(a)上記第一タンパク質及び第二タンパク質をそれらのVFTドメインのN末端部で1対のFRETパートナーのメンバーにより標識する工程;
(b)上記FRETパートナーを励起し、試験化合物の非存在下及び存在下、上記FRETシグナルを測定する工程;
(c)工程(b)において上記試験化合物の非存在下とその存在下で測定されたFRETシグナルに差異がある場合に、その試験化合物を調節化合物として選択する工程
を含む方法に関する。
(a)上記第一タンパク質及び第二タンパク質をそれらのVFTドメインのN末端部で1対のFRETパートナーのメンバーにより標識する工程;
(b)上記FRETパートナーを励起し、試験化合物の非存在下及び存在下、上記FRETシグナルを測定する工程;
(c)工程(b)において上記試験化合物の非存在下とその存在下で測定されたFRETシグナルに差異がある場合に、その試験化合物を調節化合物として選択する工程
を含み、ここで工程(b)において上記FRETシグナルは、参照アゴニスト又はアンタゴニスト化合物の存在下とその非存在下で測定されたシグナルが異なるものとなる時間窓内で測定される、方法から構成される。
・参照アゴニスト化合物の存在下とその非存在下でのシグナル平均値の差異が20%よりも大きい場合、又は
・アゴニスト化合物の存在下とその非存在下でのシグナル平均値が最大標準偏差(参照アゴニスト化合物の非存在下で測定されるシグナルの標準偏差か、又はその化合物の存在下で測定されるシグナルの標準偏差のいずれか)に対して3倍超異なる場合。
(i)上記FRETパートナー対のフェルスター(Forster)半径(R0)は20〜55Åであり、かつ、工程(b)において上記シグナルは、励起してから10〜100μ秒の遅延時間、及び100〜500μ秒の積分時間で測定されるか;又は、
(ii)上記対のフェルスター半径(R0)は55Åよりも大きく、かつ、工程(b)において上記シグナルは、励起してから180〜800μ秒の遅延時間、及び200〜2000μ秒の積分時間で測定される
ことを特徴とする。
「FRETパートナー対」:この表現は、エネルギードナー蛍光化合物(以下「ドナー蛍光化合物」)とエネルギーアクセプター化合物(以下「アクセプター化合物」)とからなる対をいう。上記化合物が互いに接近していたり、ドナー蛍光化合物の励起波長で励起されていたりする場合、上記化合物はFRETシグナルを放射する。2つの蛍光化合物をFRETパートナーとするには、ドナー蛍光化合物の発光スペクトルがアクセプター化合物の励起スペクトルと部分的に重なっていなければならないことが知られている。
(a)上記第一タンパク質及び第二タンパク質をそれらのVFTドメインのN末端部で1対のFRETパートナーのメンバーにより標識する工程、ここで上記対のフェルスター(Forster)半径(R0)は20〜55Åである;
(b)試験化合物の非存在下及び存在下、上記FRETシグナルを測定する工程;
(c)上記試験化合物の存在下又は非存在下でのFRETシグナルの変化と、上記ダイマーの公知の調節化合物の存在下又は非存在下で測定されたものとを比較することにより、上記試験化合物を調節化合物として選択する工程
を含む。
本発明の方法は、VFTドメインタンパク質のダイマーを含む細胞膜を含む測定媒体中で行われる。従って、測定媒体は、FRETパートナー化合物の添加時に、付着した状態若しくは懸濁した状態の無傷生細胞を含んでいるか、又は、細胞溶解や分画遠心分離などにより得た、懸濁した状態の細胞膜の調製物を含んでいる。膜懸濁物の調製法は当業者に公知である。本発明の方法は無傷生細胞について行うのが好ましい。
本発明の必須の特徴の1つは、検討するダイマーの構成タンパク質のそれぞれを1対のFRETパートナーのメンバーにより標識することである。
本発明の別の必須の特徴は、エネルギードナー又はエネルギーアクセプターによるVFTドメインを含む膜貫通タンパク質の標識が、ダイマーを構成する2つのサブユニットそれぞれのVFTドメインのN末端部で行われるということにある。
ドナー又はアクセプターは、少なくとも1つがタンパク質性である1対の結合パートナーによりVFTドメインタンパク質と結合させることができる。この手法では、VFTドメインタンパク質を、従来の分子生物学的方法によってタンパク質性の結合パートナーと融合させる(VFTドメインタンパク質をコードする核酸配列を含んでおり、タンパク質結合パートナーをコードする核酸配列と融合させた発現ベクターを構築し、その発現ベクターを細胞に導入する)。本発明において、結合パートナーは、VFTドメインタンパク質のN末端部、好ましくはその末端に存在する。
この手法では、ドナー又はアクセプターは、共有結合によりVFTドメインタンパク質と結合する。幾つかの方法が開示されており、これを行うのに必要な試薬は市販されている。この結合に、以下の方法のいずれかを用いることができる。
自殺酵素は、基質と迅速に共有結合できるようにする特定の変異により酵素活性が変化したタンパク質である。上記酵素は、各々が1つの蛍光分子としか結合できず、基質の結合により酵素活性がブロックされるため、自殺酵素と呼ばれている。結果的に、この酵素は、1つのタンパク質に1つの蛍光分子の割合で対象のタンパク質を特異的に標識するのに選択される道具となる。例えば以下の酵素が挙げられるが、これに限定されるものではない。
エネルギードナーの吸収波長で測定媒体を励起することができ、かつドナー化合物及び/又はアクセプター化合物(蛍光性を有する場合)の発光波長での測定媒体からの発光を測定することができる蛍光計を使用して従来法によりFRETシグナルを測定する。
・アクセプター(蛍光性を有する場合)の発光波長の発光量が増加し、
・ドナーの発光波長の発光量が減少、すなわち、ドナー化合物の蛍光の寿命が減少する。
本発明は、本発明の方法を行うための構成要素のキット、すなわち、上述の試薬を、場合によっては本発明におけるその使用上の指示を伴って、含むキットにも関する。特に、上記キットは、
・同一か又は異なる第一タンパク質及び第二タンパク質で構成される、VFTドメインタンパク質のダイマーを含む細胞膜、
・1対のFRETパートナー、
・上記第一タンパク質のVFTドメインのN末端部を上記FRETパートナー対の一方のメンバーにより標識する手段、及び、上記第二タンパク質のVFTドメインのN末端部を上記FRETパートナー対のもう一方のメンバーにより標識する手段、
・上記FRETシグナルは、参照アゴニスト又はアンタゴニスト化合物の存在下とその非存在下で測定されたシグナルが異なるものとなる時間窓内で測定されなければならないという指示
を含む。
(i)フェルスター(Forster)半径(R0)が20〜55Åである1対のFRETパートナーと、FRETシグナルが、励起してから20〜100μ秒の遅延時間、及び100〜500μ秒の積分時間で測定されなければならないという指示とを含むか;又は、
(ii)フェルスター半径(R0)が55Åよりも大きい1対のFRETパートナーと、FRETシグナルが、励起してから180〜800μ秒の遅延時間、及び200〜2000μ秒の積分時間で測定されなければならないという指示とを含む。
モジュレータ(アゴニスト、アンタゴニスト、アロステリックモジュレータ):
DCG IV、L−グルタミン酸、及びLY341495は、Tocris Biosciencesカタログにおいて入手可能である。4−MPPTS(mGluR2受容体のポジティブアロステリックモジュレータ、LY487379ともいう)はAddex Pharmaceuticalsより提供されたものであり、その合成法は米国特許第6800651号明細書に記載されている。
DMEM、DMEM+Glutamax、MEM−NEAA、MEM−ペニシリン/ストレプトマイシン、トリプシン/EDTAはGibcoカタログで入手可能である。使用したウシ胎仔血清はLonzaブランド(南アメリカ型(バッチ:7SB0017))である。
BG−Lumi4−Tb:ベンジルグアニン−Lumi4−Tb結合体(Cisbio Bioassaysから販売、Tag−Lite(登録商標)SNAP−Lumi4Tb)。BGはSnap−tag酵素の基質である。
BC−フルオレセイン:ベンジルシトシン−フルオレセイン結合体(NEB社から商品名CLIP−Vista Greenで販売)。BCはClip−tag酵素の基質である。
BG−フルオレセイン:ベンジルグアニン−フルオレセイン結合体(NEB社から商品名SNAP−Vista Green(New England Biolabs)で販売)。
Tag−Lite(登録商標)SNAP−Green:ベンジルグアニン−フルオレセイン結合体(Cisbio Bioassays社から販売)。
Tag−Lite(登録商標)SNAP−red:ベンジルグアニン−シアニン誘導体結合体(発光ピーク:670nm、Cisbio Bioassaysから販売)。
DMSO、Tris(Trizma Base)、NaCl、KH2PO4、MgSO4、KCl、及びCaCl2はSigma−Aldrichカタログで入手可能である。
HA−Snaptag−[GPCR1]融合タンパク質、及びFlag−Cliptag−[GPCR2]融合タンパク質(GPCR1及びGPCR2は、細胞により発現させることを意図したGPCRを表す)をコードするDNAを含む発現ベクターは、従来の分子生物学的方法によりprK5ベクターから作製し、その配列はシークエンシングにより確認した。4661塩基対を含む上記配列は配列番号4の配列である。このプラスミドの特徴的な制限部位の位置を以下に示す。
・mGluR5のシグナルペプチド :937−1002;
・HAエピトープ :1009−1035;
・Snap−tag :1042−1590;
・mGluR2 :1597−4149
・mGluR5のシグナルペプチド :937−1002;
・FLAG :1015−1038;
・Clip−tag :1045−1593;
・mGluR2 :1600−4152
エレクトロポレーションによるトランスフェクト
Cos−7細胞を完全培地中、37℃、5%CO2で培養した。コンフルエンス状態で、細胞をPBSで一度洗浄し、トリプシン−EDTA溶液により10分間37℃で分離した。1000万細胞の懸濁液を5μgの全DNA(prK5プラスミド(3.5μg)、prK5 Flag−Cliptag−mGluR2プラスミド(1.2μg)、及びprK5 HA−Snaptag−mGluR2プラスミド(0.3μg))と混合して最終体積300μLのエレクトロポレーションバッファとした後、4mmエレクトロポレーションキュベット(Eurogentec)中、GenePulser IIエレクトロポレータ(BIO−RAD)を使用して280V、900μFの電気ショックを与えた。その後、細胞を96ウェルプレート(Greiner CellStar 96ウェルプレート)に150,000細胞/ウェルで播種し、完全培地の最終体積を100μL/ウェルとしてから、24時間インキュベートした。
トランスフェクトしてから24時間後、標識バッファ(50μL)+BG−Lumi4−Tb(0.3μM)、及びBC−フルオレセイン(1μM)の存在下、細胞を2時間、37℃でインキュベートした。
フルオレセイン(アクセプター)の波長で放射された蛍光をAnalyst ADマイクロプレートリーダ(Molecular Devices)により、TRFモード、遅延時間50μ秒、積分時間450μ秒、励起フィルタ320nm帯域幅(BW)25、BB/UVダイクロイックミラー、蛍光フィルタ520nmBW10で読み取った。以下、このFRETシグナルをTRF520と称す。一回目は、プレートをグルタミン酸塩の非存在下で読み取った。
図1は、グルタミン酸塩濃度に対するTRF520のシグナルを示す。このシグナルは、1回目の読み取り時、すなわちグルタミン酸塩の添加前に読み取られたTRF520の百分率として示されている。すなわち以下の式により算出されるものである。
実施例1の記載と同じ試薬及びプラスミドを使用した。公知のアンタゴニスト化合物LY341495(Tocris Bioscience)を使用した。
実施例1の記載と同じプロトコルを使用した。最終濃度は、[グルタミン酸塩]=100μM(全てのウェル)、[LY341495]=1000nM、316μM、100nM、31.6nM、10nM、3.16nM、又は1nM。
図2は、一定濃度のグルタミン酸塩の存在下での、LY341495アンタゴニスト濃度に対するTRF520のシグナルを示す(対照曲線:グルタミン酸塩の非存在下)。
実施例1の記載と同じ試薬及びプラスミドを使用した。使用した公知の部分的アゴニスト化合物はDCG−IV(Tocris Bioscience)であり、参照完全アゴニストであるグルタミン酸塩と比較した。
実施例1のプロトコルを使用した。[DCG−IV]の最終濃度は以下である:100μM、31.6μM、10μM、3.16μM、1μM、0.316μM、又は0.1μM。
図3は、グルタミン酸塩(完全アゴニスト)又はDCG−IV(部分的アゴニスト)の濃度に対するTRF520のシグナルを示す。
実施例1の試薬及びプラスミドを使用した。公知のポジティブアロステリックモジュレータ化合物である4−MPPTS(=mGluR2 PAM、LY487379ともいう)を使用した。
実施例1に記載のプロトコルを使用した。
図4は、4−MPPTSの存在下又は非存在下(対照)での、グルタミン酸塩濃度に対するTRF520のシグナルの変化を示す。
・mGlu2の場合、実施例1と同じ、
・mGlu3の場合、3.5μg(prK5プラスミド)、1.2μg(prK5 Flag−Cliptag−mGluR3プラスミド)、及び0.3μg(prK5 HA−Snaptag−mGluR3プラスミド)、
・mGlu4の場合、3.5μg(prK5プラスミド)、1.2μg(prK5 Flag−Cliptag−mGluR4プラスミド)、及び0.3μg(prK5 HA−Snaptag−mGluR4プラスミド)である点。
・標識されたmGluR3ホモダイマーを発現させるため、3.5μg(prK5プラスミド)、1.2μg(prK5 Flag−Cliptag−mGluR3プラスミド)、及び0.3μg(prK5−HA−Snaptag−mGluR3プラスミド)を使用した。
・LY341495アンタゴニストの濃度を上昇させて、最終濃度:1μM、320nM、100nM、32nM、10nM、3.2nM、及び1nMとした。
図7は、LY341495濃度に対してTRF520シグナルを示す。
Claims (13)
- 測定媒体中に存在する細胞膜で発現するVFTドメインタンパク質のダイマーの活性状態を調節する効果を有する化合物を選択する方法であって、ここで前記ダイマーは、同一か又は異なる第一タンパク質及び第二タンパク質で構成されており、第一タンパク質及び第二タンパク質はそれぞれVFTドメインを有しており、
前記方法は以下の工程:
(a)前記第一タンパク質をそのVFTドメインのN末端部で1対のFRETパートナーのメンバーにより標識する工程;
(b)前記第二タンパク質をそのVFTドメインのN末端部でその1対のFRETパートナーの他方のメンバーにより標識する工程;
(c)前記FRETパートナーを励起し、試験化合物の非存在下及び存在下、FRETシグナルを測定する工程;
(d)工程(c)において前記試験化合物の非存在下とその存在下で測定されたFRETシグナルに差異がある場合に、その試験化合物を調節化合物として選択する工程
を含み、ここで工程(c)において前記FRETシグナルは、参照化合物の存在下とその非存在下で測定されたシグナルが異なるものとなる時間窓内で測定され、前記参照化合物が前記ダイマーのアゴニスト又はアンタゴニストである、方法。 - 工程(c)において前記シグナルは、励起してから180〜800μ秒の遅延時間、及び200〜1000μ秒の積分時間で測定される
ことを特徴とする請求項1に記載の方法。 - (i)前記FRETパートナー対のフェルスター半径(R0)は20〜55Åであり、かつ、工程(c)において前記FRETシグナルは、励起してから20〜100μ秒の遅延時間、及び100〜500μ秒の積分時間で測定されるか;又は、
(ii)前記対のフェルスター半径(R0)は55Åよりも大きく、かつ、工程(c)において前記FRETシグナルは、励起してから180〜800μ秒の遅延時間、及び200〜1000μ秒の積分時間で測定される
ことを特徴とする請求項1に記載の方法。 - 前記試験化合物の存在下及び非存在下でのFRET測定は、前記ダイマーの公知のアゴニストの存在下で行われる
ことを特徴とする請求項1〜3のいずれか一項に記載の方法。 - 前記第一タンパク質及び第二タンパク質は、VFTドメインを含むクラスCのGタンパク質共役受容体である
ことを特徴とする請求項1〜4のいずれか一項に記載の方法。 - 前記VFTドメインタンパク質のダイマーは、代謝型グルタミン酸受容体、γ−アミノ酪酸受容体、甘味知覚関連受容体、旨味知覚関連受容体、細胞外カルシウム感知受容体、又は塩基性アミノ酸受容体から選択される
ことを特徴とする請求項1〜5のいずれか一項に記載の方法。 - 前記VFTドメインタンパク質のダイマーは、代謝型グルタミン酸受容体であるか、又は、TAS1R2−TAS1R3、若しくはTAS1R1−TAS1R3から選択される
ことを特徴とする請求項1〜5のいずれか一項に記載の方法。 - 前記第一タンパク質及び第二タンパク質の前記FRETパートナー対のメンバーによる標識は、1対の結合パートナーによる間接標識か、又は共有結合による直接標識のいずれかである
ことを特徴とする請求項1〜7のいずれか一項に記載の方法。 - 前記第一タンパク質及び第二タンパク質はそれぞれ自殺酵素との融合タンパク質の形態で発現し、前記第一タンパク質及び第二タンパク質の標識は、前記自殺酵素の基質にそれぞれ共有結合している前記FRETパートナー対の各メンバーを前記測定媒体に添加することにより行われる
ことを特徴とする請求項8に記載の方法。 - 前記第一タンパク質及び第二タンパク質はそれぞれ、異なる自殺酵素との融合タンパク質の形態で発現する
ことを特徴とする請求項9に記載の方法。 - 前記第一タンパク質及び第二タンパク質はそれぞれ、同一の自殺酵素との融合タンパク質の形態で発現する
ことを特徴とする請求項9に記載の方法。 - 前記自殺酵素は、O6−アルキルグアニンDNAアルキルトランスフェラーゼ変異体、デハロゲナーゼ変異体、又はアシルキャリアタンパク質フラグメントから選択される
ことを特徴とする請求項9〜11のいずれか一項に記載の方法。 - 前記FRETパートナー対は、
(i)一方は、ユーロピウムキレート、テルビウムキレート、ユーロピウムクリプテート、及びテルビウムクリプテートから選択される蛍光ドナー化合物、
(ii)もう一方は、ローダミン、シアニン、スクアライン、クマリン、プロフラビン、アクリジン、フルオレセイン、ニトロベンゾオキサジアゾール、シアン蛍光タンパク質、緑色蛍光タンパク質、黄色蛍光タンパク質、オレンジ色・赤色蛍光タンパク質、及び近赤外領域で蛍光性を有するタンパク質から選択される蛍光性有機分子であるアクセプター蛍光化合物
で構成される
ことを特徴とする請求項1〜12のいずれか一項に記載の方法。
