸郭、股、骨盤、耳、鼻、靭帯、気管支、又は椎間板の一部に近似するような形をとる、請求項1に記載の足場。
- 前記足場が円柱状の形状で、並びに5〜40mmの直径、及び5〜25mmの高さを有する、請求項1に記載の足場。
- 前記生体適合性ポリマーが、前記足場の一部と一体となるポリマー層を形成する、ポリマーコーティングの形状である、請求項1に記載の足場。
- 前記ポリマーコーティングが、前記足場の末端に存在し、及び前記コーティングが、0.1〜10mmの間の厚さを有する、請求項11に記載の足場。
- 請求項1の足場、及び組織修復における前記足場の利用のための指示書、を含む軟骨又は骨の修復のためのキット。
