Description
− R1、R2およびR3は、同一であっても異なっていてもよく、各グルコース単位に対して独立に選択され、ヒドロキシル、アルコキシ、アシルオキシまたはスルホニルオキシ基を表し;
− Rは、各グルコース単位に対して独立に選択され、カルボキシル、アルコキシカルボニル、アシル基、または-CH2R4基(ここでR4は、ヒドロキシル、アルコキシ、アシルオキシ、スルホニルオキシ、スルフィニル基を表す)を表し;
− nは、平均分子量が500〜4 000 000ダルトンになるように選択され;
R基の少なくとも0.1%が、当該グルコース単位の各々に対して独立に選択され、カルボキシル、アルコキシカルボニル、アシル基を表す。
− 以下の記載で使用される「酸化度」および「分子量」の表現は、分子単独であるか分子の混合物であるかの区別なく、本ケースでは平均値を表す;
− 以下の記載で使用される「β−(1,3)−グルクロナンおよび/またはβ−(1,3)−グルコグルクロナンの酸化度」は、グルクロンユニット、または前記β−(1,3)−グルクロナンおよび/またはβ−(1,3)−グルコグルクロナンの一連のユニットに存在するグルクロン酸に由来するユニットのパーセンテージを指す。より正確には、本発明に従った前述の酸化度は、全体のR基に対する、カルボキシル、アルコキシカルボニルまたはアシル基を表すR基のパーセンテージにより特徴づけられる;
− 「アルキル基」は、飽和の、一価の、直鎖または分枝の、1〜6個の炭素原子を含む炭化水素系鎖を意味し、その代表エレメントは、たとえば、メチル、エチル、n−プロピル、イソプロピル、n−ブチル、sec−ブチル、イソブチル、tert−ブチル、ペンチル、ヘキシル基である;

13C NMRスペクトルを、Avance 400 Bruker NMRスペクトロメーター (9.4 T, 400 MHz) で記録した。30000アキュムレーションを30℃で実施した。
カードラン (10 g) を蒸留水 (1 L) 中に、0℃の温度でpH 10で溶解する。TEMPO (43.3 g)、NaBr (0.95 g) およびNaClO (13%で50 mL)を反応混合物に添加する。酸化反応を、メタノールを添加することにより1時間後に停止させ、その後、塩酸 (2M) で中和する。形成されたβ−(1,3)−グルクロナンを氷冷アルコールで沈殿させる。その後、この陰イオンポリサッカリドを遠心分離 (15000 g, 20 min, 4℃) により回収し、最後に凍結乾燥 (24 h) により乾燥させる。
カードラン (10 g) を蒸留水 (1 L) 中に、0℃の温度でpH 10で溶解する。TEMPO (43.3 g)、NaBr (0.95 g) およびNaClO (13%で50 mL)を反応混合物に添加する。酸化反応を、メタノールを添加することにより30分後に停止させ、その後、塩酸 (2M) で中和する。形成されたβ−(1,3)−グルコグルクロナンを氷冷アルコールで沈殿させる。その後、この部分的に酸化されたポリサッカリドを遠心分離 (15000 g, 20 min, 4℃) により回収し、最後に凍結乾燥 (24 h) により乾燥させる。
β−(1,3)−グルクロナンの溶液(水中10 g/L)を100℃の温度で20分間加熱する。冷却後、溶液を酸性化し(pH 2)、その後、遠心分離する (15000 g, 20 min, 4℃)。オリゴサッカリドを含有する上清を回収し、中和し、凍結乾燥により乾燥させる。
β−(1,3)−グルクロナンの溶液(ピリジン中10 g/L)に無水酢酸を添加し(1体積)、室温で少なくとも24時間(24-48時間)維持し、その後、真空下で蒸発させる。このようにして得た過アセチル化β−(1,3)−グルクロナンを回収する。
β−(1,3)−グルクロナンの溶液(DMF中10 g/L)にSO3/DMFの溶液を添加し(1体積)、室温で少なくとも24時間(24-48時間)維持し、その後、凍結乾燥させる前に中和する。硫酸化度は、電位差の決定により測定される。
β−(1,3)−グルクロナン化合物またはβ−(1,3)−グルコグルクロナン化合物の混合物の生物学的活性を、分子生物学のオリジナル方法(転写技術)により選別し、美容学で慣用的に使用される方法論により検出した。
これら化合物の一または複数の非存在下または存在下においてインビトロで培養したヒト繊維芽細胞で実施した転写研究により、これら分子が、同じ名称のアジポカイン(adipokine)をコードするANGPTL4遺伝子を過剰発現に導くことが示された。脂肪組織で局所的に産生されるこのアジポカインは、酵素リポタンパク質リパーゼ(LPL)の活性を阻害し、これによりリポタンパク質から含脂肪細胞への脂肪酸の移動を妨害し、これにより脂質生成を大きく減少させる。同時に、このアジポカインは、含脂肪細胞に存在する酵素ATGL(脂肪トリグリセリドリパーゼ)を活性化し、これにより脂肪分解および含脂肪細胞からの脂肪酸の放出を大きく増大させる。
これら化合物の一または複数の非存在下または存在下においてインビトロで培養したヒト繊維芽細胞で実施した転写研究により、これら分子が、遺伝子MMP、EGR1、WNT11、PDGFD、TFPI2、PTGS2を過剰発現に導き、瘢痕形成および組織リモデリングのプロセスに関与する同じ名称のタンパク質をコードするHAPLN1遺伝子を発現低下に導くことが示された。実際、金属タンパク分解酵素(MMP)の(TEPI2およびPDGFDの制御下の)一過性誘導およびHAPLN1の(TFPI2の制御下の)一過性阻害は、主にコラーゲン繊維に対する異化作用により、細胞外マトリクスの(瘢痕形成を必要とする)ストレス部位を一過的に局所的に不安定にする。TFPI2により制御される細胞外マトリクスの分解後、これにより、ダメージを受けたコラーゲン繊維は除去され、組織リモデリングのプロセスに必須の細胞(繊維芽細胞、ケラチノサイト、メラノサイトなど)は(PTGS2により制御される)移動および増殖をし、タンパク質EGR1およびWNT11は、コラーゲン繊維の合成を刺激し、その結果、新たな細胞外マトリクスを再構築し、安定化させる。したがって、これらタンパク質の発現の誘導により、β−(1,3)−グルクロナン化合物またはβ−(1,3)−グルコグルクロナン化合物の混合物は、化粧品、とりわけ、創傷治癒、抗老化、抗しわ、コンシーリング、リフティング、引き締め、および潤い製品、並びに卵型の顔を維持することを助ける製品で使用することができる、創傷治癒活性および組織リモデリングに対する作用を有する。
β−(1,3)−グルクロナン化合物またはβ−(1,3)−グルコグルクロナン化合物の混合物の非存在下または存在下においてインビトロで培養したヒト繊維芽細胞で実施した転写研究により、これら分子が、化粧品のターゲット細胞である繊維芽細胞、ケラチノサイトおよびメラノサイトの(過剰発現されるTFPI2の制御下の)活性化、増殖および分化を担う、同じ名称のタンパク質をコードする遺伝子EGR1、PTGS2、PDGFD、NPTX1を過剰発現に導くことが示された。
これら化合物の一または複数の非存在下または存在下においてインビトロで培養したヒト繊維芽細胞で実施した転写研究により、これら分子が、抗炎症活性を担う同じ名称のタンパク質をコードする遺伝子NPTX1、PTGS2およびAPOEを過剰発現に導くことが示された。実際、タンパク質PTGS2は、プロスタグランジンE2のプロスタグランジンD2(炎症前)へのスイッチを阻害することにより、炎症の負の調節因子の役割を果たす。更に、APOEは、炎症部位への単球の流入を減少させることにより炎症の負の調節因子として作用する。NPTX1は、その役割のため、T細胞の活性化を阻害する。したがって、これらタンパク質の発現の誘導により、β−(1,3)−グルクロナン化合物またはβ−(1,3)−グルコグルクロナン化合物の混合物は、化粧品、とりわけ、抗炎症、抗赤味および/または緩和製品で使用することができる抗炎症活性を有する。
これら化合物の一または複数の非存在下または存在下においてインビトロで培養したヒト繊維芽細胞で実施した転写研究により、これら分子が、抗酸化および抗ラジカル活性を担う同じ名称のタンパク質をコードする遺伝子ALDH3A1、EPXH1およびHSPA6を過剰発現に導くことが示された。実際、タンパク質ALDH3A1は、フリーラジカルの直接的なトラッピング、酸化還元バランスの維持、グルタチオン状態の維持(抗酸化分子)、並びに脂質および過酸化アルデヒドの無毒化に同時に関与する。更に、タンパク質EPXH1は、過酸化脂質の代謝および求電子体(酸化スチレン、酸化エチレン、ガンマ照射など)の無毒化に関与する。タンパク質HSPA6は、その役割のため、細胞レベルにおいて全般的に酸化ストレスに対して活性である。したがって、これらタンパク質の発現の誘導により、β−(1,3)−グルクロナン化合物またはβ−(1,3)−グルコグルクロナン化合物の混合物は、有効成分として、および/または他の化合物と組み合わせて、抗酸化および/または抗ラジカル活性を有する美容用組成物の調製のために使用することができる。
β−(1,3)−グルクロナン化合物またはβ−(1,3)−グルコグルクロナン化合物は、一価および二価の陽イオンをキレーティングおよび金属イオン封鎖することができる多価陰イオン分子である。このことは、一価の陽イオン(特にNa+およびCu+)および/または二価の陽イオン(特にCu2+、Ca2+、Mg2+およびZn2+など)を、攪拌しながら、β−(1,3)−グルクロナン化合物またはβ−(1,3)−グルコグルクロナン化合物の混合物の存在下で溶解させることにより実証された。化合物を回収し、洗浄した後、原子吸光による調査により、β−(1,3)−グルクロナン化合物またはβ−(1,3)−グルコグルクロナン化合物の混合物は、一価および二価の陽イオンをキレーティングおよび金属イオン封鎖することが示された。
Claims (9)
- 式(I)のβ−(1,3)−グルクロナンまたはβ−(1,3)−グルコグルクロナン、並びにこれらの薬学的に許容可能な塩の一または複数を有効成分として含む美容用組成物:
− R1、R2およびR3は、同一であっても異なっていてもよく、各グルコース単位に対して独立に選択され、ヒドロキシル、アルコキシ、アシルオキシまたはスルホニルオキシ基を表し;
− Rは、各グルコース単位に対して独立に選択され、カルボキシル、アルコキシカルボニル、アシル基、または-CH2R4基(ここでR4は、ヒドロキシル、アルコキシ、アシルオキシ、スルホニルオキシ、スルフィニル基を表す)を表し;
− nは、平均分子量が500〜4 000 000ダルトンになるように選択され;
R基の少なくとも0.1%が、当該グルコース単位の各々に対して独立に選択され、カルボキシル、アルコキシカルボニル、アシル基を表す。 - R1、R2およびR3は、同一であっても異なっていてもよく、各グルコース単位に対して独立に選択され、ヒドロキシル、アルコキシ、またはスルホニルオキシ基を表すことを特徴とする、請求項1に記載の組成物。
- Rは、各グルコース単位に対して独立に選択され、カルボキシルまたはアシル基、または-CH2R4基(ここでR4は、ヒドロキシル、アルコキシ、スルホニルオキシ基を表す)を表すことを特徴とする、請求項1または2に記載の組成物。
- nは、平均分子量が500〜4 000 000ダルトンになるように選択されることを特徴とする、請求項1〜3の何れか1項に記載の組成物。
- 式(I)の化合物が、少なくとも5%の酸化度を有することを特徴とする、請求項1〜4の何れか1項に記載の組成物。
- 式(I)の化合物が、様々な酸化度を有し、以下のものから選択されることを特徴とする、請求項1に記載の組成物:
− 500〜2 500 000ダルトンの分子量を有するβ−(1,3)−D−ポリグルクロン酸;
− 600〜3 500 000ダルトンの分子量を有する過アセチル化β−(1,3)−D−ポリグルクロン酸;および
− C2およびC4の位置で硫酸化された、800〜4 000 000ダルトンの分子量を有するβ−(1,3)−D−ポリグルクロン酸。 - 前記組成物が、痩身組成物、抗セルライト組成物または抗ミカン肌組成物である、請求項1〜7の何れか1項に記載の組成物。
- 痩身組成物、抗セルライト組成物または抗ミカン肌組成物の製造のための、請求項1〜7の何れか1項に規定されるβ−(1,3)−グルクロナンまたはβ−(1,3)−グルコグルクロナンの使用。
