Description
本発明は、様々な抗原からの挿入されたエピトープを提示する組換え分子を含む免疫原性組成物およびワクチン組成物に関する。特に、本発明は、心血管疾患の発生(development)に関与する抗原および病原体に対する免疫応答を誘発するためのかかる組成物に関する。
心血管疾患(CVD)は、全ての死のほぼ 50% を占め、死および身体障害の遍在する原因となっている。心臓発作(CHD)および脳卒中がその最も重要な症状のうちの2つであり、他には閉塞性末梢血管疾患(PVD)および静脈血栓塞栓症がある。CVD は現在、全ての死の 51%、即ち全ての型の癌を合わせた(21%)よりも2.5倍多くを占めている。
血小板由来増殖因子(PDGF)は、ジスルフィド結合した A および B ポリペプチド鎖からなるホモ二量体およびヘテロ二量体タンパク質のファミリーを含む (Waterfield et al、Nature 1983、304、35-39; Uutela et al、Circulation 2001、103、2242-2247)。いくつかの細胞型は PDGF 遺伝子の協調した発現を示すが、一般的には、各々の発現は独立している (Heldin et al,Physiol Rev 1999、79、1283-1316)。広範囲の細胞外の刺激が各々の発現に影響を与えるため、合成レベルは細胞特異的でありうると考えられる。
PDGF は、胚発生において基礎的な機能を有している(ノックアウトマウスにおいて周産期死亡が起こる)。PDGF-B 鎖ノックアウトマウスモデルにおいては、例えば、心臓の欠陥ならびに不完全な腎臓および血管が見出された(Leveen et al、Genes Dev 1994、8、1875-1887)。結合組織を生産する細胞型は、特に PDGF に依存している。結合組織の形成における PDGF の重要な役割は、成人の創傷治癒においても重要である(Werner & Grose、Physiol Rev 2003、83、835-870)。
腫瘍増殖および線維性疾患における PDGF の関与の報告が続いているが、アテローム性動脈硬化は研究的興味の主要な的である。
HSP の一般的バックグラウンド: 熱ショックタンパク質は、他のタンパク質の組立て(assembly)、フォールディングおよび転位置を助ける一群のタンパク質である。さらに、それらは熱傷または他の形態のストレスから細胞を保護する。原核および真核の全ての細胞は、これらのタンパク質を合成することによって様々な細胞ストレスに応答することができる。熱ショックタンパク質は高度に保存され、普遍的に分布し、かつ、ウイルス性および細菌性感染の重要な側面、自己免疫疾患および癌免疫に関与する。
酸化型 LDL (OxLDL): OxLDL は、それがコレステロールを有するマクロファージのロード(loading)を説明することからアテローム性動脈硬化のシグナル伝達に関与すると当初は考えられていたが、oxLDL が潜在的にアテローム生成誘発性の(pro-atherogenic)他の多くの性質を有することがすぐに明らかとなった。興味は、(a) 最小限に(minimally) oxLDL がマクロファージ化学誘引物質(Cushing et al、Proc Natl Acad Sci USA. 1990、87,5134-518)、接着分子(Berliner et al、J Clin Invest. 1990、85、1260-1266)およびサイトカイン(Rajavashisth et al、Nature. 1990、344、254-257)の内皮性発現を刺激すること、ならびに (b) 大いに(extensively) oxLDL が、細胞コレステロール含量によって調節されない様式でマクロファージのスカベンジャー受容体によって取り込まれること(Henriksen et al、Proc Natl Acad Sci USA. 1981、78、6499-6503; Witztum JL. Role of oxidized low density lipoprotein in atherogenesis. Br Heart J. 1993、69、S12-S8)を実証したインビトロの研究に部分的に基づいている。
OxLDL は、タンパク質部分および脂質部分の両方の上に様々な酸化ネオエピトープ(oxidation neoepitope)を含有する。かかるエピトープの生成において特に興味深いものは、ホスファチジルコリン(PC)の反応性分解産物、例えば LDL タンパク質および脂質を修飾して共有結合付加体を形成させることができる 1-パルミトイル-2-(5-オキソバレロイル)-sn-グリセロ-3-ホスホリルコリン(POVPC)である。POVPC およびマロンジアルデヒド(malonodialdehye)等の物質による修飾は、ネオ自己エピトープ(neo-self epitope)の形成をもたらすと考えられる。これらの修飾は、ホスホリルコリン含有酸化リン脂質それ自体と共に、スカベンジャーおよび Toll 受容体との LDL の相互作用に寄与すると考えられる。
ヒトサイトメガロウイルス(HCMV)は、世界中の人口の大部分に感染する種特異的ベータヘルペスウイルスである(Stern、J.lL.et al.、The Journal of Immunology、2008; 180: 6577-6585)。CMV 感染はまた、後期の移植片不全(graft failure)の根底にある慢性の同種移植脈管障害(移植血管の動脈硬化によって特徴付けられる)の発生を促進する (Grattan MT、et al.、JAMA 1989; 261: 3561-3566; Everett JP、et al.、J. Heart Lung Transplant. 1992; 11: S133-S137)。インテグリン αvβ6 による TGF-β1 の活性化は病理学的変化に寄与し、慢性的に CMV に感染している組織における内皮細胞の機能を障害し得る (Tabata T、et al.、American Journal of Pathology. 2008;172:1127-1140)。マウス CMV(MCMV)の単回高容量による感染は、apoE-/- マウスにおいて、循環するインターフェロン-γ(IFN-γ)および腫瘍壊死因子-αのレベルの初期の上昇に続いてアテローム性動脈硬化の病変の進行を悪化させることが実証されている (Hsich E、et al.、Atherosclerosis 2001;156: 23-8; Burnett MS、et al.、J Infect Dis 2001;183: 226-31)。動物モデルによって、CMV による感染が動脈硬化を加速させ得ることが確認されている(Stassen FR、et al.、J Clin Virol 2006; 35:349-353)。
Cpn 感染とアテローム性動脈硬化とをつなぐ最初の証拠は、冠動脈心疾患および急性心筋梗塞を有する個人が、心血管疾患を有さない個人よりも有意に高い Cpn 特異的 IgG 抗体の力価を有することが見出された、という知見から得られた (Saikku P、et al.、Lancet. 1988; 2: 983-986)。その後の研究により、Cpn 感染は、動脈の脂肪線条ならびに進行したアテローム性動脈硬化のプラークにおいて常在することが見出された(Shor A、et al.、S Afr Med. J. 1992; 82: 158-161; Kuo CC、et al.、J Infect Dis. 1993; 167: 841-849)。Cpn はアテローム性の組織のおよそ 50% から 60% において存在すると推定された一方、アテローム性動脈硬化の証拠を有さない患者からのサンプルにおける有病率は 5% 未満である (Shor A、Phillips JI. JAMA. 1999; 282: 2071-2073)。アテローム性動脈硬化の病変から該細菌を単離および培養することも可能であり(Ramirez JA. Ann Intern Med. 1996; 125: 979-982)、該病原体が生存可能なだけでなく活発に複製することもできることが示される。したがって、現在の主な証拠は、アテローム性動脈硬化における Cpn 感染の因果的役割を大いに示唆している。症例対照研究によって、特定の抗 Cpn 抗体のレベルが対照患者よりも CVD を有する患者において有意に高いことが明らかにされ、追跡研究によって、Cpn に対する高い抗体力価が臨床イベントのリスクの増大と関連することが示された (Virok D、et al.、Stroke 2001; 32:1973-6)。
アテローム性動脈硬化と主要な歯周病病原体であるポルフィロモナス・ジンジバリスとの間の有意な関連が示されている (Ford et al.、J Dent Res. 2007; 86: 35-40)。ポルフィロモナス・ジンジバリスは、高脂血症の動物においてアテローム性動脈硬化の病変の発生を加速することが示されている (Koizumi et al.、Infect Immun. 2008; 76: 2958-65)。
HSP は、タンパク質合成の間ならびにタンパク質の細胞内分解および除去の間に細胞内において生成される多数のペプチドと非共有結合性に結合する本来的な能力を有している。これらのペプチド/HSP 複合体のいくつかは表現型特異的であり、異常な細胞に対する免疫応答の生成のために用いられ得る (Xu、Thromb Vasc Biol 2002、22、1547-1559)。実際に、HSP の免疫学的機能は、特定の HSP (hsp70、hsp90 および gp96) が腫瘍から精製された場合に、その特定の腫瘍に対する免疫学的反応を誘発し得ることが観察された時に初めて発見された (Blachere et al、J Immunother 1993、14、352-356; Nieland et al、Proc Natl Acad Sci U S A 1996、93、6135-6139)。その後の研究により、免疫原性は HSP と複合体化した腫瘍特異的抗原性ペプチドに起因することが示された。提示のために抗原性ペプチドを保持することに加えて、HSP/抗原複合体はマクロファージおよび樹状細胞による炎症誘発性サイトカインの分泌ならびに樹状細胞の成熟を刺激した。これらの免疫学的特性は、癌および感染性疾患に対する免疫増強剤(immuno-adjuvant)としての HSP の使用における大きな興味を生み出した(Nieland et al、Proc Natl Acad Sci U S A 1996、93、6135-6139; Pockley、Expert Rev Mol Med、2001、1-21; Manjili、Front Biosci 2002、7、d43-52; Lamb、Atherosclerosis 2003、167、177-185; Lewis、Proc Natl Acad Sci U S A、2004、101、14653-14656)。HSP/抗原性ペプチドは切除された腫瘍組織から単離し、免疫化(自己免疫化)のために用いることができるため、これは個々の腫瘍に対して適用できるアプローチである。
ヒトおよび動物モデルについての疫学的研究により、冠動脈疾患(CAD)と口腔感染、特に歯周炎との間の関連の証拠が示されている (Matilla、BMJ 1989、779; de Stefano、BMJ 1993、306、Beck、J. Periodontol、1996 1123; Scannapeco、Ann. Periodontal 2003、38)。歯周炎は、歯肉膨張(gum inflation)(歯肉炎)および歯肉の進行性脱離(重度の歯周炎)から、歯の喪失につながる歯槽骨(bone alveolar)の密度の減少までの範囲を含む、慢性の破壊的炎症状態である。口腔の細菌、例えばポルフィロモナス・ジンジバリス、アクチノバチルス・アクチノミセタムコミタンス(Actinobacillus actinomycetemcomitans)、タンネレラ・フォーサイセンシス(Tannerella forsythensis)、トレポネーマ・デンティコラ(Treponema denticola)およびカンピロバクター・レクタス(Campylobacter rectus)は、該疾患の原因と関係づけられている。したがって、歯周組織の感染は、アテローム形成に寄与するサイトカインおよびリポ多糖のような炎症メディエーターの慢性的な貯蔵所を提供する。CAD の発病における伝統的な危険因子の役割は良く確立されているが、先進国および発展途上国の両方における該疾患の高い発生率および世界的な有病率を完全には説明できない。炎症は CAD において中心的役割を果たし、病原体は宿主における慢性の組織的炎症性応答を誘発することが伝統的に知られている(Danesh、Lancet 1997、350; Hoffmeister、J. Am Coll Cardiol、2000; 35-112)。
第1の側面において、本発明は、
i) キャリア部分;
ii) 抗アテローム応答を誘発することができる第一のエピトープ; および
iii) 抗アテローム応答を誘発することができる第二のエピトープ
を含み、該第一および第二のエピトープが互いに異なることを特徴とする組換えタンパク質を提供する。
a. 該対象とする1以上のエピトープを含むデンドロアスピン足場タンパク質を構築し、該タンパク質を発現させる工程、
b. 真核細胞を該デンドロアスピン足場タンパク質と共にインキュベートする工程、
c. 該真核細胞を用いてミクロソームを調製する工程、
d. 該ミクロソームおよびデンドロアスピン足場タンパク質を1以上の医薬上許容される成分と組み合わせて、経口投与可能な(orally administerable)調製物を生産する工程、
e. 該調製物を哺乳類またはヒトに経口投与する工程。
本発明を、以下の図を参照して詳細に説明する。
1) PDGF エピトープの選択
1-a) Atherovax の開発において用いたペプチド配列
PDGF-BB のループ I および III における残基に対応する一連のペプチドを、研究所において、 Atherovax の開発において抗原として調製した (Oefner et al、EMBO J 1992、11、3921-3926; Sun et al、Annu Rev Biophys Biomol Struct 1995、24、269-291; LaRochelle et al、J Biol Chem 1992、267,17074-17077; Brennand et al、FEBS Lett 1997、413、70-74; Patel et al、J Pept Res、1999、53、68-74)。さらに、該ペプチドのうち特定のものは、真皮の線維芽細胞において DNA 合成を刺激することが示された。これらに基づいて一連の環状ペプチドをも調製し、そのうちの1つは PDGF に誘導される DNA 合成を阻害することができ、かつ、アポトーシスを誘導することができた。該環状ペプチドは、その細胞増殖を阻害する能力と細胞上における PDGF 受容体の存在との間の相関を示した。
ペプチドは、Fmoc ストラテジーを用い、連続フロー固相合成によって合成される。切断および脱保護の前に、N,N-ジメチルホルムアミド中の 45% 無水酢酸を用いて 30 分間、N-末端の封鎖を行う。アリルに基づく Glu の保護を用いて、線上で(on line)ペプチドの環化を行う。N-Fmoc 基の除去の後、製造者の説明書に従い、O-(7 アザベンゾトリアゾール-1-イル)-1,1,3,3-テトラメチルウロニウム ヘキサフルオロフォスフェートおよび 1-ヒドロキシ-7-アザベンゾトリアゾール(azabenzotrizole)を用いて頭-尾(head to tail)の環化を行う。0.1% トリフルオロ酢酸中の 10-60% アセトニトリルの 30 分の勾配を用いる Vydac C18 カラム(22×250 mm)上での逆相 HPLC によって、ペプチドを精製する。精製されたペプチドの同定および性質決定は、分析的逆相 HPLC および ESI 質量分析を用いて行う。ペプチド配列の例および定めた合成ペプチドの番号を、表 1 に示す。
Wick および同僚による先駆的な研究(Wick et al、Immunol Today. 1995、16、27-33)によって、ヒトおよび実験的アテローム性動脈硬化における、HSP65/60 に対する抗体応答および細胞媒介性(cell-mediated)免疫反応性が明らかにされた。oxLDL とは対照的に、HSP65 による非経口の免疫化はアテローム性動脈硬化を悪化させ(Xu et al、Arterioscl Thromb. 1992、12、789-799)、HSP65-反応性の細胞の移植も、高コレステロール血症の動物モデルにおいて疾患を加速する (Shoenfeld et al、J Autoimmun. 2000、15、199-202)。近年の研究により、ヒト HSP60 および bovis HSP65 (Uray et al、Int Immunol. 2003、15、1229-1236)が病原性の原因であるエピトープを有し得、かつ、例えば関節炎(Ulmansky et al、J Immunol. 2002、168、6463-6469)およびアテローム性動脈硬化(Perschinka et al、Arterioscler Thromb Vasc Biol. 2003、23、1060-1065)において疾患誘導に対する抵抗性を与えることが示唆されている。ヒト HSP60 および bovis HSP65 のエピトープは、その抗原性について試験されている。エピトープ選択は、β-ターン二次構造の高い可能性および疎水性の低い可能性を有するタンパク質セグメントに基づいていた(Uray et al、Int Immunol. 2003,15,1229-1236)。ペプチド配列の例および定義した合成されるペプチドの番号を、表 2 に示す。
3-a) 組換えデンドロアスピン足場免疫原の発現
PDGF および/またはストレプトコッカス・ニューモニエ 、CMV、ApoB または M bovis hsp60 由来のペプチドがループ 2 および/または 3 の中に移植されたデンドロアスピン足場を含む多数の組換え免疫原(表 3 に示す)を作成した。該デンドロアスピン分子は、その上へペプチド配列を移植するのに有用な足場を提供する。それは、移植されたペプチド配列の発現および提示を可能にする3つの露出したループを有する(図 1 参照)。かかる組換えキメラデンドロアスピンコンストラクトは、標準的な方法によって、原核生物の発現系においてグルタチオン S-トランスフェラーゼ融合タンパク質として都合良く発現される。
組換えタンパク質の発現を、先に記載した通りに行った(Lu et al、2001; BJ)。簡潔には、500 ml の培養液(2YT 培地プラス終濃度 100 μg/ml のアンピシリン)に 1 ml の種培養を接種した。該培養を、37℃において振盪しながら(250 rpm)終夜増殖させた。1 リットルの新鮮な 2YTamp を添加し、該培養を 37℃においてさらに 3 時間インキュベートした。IPTG を 0.1 mM の終濃度まで添加し、該培養を、Sorvall (GS-3) ローター中における 4℃での沈降(6000 rpm 15 分)の前に、30℃においてさらに 4 時間インキュベートした。ペレットを -20℃において保管した。
親和性精製: GST-組換えタンパク質を、グルタチオン-セファロース 4B カラムを用いるアフィニティークロマトグラフィーによって、遠心分離後に超音波処理した細胞の上清から精製した。グルタチオンセファロース 4B は、グルタチオン S-トランスフェラーゼの組換え誘導体または他のグルタチオン依存性タンパク質の迅速な一段階精製のために設計されている。細胞を、以下の阻害剤を有する PBS 中に溶解した: トリプシン阻害剤 (50 mg/ml)、ペプスタチン (10 mg/ml)、アプロチニン (10 ml ストック)、EDTA (0.5 M) および PMSF (EtOH 中における 100 mM)。超音波処理の後(氷上で 4 回超音波処理、30 ml 容量の細胞混合物に対し、各回 50 秒)、10 % の Triton-X 100 溶液を添加し、氷上に 10 分間置いた。Sorvall RC-5B 遠心機においてローター SS-34 を用い、16,000 x g で 30 分間遠心分離を行った。次いで上清を回収し、精製カラム上にロードした。該カラムを、PBS およびトリス-HCl(pH 8.0)バッファーで洗浄した。50 μM のグルタチオンを含有するバッファーを用いて、タンパク質を溶出した。タンパク質産物のピークレベルを回収し、プールし、4 ℃における 12 時間の第Xa因子による消化(1:300、w/w、第Xa因子:融合タンパク質)によって、突然変異体-ADAM を GST-融合タンパク質から遊離させた。産物を、逆相 HPLC によって精製した。
第Xa因子切断の後、画分を Vydac C18 逆相 HPLC 分析カラム(TP104)上にロードし、0.1 % TFA を含有する 0-26 % のアセトニトリルの勾配(毎分 1.78 %)およびその後の 0.1 % TFA 中の 26-36 % アセトニトリルの勾配(毎分 0.25 %)を用いて溶出した。活性な画分を機能アッセイによって同定し、均一性のために再度クロマトグラフに供した。精製されたタンパク質を、最初に質量分析によって確認した。
細胞遊走阻害アッセイ:
孔径が 8 μm、インサートの直径が 6.5 mm の Transwell プレート(Corning Life Sciences)を用いて、細胞遊走阻害アッセイを行った。コンフルエントな細胞を、実験の前に 0.5 % BSA を有する無血清 DMEM 培地中で 24 時間維持した。プレートも、24 時間前に 100 μg/ml の Invitrogen の溶液 100 μl を用いてコートした。トリプシンを用いて細胞を回収し、細胞数を決定し、遠心し、1 x 10 6 細胞/ml の密度で再懸濁した。次いで、細胞の添加の前に、両方のチャンバーを 30 分間プレインキュベートした。100 μl の容量の細胞を上部チャンバーに置き、化学誘引物質 PDGF (A15 を伴うまたは伴わない)を下部チャンバーに添加した。37℃、5 % CO2 における 16 時間のインキュベーション期間の後、膜の上面からの細胞を除去し、下面からの遊走した細胞をメタノール中で固定し、Diff-Quik 溶液を用いて染色した。次いで、膜を顕微鏡スライド上に乗せ、5 つのランダムな視野において 40 x の倍率で計数した。
図 2 は、20ng ml-1 の PDGF 単独(100%)と比較した、遊走する細胞のパーセンテージとして表される細胞遊走阻害アッセイの結果を示す。したがって、低い値は、PDGF 受容体結合ドメインを提示する組換えタンパク質(10μM)による PDGF の効果の競合的阻害の成功を示す。効果的であることが知られている環状 PDGF ループ I ペプチド(TROM と称する)を、より高い濃度(200μM)で比較している。
細胞増殖アッセイ-MTT アッセイ:
黄色のテトラゾリウム MTT (3-(4,5-ジメチルチアゾリル-2)-2,5-ジフェニルテトラゾリウムブロミド) は、代謝的に活性な細胞によって、部分的には脱水素酵素の作用によって還元されて、還元当量(reducing equivalent)、例えば NADH および NADPH を生成する。その結果得られる細胞内の紫色のホルマザンは可溶化することができ、分光光度的手段によって定量することができる。このプロトコールは、MTT アッセイキットのためのマニュアルではあるが、一般的な用途に用いることができる。MTT ストック溶液: フェノールレッドを含まない RPMI-1640 中における 5mg/ml の MTT (Promega)。この溶液は、0.2 mm のフィルターを通して濾過し、2-8℃において保管する。MTT 使用溶液: 該 5mg/ml ストック(フェノールレッドを含まない RPMI 中の MTT)の 1:10 希釈。1. 培養細胞を、フェノールレッドを含まない温かい RPMI-1640 で洗浄する。 2. MTT 使用溶液を調製する。 3. MTT 使用溶液をアッセイされるウェルに添加する、例えば 12-ウェルプレートの各ウェルに対し 1.0 ml。37℃において 30 分から 3 時間(この時間は細胞密度および細胞型に依存する)インキュベートする。 4. 付着した細胞を扱う場合には、インキュベーション期間の終わりに、培地を除去することができる。 5. 変換された色素を、1ml の酸性イソプロパノール(無水イソプロパノール中における 0.04 M HCl)を用いて可溶化する。ピペットで数回上下させて、変換された色素を完全に溶解させる。 6. 細胞を伴う該色素溶液を 1.5 ml のエッペンドルフチューブへ移し、13,000 rpm で 2 分間遠心する。 7. 上清を新しいエッペンドルフチューブへ移す。変換された色素の吸光度を、650nm のバックグラウンド減算を用いて、570nm の波長において測定する。測定には、分光光度計および使い捨てのプラスチックキュベットを用いる。
図 3 は、cPDGF と比較した、5 つの免疫原コンストラクトの、PDGF に誘導される増殖に対する阻害効果を比較する。高い値は、最大の競合的阻害を示す。
抗体および抗原の力価アッセイ:
本発明者らは、組換えタンパク質(抗原)の特性決定のために、抗体および抗原の ELISA に基づくアッセイを用いる。ELISA アッセイを行うため、親和性カラムの後に GST-タンパク質を透析してトリスおよび NaCl を除去した。GST-タンパク質は、高濃度の塩(NaCl)を含有するため、凍結乾燥することができない。ELISA アッセイは、以下の工程を含む: 1. 予め決定した最大濃度の抗原を、マイクロタイタープレートへ吸収させる。コーティングバッファー中において希釈し、ウェルあたり 100 μl を 4℃において終夜インキュベートする。 2. 抗原溶液を吸引し、ウェルあたり 100 μl のブロッキング溶液で 15 分間インキュベートすることによってプラスチック上の非結合部位をブロッキングする。 3. 結合していない抗原を洗い流し、洗浄バッファーを用いてタンパク質を3回ブロックする。100倍希釈の抗体をプレートに添加する。 4. 結合していない抗体を、洗浄バッファーを用いて3回洗い流す。 5. HRP 標識二次抗体を推奨されたまたは予め決定した希釈において添加する。ウェルあたり 100 μl を周囲温度(ambient temperature)において1時間インキュベートする。 6. 結合していない標識を、洗浄バッファーを用いて3回洗い流す。 7. 酵素および基質を添加する。ウェルあたり 100 μl を、周囲温度において色が十分に発生するまでインキュベートする(理想的にはおよそ 30 分)。 8. 洗浄は行わない。50 μl の適切な停止溶液を添加し、マイクロタイタープレートを読み取るよう設計された分光光度計において視覚的または定量的に評価する。
図 4 A は、PDGF ループ I コンストラクト(表 3 に示すコンストラクト V2、V3、V4 および V6)の、抗 PDGF BB ポリクローナル抗体との飽和可能な(saturable)反応性を示す。PDGF ループ III コンストラクトである V1 は非反応性である。
4-a) エピトープの選択
一連のエピトープを、アテローム性動脈硬化のタンパク質抗原(oxLDL および HSP)およびアテローム性動脈硬化の病原体(HCMV、Cpn およびポルフィロモナス・ジンジバリス(p. gingivalis))から得た。該エピトープの例を表 4 に示す。該エピトープを、上記セクション 1-b に記載した通りに調製した。
試験したエピトープの組み合わせの例を、表 5 に示す。
5-a) 動物モデルにおける免疫応答
組換え免疫原の、BALB/C マウスモデル、C57/BL6 マウスモデルおよび Apobtm2Sgy Ldlrtm1Her/J KO-マウスモデルにおいて免疫応答を生み出す能力。マウスを、反復性の複数部位免疫化ストラテジー(repetitive, multiple site immunization strategy)(RIMMS)を用いて免疫化した。10 頭の 6-8 週齢の BALB/c マウスの群に、それぞれ 5 回、フロイントアジュバントと混合された 4 μg の組換え免疫原を接種した。免疫化は、0、2、5、7 および 9 日目に、8 つの部位へ、皮下に行った。12 および 21 日目に、抗体応答を試験するために血液サンプルを回収する。組換え分子を最初に Balb/C または C57/BL6 マウスにおいて試験して免疫応答を観察し、その後、それらの免疫応答に基づいて、該分子を KO マウスにおいてさらに試験し、アテローム性動脈硬化に対するこれらのペプチドの効果を調査した。
ApoB-100 を発現する LDL 受容体欠損マウスを、ApoB1(IEIGLEGKGFEPTLEALFGK、配列番号25)および hHSP60(AELKKQSKPVT、配列番号26)に由来する2つのペプチドの組み合わせを用いて免疫化した。二特異性組換え免疫原を用いる免疫化は、アテローム性動脈硬化の病変の進行からの相乗的な保護を提供することが示された。
Apobtm2SgyLdlrtm1Her/J マウスを、キャリアタンパク質としてのキーホールリンペットヘモシアニン(KLH)と結合した ApoB および hHSP エピトープの組み合わせを用いて免疫化した。該ペプチドは、組み合わせて二特異性免疫原として、または完全/不完全フロイントアジュバントと共に単一の形態において、与えた。マウスに高脂肪の食餌を 7 週間与え、その後屠殺した。ELISA によって、ペプチド-特異的抗体応答を血清中において検出した。以下の表 7 に示す通り、両方のペプチドの組み合わせを用いる免疫化によって誘導された抗体レベルは、ApoB または hHSP ペプチドのいずれかを用いる免疫化によって得られた値と同様の OD 値を示した。
図 5A-D に示される通り、大動脈の組織を組織学的に解析した。結果: 組織学的解析により、大動脈洞におけるアテローム性動脈硬化の病変は、対照マウス(図 5A)におけるよりも、2つのペプチドの組み合わせを用いて免疫化したマウス(図 5D)において、サイズが 41.34% 小さいことが明らかとなった。病変は、ApoB1 ペプチド単独での免疫化の後では対照マウスよりも 14.66% 小さく(図 5B)、hHSP60 ペプチド単独での免疫化の後では対照マウスよりも 21.15% 小さかった(図 5C)。
Apobtm2SgyLdlrtm1Her/J マウスにおけるアテローム性動脈硬化の病変へのマクロファージの浸潤を、抗 CD68 を用いる染色によって定量した。組織サンプルの切片(厚さ 5 μm)をヘマトキシリン・エオシン(H.E.)を用いて染色し、光学顕微鏡で観察した。マクロファージのレベルを、アジュバントのみを用いて免疫化したマウス(対照)、ApoB のみを用いて免疫化したマウス、hHSP60 のみを用いて免疫化したマウスおよび二特異性 ApoB1 + hHSP60 免疫原を用いて免疫化したマウスから得たサンプルにおいて比較した。
BALB/c マウス(雌、6 週齢)を、デンドロアスピン足場中に ApoB1 ペプチド1、hHSP60、cpn 1 および cpn 2 を含む四重特異性 GST 結合免疫原を用いて免疫化した。
BALB/c マウス(雌、6 週齢)を、デンドロアスピン足場中に CETP ペプチド + ポルフィロモナス・ジンジバリス(p.gingivalis)からの選択されたエピトープを含む二特異性 GST 結合組換え免疫原を用いて免疫化した。
6-a) アプローチの概要
ワクチンの開発の広い特徴は以下の通りである。抗原を用いてマウスを免疫化し、生じた抗体を親和性カラムによって精製する。結合に関与する可変領域内の配列を、ハイブリドーマから単離された mRNA または単離された抗体の可変領域の mRNA をシークエンスすることによって同定する。これらの配列を、組換え抗体発現系へ組み込む。該抗体のアテローム性動脈硬化を阻止する能力を、動物モデルにおいて試験する。一揃えの HSP60 ペプチド(直鎖状または環状)を用いて BalbC マウスを免疫化する。最も高い阻害力価を生み出すペプチドを、その後マウスを免疫化するために用いる。必要であれば、組換え抗体ライブラリーをこの段階で用いる。上記の通り、抗体の親和性に関与する主要な配列を、DNA ワクチンの開発において用いる。上記の両方の組み合わせから、二特異性ワクチンを開発する。
二特異性分子を生成するために用いられる単鎖抗体(single chain variable fragment)(scFv) および Fab 組換え抗体の生産のために、融合が用いられる。ScFvs は、そのサイズの小ささに起因する、モノクローナル抗体を超えるいくつかの利点、例えば少ない腎臓取り込み、迅速な血液クリアランスおよびヒト免疫系による負の応答の低さを提供する。
これは、高い親和性および選択性を有する抗体を発現するハイブリドーマからの mRNA の単離を含む。mRNA の単離は、単一のチューブにおいて、ポリアデニル化された RNA を、組織ライセートから直接、オリゴ(dT)25 結合磁性粒子へハイブリダイズさせることによって行う。MPG(登録商標) Direct mRNA Purification Kit (MDRK1010) または MPG(登録商標) Guanidine Direct mRNA Purification Kit (MGRK1010) のいずれかを、製造者の説明書に従って使用する。次いで、単離された mRNA を、分光光度法、ゲル電気泳動および完全溶液相(ultimately solution phase)RT-PCR によって、収量、純度および完全性(integrity)について解析する。モノクローナルからの mRNA をクローニングして、可変重鎖(Vh)および軽鎖(Vl)がその後そこから発現ベクターへとサブクローン化される cDNA ベクターを作成する。
供給者の説明書に従い、Superscript 逆転写酵素(GIBCO/BRL)を用いて逆転写を行う。5%(vol/vol)のジメチルスルホキシドの存在下で、Taq DNA ポリメラーゼ(GIBCO/BRL)を用いて PCR を行う (94℃において 4 分、次いで、94℃で30秒、37℃で30秒および 72℃で2分の 3 サイクル、次いで、60℃において同様のサイクルを 10 回、72℃における伸長が1サイクル毎に 15 秒ずつ延長される、37℃の代わりに 60℃における同様のサイクルを 20 回、および 72℃における 10 分、で終了する)。
5巡目のリボソームディスプレイの後、PCR 産物を適当なベクター(例えば pTFT74)へクローニングした。S-30 大腸菌系におけるインビトロの共役した転写-翻訳を、50 μg/ml のプラスミド DNA および以下の改変を伴う上記と同様の条件を用いて行う。共役した転写-翻訳は 37℃において 30 分間行い、反応混合物には 2000 単位/ml の T7 RNA ポリメラーゼおよび 0.5 mM の UTP および TTP を補充する。混合物は、[35S]メチオニンを 50 αCi/ml (1 αCi = 37 kBq) で含み、メチオニンを除く各アミノ酸を 0.35 mM で含んだ。翻訳の後、反応混合物を PBS を用いて 4 倍に希釈し、マイクロタイターウェル中において、固定化された抗原ペプチドと結合させる。穏やかな振盪を伴う 60 分のインキュベーションの後、0.1% Tween 20 を含有する PBS を用いてマイクロタイターウェルを5回洗浄し、結合した放射性タンパク質を 0.1 M のトリエチルアミンを用いて溶出させる。溶出したタンパク質(抗体)を、シンチレーションカウンターにおいて定量する。
ラージスケールのタンパク質発現のために、選択された抗体配列を pGEX-3X または pET32b ベクター中にクローニングし、大腸菌を形質転換する。標準的な方法に従って、GST 融合タンパク質の発現および精製を行う。抗体を、真核細胞発現ベクター中にもクローニングし、真核細胞、例えば酵母において発現させる。
ファージディスプレイは、ヒトの抗体タンパク質をその表面上に‘提示’するよう、ファージを作る方法である。ヒトの抗体遺伝子ライブラリーからの遺伝子を、ファージの集団へ挿入する。各々のファージは異なる抗体の遺伝子を保有し、それにより、異なる抗体をその表面上に提示する。これらの遺伝子は回収することができ、抗体治療薬の前向きの開発および潜在的な製造における使用のために利用することができる。
“リボソームディスプレイ”により、無細胞系において、いかなる細胞、ベクター、ファージまたは形質転換をも用いずに、機能的全タンパク質をその結合機能について濃縮することができる新規な方法が開発された。この技術は、mRNA およびタンパク質産物の両方がリボソームから離れないインビトロの翻訳に基づいている。この技術は、リボソームディスプレイライブラリーの構築、抗体ライブラリーの一本鎖断片 scFv のインビトロの翻訳、リボソーム複合体の親和性による選択および RNA の単離、抗体の一本鎖断片の ELISA、修正された(corrected)突然変異体の存在についてのライブラリーの PCR 解析、タンパク質の発現、抗原のアフィニティークロマトグラフィー、ウエスタンブロット、競合 BIAcore による溶液中における抗原解離定数の決定等を含む。
選択されたエピトープまたは適切な抗体(二官能性抗体を含む)を発現させるために、DNA ワクチン接種を用いる。実験的には、これは哺乳類細胞トランスフェクションキットプロトコール(www.SpecialtyMedia.com)を用いて行う。ヒトの HSP65 またはマイコバクテリアの HSP65 および PDGF をコードする pcDNA (Invitrogen) ベクターを構築し、抗原の発現および DNA ワクチン接種のために用いる。トランスフェクションを、DNA を CaCl2 およびリン酸緩衝液と直接混合して微細な沈殿を形成させ、それを培養細胞上に分散させることを含むリン酸カルシウム媒介法によって行う。該リン酸カルシウムトランスフェクションのプロトコールは、様々な細胞型の一過性のおよび安定なトランスフェクションの両方のために、日常的に用いられる。
裸の(naked) DNA の遺伝子銃または筋肉内(i.m.)注入プロトコールは、筋肉内の免疫化のために用いられる。
7-a) ELISA: 各々の抗体の特性の追跡および定量を、標準的な ELISA の技法を用いて、各々の抗原に対してチェックする。
15,000 共鳴単位(resonance unit)の BSA-抗原複合体または対照としての BSA のみによってコートされたセンサーチップ CM5 (Pharmacia) を用いて、質量輸送(mass-transport)下において BIAcore 解析を行う。各々の結合-再生サイクルを、HBST(20 mM Hepes、pH 7.2/150 mM NaCl/0.005% Tween 20)を用い、25 μl/分の一定の流速を用いて行う。様々な量の抗原を含有する、HBST 中における 250 μl の抗体のサンプルを、該システムのサンプルループを用いて注入し、その後、HBST 中の 6 M の塩化グアニジウムを 20 μl 注入することによって表面を再生する。注入前における、4℃での、少なくとも 1 時間の、一連の濃度における抗体と抗原ペプチドとの共インキュベーションによって阻害試験を行う。データを、BIAEVALUATION ソフトウェア (Pharmacia) および KALEIDAGRAPH (Synergy Software、Reading、PA) を用いて評価する。直線状センソグラム(sensogram)の結合相の傾きを、対応する全抗原濃度に対してプロットする。
通常飼料における 10 週齢の雄の LDL 受容体(LDLR)欠損マウスを、適切な量の熱ショックタンパク質-PDGFペプチド複合体(前のセクションに記載した免疫原性実験から決定される)を用いて免疫化する。これらの動物の応答を、ペプチド単独、HSP 単独または緩衝液単独で免疫化した動物と比較する。2回目の免疫化を、1 週間後に行う。その後、動物を2つの群に分け、一方に通常の飼料を与え、他方に高コレステロールの食餌(1.25% コレステロール、0% コール酸塩(cholate); Research Diets)を与える。14 週間後に、マウスを殺し、血液を採取し、大動脈を除去し、0.9% NaCl を用いて灌流する。大動脈を 3 つの部分に解剖する - 基部および弓部はクライオスタット切片作成のために Tissue-Tek(登録商標) OCT Compound 中において急速凍結させ、腹部は 2% パラホルムアルデヒド中で固定する。定量化をコンピューター画像解析(Zeiss software)によって行う標準化された方法に従い、大動脈基部および胸腹部大動脈においてアテローム性動脈硬化の程度を評価する。これは、大動脈基部における互いに 50 μm 離れた 6 つの切片中の病変の平均領域の算出; およびこの脂質沈着の領域(スーダン IV によって染色される)を全弁表面で除算することを含む。スーダン IV を用いた標本において病変領域を染色し、腹側正中線に沿った縦方向の切開によって胸腹部大動脈を試験する。スーダン IV で染色された領域を全胸腹部表面で除算することによって、脂質沈着のパーセンテージを算出する。
8-a) インビトロの泡沫細胞モデル
これは、脂質がロードされた、ホルボールエステルによって分化した THP-1 マクロファージ (0.1μM/24h) および J774 マウスマクロファージ細胞株を用いる Kellner-Weibel らの方法に基づいている(Kellner-Weibel et al、Arterioscler Thromb Vasc Biol 1998、18、423-431)。ヒト LDL を遠心分離によって血漿から分画し、Basu らの方法 (Basu et al、Proc Natl Acad Sci U S A 1976、73、3178-3182) によってアセチル化する。いくつかの実験において、単核白血球を調製するために、採取したばかりの(freshly drawn)血液から密度勾配を用いて単球画分を単離し、そこから磁性ビーズを用いるリンパ球のネガティブ除去(negative depletion)によって単球を分離する。細胞を、細胞における遊離コレステロールの迅速な蓄積をもたらすアシル補酵素 A:コレステロールアシルトランスフェラーゼ阻害剤(ACAT 阻害剤、58035)の存在下で培養する。
20mM トリス-HCl (pH 7.9)、0.5 M NaCl、5mM イミダゾール、0.1% ノニデット(Nonidet) P-40 およびプロテアーゼ阻害剤を含有するバッファー中において超音波処理によって細胞を溶解し(100 ml の溶解バッファーあたり 109 細胞)、その後氷上で 0 分インキュベートする。ライセートを 10000xg で 1 時間遠心し、この工程からの上清を 100000g における 2 時間のさらなる遠心分離(Beckman Ti75 ローター)に供する。その後、個々の HSP の精製を上記の通りに行う (精製された熱ショックタンパク質)。
精製の間、精製された HSP を洗浄して、ゆるく結合したあらゆるペプチドを除去する。次いで、結合したペプチドを、トリフルオロ酢酸を用いてタンパク質から溶離させ、表面増強レーザー脱離/イオン化(SELDI)質量分析を用いて解析する。(Ciphergen Biomarker System)。利用できるチップのタイプ(正または負の荷電、疎水性および金属キレート化を介する結合)の各々において、ペプチド画分を解析する。泡沫細胞の表現型において存在しない細胞と比較して増強したレベルを示すペプチドを、マトリックス支援レーザーによる一次構造の完全な同定のために選択する。切除された腫瘍組織から HSP/抗原性ペプチドを単離し、免疫化(自己免疫化)のために用いることができるため、これは個々の病変に適用できるアプローチである。
セクション中において PDGF ペプチドについて説明した手順に従い、HSP を、泡沫細胞に特異的であると同定されたペプチドの各々と複合体化する(酵素結合免疫吸着スポットアッセイ)。泡沫細胞の形成を阻止するかまたは確立された脂肪線条を逆行させる、泡沫細胞に対するワクチンとして作用するこれらの複合体の能力についても、上記と同様の方法(熱ショックタンパク質-PDGF 複合体の免疫原性の決定)によって決定する。
DNA ワクチンの臨床の場への移行は、その限られた効力によって阻まれてきた。これは、プラスミド DNA の筋肉内または皮内注入の後、ミオサイトおよびケラチノサイトが優性にトランスフェクトされるためである。これらの細胞は、強い一次免疫応答を生み出す能力を欠いている。この欠点を克服するために提案された1つの方法は、骨髄由来の抗原提示細胞、特に樹状細胞を標的とすることである (Hauser et al、Gene Ther 2004、11、924-932; Liao et al、Mol Ther 2004; 9:757-64)。これは、抗原と HSP とのキメラ融合遺伝子を調製することによって可能であることが最近示された。このアプローチによって、強い T 細胞および B 細胞応答を生み出すことが可能であった。
抗原提示細胞(APC)、例えばマクロファージからのミクロソームは、その中でプロセスされた抗原 ペプチドが MHC と複合体化し、APC の表面上における最終的な提示のための他の因子と相互作用する細胞内コンパートメントである、細胞を含まない小胞体(ER)の膜小胞からなる。表面上に現れると、それらは T 細胞と相互作用し、該 T 細胞を刺激することができる。該技術の特に有用な点は、ER がインビトロにおいて既知の‘保護性’ペプチドを効率的に充填し、その結果、APC を模倣する、高レベルの、膜中に提示された正しく組立てられた MHC-ペプチド複合体を有するミクロソームをもたらすことである。ミクロソームは、レシピエントの中に注入することができ、そこにおいて直接的に T 細胞と相互作用し、該 T 細胞を刺激する。
末梢血単核細胞(PBMC)を、健康な血液ドナーの新鮮なバフィーコートから、フィコール(Ficoll)密度遠心分離(Ficoll-Paque、Pharmacia、Uppsala、Sweden)によって調製する。バフィーコートを 0.9% NaCl において 1:2 に希釈し、35 ml を 15 ml のフィコールパック(Ficoll Paque)上に層状にし、400 g において 25 分間遠心した。細胞を、RPMI 1640 (Gibco、Grand Island、NY) 中において2回洗浄した。いくつかの実験のために、37℃における 2 時間のインキュベーションによって PBMC から接着性の細胞を枯渇させた。
樹状細胞(DC)を調製するため、PBMC を大きな細胞培養フラスコ中に移し、50 ml の RPMI 培地/3%/ヒト血清中において、加湿された細胞培養インキュベーター中で 37℃において 1 時間インキュベートする。1 時間後、懸濁状態の細胞をリンパ球画分として回収する一方、接着画分には単球が濃縮される。
抗-CD4-MACS(登録商標) 磁性ビーズカラムによって、リンパ球集団を CD4+ 細胞について濃縮する。MACS MicroBeads は、直径およそ 50 ナノメートルの超常磁性の粒子である。それは生分解性のマトリックスで構成されており、そのため、分離工程の後にそれを細胞から除去する必要がない。精製された CD4+ T 細胞を、20-50 IU/ml の組換えヒト IL-2 と共に培養状態に維持するか、またはさらなる使用の時まで凍結させる。成熟し、ロードされた DC を、T:DC が 10:1 の割合における自己 CD4+ T 細胞との共培養の前に、マイトマイシン C と共にインキュベートし、24 時間インキュベートする - その 4 日後、応答を媒介した細胞を、細胞内染色または Elispot によって測定される IFN-γ(Th1) および IL-4(Th2)についてのサイトカインプロファイル解析によって測定する。T 細胞の増殖を、CFSE 標識、BrdU 処理またはチミジン取り込みアッセイによって測定する。
DC 培養の 9 日目において、ロードされた DC 群(T 細胞とのインキュベーションの前に 20ug/ml のマイトマイシン C と共にインキュベートされた DC)を、ロードされた生きた DC としての使用、またはロードされた DC の超音波処理物(ミクロソームおよび原形質膜)としての使用のために、2 つのさらなる群へ分ける。

超音波処理物の調製のために、以下の手順を用いる。

RM バッファー(pH 7.2 の 50 mM の HEPES、250 mM のスクロース、50 mM の酢酸カリウム、2 mM の酢酸マグネシウム)中においてペレットをホモジナイズし、再懸濁させる(200ul/108 細胞)
8. 280nm において OD をチェックし、OD 60 の終濃度を作る
9. 分注されたチューブを、液体窒素中で急速凍結させる
10. 超音波処理物を、液体窒素中または -80℃において保存する
ミクロソーム画分は、細胞が破壊される際に小胞体から形成される、直径 20-200nm の不均一な小胞のセットを含む。該小胞は分画遠心によって単離され、3つの構造的特徴により構成される: 粗面小胞、滑面小胞およびリボソーム。多くの酵素活性が、ミクロソーム画分と関連する。ミクロソームは、操作上、低速の遠心分離によって核およびミトコンドリア画分が除去された後に、超遠心分離によって組織ホモジネートから得られる粒子状の画分として定義される。電子顕微鏡により、ミクロソームは、主として小胞と呼ばれる閉じた嚢状の膜によって構成されることが示されている。ほとんどの小胞は、粗面および滑面小胞体(ER)に由来する。ゴルジ体、ペルオキシソーム、エンドソーム、トランスゴルジ網(trans Golgi network)および他の中間コンパートメント(intermediate compartment)に由来する膜小胞は、ミクロソームのマイナーな構成成分を含む。肝臓のミクロソームは、粗面および滑面 ER 小胞をおよそ 2:1 の比で含有し、かつ、タンパク質分泌経路の構成成分に加えて、脂質/リポタンパク質の生合成および薬物代謝に関与する多数のタンパク質を含有する。該 ER は、代謝的に活性な細胞において圧倒的に豊富に存在する膜である。湿った組織1グラムあたり約 2-3 mg のミクロソームタンパク質を、肝臓から得ることができる。このように、ミクロソームは、酵素の構造、タンパク質-タンパク質および脂質-タンパク質の相互作用ならびに膜結合酵素の機能的特性の間の関係を研究するために理想的な調製物である。最も豊富なミクロソームタンパク質の多くが広く研究されてきたが、さらに多くのタンパク質が未だ単離および特徴付けされていないままである。

研究のために組織を選択した後、ホモジナイズバッファーの組成物、ホモジナイズの方法、ならびに低速遠心分離工程の時間および強さが、ミクロソームの調製における主要な変数である。実験処理あたりおよそ 10 g の肝臓を、室温のホモジナイゼーションバッファー(0.125 M の塩化カリウムおよび 1.0 mM の EDTA を含む、pH 7.4 の 0.1 M のリン酸カリウム緩衝液)中において解凍した。25 ml の冷却したホモジナイゼーションバッファー(バッファーの実験においては 0.25 M のスクロースを添加または非添加)へ移した後、ハサミを用いて肝臓を完全に細かく刻み、テフロンガラス(Teflon-glass)のホモジナイザーを用いて 10 ストローク(ストローク実験においては 6、8、または 10 ストローク)によってホモジナイズした(870 rpm)。ガラスチューブの底にある物質に対してより大きな圧力および時間が費やされる最初の2ストロークを除き、通過の間およそ 15 秒続くストロークは、等しくかつ一定であった。全てのホモジナイゼーションの間、チューブを氷および水の小さなバケツ中に沈めた。ホモジネートを、サンプル重の 4 倍容量(およそ 40 ml)まで希釈した。次いで、サンプルを、Sorvall SA-600 ローターを備える Sorvall RC-5B 中において 12,000g(力の実験においては 9,000、10,500、または 12,000g)で 20 分間遠心した (Sorvall、Newton、CT)。最初の遠心分離からの上清を除去し、ミトコンドリアのペレットを 25 ml 中に再懸濁し、遠心分離を繰り返した。上清を合わせ、Sorvall T-1270 ローターを備える Sorvall Ultra Pro 80 中において 138,000g で 60 分間遠心した。上部の脂質層を除去し、サイトゾルの上清を回収した。ミクロソームのペレットを、3回のホモジナイゼーションストロークによって 0.125 M の KCl、0.1 M のトリス(pH 7.4)中に再懸濁させ、138,000g における 60 分間の遠心分離を繰り返した。ミクロソームのペレットを、6回のストロークによってインキュベーションバッファー中に再懸濁させ、最終容量を 26 ml とした。サンプルを -70℃において保存した。

単球の DC への分化は、外来性の IL-4 および GMCSF を用いて 7 日間、インビトロにおいて行う。得られる細胞集団を、細胞表面分子である CD11c、CD123、HLA-DR についての多色染色によって特性決定する。一般的に、培養状態の細胞の 90% が DC マーカーに関して陽性である一方、5% がリンパ球マーカーである CD3 および CD5 に関して陽性である。
