Description
本発明は、濃透析酸溶液(dialysis acid concentrate solution)の調製中に使用するための、並びに使用準備が完了した(ready-for-use)透析液にする水及び重炭酸塩含有濃縮物との更なる混合のための透析酸前駆組成物に関する。本発明は、水及び重炭酸塩濃縮物によって希釈して、使用準備が完了した透析液を製造するための濃透析酸溶液を提供する方法に更に関する。
ヒトの腎臓が正常に働かないと、尿毒症が発現する。透析は、尿毒症に対しての、十分に確立された治療技術である。本質的に、透析は、腎臓の機能を人工的に取って代わる。透析には、血液透析及び腹膜透析という2つの異なるタイプが存在する。
本発明の1つの目的は、更に改善された安定性と、抑えられた化学的劣化と、高められた保存可能期間とを示す透析酸前駆組成物を提供することにある。
(a)塩化ナトリウム、少なくとも1種の乾燥酸、及び少なくとも1種のマグネシウム塩、任意にカリウム塩、カルシウム塩及びグルコースを含む透析酸前駆組成物を提供すること、ここで前記少なくとも1種のマグネシウム塩及び前記任意のグルコース(即ちグルコースが存在する場合)は、前記透析酸前駆組成物中の無水成分として存在する、
(b)前記透析酸前駆組成物を、水蒸気透過率が38℃/90%RHで0.3g/m2/日未満である密封された耐湿性容器中に提供すること、及び
(c)定められた体積の水を前記容器内の前記透析酸前駆組成物中に加え、これらを混合して、それにより前記透析酸濃縮物を溶液として提供すること
を含む。
塩化カリウム、塩化マグネシウム、塩化カルシウム、グルコース、塩化ナトリウム、重炭酸ナトリウム並びにクエン酸及びグルコノ−δ−ラクトンなどの乾燥酸など、標準透析液の乾燥粉末成分の多種多様な異なる組み合わせ及び分割物を調製し、強制(forced)安定性試験を行った。1ヶ月、4ヶ月及び10ヶ月の保存期間の後に、ケーキング、塊形成、変色及び溶解速度などの事項を調べた。
以下の例は、例としてのものであり、限定するものではなく、本発明の態様に従った種々の透析酸前駆組成物を確認する。
本発明の態様によるもの及び比較物の両方の、異なる乾燥粉末組成物の安定性を調べるために試験を行った。ケーキング、塊形成及び変色などのパラメータを評価した。
プラスチックフィルムを溶着して、1室を有する袋とした。
230Lの透析流体の製造に必要な量の塩化カリウム、無水塩化マグネシウム、塩化カルシウム二水和物、無水グルコース、クエン酸及び塩化ナトリウムの粉末成分を、水蒸気透過率が38℃/90%RHで0.11g/m2/日であるプラスチックバッグに充填した。バッグを密封し、それぞれ、30℃、65%RHで、及び40℃、75%RHでインキュベートした。
230Lの透析流体の製造に必要な量の塩化カリウム、無水塩化マグネシウム、無水塩化カルシウム、無水グルコース、クエン酸及び塩化ナトリウムの粉末成分を、水蒸気透過率が38℃/90%RHで0.11g/m2/日であるプラスチック袋に充填した。バッグを密封し、それぞれ、30℃、65%RHで、及び40℃、75%RHでインキュベートした。
230Lの透析流体の製造に必要な量の塩化カリウム、無水塩化マグネシウム、塩化カルシウム脱水(dehydrate)、無水グルコース、クエン酸及び塩化ナトリウムの粉末成分を、38℃/90%RHで2.7g/m2/日の水蒸気透過率を有するプラスチックバッグに充填した。バッグを密封し、それぞれ、30℃、65%RHで、及び40℃、75%RHでインキュベートした。
組成物1及び2は、少なくとも6ヶ月にわたって安定性を維持したことが立証されたが、一方で比較組成物3は、1ヶ月に満たない内に褐色の塊が生じたため、安定性維持が立証されなかった。
Claims (10)
- 濃透析酸溶液の調製中の使用のため、並びに使用準備が完了した透析液にする水及び重炭酸塩含有濃縮物との混合のための透析酸前駆組成物であって、前記透析酸前駆組成物は、塩化ナトリウム、少なくとも1種の乾燥酸及び少なくとも1種のマグネシウム塩、並びに任意にカリウム塩、カルシウム塩及びグルコースを含む粉末成分からなり、前記少なくとも1種のマグネシウム塩及び前記任意のグルコースは、前記透析酸前駆組成物中の無水成分として存在し、前記透析酸前駆組成物は、水蒸気透過率が38℃/90%RHで0.3g/m2/日未満である耐湿性容器中に密封されている透析酸前駆組成物。
- 前記少なくとも1種の乾燥酸は、乳酸、クエン酸、グルコン酸、グルコノ−δ−ラクトン、N−アセチルシステイン及びα−リポ酸からなる群より選択される請求項1に記載の透析酸前駆組成物。
- 前記透析酸前駆組成物中の前記少なくとも1種のマグネシウム塩は、無水塩化マグネシウム、グルコン酸マグネシウム、クエン酸マグネシウム、乳酸マグネシウム、及びα−ケトグルタル酸マグネシウムからなる群より選択される請求項1又は2に記載の透析酸前駆組成物。
- 前記透析酸前駆組成物中の前記カルシウム塩は、塩化カルシウム二水和物、塩化カルシウム一水和物、無水塩化カルシウム、グルコン酸カルシウム、クエン酸カルシウム、乳酸カルシウム、及びα−ケトグルタル酸カルシウムからなる群より選択される少なくとも1種である請求項1〜3の何れか1項に記載の透析酸前駆組成物。
- 前記耐湿性容器は、水蒸気透過率が38℃/90%RHで0.2g/m2/日未満である請求項1〜4の何れか1項に記載の透析酸前駆組成物。
- 前記耐湿性容器は、水蒸気透過率が38℃/90%RHで0.1g/m2/日未満である請求項1〜5の何れか1項に記載の透析酸前駆組成物。
- 前記透析酸前駆組成物は、定められた体積の水と前記耐湿性容器内部で混合されて、濃透析酸溶液を提供するように構成されている請求項1〜6の何れか1項に記載の透析酸前駆組成物。
- 水及び重炭酸塩含有濃縮物で希釈して使用準備が完了した透析液を製造するための透析酸濃縮物液を提供する方法であって:
(a)塩化ナトリウム、少なくとも1種の乾燥酸、及び少なくとも1種のマグネシウム塩、任意にカリウム塩、カルシウム塩及びグルコースを含む透析酸前駆組成物を提供すること、ここで、前記少なくとも1種のマグネシウム塩及び前記任意のグルコースは、前記透析酸前駆組成物中の無水成分として存在する、
(b)前記透析酸前駆組成物を、水蒸気透過率が38℃/90%RHで0.3g/m2/日未満である密封された耐湿性容器内に提供すること、及び
(c)定められた体積の水を前記容器内の前記透析酸前駆組成物中に加え、これらを混合して、それにより前記透析酸濃縮物を溶液として提供すること
を含む方法。 - 請求項1〜7の何れか1項に記載の透析酸前駆組成物の、濃透析酸溶液を調製するための使用。
- 請求項1〜7の何れか1項に記載の透析酸前駆組成物の、透析液、注入液、補充液、リンス液又はプライミング液を調製するための使用。
