Description
本発明は、特に、哺乳動物において、感染性病理因子に対して、特に、ワクチンとして使用できる免疫性組成物に関する。
病原体(agents pathogenes)によって誘起される感染時に上記病原体の若干の領域によって果たされる主要な役割は公知である。
従って、本発明の課題は、細胞感染に依存する感染性病理因子に対して、哺乳動物に使用できる免疫性組成物およびワクチン組成物を提供することにある。
・上記の如き感染性病原体の1つおよび/または複数の標的受容体を発現する第1手段を、上記標的を認識する病原体の領域を少なくとも発現する第2手段とともに、複合体を形成するよう第1,第2手段の相互作用を可能とする条件において、インキュベーションを行うことによって得られる調製体から生成され、
・上記インキュベーション段階は、異なる融合段階に対応する複合体を生ずるよう異なる期間(duree)に従って(それゆえ新たに脱マスクされたエビトープの暴露およびコンホメーションによって)遂行され、
・抗体の形成が望ましいエピトープ(すなわち形成されるべき抗体に対するエピトープ)の異なる暴露およびコンホメーションによって複合体を固定するよう、生成した複合体を固定剤(仏agent fixateur、英binding agent)と異なる期間にわたって接触させ、
・上記第1,第2手段が、哺乳動物に許容される(仏tolerer、英tolerate)ことを特徴とする。
細胞性免疫組成物の調製
一方では、ワクチンを投与すべき患者から採取され、JVI 1998,71:8273−8280のRileyらの方法にもとづき3−6日間PMA/IL2で励起(stimulation)した後CD4/CCR5およびCXCR4を発現するヒトの自家移植細胞を使用し、他方では、HIV−1の1次分離体またはこの種の分離体によって感染された細胞を使用した。
それぞれ15,30,45,60,120,180,240および300分間の各期間にわたって37℃においてインキュベーションを行うため、これら2つの集団を各ペトリボックスに分配した。各実験のため、3×106〜108の自家移植細胞および、例えば、HIVの2つの共受容体CXCR4およびCCR5を利用するエイズ・リサーチ・アンド・リファレンス・プログラム,NIH(EUA)から得た1次分離体92HT593またはACH168.10を使用した。
所定の期間の終了後、AT−2を600−1000μmの割合でPBS溶液として加えた。固定は、4℃において、Rossioら(JVI,1998,72,7992参照)が推奨している如き時間、即ち、1−12時間にわたって実施した。−80℃で保存するため、形成された細胞複合体をPBS(リン酸緩衝溶液)で洗浄、回収し、106〜108細胞/0,1mlの割合でPBS/(DM50 10%)中に懸濁させた。
凍結した病原体を解凍し、複数回洗浄し、次いで、同量のアジュバント、例えば、Ribi(R−700またはR−730)を加えた。
実施例1と同様に操作した。但し、一方では、CD4およびCCR5受容体および/またはCXCR4を表面に担持するバキュロウイルス系を使用し,他方では、gp120モノマーまたはgp120オリゴマーおよびgp41オリゴマーを担持する疱瘡ウイルスまたはセムリキ森林ウイルスを使用した。換言すれば、それ自体の膜内にタンパク質を発現できるバキュロウイルス系を使用した(Boublickら、Biotechnology,1995,13:1079-84)。
実施例1と同様に操作した。但し、CD4およびCCR5および/またはCXCR4を表面に担持するリポソームを使用した。上記リポソームの調製は、機能メンブランタンパク質またはトランスメンブランタンパク質を担持するリポソームを記載したRigaudらの情報(Biochim.Acta Phys.1995,1231:223-246またはPitardらの情報(Eur.J.Biochem.1996,235,3769-3778参照)に依拠した。これらを融合のため、HIV−1エンベロープまたはHIV−2エンベロープとともにインキュベーションを行った。異なる組換えエンベロープを含むHIVウイルス(プソイドタイプの補完ウイルスの利用)またはHIV組換えエンベロープを担持するウイルスベクターが対象となる。リポソームへの細胞膜のトランスメンブラン受容体の通過のために、細胞当り106−107ヶの受容体コピーを得ることができる発現性の強いベクター(例えば、バキュロウイルスまたはSaccharomyces cerevisae)を用いた。
実施例3の免疫性組成物を使用した。この場合、あらかじめ、上記組成物に対して、その抗体応答形成能を評価するため動物実験を行った。この組成物をPBS中で複数回洗浄し、生理学的血清中に懸濁させ、次いで、上記組成物にアジュバントとしてRibiを添加した。
上記調製体を上記動物(ハツカネズミ、サル、場合によっては、ヒト)に0,05ml−1mlの割合で注射した。更に4−6ヶ月後、追加注射を行った。
1.pcDNA3 CCR5は、ヒトのCCR5の完全なシークエンスを含む(遺伝子バンクのアクセスナンバー:NM000579)。PCRによってEco−RIサイトとTGAサイトとの間のC−末端領域を増強した。ATG(+10)から出発して、EcoRIサイトは、位置793にあり、Stop遺伝暗号は、位置1066にある。
2.シークエンス化のため、上記フラグメントをプラスミドpUC18においてクローニングし、次いで、フラグメントをEcoRI−XbalのpcDNA3CCR5に再挿入した。
3.かくして、プラスミドpcDNA3C CR5を改質し、pcDNA3 CCR5−6HISとした。
4.BamHI−BamHIをカットすることによってpcDNA3から遺伝子CCR5およびC−末端のその6HISタッグを抽出した。
5.次いで、BgI II−bgl Iによって、プロモータp10の下流(仏aval)において遺伝子CCR5−6HISを伝達ベクター(バキュロ)p119に導入した。
6.伝達ベクターp119にCCR5−6HIS遺伝子を添加した後、上記伝達ベクターをウイルスAcSLP10のDNAとともに昆虫(Autographa californica)のSf9細胞に共移入した。
7.CCR5−6HISが、上記細胞の表面に発現された。上記CCR5−6HISの密度をスキャッチャード(REF)によって評価した。
8.上記受容体の機能特徴を、上記細胞に対するウイルスHIVまたはHIVgp120の固定能によって評価した。
1.CD4遺伝子を含むpGEM−Tプラスミドに6つのヒスチジンタッグを加えた。ATG(+1)から出発して、Bsu361サイトは、位置1087にあり、Stop遺伝暗号は、位置1375にある。
2.C−末端に6つのヒスチジンタッグを加えたCD4遺伝子を、BgI II−bglIによって、pGEMAcI16T伝達ベクターのポリエドリン(polyedrine)のプロモータの下流に導入した。
3.pGEMAcI16T CD4−6HIS伝達ベクターを、AcSLP10ウイルスのDNAとともに、昆虫(Autographa californica)のSf9細胞に共移入した。
4.CD4−6HISが、上記細胞の表面に発現された。上記CCR5−6HISの密度をスキャッチャード(Cahoreauら、1992,Biochimie,74,1053-1065))によって評価した。
5.上記受容体の機能特徴を、上記細胞に対するウイルスまたはHIVgp120の固定能によって評価した。
1.CCR5−6HIS遺伝子およびCD4−6HIS遺伝子を、それぞれ、ベクターp119およびpGEMAcI16Tにおいてクローニングした。
2.p10プロモータおよびCD4−6HISの監視下(仏controle)のCCR5−6HISの発現をポリエドリンのプロモータの監視下で実施した。
3.2つの伝達ベクターを、AcSLP10ウイルスのDNAとともに、昆虫(Autographa californica)のSf9細胞に共移入した。
4.かくして、CCR5−6HISおよびCD4−6HIS対の等割合のダブル組換え体を表面に発現するSf9細胞が得られた。
かくして、Sf9細胞膜から、下記を含むリポソーム内に受容体を再構成できる:
1.CCR5のみ,
2.CD4のみ,
3,CCR5およびCD4を個別に発現する細胞から選択した割合のCCR5およびCD4,
4.同量のCCR5およびCD4を同時に発現する細胞から選択した割合のCCR5およびCD4。
HIVの共受容体と融合するHIVエンベロープを調製し、次いで、固定剤(例えば、パラホルアルデヒドまたはグルタルアルデヒド)によって融合を停止し、huCD4/huCCR5トランスジェニックマウスまたはマカックモデルまたは他の真猿類に上記免疫性対を注射した。結果に依存して、上記製剤をヒトに注射できる。
CXCR4について同一の系を用意した。
Rigaudら(1988,Biochemistry,27、2677-2688),Paternostreら(Biochemistry 1988,27,2668-2677)およびGaymardら(J.Biol.Chem.1996,271,22863-22870)が誘導した方法にもとづきプロテオリポソームを得るため、受容体CCR5(またはCXCR4)および/またはCD4を超発現(仏surrexpriment、英overexpress)するAutographa californicaのSf9細胞を適切な洗剤によって消化した。
1.Sf9細胞の表面に発現された場合
a)FACSおよび共焦(仏confocale)顕微鏡によって解析した細胞表面の受容体の存在:
I.特殊な抗−CD4または抗−CCR5抗体による
II.特殊抗体によってマーキングされたgp120による
III.変異したまたは変異してないエンベロープを担持するHIV−1による
IV.第1に、Sf9細胞の表面における受容体の機能性を特徴づけ、スキャッチャード(Cahoreauら、既述)によって、細胞膜の脂質(仏lipidique)状態が認められる細胞毎に分子数を定量する。
b)Robert Blumenthalの誘導した方法(NIH,個人的コミニュケーシュン,2000)およびVidalらの誘導した方法(1996,J.Biol.Chem.270,17823-17829)による融合の特殊蛍光の共焦解析
I.(場合によっては変異した)HIV−1エンベロープを発現する細胞との接触後
II.HIV−1(または場合によっては変異したエンベロープを担持するプソイドタイプ・ウイルス)との接触後
III.ウイルスプソイド粒子による。
2.対応するプレテオリポソームにおいて

Bac−CCR5:原プラスミドの変異したフラグメントの再組込みのために(遺伝暗号の変性(仏degenerescence)によって形成された)StulサイトとXbalサイトの上記オリゴヌクレオチドへの導入。

増強されたフラグメントEcoRI−XbalをEcoRI−XbalのベクターpUC中でクローニングし、次いで、シークエンス化した。次いで、変異したフラグメントをEcoRI−Xbal原プラスミド中に再挿入した。
このように改質したプラスミドをStulおよびXbaによってカットし、次いで、上記のStul−Xbal DNAフラグメントと結合した。このフラグメントは、6つのヒスチジン遺伝暗号および1つのStop遺伝暗号TAAを与える。

方法:
1.フラスコ内の細胞の計数。
2.1000rpmで5min遠心分離。
3.媒体5ml中への細胞の懸濁(例えば、細胞0.5×106/ml)。
4.カルセイン1μl/細胞5ml(約2.5mmolカルセイン)の添加。
5.良好な混合を達成するための渦流(vortex)
6.遮光状態でCO25−7%中で37℃において45minインキュベーション。
7.1000rpmで5min遠心分離。
8.媒体(またはPBS)10mlで2回洗浄。
9.媒体(またはPBS)5ml中にまたは細胞0.5×106/mlの濃度で細胞の懸濁。
10.遮光状態でCO25−7%中で37℃において30minインキュベーション。
11.媒体(またはPBS)10mlで2回洗浄。
12.媒体(またはPBS)5ml中にまたは細胞0.5〜1×106/渦流mlの濃度で細胞の懸濁。
CMTMRは、生きている細胞膜を経て自由に拡散するクロロメチルの蛍光性誘導体である。細胞内で、僅かにチオ反応性のゾンデ(プローブ)は、グルタチオン−S−トランスフェラーゼにもとづき反応して、細胞膜を透過しない蛍光性着色付加生成物を生成する。
CMTMRによる細胞の着色によって、不動の各周辺領域、ERおよびGolgiでのタンパク質との反応から生ずる鮮明な蛍光が生じ、且つまた小さい融合気孔を経て拡散できる細胞質ゾル中の蛍光性グルタチオン付加生成物(PM〜600Da)に起因するより弱い蛍光が生ずる。
1.蛍光性細胞質ゾンデCMTMR(5−(((4−クロロメチル)べンゾイル)アミノ)テトラメチルローダミン)および(6−(((4−クロロメチル)べンゾイル)アミノ)テトラメチルローダミン)(Molecular Probe,cat#C-2927,ex/em541/565nm)をDMSOに10mMの濃度で溶解した。
10μlまたは20μlの分別量を採取し、−20℃に保存した。加熱によって不活性化したSVF10%(100μ/ペニシリンml,100μg/ストレプトマイシンml(D10))を補足したDMEMでCMTMRの希釈物(1:500)を調製した。
2.(微小ウエル上で)標的細胞から媒体を除去し、CMTMRの希釈溶液1mlを塗布した。37℃において45−60min試料のインキュベーションを行った。着色溶液を1mlのD10で置換し、更に15−30min、インキュベーションを続行した。
融合の実施
1.CMTMRでマーキングした標的細胞に、カルセインでマーキングしたエフェクター細胞(2ml)を加えた。37℃において3−5時間、2つの細胞集団のインキュベーションを行った。
2.インキュベーション終了後、媒体を1mlのD−PBSで置換し、40倍の油浸レンズで相および蛍光を撮影した。
カルセインで着色された細胞の観察のため、FITC(励起装置:BP470−490;プリズムDM505;伝送器:BA515−550)を使用し、CMTMRのためにローダミン(励起装置:BP530−550;プリズムDM570;伝送器:BA590)を使用した。かくして、着色剤の蛍光を観察する際に生ずるオーバフローが避けられた。各試料についてランダムに選択した6−10の異なるフィルタによって像を統合した。
3.ソフトウエアMetamorph(Universal Imaging Inc.)を使用して像を重畳させ、計数してデータを解析した。CMTMRについてプラスの全細胞数および2つの蛍光ゾンデについてプラスの全細胞数を計数した。マーキングした細胞が相互に重畳するが融合してないプラスの比率を区別するため、高光沢像を使用する。融合%は、下式にもとづき計算する:
融合%=100×[2つの着色剤についてプラスの細胞数]/[全標的細胞数]
Claims (7)
- 免疫性組成物であって、該免疫性組成物が、
・CD4、および、CCR5および/またはCXCR4の組み合わせを発現する、細胞、ベクターおよびリポソームからなる群から選択される第1手段を、HIVウイルスのgp120またはgp160エンベロープタンパク質の少なくとも保存領域を発現する第2手段とともに、該第1および第2手段との間で複合体を形成するよう第1,第2手段の相互作用を可能とする条件において、インキュベーションを行うことによって得られる調製体から生成され、
・上記インキュベーション段階は、融合段階に対応する複合体を生ずるような期間に従って遂行され、
・生成した複合体を固定剤と、抗体が形成されるべきエピトープの暴露およびコンホメーションによって複合体を固定するような期間にわたって接触させ、
・上記第1,第2手段が、哺乳動物に許容されることを特徴とする、免疫性組成物。
- 第1手段が、哺乳動物の自家移植細胞、特に、ワクチンを投与すべき患者から採取したヒトの健全細胞であることを特徴とする請求項1に記載の組成物。
- 第1手段が、表面に1つまたは複数の標的受容体を発現するベクターであることを特徴とする請求項1に記載の組成物。
- 第1手段が、表面に1つまたは複数の標的受容体を担持するリポソーム、特に、表面に請求項3に定義の如きベクターを担持するリポソームであることを特徴とする請求項1に記載の組成物。
- 調製体が、インキュベーション後、アルジドリチオール−2で固定されていることを特徴とする請求項1−4の1つに記載の組成物。
- 請求項1−5の1つに記載の組成物に対して形成された血清および抗体。
- 哺乳動物への投与のための、感染性病理因子に対するワクチン組成物において、該組成物が、請求項1−5の1つに記載の有効量の免疫組成物を、哺乳動物への投与のために許容される不活性ビヒクルと共に、場合によっては更にアジュバントと共に、含むことを特徴とするワクチン組成物。
