Description

本発明は、インフルエンザウイルス様粒子の分野に関する。特に、キメラインフルエンザウイルス様粒子として本明細書において開示されている。
インフルエンザ AおよびBは、流行性のヒト疾患を引き起こす2種類のインフルエンザウイルスである(111)。インフルエンザ Aウイルスは、2つの表面抗原:ヘマグルチニン(HA)およびノイラミニダーゼ(NA)に基づいて、さらにサブタイプに分類される。インフルエンザ Bウイルスは、サブタイプには分類されないが、種が長い時間をかけて分かれる連続変異を受けていく。1977年以来、インフルエンザ A(H1N1)ウイルス、インフルエンザ A(H3N2)、およびインフルエンザ Bウイルスは、世界的規模に循環している。ヒト A(H3N2)およびA(H1N1)ウイルスの間において遺伝的な再会合の後におそらく出現した、インフルエンザ A(H1N2)ウイルスは、多くの国において最近、検出されている。インフルエンザ AおよびBウイルスは、抗原特性に基づく群に、さらに分けられる。新たなインフルエンザウイルスのバリアントは、ウイルス複製の間に起こる点変異の結果として生じる頻繁な抗原変化(すなわち、抗原連続変異)の結果として、生じる。インフルエンザ Bウイルスは、インフルエンザ Aウイルスよりも緩やかな速度において抗原連続変異を受ける。抗原連続変異を介した抗原バリアントの頻繁な発生は、季節的な流行に対するウイルス学的な原理であり、かつ毎年のインフルエンザワクチンに少なくとも1つの株が編入される理由である。
本発明は、ワクチン製造にとって十分な量に生成され得、鼻腔内を含む種々の種々の手段によって送達され得、新たな株のウイルスに対してそれらが生じるときに素早く適合され得、かつ免疫原性の増強に必要とされるようなVLPの一部として免疫賦活剤を含み得る、キメラインフルエンザVLPの製造および利用に関して、本明細書に開示されているような種々の方法および組成物を提供することによって、これらの必要性を満たす。さらに、本明細書に開示されているインフルエンザVLPは、インフルエンザに対する異種亜型防御を提供することができる。全体に渡って使用されるように、インフルエンザは、特に指定がない限り、一般的にインフルエンザ AおよびBのいずれか、または両方を指す。

本発明は、キメラインフルエンザVLPの形成を基準にして、好ましくはマウス白血病ウイルス(MLV)から得られるgagポリペプチドを包含する。VLPを生成する好ましい方法は、バキュロウイルス発現系において種々のレトロウイルスから取得され得るgag VLPの顕著な収量(23、24、46、49、52−58)といった理由から、好ましくはインフルエンザHAポリペプチド抗原、およびインフルエンザNAポリペプチド抗原の共発現を含めた、昆虫細胞における発現によるものである。Gagポリペプチドは、機能的なgagタンパク質が、リボソームフレームシフトに起因する、レトロウイルスプロテアーゼ活性、逆転写酵素活性、およびインテグラーゼ活性を含む巨大なC末端伸張を天然に有する、天然のレトロウイルスアッセンブリ過程において、固有にC末端伸張を含む。人工的な伸張を用いた機能的なgagタンパク質の産生は、RSV gag(59)およびMLV gag(60)の両方に関して達成されている。gagC末端の操作におけるこの自由度は、本明細書に開示されている、インフルエンザキメラVLPにおける他のポリペプチド(例えば、他の抗原および免疫刺激性タンパク質の配列(例えば、TLRアゴニストであるフラゲリン))の包含にとって重要な部位を提供する。
本明細書において使用されるときに、“Gagポリペプチド”は、本明細書に記載されているウイルス様粒子の形成を担う、レトロウイルス由来の構造ポリペプチドである。ある実施形態において、gagポリペプチドは、ある特定の性質(例えば、RNAをパッケージする性質、または粒子形成および出芽の効率)に影響を与えるために、わざと変異され得る。当該変異の一例は、RNAを組み込むgag由来VLPの能力に影響を与える、アミノ酸変更である。VLPの出芽効率を向上または修飾する他のアミノ酸変更が、なされ得る。レトロウイルスのゲノムは、3つの主要な遺伝子産物をコードする:構造タンパク質をコードするgag遺伝子、核酸分解酵素およびインテグラーゼ関連機能である逆転写酵素、および関連タンパク質分解性ポリペプチドをコードするpol遺伝子、感染細胞の表面上、および成熟した放出ウイルス粒子の表面上にもまた検出される糖タンパク質膜タンパク質をコードするenv。すべてのレトロウイルスのgag遺伝子は、全体として構造的な類似性を有し、レトロウイルスの各群の内部において、アミノ酸レベルにおいて保存されている。gag遺伝子は、逆転写酵素を除いて、コアタンパク質を生じる。MLVに関して、Gag前駆体タンパク質は、Pr65Gagであり、かつ前駆体における順番が、NH2−p15−pp12−p30−COOHである4つのタンパク質に切断される。これらの切断は、ウイルスプロテアーゼによって媒介され、かつウイルスに依存してウイルス放出の前または後に生じ得る。MLV Gagタンパク質は、糖鎖付加形態および非糖鎖付加形態において存在する。糖鎖付加形態は、非糖鎖付加Pr65GagにとってのAUGコドンから上流に位置する別のインフレーム開始コドンから合成される、gPr80Gagから切断される。糖鎖付加Pr65Gagを合成しないMLVの欠失変異体は、依然として感染性であり、かつ非糖鎖付加Gagは、依然としてウイルス様粒子を形成でき、従って糖鎖付加現象の重要性に関して疑問を生じる。ウイルスにコードされるプロテアーゼによる、HIV−1 Gag前駆体 pr55Gagの転写後切断は、N−ミリストイル化および内的なリン酸化p17 マトリックスタンパク質(p17MA)、リン酸化p24 カプシドタンパク質(p24CA)、ならびにp9およびp6にさらに切断されるヌクレオカプシドタンパク質 p15(p15NC)を生じる。
VLPは、gagポリペプチドおよびノイラミニダーゼポリペプチドを含み、かつヘマグルチニンポリペプチドをさらに含み得るアセンブリしたVLPを好ましく生じる、当業者に利用可能なあらゆる方法によって容易にアッセンブリされ得る。好ましい実施形態において、ポリペプチドは、利用可能なあらゆるタンパク質発現系、好ましくは脂質におけるラフト−脂質ドメインを含む細胞から作られる系(例えば、哺乳類細胞発現系および昆虫細胞発現系)において共発現され得る。
調合物
本明細書に記載されているVLPの好ましい利用は、ワクチン調製物としてである。典型的に、当該ワクチンは、液体溶液または懸濁液;また注射の前に液体が調製され得る溶液にいれるか、または懸濁液にいれるために好適な固体の形態のいずれかとしての注射可能物質として調製される。また、当該調製物は、乳化され得るか、または乾燥粉末として製造される。活性な免疫原性成分は、当該活性成分と薬学的に受容可能であり、かつ適合性である賦形剤としばしば混合される。好適な賦形剤は、例えば、水、生理食塩水、デキストロース、スクロース、グリセロール、もしくはエタノールなど、およびこれらの組み合わせである。さらに必要に応じて、ワクチンは、補助剤(例えば、湿潤剤もしくは乳化剤、pH緩衝剤、またはワクチンの有効性を増強する免疫賦活剤)を含有し得る。

本明細書に開示されているVLPは、インフルエンザの特定の株に関しておよび/またはインフルエンザの複数の株に渡って、免疫原性を増加させるために、インフルエンザから得られる付加的な抗原を含み得る。
(実施例1−キメラインフルエンザVLPの産生)
MLVgagコーディング配列は、完全なモロニー(Moloney)マウス白血病ウイルスの広宿主性プロウイルス配列を含有するプラスミド pAMS(ATCC)から、PCRによって得られた。gagコーディング配列は、pFastBacI(インビトロジェン(Invitorgen))の中に、多角体プロモータの後ろに挿入され、かつ結果のプラスミドは、バキュロウイルスゲノムへの組み換えのために、DH10Bac形質転換受容性細胞の中に形質転換された。それから、高分子量バックミドDNAは、精製され、かつgag発現組み換えバキュロウイルス生成のために、Sf9細胞にトランスフェクトされた。A/PR/8/34(H1N1)のヘマグルチニンおよびノイラミニダーゼのそれぞれをコードする、2つの他の組み換えバキュロウイルスは、ウイルスRNAからのHAおよびNAコーディング配列のRT−PCR後に、同様の方法において産生された。最後に、3つの産物のすべて(HA−gag−NA)をコードする単一のバキュロウイルスベクターは、個々のpFastBacIプラスミドからのHA、gagおよびNA発現ユニット(多角体プロモータ−コーディング配列−ポリA部位)を、単一のpFastBacIベクター内に一体化することによって産生された。最初の分析用に、gagまたはHAまたはgag−HA−NAをコードする組み換えバキュロウイルスは、6ウェルプレートにおけるSf9細胞内に、1を越えるMOIにおいて感染させられた。感染後の3日目に、培養上清が、壊死組織片を除去されて、それから20%スクロース緩衝によって100000×gにおいてペレット化された。ペレットは、gag特異的抗血清およびH1N1特異的抗血清を用いた、ウエスタンブロッティング分析によって分析された(図1AおよびBを参照すればよい)。
HA−gag−NA VLPは、インフルエンザウイルス A/PR/8/34のMLV gagおよびHA産物およびNA産物をコードする、3重発現バキュロウイルスベクターを用いて感染させたSf9細胞の培地から調製された。培地は、細胞生存度が10%未満に下がったときに回収され、かつ2000rpmにおける15分に渡る遠心分離によって細胞片を除去された。不純物を除去した40mlの培地(MLA−80 超遠心チューブ毎に5ml)は、pH7.4のトリス緩衝化生理食塩水(TBS)において、1mlの20%スクロース緩衝の上に層状化され、それからMLA−80 ローターにおいて45000rpm、10℃において2時間に渡って遠心分離された。ペレット化された物質は、TBSにおいて再懸濁化され、かつ20−60%のスクロース段階勾配の上に層状化され、かつMLA−80 ローターにおいて、36000rpm、10℃において2時間に渡って遠心分離された。勾配は、0.5mlの画分に分画され、かつSDS−PAGEゲル上において、p65 gagの存在に関してアッセイされた。VLPを含有するプールした画分(2つの勾配から得られる約3ml)は、濃縮され、かつ緩衝液は、遠心分離微量濃縮器を用いてTBSに交換した。VLPの収量は、SDS−PAGEによる少量の分量の分析によって、評価された。
実施例1に記載されているように、A/PR/8/34のMLV gagおよびHA産物およびNA産物を発現する個々のバキュロウイルスが、産生されている。またさらに、3つの産物のすべてをコードする3重発現バキュロウイルスベクターが、構築されており、かつ感染昆虫細胞から得られるペレット化された培養上清のスクロース密度勾配遠心分離は、HAを有するVLPが形成されていることを示すウエスタンブロッティングによって検出されるように、gagおよびHAの一致したバンドを示した。HA、gag、およびNAのコーディングエレメントが、それ自身のプロモータ(Pr)およびポリアデニル化配列(pA)を伴って、頭−尾様式に配置される、3重発現ベクターにおけるコーディング配列の配置は、図4に示されている。単一のベクター内にすべてのコーディング配列を組み合わせることは、別々のウイルスを用いた共感染を実施する必要、および3つの別々のウイルスの一致した感染多重度を達成する関連した困難性を回避する。
バンド化されたVLPのスクロース密度勾配画分は、MLV gagと同時に結合するHA抗原活性が、機能的なHAであることを証明するために、ニワトリまたはテンジクネズミのRBCを採用する標準的なアッセイを用いた赤血球凝集反応活性に関して、ふるい分けされる。また、孵化鶏卵において培養されたインフルエンザウイルス A/PR/8/34は、スクロース勾配上において層状化され、かつ陽性対象として赤血球凝集反応アッセイにかけられる。
スクロース密度勾配精製VLPが、セファロース(Spharose) CL−4Bを用いたサイズ排除クロマトグラフィーにかけられ、かつ画分がウエスタンブロットによって、MLV gagおよびHAに関して観察される。VLPは、空隙容量において溶出し、かつMLV gag、HAおよびNAを含有する。A/PR/8/34 ウイルスは、対照としてクロマトグラフにかけられる。
スクロース勾配からのVLP試料は、2%のグルタルアルデヒドを用いて処理され、EMグリッド上に吸着され、リンタングステン酸ナトリウムを用いて陰性染色され、かつ電子顕微鏡によって調べられる。
VLP免疫原性は、参考文献(44)および上述の実施例2に記載されている炭疽病防御抗原(PA)調合物と類似の鼻腔内的なキトサン/MPL調合物を用いて、メスのBalb/cマウスにおいて評価される。VLPは、それらが後に、トリス緩衝化生理食塩水に再懸濁され、かつ20−60%スクロース密度勾配上に層状化される、100000×gにおいて1時間に渡って20%のスクロース緩衝を介して、VLP含有培養培地をペレット化することによって精製される。VLP含有画分は、ウエスタンブロットまたは赤血球凝集反応アッセイによって同定され、かつ貯蔵される。VLP試料は、PBS内において透析され、かつ遠心分離微量濃縮器を用いてか、または100000×gにおいて遠心分離によって濃縮される。
本実施例4は、適応性の免疫応答の強度を高めるためのTLR5アゴニストであるフラゲリンの組み込みによって、ならびに連続変異バリアントおよび異種亜型に対する防御を向上するためのM2外部ドメインエピトープの組み込みによって、向上した免疫原性および防御に対するVLP増強を証明している。
フラゲリンコーディング配列は、最近になってS.ティピムリウムのゲノムDNAからクローン化され、かつ終止コドンまでのちょうど5’までをgagコーディング配列の3’末端に挿入された。また、フラゲリンコーディング配列は、シグナル配列および成熟なコーディング配列の間における境界に位置するPstI部位を用いて、A/PR/8/34のN末端に挿入されている。HAにおけるこの配置におけるHAは、SDS−PAGEによって証明されているような予想される分子量の増加を伴う、キメラHA分子の適切な発現を導く。フラゲリン修飾gagおよびHAコーディング配列は、実施例1に記載されるような3重バキュロウイルス組み換え体(HA−gag−NA)を生成するために使用される。フラゲリン修飾gagまたはフラゲリン修飾HAを有するVLPをコードする組み換えバキュロウイルスは、産生され、かつフラゲリン配列を欠如する基本的なVLPをと比較した、免疫原性試験用のVLPを生成するために使用される。実施例3に述べたように、免疫化試験のすべては、4週間の間隔を空けて初回接種および追加接種を用いた。免疫応答的な読み出しは、全身性IgG応答および粘膜IgA応答の両方を試験する、上述のようなHAIおよびELISAアッセイを介してである。
M2外部ドメインエピトープコーディング配列は、PCRによって生成され、かつこの配列の1コピーが、単一のペプチドおよびPR/8/34 HAの成熟コーディング配列と境を接するPstI部位に挿入される。昆虫細胞におけるこのキメラ体の発現のウエスタンブロット分析は、HA特異的およびM2特異的な抗体と反応する、予想された大きさのキメラHA−M2産物を示した(図示せず)。この同じ位置においてM2の2つおよび3つの縦列コピーを含有する構築物が、進行中である。HA(M2)−gag−NA VLPをコードする3重発現バキュロウイルス組み換え体(M2の1、2または3コピーを有する)は、基本のVLPに対しての、免疫原性および攻撃防御の比較用に産生される。マウスは、上述のように鼻腔内のキトサン/MPL調合物を用いて免疫化され、かつ免疫応答は、HAIおよびELISAアッセイを介して監視される。ELISAアッセイは、HA/NAおよびM2に対する応答を別々に監視するために、完全なウイルスまたはM2外部ドメインペプチドのいずれかを採用する。さらに、免疫化された動物は、同型防御および異種亜型防御のそれぞれを監視するために、A/PR/8/34(H1N1)またはA/KH/68(H3N2)のいずれかを用いて攻撃される。攻撃後における体重低下/生存およびMDCK細胞におけるウイルスの力価測定は、上述のように実施される。以下の表2は、特定の目標2(Specific Aim 2)を目的とした動物実験の概要を示している。
A/ベトナム/1203/04(H5N1)およびA/インドネシア/5/05(H5N1)のHAおよびNA遺伝子は、H5N1 VLPの産生および続くH5N1の免疫原性研究用に取得されている。両方のH5N1株から得られるHA−gag−NAをコードする3重のバキュロウイルス発現ベクターは、実施例3に記載されているように、産生され、かつ性質決定されている。2つの3重バキュロウイルス発現ネクターによってコードされる、ベトナムおよびインドネシアのH5N1 VLPは、20−60%スクロース密度勾配における層状化によって精製され、かつVLPを含有する画分は、SDS−PAGE、ウエスタンブロット、および赤血球凝集反応アッセイによって同定された。3つのアッセイのすべては、多量のH5N1 VLPを含有するような同じピーク画分を同定した。マウスの免疫化に関して、ピーク画分が貯蔵され、トリス緩衝化生理食塩水を用いて3倍に希釈され、かつVLPが、100000×gにおける遠心沈降によって濃縮され、かつPBSにおいて再懸濁された。
鼻腔内ワクチンの乾燥粉末調合物は、液体調合物と比較して、増強された有効性を示している(42−44)。マウスは、鼻腔内の乾燥粉末送達には小さすぎ、かつ鼻腔内の乾燥粉末ワクチンがウサギモデルにおいて証明されているので、ウサギが、乾燥粉末調合物および免疫化研究に利用される。
免疫化に対して、基本的なH1N1 VLP(gag+HA(H1)+NA(N1))、およびマウス適合化H1N1(A/PR/8/34)ウイルス、およびH3N2スワーム(A/HK/68+A/Mem/85)攻撃ウイルスを採用する、免疫化ならびに攻撃研究が、実施された。図5は、生理食塩水における5μgのH1N1 VLPおよび20μgのモノホスホリル脂質A(MPL)を伴った初回免疫および追加免疫の後における、マウスにおける赤血球凝集反応阻害(HAI)力価を示す。I.P.免疫化は、4週間の間隔を空けられ、かつ血清試料は、免疫化のそれぞれの2週間後に回収された。血清試料は、受容体破壊酵素(RDE)を用いて処理され、かつ新鮮なヒヨコのRBCを採用する標準的なアッセイを用いて、HAI活性についてアッセイされた。16匹のVLP免疫化マウスのすべては、鶏卵増殖H1N1(A/PR/8/34)に対する強いHAI力価を示した。予想される通り、HAI活性は、H3N2スワーム(A/HK/68+A/Mem/85)に対して観察されなかった。また、未処理対照動物は、H1N1またはH3N2ウイルスのいずれかに対してもHAI反応性を示さなかった。

Claims (17)
- gagポリペプチド、インフルエンザのヘマグルチニンポリペプチド、およびインフルエンザのノイラミニダーゼポリペプチドを包含するキメラインフルエンザウイルス様粒子であって、
上記ヘマグルチニンポリペプチドが、インフルエンザ抗原に対して共有結合的に連結されており、
上記キメラインフルエンザウイルス様粒子は昆虫細胞から生成されている、キメラインフルエンザウイルス様粒子。 - 上記gagポリペプチドが、マウス白血病ウイルス、ヒト免疫不全ウイルス、アルファレトロウイルス、ベータレトロウイルス、ガンマレトロウイルス、デルタレトロウイルスおよびレンチウイルスからなる群から選択されるレトロウイルスから得られる請求項1に記載のキメラインフルエンザウイルス様粒子。
- 上記インフルエンザ抗原が、インフルエンザウイルスM2エピトープである請求項1に記載のキメラインフルエンザウイルス様粒子。
- 上記ウイルス様粒子との混合物において免疫賦活剤をさらに包含する請求項1に記載のキメラインフルエンザウイルス様粒子。
- 上記免疫賦活剤が、上記ウイルス様粒子の内部または外部にある請求項4に記載のキメラインフルエンザウイルス様粒子。
- 上記免疫賦活剤が、上記ウイルス様粒子の内部にあり、上記免疫賦活剤が、上記gagポリペプチドに対して共有結合的に連結されて、共有結合を形成している請求項5に記載のキメラインフルエンザウイルス様粒子。
- 上記免疫賦活剤が、上記ウイルス様粒子の外側にあり、上記ヘマグルチニンポリペプチドに対して共有結合的に連結されて、共有結合を形成している、請求項5に記載のキメラインフルエンザウイルス様粒子。
- 上記免疫賦活剤が、上記ウイルス様粒子の外側にあり、上記ノイラミニダーゼポリペプチドに対して共有結合的に連結されて、共有結合を形成している請求項5に記載のキメラインフルエンザウイルス様粒子。
- gagポリペプチドをコードする第1のヌクレオチド配列、インフルエンザのノイラミニダーゼポリペプチドをコードする第2のヌクレオチド配列、およびインフルエンザのヘマグルチニンポリペプチドをコードする第3のヌクレオチド配列を包含し、ここで、昆虫細胞における発現によって、上記ポリペプチドが、キメラウイルス様粒子を形成し、
第1、第2、および第3のヌクレオチド配列が、単一の発現ベクター内にある、キメラインフルエンザウイルス様粒子発現ベクター系。 - 第1、第2、および第3のヌクレオチド配列が、単一のプロモータに対して作動可能に連結されている請求項9に記載のキメラインフルエンザウイルス様粒子発現系。
- キメラインフルエンザウイルス様粒子の製造方法であって
(a)gagポリペプチド、インフルエンザのヘマグルチニンポリペプチド、およびインフルエンザのノイラミニダーゼポリペプチドをともに発現する1つ以上の発現ベクターを準備すること;
(b)培地における昆虫細胞内に上記1つ以上の発現ベクターを導入すること;
(c)上記gagポリペプチド、ヘマグルチニンポリペプチド、およびノイラミニダーゼポリペプチドを発現して上記キメラインフルエンザウイルス様粒子を製造すること;ならびに
(d)上記昆虫細胞が培養されている培地から上記キメラインフルエンザウイルス様粒子を回収することを包含しており、
上記発現ベクターが、バキュロウイルスベクターである、方法。 - 請求項1〜8のいずれか1項に記載のキメラインフルエンザウイルス様粒子の免疫原性のある量を包含する、薬学的組成物。
- インフルエンザの処置または予防のための医薬品を製造するための、gagポリペプチド、インフルエンザのヘマグルチニンポリペプチド、およびインフルエンザのノイラミニダーゼポリペプチドを含んでおり、昆虫細胞を用いて生成されるキメラインフルエンザウイルス様粒子の、使用。
- 対象に対する上記医薬品の投与によって、上記対象における防御免疫応答が誘導される請求項13に記載の使用。
- 処置されるかまたは予防される上記インフルエンザが、ヘマグルチニンポリペプチドの連続変異バリアントを有するインフルエンザウイルスによって引き起こされるインフルエンザ、およびノイラミニダーゼポリペプチドの連続変異バリアントを有するインフルエンザウイルスによって引き起こされるインフルエンザを含む請求項13に記載の使用。
- 上記防御免疫応答は、異種亜型防御または連続変異バリアント防御を含んでおり、
上記異種亜型防御はH1N1インフルエンザウイルスに対する防御を含んでおり、
上記ヘマグルチニンポリペプチドはH5であり、かつ上記ノイラミニダーゼポリペプチドはN1である請求項14に記載の使用。 - 上記防御免疫応答は、異種亜型防御または連続変異バリアント防御を含んでおり、上記異種亜型防御が、H3N2 インフルエンザウイルスに対する防御を含み、かつ上記ヘマグルチニンポリペプチドが、H1であり、かつ上記ノイラミニダーゼポリペプチドが、N1である請求項14に記載の使用。
