Description
本出願は、参照によりその全体が本明細書で援用される、2004年9月2日出願の米国仮特許出願第60/606,506号明細書、2004年9月9日出願の同第60/608,148号明細書、および2004年9月2日出願の同第60/606,507号明細書の優先権を主張する。本出願は、参照によりその全体が本明細書で援用される、1999年3月1日出願の米国特許出願第09/260,430号明細書、2001年9月21日出願の同第09/957,909号明細書、2002年9月23日出願の同第10/252,120号明細書、および2005年3月29日出願の同第11/091,336号明細書にも関する。
本発明は、外傷性脳損傷、炎症性腸疾患(IBD)、クローン病、潰瘍性結腸炎、関節炎、多発性硬化症、アテローム性動脈硬化症、および敗血症の治療のための化合物および方法を提供する。本発明は、全身照射および局所放射線療法を含む放射線の影響から対象を保護する方法にも関し、かつ電離放射線への偶発的または意図的な曝露の場合の移植、癌療法、および救急医療の分野に関する。
アポE(アポリポタンパク−E)が急性および慢性神経学的疾患における臨床転帰の緩和に重要な役割を果たすことについて多数の収束的な一連の証拠がある。これらの所見は、患者のアポE遺伝子型に基づくが、アポEが神経保護効果をもたらす脳卒中および外傷性脳損傷(TBI)のマウスモデルと一致する(ラスコウィッツ(Laskowitz)ら、1997年、シェング(Sheng)ら、1998年、1999年、リンチ(Lynch )ら、以下を参照)。
本発明は、アポEの残基133−149から成る切断ペプチドであるCOG133の類似体および誘導体を提供する。この切断アポEペプチドは、COG133(LRVRLASHLRKLRKRLL(配列番号1))と呼ばれ、脳虚血または脳炎症の治療または軽減に有用であるとわかった。参照によりその全体が本明細書で援用される、2002年9月23日出願の米国特許出願第10/252,120号明細書。しかし、動物モデルでは、COG133は、TBI直後に投与されると最も効果的である。本発明の化合物は、外傷性脳損傷の神経学的影響の治療および予防に対する広い治療の窓を提供する。治療の窓は、本発明の化合物がTBI後に有効に投与されうる時間を指す。治療の窓を増大させることによって、本発明の化合物はTBI後のより大きな時間間隔で投与され、TBIの神経学的影響を有効に治療または予防し、TBI後の脳炎症もしくは虚血を減少させ、または認知機能を改善することができる。また、本発明の化合物は、外傷性脳損傷の神経学的影響の治療および予防の有効性の増強、大きな治療指数、および長い治療の窓を提供する。
本発明は、脳浮腫、脳虚血およびアルツハイマーのほか、リウマチ疾患、多発性硬化症、CABG手術、アテローム性動脈硬化症、敗血症、結腸炎、および放射線保護を含む中枢神経系(CNS)疾患の治療のための化合物、組成物、および方法を提供する。本明細書に記載された化合物、組成物、および方法は、CNS疾患と関係がある症状を改善し、認知機能を改善する。
理論に拘束されることなく、損傷脳におけるアポE作用に対する少なくとも2つの明確な機序、すなわち、グリア調節および神経保護を裏づける証拠がある。脳は急性および慢性損傷に対する反応の限定されたレパートリーを有する。反応酵素種(ROS)、グルタミン酸、プロテアーゼ、および炎症性サイトカインのその後の放出によるグリア活性化は、アルツハイマー病(AD)および急性脳損傷において確認されるものなど両方の神経変性過程におけるニューロン損傷に関与していると考えられている。出願人最近、アポEが、混合グリア培養およびリポ多糖(LPS)による刺激後の精製ミクログリア培養におけるグリア活性化、酸化窒素(NO)の放出、および炎症性サイトカインの放出をダウンレギュレートすることを明らかにした(ラスコウィッツ(Laskowitz)ら、1997年を参照)。これらのin vitro所見は、マウスアポEタンパク質を発現する対応対照と比べると、LPSを注射したアポE欠乏マウスまたは閉鎖性頭部損傷にさらされた脳において大幅にアップレギュレートされた炎症性遺伝子の発現として生物学的に重要であると思われる(リンチ(Lynch)ら、2001年)。アポEとグリア活性化および炎症性サイトカイン放出との関係は、アポEがミクログリアとして周知の脳特異的マクロファージと密接に関連するマクロファージにおけるシグナル反応を誘発することを示す最近の報告に照らして特に興味深い(ミスラ(Misra)、2001年)。アポE/グリア活性化関係の別の証拠が、多発性硬化症のける障害の進行が患者が発現する特定のアポEイソフォームに依存すると思われる臨床所見によって提供される(チャップマン(Chapman)ら、1999年)。
本発明のペプチドは、当業界で周知の標準方法によって製造されうる。これらのペプチドと関係がある機能的活性を増強する本明細書に開示されたペプチドの変更は、当業者によって容易に達成されうる。例えば、本発明の方法で使用されるペプチドは、化学的に変更され、溶解度、血清安定性等などのパラメータを増強すると同時に、機能的活性を保持するために他の分子にコンジュゲートしうる。具体的には、本発明のペプチドはN末端でアセチル化され、かつ/またはC末端でアミド化され、またはアルブミン、免疫グロブリン、およびそれの断片、トランスフェリン、リポタンパク、リポソーム、α−2−マクログロブリン、およびα−1−糖タンパク質、PEG、およびデキストランを含むが、これらに限定されない血清安定性を増強する分子にコンジュゲート、複合、または融合されうる。かかる分子は米国特許第6,762,169号明細書に記載されており、これは参照によりその全体が本明細書で援用される。
以前に、COG133ペプチドが、N−メチル−D−アスパラギン酸曝露と関係がある神経細胞死およびカルシウム流入を大幅に抑制することが証明された。参照によりその全体が本明細書で援用される、米国特許出願第10/252,120号明細書。したがって、本発明のペプチドは、NMDA興奮毒性と関係がある疾患を治療するための改善された治療組成物の基礎を提供する。例えば、NMDA興奮毒性は、HIV認知症および脳障害と関係がある(ペレス(Perez)ら、2001年、ホーヒー(Haughey)ら、2001年、ドーブル(Doble)、1999年)。
本発明の化合物および治療は自由形態または塩の形態でありうるが、ここで塩は医薬として許容可能である。
かつ具体的には、治療される対象の脳に提供する、当業界で周知である投与の経路の使用
を指す。
実施例1.改善されたペプチド類似体のデザインおよび特性
レトロインベルソペプチド
D−アミノ酸によるL−アミノ酸の置換を含むペプチド類似体をアポE130−150活性の特性の性質を調査するために製造した。出願人はすべてのL−アミノ酸をすべてのD−アミノ酸ペプチドと比較し、アポE130−150のレトロ−インベルソ類似体が活性であったかどうか試験した。レトロ−インベルソ類似体は、D−アミノ酸のみ(全−DアポE150−130)で製造された逆配列(すなわち、アポE150−130)であった。ペスカロロ(Pescarolo)ら(2001年)によって報告された実験とは逆に、出願人は、レトロ−インベルソペプチドが0.01uMを超える濃度で信じられないほど毒性であることを見出した。したがって、酸化窒素のBV−2ミクログリア細胞産生において確認された劇的な軽減は、アッセイにおける細胞がこのレトロ−インベルソペプチドの適用で殺傷されていたためアーチファクトであった。また、アポE130−150の全−Dアミノ酸類似体は、リポ多糖(LPS)処置BV2ミクログリア細胞からの酸化窒素(NO)放出の抑制において活性なしであった。本化合物の利用の可能性は、マクロファージ、および癌治療における骨髄移植の前駆体としての免疫抑制療法の可能性としてレトロインベルソアポE133−149によって殺傷される他の細胞を全滅させたいと思う場合であろう。LPS処置BV2細胞からのNOおよびTNFα放出を抑制する全−Lアミノ酸アポE130−150ペプチドの活性、およびアポE130−150の全D−アミノ酸類似体の活性の欠如は、適切な細胞受容体への全−Lアミノ酸ペプチドの立体特異的結合と一致している。このデータに基づき、レトロ−インベルソ法の別の追求が、免疫抑制パラダイムへの今後の試験において拡大される必要がある。
次いで、出願人は体系的にアポE133−149(COG133)における各々アミン素をアラニンと置換し、次いで各々アポEペプチド類似体の活性を測定した。これらの置換に使用される略語は当業者に周知であり、例えば、アポE133−149ペプチドの149位でのロイシン(L)を意味するL149A(133−LRVRLASHLRKLRKRLL−149)(配列番号1)はアラニン(A)と置換されており、L149A類似体(133−LRVRLASHLRKLRKRLA−149)(配列番号65)を示す。
最大の活性を保存しながらアポE130−150のサイズを最小限に抑える努力において、ペプチドは漸次、アミノ末端およびカルボキシ末端から切断された。カルボキシ末端から開始することにより、アポE130−149はアポE130−150親ペプチドの活性を維持した。対照的に、アポE130−148およびアポE130−147は25μMで活性を示さなかった。アミノ末端から開始することにより、アポE133−149はアポE130−150親ペプチドの活性を維持した。アポE139−149は25μMでも活性を示さなかった。残りの介在ペプチドのうち、アポE134−149が最も活性であったが、アポE130−150よりも効力が2.5倍低かった。アポE135−149およびアポE136−149はアポE130−150よりも効力が5倍および8倍低かったが、アポE137−149およびアポE138−149は25μMでも活性を示さなかった。これらの活性測定値から、アポE133−149(COG133)は、アポE130−150親ペプチドの完全な活性を維持する最も短いアポEペプチドであった。出願人は、新しいペプチド類似体の基礎をアポE133−149(COG133)に置き、アポEペプチドの薬理学的活性に重要である残基の構造活性関係をさらに精緻化した。

炎症抑制アッセイのために、炎症の抑制の公開された細胞ベースのモデルを、COG133またはCOG133の類似体の非存在または存在下にリポ多糖(LPS)で刺激されたBV−2マウスミクログリア細胞を用いて使用されうる(ラスクウィッツ(Laskwitz)ら、2001年)。一般的に、BV−2細胞は完全培地における96ウェルプレートにプレーティングされ、次いで培地は翌日、血清を含まない培地または軽減された血清(1%)培地と交換される。細胞は、LPSの添加によって、または陽性対照としてLPS+さまざまな濃度のCOG133の添加によって刺激される。LPS+さまざまな濃度のCOG133の類似体もウェルを分離するために添加される。さまざまな濃度のCOG133の類似体もウェルを分離し、ペプチドのみ(LPSの非存在下)の活性を制御するために添加される。ペプチドの各々濃度(標準初期濃度は0.1、0.5、1、3、10、および25μM)は、少なくとも6ウェルの細胞に添加され(例えば、6ウェルは0.1μMペプチドになり、6ウェルは0.5μMペプチド等になる)、データは6ウェルすべてから平均化される。24時間のインキュベーション後、馴化培地中のTNFαおよび亜硫酸塩レベルをラスコウィッツ(Laskowitz)ら(2001年)に記載されたELISAおよびグリース(Griess)アッセイで測定される。細胞生死はさらにMTTアッセイで測定される(ラスコウィッツ(Laskowitz)ら2001年)。各々の試験ペプチドのEC50は、t検定および/またはANOVAによって生物学的活性についてCOG133のEC50と比較され、ここでp<0.05が有意とみなされる。
受容体結合は公開方法の修正として試験されうる(ミスラ(Misra)ら、2001年)。出願人の修正は、125I−ストレプトアビジン(ISA、アマシャム(Amersham))で検出されうるビオチン−LRVRLASHLRKLRKRLL−アミドを合成することによってアポEペプチドでビオチン標識を使用することである。また、出願人は、より一貫した多数の計数の特定のペプチドの結合を提供する助けとなる、4℃下に6ウェルディッシュ(ヌンク(Nunc))でウェルあたり250,000個の細胞への結合を使用する。125I−ストレプトアビジンは、ストレプトアビジン‐セイヨウワサビペルオキシダーゼコンジュゲート、またはCDPStar(ロシュ・アプライド・サイエンス(Roche Applied Science)を検出試薬として使用してストレプトアビジン‐アルカリホスファターゼコンジュゲートと置換されうる。
次いで、候補の化合物が、損傷後30分で静脈内投与による外傷性脳損傷のわれわれの閉鎖性頭部損傷モデルにおいてスクリーニングされる。新規化合物が最初にその最大耐量(0.5×MTD)の半分で試験される。最大耐量(MTD)は、マウスの死亡をもたらさない尾静脈注射で投与されるペプチドの用量である。死亡は、呼吸の完全欠如、および10分もしくはそれ以上の間のテイルピンチおよび/またはトーピンチなど外部刺激に対する完全無反応として規定される。動物には1mg/kgのペプチド類似体または非ペプチド類似体の初期用量が投与され、継続的に15分間観察され、次いで15分間隔で2時間、次いで1時間間隔で4時間以上観察される。動物は注射後24時間の時点でも観察される。
TBIの実験モデルは、TBIと関係がある複雑な生理的、行動的、および組織学的変化を評価し、理解する過程において重要な役割を果たす。この相互作用ネットワークをさらに解明するために、TBIの既存の前臨床モデルがヒトTBIの臨床続発症を厳密に模倣するようにデザインされている。TBIを生成する最も広く使用されている実験法の1つでは、堅いインパクタを使用し、機械的エネルギーを発生させ、衝撃の送達中に通常、拘束され続ける動物の無傷頭蓋に衝撃を与える。現在、この種類の損傷をもたらす最も一般的な方法では、鋼のチップインパクタを頭蓋内へ推進し、下記の通り、外傷性脳損傷と呼ばれる機械的エネルギー源として加圧空気が使用される。マウスを含むいくつかの種における使用に適合させ(スミス(Smith)ら、1995年)、空気圧作動デバイスによる変形パラメータ(衝撃の時間、粘度、および深度)を制御する能力、およびリバウンド損傷のリスクの欠如(ライトホール(Lighthall)1988年)により、外傷性脳損傷(TBI)モデルが自由落下誘導錘の重力によって推進されるデバイスよりも優れている(フィーニィ(Feeney)ら、1981年、デイル(Dail)ら、1981年)。
C57Bl/6Jオスマウス(ザ・ジャクソン・ラボラトリー(The Jackson Laboratory)、メイン州(ME))、年齢12−16週および体重24−32gをすべての実験に使用した。麻酔を最初に30%O2/平衡N2中イソフランで誘発する。気管に挿管し、30%O2/平衡N2中1.6%イソフランで肺を機械的に換気する。温度を直腸プローブでモニタリングし、加熱ランプで36.5℃下に維持する。右内頸静脈にシリコーンカテーテルを挿入する。この損傷モデルはラットの閉鎖性頭蓋外傷の既述モデル(マルマロウ(Marmarou)ら、1994年)から、既述通り(リンチ(Lynch)ら、2002年)に適合させた。次いで、挿管動物を成形アクリル鋳造物上に腹臥位に配置する(図6A)。動物をアクリルモールド上に配置することにより動物が安定し、衝撃法の間の移動を阻止し、より均質の損傷を作成する助けとなる。頭皮を切開し、頭蓋を露出させる。凹面の3mm金属円盤を接着剤で(頭蓋に対して並列に凹表面とともに)頭蓋表面に固定する。モデルの開発中、この円盤の配置が頭蓋骨折の発生を軽減する助けとなり、よりびまん性の脳損傷が生じることがわかった。円盤はブレグマに対して尾側正中線に直接配置した(図6B)。さらに、この正中腺損傷は、びまん性軸索損傷のより臨床的に重要なモデルであることが判定された。この損傷は結果として、フルオロジェード(ヒストケム社(HistoChem,Inc.)染色によって測定される、同等の機能的欠損による片側損傷と比べ比較的軽度の両側海馬損傷が生じる。マウスは定位フレームに配置される。定位フレームは動物を安定化し、衝撃時の鉛直変位(3mm)を正確に較正する助けとなるが、イヤーバーははるかに高い頭蓋骨折率(または頭部が衝撃中にイヤーバーの軸の周りを回転する場合には脳幹損傷)により使用すべきではない。ピストンは、アクリルモールドにおける頭蓋を衝撃して最大およそ3mm移動させるために6.8±0.2m/sで放出させる(図6C)。吸引イソフルランは衝撃直後に0.7%に減少される。頭皮をリドカインで浸潤し、縫合で閉鎖した。眼軟膏を保護用に眼に塗った。頭蓋骨折の発生率は、金属円盤が衝撃の拡散を助けるため低く(およそ10%)、イヤーバー固定を行わないことにより全頭部の転位が可能である。陥没頭蓋骨骨折が確認された場合は(発生率およそ10%)、マウスを除外した。動物を自然換気に戻させ、次いで抜管した。代理の生理学的対照群の使用は長期の転帰を含む実験に必要である。動脈カテーテルの配置は生理学的パラメータを測定するために必要であり、他の配置は大腿神経叢を損傷し、運動障害をもたらしうる。他の配置は運動技能(モリスの水迷路における泳ぎを含む)を含む行動試験を解釈不能にする。これらのマウスにおいて、ベースライン、損傷直後、および損傷後15分での動脈血圧、血液ガス、および血糖を回復期間中にモニタリングする。
この正中線閉鎖性頭部損傷は結果として、フルオロジェード染色によって視覚化された海馬のCA3部位における再現可能な病変をもたらす。しかし、片側液体打撃モデルとは異なり、このパラダイムは結果としてよりびまん性の損傷をもたらし、行動の欠陥に対して比例して海馬ニューロン損傷が少ない。出願人は、この正中線損傷が結果として放射線学的脳浮腫および炎症性サイトカインのアップレギュレーションをもたらすことも明らかにした。これは、これがびまん性軸索損傷の臨床的に重要なモデルであるという主張を裏づける。
このパラダイムは軽度‐中等度閉鎖性頭部損傷としてデザインされた。マウスは最初に、下記の通り損傷後1−5日目の回転棒および神経学的重篤度スコアによって評価された運動および小脳障害を有する。頭部損傷の生存者において一般的なより適切な長期の神経学的障害をモデル化するために、損傷後21‐25日目にモリスの水迷路に記憶が評価される。具体的には、これにより90秒間隔以内で直径105cmのプールにおける直径7.5cmの隠されたプラットフォームを見つけ出す能力が試験される。
外傷性脳損傷(TBI)、ペプチドの静脈内投与、および行動性能は上記および発表(リンチ(Lynch)ら、2002年、リンチ(Lynch)ら、提出済み)通りに実行される。手短に言えば、回転棒でのベースライン性能をマウスが挿管される前に確認し、頭皮上の皮膚を反映し、空気インパクタからの制御された皮質衝撃を正中線に与える。皮膚は外科クリップで閉鎖され、動物は麻酔から覚めるまでつねに、さらにその後の4時間にわたって1時間ごとにモニタリングされる。衝撃後30分の時点で、生理食塩水賦形剤または試験品(COG133、COG133の類似体、またはCOG133の非ペプチド類似体)が尾静脈注射によって投与される。回転棒での性能は衝撃後24時間、次いで5日間毎日試験され、データは記載通り示されている。マウス12匹の群を0.5×MTDでの各々の化合物のために使用される。陰性対照群には生理食塩水賦形剤のみが投与され、陽性対照群には尾静脈注射によって100ul量の生理食塩水中4mg/kgのCOG133が投与される。新規ペプチドおよび非ペプチド類似体も尾静脈注射によって投与される。
われわれのマウスTBIモデルにおいて外傷性脳損傷後60、90、および180分の時点で類似体をさらに試験することができる。このより厳密な遅延試験において、新規化合物の有効性を、上記の通り再現可能な外傷性脳損傷(TBI)を与える標準閉鎖性頭部損傷後60分の時点で尾静脈注射によって投与されるCOG133と比較されうる。化合物が、反復測定ANOVAで分析される回転棒性能によって判定される通りTBI遅延後60分の時点でCOG133より大幅に優れていることが判明する場合は、これはTBI遅延後90分で試験される。化合物が、反復測定ANOVAで分析される回転棒性能によって判定される通りTBI遅延後90分の時点でCOG133より大幅に優れていることが判明する場合は、これはTBI遅延後180分で試験される。
認知および学習異常は臨床集団におけるTBIの一般的な長期の続発症である。われわれのモデルの感度および臨床上の重要性を最大限にするために、神経学的機能における学習、保持、および行動障害が評価されうる。このために、モリス水迷路タスクにおける性能の変化が試験されうる(モリス(Morris)、1984年)。このタスクでは自然に泳いでいる齧歯類の平均を取り、視空間合図を作業および基準記憶へ組込む動物の能力を測定する。このタスクでの性能はヒト臨床集団において見られる神経精神障害の相関でありうる。理論上、マウスは水泳によるタスクから任意に、またはプールの全体を通じて非系統的な通路で逃げることもできるが、しかし、作業記憶が無傷である場合は、マウスは試験チャンバーの固定物から遠位の合図を使用し、プラットフォームの相対位置を学習する。したがって、プラットフォームを見出すタイムレイテンシーは実行の関数として減少し、学習能力の指数として使用されうる。
一部のペプチドは、トリプシン様プロテアーゼによって開裂されうる。ペプチドの開裂は、液体クロマトグラフィー−エレクトロスプレイイオン化質量分析(LC/MS)法においてトリプシンおよび脳ホモジネートを使用して測定されうる。手短に言えば、ビードアガロース(パース(Pierce))に結合したトリプシンを0.1M炭酸アンモニウム、pH8.0で2回洗浄し、その後にカルシウム、マグネシウムを含まないリン酸緩衝生理食塩水、pH8.0(PBS−8)で2回洗浄する。20ug/mlのペプチド基質を含有するPBS−8中に樹脂を再懸濁し、0、0.1、1、5、および20時間37℃下に消化させる。3通りの試料を各々時点で評価する。試料を一時的に遠心分離し、樹脂を含まない上清をATI(アセトニトリル、TFA、内部標準)で抽出し、処理して以下の条件でLC/MS定量にかける。

各々の時点で評価する。各々実験において、無傷、非分解ペプチドのモル、および主要な
代謝産物(無傷、非分解ペプチドのモル量の少なくとも20%である断片)を各時点で測
定する。均質化脳も使用し、ペプチドの脳への取込みをLC/MS法を使用して測定する
ことができる。
図4は、4mg/kgまたは1mg/kgのCOG432、(COG432:Ac−ASHLRKLAibKRLL(配列番号6))または対照(生理食塩水)によるTBI後の回転棒試験の結果を示す。縦軸は、回転棒性能を示す(100%はTBI前の回転棒性能)。COG432、4mg/kg、または生理食塩水による処置を外傷性脳損傷後2時間の時点で開始し、動物をTBI後1日目に開始して次の5日間毎日試験した。TBI後5日までに、4mg/kgのCOG432ペプチドで処置した動物は回転棒試験によって測定されるように約80%の機能まで回復していた。生理食塩水賦形剤のみで処置した対照マウスが回復した機能は50%未満であった。
われわれは、L140およびR145の位置で2つのアミノイソブチル酸(Aib)置換を含有する類似体であるCOG1410を合成した。Aibは、ペプチドに存在するアミノ酸の種類に関係なくらせん構造を形成することが証明されている非天然アミノ酸である(マーシャル(Marshall)ら、1990年)。また、Aibは、他のアミノ酸に対して結合時に配座エントロピー喪失の軽減を示すため結合親和性を改善する(ラトナパルキー(Ratnaparkhi)ら、2000年)。
図7に示されているように、COG1410は、酸化窒素(9A)およびTNFa(9B)放出の抑制のわれわれの細胞ベースのアッセイにおいてCOG133よりも大幅に強力であった。また、in vivoでの予備スクリーニングは、COG1410がTBI後120分で投与すると神経保護性であることを示したが、これはこの時点で神経保護活性を欠いているCOG133とは異なる(図8)。
予備用量反応試験は、COG1410の最小有効量(MED、賦形剤処置対照と比べ性能における統計的に有意な改善を示す最低用量)は、COG133のものとほぼ同じであり、それぞれ、0.3mg/kg対0.4mg/kgであった。しかし、COG1410の最大耐量(MTD、24時間でマウスの死亡をもたらさない最高用量)は、COG133の1.4mg/kgのMTDに対して8mg/kgであった。COG1410の治療指数(TI、MTDのMEDに対する比)は26であり、COG133の3.5のTIよりも大幅に良好である。TIが高いほど、薬物は安全であるとみなされる。COG1410のこの高いTIは、有利かつ所望の保護硬化をもたらすよりも毒性反応を引き起こすためにはるかに高い用量を取ることを示す。
考慮する多くの薬物動態パラメータのうち、血漿半減期およびタンパク質分解に対する耐性が、液体クロマトグラフィー−エレクトロスプレイイオン化質量分析(LC/MS)法で正確に測定されうるペプチドの2つの特性である。LCMS−LLCを用いて、われわれは、ペプチド量および血液血漿およびマウス脳抽出物におけるペプチド断片を測定するLC/MS法を開発した。
COG1410によるこれらのデータは、増大した安定性、増強された効力およびBBB透過性、減少した毒性、増大した治療指数、およびTBIの治療に対する拡大した治療の窓を有する化合物を発現する実現可能性を示す。分解に対する増大した耐性など、さらに優れた医薬特性を有する追加の類似体が、上記のガイドラインを使用して制限された数の誘導体の合成によって達成可能である。
COG1410は、多くの基準によってCOG133に対して大幅な改善である。われわれの予備データは、アルファらせん構造がきわめて重要であり、このらせん構造の強化がCOG1410をもたらすことを示した。らせん構造を促進するAibのようなアミノ酸を含めるわれわれの初期の方法は、潜在的にアラニンスキャニング作業から可能である2つの新しい位置に拡大される(実施例1を参照)。COG1410のらせん性質へのさらなる強化は、ペプチドの生物活性を直接与えるとは思われないらせんの表面でアミノ酸の側鎖の誘導体化によって達成されうる。これらの誘導体の全体的な性質は、その柔軟性を抑制し、らせん構造に残る傾向を増大させる選択アミノ酸間のオレフィン架橋を含めるべきである。
閉環メタセシス(RCM)法を使用することにより、オレフィン側鎖が組込まれ、配列:ASHLRKLRKRLLにおける選択残基で側鎖と側鎖を共有結合しうる。COG1410は、らせんの一面上の荷電残基のすべておよびらせんの対向面上の疎水性残基のすべてを有する両親媒性アルファらせんを形成するように思われる。大部分の荷電残基のアラニン置換は大幅に抗炎症活性を軽減したため(表1)、荷電残基は側鎖結合法の候補ではない(Aib置換が生物活性を増強したR145を除く)。これは疎水性残基の大部分を側鎖結合法による変更に利用可能なままにする。したがって、以下の架橋結合分子が合成されうるが、ここでXおよびZで示されたアミノ酸は、架橋結合オレフィン架橋に結合される側鎖を含有するアミノ酸でありうる。すなわち、ASHXRKLZKRLL(配列番号60)、XSHLRKLZKRLL(配列番号61)、ASXLRKLRKZLL(配列番号62)、およびASHXRKLRKRZL(配列番号63)。
LC/MSによるわれわれの予備データにより、COG1410の潜在的に重要な代謝産物としてアセチル−AS−Aib−LRKL−Aib−KRが同定された。この断片は、トリプシン様プロテアーゼによる細胞内タンパク質分解開裂および/またはカルボキシペプチダーゼによる細胞外タンパク質分解開裂によって生成されうる。エンドプロテアーゼの可能性に対処するために、オルニチン、ニトロアルギニン、ホモアルギニン、またはジメチル‐アルギニンをアルギニンに対して147の位置で自動ペプチド合成器で標準Fmoc化学によって置換するCOG1410の誘導体が合成されうる。エクソプロテアーゼの可能性に対処するために、バリン、メチオニン、またはイソロイシンをロイシンに対して149の位置で置換するCOG1410の誘導体が合成されうる。
多発性硬化症(MS)の信頼できる動物モデルの1つとして、実験自己免疫脳脊髄炎(EAE)は中枢神経系(CNS)において重篤な脱髄を引き起こす炎症性疾患である(ペンダー(Pender)とウォルフェ(Wolfe)2002年)。EAEおよびMSは、ミクログリア活性化、および主にTリンパ球およびマクロファージから成る炎症性細胞のCNSへの顕著な浸潤を含む共通の組織学的特徴を共有する(へーバー・カッツ(Heber‐Katz)1993年)(へマー(Hemmer)、アルケロス(Archelos)ら、2002年)。これらの活性化エフェクタ細胞は、TNFα、IL−1β、IFNγ、およびリンホトキシンなど一連の前炎症性サイトカインを放出する。次いで、これらの因子はさらに浸潤細胞の蓄積を促進するが、これらは炎症、および組織損傷と関係がある(エング(Eng)、ジルニカー(Ghirnikar)、1996年)(ベンベニスト(Benveniste)1997年)(ウィーマン(Wiemann)、ファン(Van)ら、1998年)。
マウス
メスC57BL/6Jマウス(12週齢)をジャクソン・ラボラトリーズ(Jackson Laboratories)から購入し、デューク(Duke)大学実験動物施設に収容した。動物ケアおよび実験法はデューク大学動物ケアおよび使用委員会によって承認された規則に一致した。
ミエリンオリゴデンドロサイト糖タンパク質(MOG)ペプチドは、マウスMOGタンパク質の残基35‐55(MOG35‐55、MEVGWYRSPFSRVVHLYRNGK(配列番号64))由来であり、アポE類似ペプチド(COG133)は、アセチル−LRVRLASHLRKLRKRLL−アミドの配列を有するヒトアポリポタンパク‐Eの残基133〜149由来である。アポE逆ペプチド(COG−149で示される)は、ここではCOG133のスクランブル対照として処置される。アンテナペディア結合ペプチド(COG134、akaCOG4502として示される)は、アンテナぺディアプレフィックスペプチドをCOG133と結合する。ペプチドはすべて、RP‐HPLCによりメリフィールド固相化学および精製によりFMOC試薬を使用してUNCでのペプチド合成施設によって合成された。以下の試薬をシグマ(Sigma)から購入した。すなわち、百日咳毒素、LPS、およびIFN‐γ。TNFαおよびIL‐6の定量ELISAキットをバイオソース(Biosource)から得た。
EAEをファインスタイン(Feinstein)らの方法に従ってマウスにおいて誘発した(ファインスタイン(Feinstein)、ガレア(Galea)ら、2002年)1)2回のs.c.注射、5mg/mlのヒト型結核菌(Mycobacterium tuberculosis)H37RA(ジフコ(Difco)、St.Louis、ミズーリ州(MO))を含有する同等量のCFA中に乳化したPBS 0.1ml中MOG35‐55ペプチド300μgを後足の各々の側面に0日目に投与した。2)百日咳毒素(PBS 0.1ml中マウス1匹につき200ng)を最初のMOG注射の直後および48時間後にi.p.投与した。3)同一の乳剤、投与経路、および位置による追加免疫を7日目に投与した。
最初のMOG注射(0日)後、マウスを無作為に1群マウス15匹の3群に分け、対照群、アポE133‐149(COG133)処置群、およびアポE逆ペプチド処置群とした。生理食塩水中COG133、生理食塩水中逆ペプチド、または生理食塩水を6、7、8、10、12、14、16、18、20、および22日に100μl量中1mg/kgの用量で静脈内注射し、各々の処置群に合計10用量を投与した。
脳炎誘発性チャレンジの後、マウスを毎日モニタリングし、神経学的障害を以下の通り臨床スコア(C.S.)によって評価した。すなわち、0、EAEの臨床徴候なし、1、だらりとした尾、2、たるんだ尾と異常な歩き方(後肢の運動失調および/または麻痺)、3、重度の後肢麻痺(hind bodyによる)、4、完全麻痺、5、瀕死または死亡。
このために、30dpi(免疫後の日数)で臨床スコアが0のマウス3匹および臨床スコアが4のマウス3匹を腹膜マクロファージ培養の調製用に使用した。PBSを含有するヘパリン(10U/ml)3mlによる腹膜洗浄後にマクロファージを回収し、次いで96ウェルマイクロプレートに1×105細胞/ウェルの密度で10%ウシ胎仔血清、2mM L‐グルタミン血清、1%HEPES、および100U/mlペニシリン、および100μg/mlストレプトマイシンを含有するダルベッコMEM中、5%CO2による加湿インキュベータ中37℃下に接種した。24時間後、血清含有培地を除去し、血清を含まない培地で細胞を1回洗浄した。免疫促進剤に対するマクロファージの分化反応を検査するために、臨床スコア0または4のマウスから得られたマクロファージを連続濃度のIFN‐γ/LPSまたはMOG35‐55ペプチドに曝露した。IFN‐γ/LPSまたはMOG誘発サイトカイン産生に対するCOG133またはCOG134の効果を調査するために、これらペプチドの指定濃度をIFN‐γ/LPSまたはMOG治療前30分に適用した。培地を45時間または72時間の時点で回収し、以下の詳述通り、TNF−γおよびIL−6または酸化窒素(NO)についてELISAによって分析した。
酸化窒素放出の安定した最終産物として、50μl培地試料をシーバース(Sievers)280NOA分析器(ボルダー(Boulder)、コロラド州(CO)、米国(USA))ヘ注入することによって馴化培地中の亜硫酸塩レベルを測定した。総タンパク質(μg/ウェル)を標準としてBSAによるメーカーの指示に従いBCA法(パース(Pierce)、Rockford、イリノイ州(IL)、米国(USA))を使用して測定した。BCA値をOD562でモレキュラー・デバイセス・サーモマックス・マイクロプレート・リーダー(Molecular Devices Thermomax Microplate Reader(メンロ・パーク(Menlo Park)、カリフォルニア州(CA)、米国(USA))を使用して測定した。酸化窒素レベルはμM NO2−/mgタンパク質で表される。
サイトカインアッセイのために、TNF−γ/LPSまたはMOG処置後45時間に回収し、選択ペアのモノクローナル抗体を使用する定量ELISAを(メーカーのバイオソース(Biosource)による推奨通り)実行し、サイトカイン、TNF−αおよびIL−6を定量した。TNF−αELISAのために、96ウェルELISAプレートを捕捉抗体としてのTNF−αに対するウサギ抗マウスポリクローナルで18時間、2−8℃でプレコートし、次いで2時間、室温で阻止した。ウェルを試料またはマウスTNF−α標準でインキュベートした。室温下に2時間のインキュベーション後、ウェルを洗浄し、その後にハムスター抗マウスTNF−αビオチン化二次抗体およびセイヨウワサビペルオキシダーゼコンジュゲートを添加した。次いで、テトラメチルベンジジン(TMB)溶液を添加し、ペルオキシダーゼ触媒変色を2N H2SO4による酸性化によって停止した。プレートを波長450nmでスキャンニングし、吸光度を測定した。結果は平均濃度(pg/ml)±SEMで表されている。
マウスを麻酔し、出血させ、PBS 25mlおよび緩衝PBS中4%パラホルムアルデヒド25mlで灌流した。脳および脊髄を臨床スコアが0のマウス3匹および臨床スコアが4の別のマウス3匹から免疫後30日に切除した。組織をさらに24時間4%パラホルムアルデヒド中に後固定し、次いで0.1%アジ化ナトリウムとともに1×PBS中に保存した。固定組織をパラフィン中に埋設し、5μM厚切片を脳、脳幹、および脊髄の3つの異なるレベル(頸部、胸部、および腰部)から切断した。切片を炎症の証拠のためにヘマトキシリン/エオシン、および(脱髄ために)ルクソールファーストブルーで染色した。炎症の重篤度を以下の基準を使用して評価した。すなわち、0、炎症なし、1、血管周囲および髄膜でのみの細胞浸潤、2、実質でのみの軽度浸潤(1−10/切片)、3実質での中等度浸潤(11‐100/切片)、4、実質での顕著な細胞浸潤(>100切片)。
回転棒データ、サイトカイン、および亜硫酸塩濃度をANOVAの後、ダネットの比較によって分析した。疾患スコアをマン・ホイットニー検定によって分析した。
COG133は活性EAEの発現を軽減する
COG133の推定の保護効果を検査するために、1mg/kgCOG133、もしく
は逆ペプチド、または生理食塩水の10用量の静脈内投与を尾静脈注射によって行い、6
日目に開始して22日目に終了した。臨床徴候を毎日、回転棒での行動を1日おきに検査
した。COG133処置群の平均最大臨床スコアは、生理食塩水またはスクランブルペプ
チド処置群のものより大幅に低かった(P<0.05)(図10)。この結果は、COG
133で処置した動物が疾患の重篤な臨床徴候をあまり示さないように思われるというわ
れわれの一般的な所見に対応する。
EAEの病因におけるTNF−αおよびIL−6およびフリーラジカル、酸化窒素、またはNOなどサイトカインの役割を検査するために、マクロファージを臨床重篤度が異なる(C.S.0および4)マウスから採集し、次いでさまざまな免疫促進剤、すなわち、LPSおよびIFN−γまたはMOGペプチドでチャレンジした。MOGペプチドはLPS+IFN−γと比べ比較的低刺激性の免疫促進剤であるが、NOの強固な産生が依然として濃度依存的にEAEマウスからのマクロファージにおいて確認された(図11A)。C.S.が4のマウスの重度障害マウスからの高濃度のMOG(20μg/ml、7.75μMに相当)処置マクロファージは、C.S.が0の非障害マウスからのMOG処置マクロファージからのものよりはるかに高い酸化窒素の放出を誘発し、NOの産生が有害な疾患の発現の大きな原因になりうることを示している。
われわれは以前、COG133が、ミクログリア(脳のマクロファージ)活性化(ラスコウィッツ(Laskowitz)ら、2001年)を抑制し、培養細胞における細胞内シグナル伝達系を開始する(ミスラ(Misra)ら、2001年)その能力における無傷アポEタンパク質の生物活性を保持することを示した。アポEがその免疫調節効果を誘発する機序を調査するために、われわれは、C57BL6マウスにおけるin vivoでの全身炎症性反応を抑制するCOG133の能力を検査した。LPSに対する炎症性反応は、末梢循環および脳における2つの明確なプロ炎症性サイトカイン、腫瘍壊死因子アルファ(TNFa)およびインターロイキン−6(IL−6)の一時的分泌および発現プロフィールを測定することによってモニタリングされた。マウスに対し、尾静脈を通じてLPS注射し、血清試料を指示時点で得るとともに、TNFaおよびIL−6のレベルをELISAで測定した。LPSと同時投与すると、COG133は1時間で血清TNFaレベルを有意に軽減し、かつ血清IL−6レベルを1および3時間で軽減した(図15)。各群において注射後24時間の時点で測定可能なTNFaまたはIL−6タンパク質はなかった。これらの結果は、COG133が全動物モデルにおけるLPS誘発炎症を抑制することができ、TNFaおよびIL6放出の抑制において特に有効であるように思われることを示す(リンチ(Lynch)ら、2003年)。
本試験において、われわれは、アポE類似ペプチドであるCOG133が、ヒトにおける多発性硬化症の特徴の大部分を模倣する実験的に誘発されたアレルギー性脳脊髄炎(EAE)の行動転帰を改善する顕著な能力を示すことを明らかにする。MSは、広範囲な炎症、限局性脱髄、およびさまざまな程度の軸索喪失によって特徴づけられる中枢神経系の慢性炎症性疾患である(コルネック(Kornek)、ストルヒ(Storch)ら、2000年)。マクロファージおよびTリンパ球の大規模な浸潤が、全能にわたって、特に脊髄における炎症の顕著な徴候として確認されうる(レイン(Raine)1994年)。浸潤および活性化マクロファージおよびTリンパ球によるプロ炎症性サイトカインの放出は、EAEにおけるCNS病因の重要なメディエーターである。実際に、大量のTNF−α、IFN−α、およびIL−1βが脱髄斑に存在する(ブロスナン(Brosnan)、カネラ(Cannella)ら、1995年)。また、浸潤免疫反応性細胞は毒性サイトカインを放出することによって直接、または間接に星状膠細胞およびミクログリアを活性化するが、これらはCNSにおけるマクロファージと考えられ、したがって、炎症性反応の二次カスケードをもたらす(プリネアス(Prineas)、クォン(Kwon)ら、2001年)。次いで、脊髄における炎症は増悪中に顕著となり、運動の顕著な障害をもたらし、場合により、マウス対象の死亡をもたらす。したがって、抗炎症性介入は、疾患の有害な過程を防ぐ有望な方法として相当な関心を引出した(リークマン(Rieckmann)とマウラー(Maurer)2002年)。
自己免疫モデル、EAE、およびEANにおけるアポE欠乏の分離in vivo試験は、1)炎症性反応の規模、および2)慢性、Th1介在、自己免疫反応において持続される組織破壊の量を阻害することによって、アポEが大きな疾患重篤度および死亡からマウスを保護することを示した。したがって、アポE類似ペプチドは、関節リウマチのコラーゲン誘発モデルにおける有効性について試験されうる。
パンヌス組織は、関節リウマチを有する患者およびCIAを有するマウスの骨関節内で過剰増殖を受けた滑膜細胞から成る。この組織は、そのサイトカイン、MMP、および酸化窒素の産生のため軟骨および骨の破壊の主要な原因である(ピリンジャー(Pillinger)ら、2000年)。疾患過程に生理学的に関連する細胞を使用してRAに対する候補の療法の試験は、その処置がin vivoで有する潜在的な利点の判定の助けとなる。
アポE類似物はTBIのマウスモデルにおいて神経保護性である
TBI後30分に、マウスを406μg/kgCOG133または生理食塩水賦形剤で処置した。損傷後24時間、生理食塩水注射動物は回転棒試験によって測定される通り運動神経および平衡の顕著な障害を示した(図15A)。この運動障害は体重喪失と関係があった(データは示さず)。COG133処置マウスは、それらの生理食塩水処置対照よりも毎日、有意に優れて機能した、p<0.01(リンチ(Lynch)ら、2001年)。この回復の機能は、生理食塩水処置マウスと比べCOG133処置マウスの海馬におけるニューロン死の減少と相関した、p<0.05、データは示さず。これらの結果は、COG133の静脈内投与が回転棒での性能を改善し、頭部外傷と関係があるニューロン死を予防したことを強く示す。
先の研究では出願人は、COG133がBV2マウスミクログリア細胞におけるLPS刺激亜硫酸塩分泌の用量依存抑制を示したことを明らかにした。図16に示されているように、50%抑制がおよそ3μM COG133の用量で達成された。出願人の予備研究のために、2つの最も一般的なPTD、TATおよびペネトランを使用した。図17に示されているように、TAT−COG133コンジュゲートは、25−1000nMの範囲では効果的ではなかった。1000nMよりも高い容量は結果として細胞の90%以上の死をもたらした(データは示さず)。図18は、ぺネトラチン−COG133コンジュゲートが有意により強力であったことを示す。ぺネトラチン−COG133(COG4502)による50パーセント抑制がおよそ30nMで生じたが、COG133のみでは本実験で使用された用量範囲内では50%抑制は達成されなかった。50%の抑制が3μMで達成された図16におけるデータと図18におけるデータの比較は、COG133の有効性がぺネトラチンにコンジュゲートすると(COG4502)約100倍に増大したことを示す。これらの予備データは、TAT−COG133は細胞傷害性であったが、ぺネトラチン−COG133(COG4502)はBV2ミトコンドリアにおけるCOG133の有効性を有意に増大したことを示す。
マウスにはCOG133、TAT−COG133、またはペネトラチン−COG133(COG4502)をTBI後のさまざまな時点で投与し、回転棒タスクでの性能によって効果を分析した。図20に示された結果は、COG133のペネトラチンとのコンジュゲートがCOG133の治療の窓を増大せさうるが、TAT−COG133による処置は効果的ではなく、生理食塩水と同様であったことを示す(データは示さず)。これらのデータは、BV2細胞におけるCOGコンジュゲートの活性をスクリーニングし、それによってCOG活性の一次スクリーンとしてのBV2細胞の有効性を確立することにより得られた結果と一致する。
実験デザイン:独立変数がPTDであり、依存変数が培養培地に存在する酸化窒素のレベルとなる無作為化デザインが使用される。
図23に示されたデータは、SynB3−COG133コンジュゲートが、外傷性脳損傷後2時間に1回尾静脈注射によって投与するとマウスにおける頭部外傷からの転帰を有意に改善する(回転棒レイテンシーによって測定)ことを示す。
アポEホロ−タンパク質由来のペプチドによりホロ−アポE−タンパク質と同じ抗炎症活性が得られるか試験するために、われわれは最初、COG133(aka.アポE133−149)またはアポE130−149の添加が結果としてリポ多糖(LPS)で刺激したBV2ミクログリア細胞系からのTNFα放出の用量依存阻害をもたらすことを示した(ラスコウィッツ(Laskowitz)ら、2001年)。対照的に、スクランブルペプチドで処置した細胞は、TNFα放出を阻害しなかった。同様に、COG133およびアポE130−149はLPS処置BV2細胞からの酸化窒素放出を有意に軽減したが、スクランブルペプチド処置は酸化窒素放出を阻害しなかった。これらのデータは、マクロファージ細胞のLPS誘発死激後のサイトカイン(TNFα)およびフリーラジカル(NO)の放出がCOG133の存在下に有意に軽減され、COG133が抗炎症剤として機能しうることを示しているのを明らかにした。参照によりその全体が本明細書で援用される、米国特許第10/252,120号明細書を参照。
アポEの抗炎症効果がTBIおよび放射線療法を受ける対象の保護に有用であることを確認するために、われわれは、マウスアポEタンパク質を発現する野生型C57B1/6マウスにおけるTBIの7Gyおよび8Gyの効果をアポEノックアウトマウス(ジャクソン・ラボ社(Jackson Labs)、Bar Harbor、メイン州(ME))におけるTBIの7Gyおよび8Gyの効果と比較した。マウス10匹の群を0の時点で照射し、その生存を翌36日にわたって追跡した。図22に示されているように、7Gyへ曝露された野生型マウスのうち1匹も30日の時間的経過中に死亡しなかったが、アポEノックアウトマウス10匹のうち2匹が照射後13日で死亡し(20%死亡/80%生存)、30日の時間的経過中にさらなる死亡はなかった。野生型マウス10匹の群を8GyのTBIへ曝露した場合、4匹が照射後21日目に死亡し、さらに2匹が23日目に死亡したが、36日までにそれ以上の死亡はなかった。アポEノックアウトマウス10匹の群を8GyのTBIへ曝露した場合、6匹が照射後10日で死亡し、さらに2匹が11日目に死亡し、最後の2匹は12日目に死亡した。
TBIが炎症を刺激し、アポE類似物であるCOG133が丸ごとの動物における炎症を調節しうるという情報を組合わせることにより、われわれは、全身照射へ曝露された野生型マウスにおける生存を改善するCOG133の能力を試験した。
全体としては、マウスをワイヤハング試験で試験し、マウスがその肢の強さを有することを示す。マウスが強さを有する場合は、次いで、運動強度および調整の行動性能の統合試験である回転棒で試験される。ストレスをかける前にこれらの試験にパスしない動物は、われわれのその後の研究で使用されない。TBI+COG133を受けた動物がTBIのみを受けたものより有意に優れて機能する場合は、これはCOG133が死亡を改善するだけではなく、病的状態も改善することを示す。
アポEタンパク質の抗炎症性特性に基づき、われわれは、299アミノ酸、アポEホロタンパク質の受容体結合部位に位置したアミノ酸残基(133−149)由来のペプチドであるCOG133を開発した(ラスコウィッツ(Laskowitz)DTら、2001年)。われわれは、培養マクロファージ(ラスコウィッツ(Laskowitz)DTら、2001年)およびC57B1/6マウス(リンチ(Lynch)J.R.ら、2003年)を使用することにより、COG133がアポEタンパク質の抗炎症性特性を保持することを明らかにした。炎症をLPS注射±COG133によってナイーブマウスにおいて誘発し、注射後0、1、3、および24時間の時点で血清を回収した(図24AおよびB)。サイトカインELISAキット(パース(Pierce)を使用することによって、COG133処置動物は、生理食塩水対照と比べ、有意に低い血清TNF−αおよび有意に低い血清IL−6を有した(リンチ(Lynch)J.R.ら、2003年)。われわれもリンチ(Lynch)らにおいて、TNF−αおよびIL−6の脳レベルが、定量的RT−PCRによってタンパク質レベルまたはmRNAレベルのいずれかの測定によって、生理食塩水対照と比べCOG133処置動物において有意に低いことを報告した。新鮮ヒト血液±COG133のLPS刺激も、COG133が生理食塩水処置対照と比べ、酸化窒素およびTNF−αレベルを有意に軽減したことを示した。これらのデータは、COG133がin vitro、in vivo、およびエクスビボで炎症を軽減することを示し、これは、COG133がヒト疾患における炎症を効果的に軽減しうるというわれわれの考えを裏づける。
マウスにおけるMOG誘発実験アレルギー性脳脊髄炎(EAE)は、MSおよびEAEの臨床的および組織病理学的特徴が多くの基本的な点で同様とみなされるため多発性硬化症(MS)と関係がある炎症の最もしばしば使用されるマウスモデルである。これらには、CNSの全体を通じて多発性病変の臨床症状、発生率、脱髄性斑、血管周囲炎症、およびTNF−α、IL−6、およびNOのような炎症原を放出する侵入炎症性細胞を関与が含まれる(ランゾホフ(Ransohoff)R.M.、1999年)。したがって、このモデルは、COG133がその抗炎症性特性によりMSの障害を改善しうるというわれわれ仮説を試験するために選択された。
COG133をテンプレートとして使用することにより、われわれは、増強された所望の医薬特性を有する新しい類似体を生成し続けている。図26に示されているように、COG1410は、非天然アミノ酸を含有し、COG133親化合物よりも優れたLPS−刺激、BV2ミクログリア細胞におけるNO放出を抑制することができる類似体である。同様に、COG4502は、エキストラアミノ酸のプレフィックスに引き続きCOG133配列を含有し、COG133親よりもはるかに優れてこの系でNO放出を抑制することができる別の類似体である。より生理的な系におけるCOG4502の有用性をさらに検証するために、われわれは、マウス腹膜マクロファージを単離し、それらをCOG4502の存在または非存在下にLPSで刺激した。図27に示されているように、COG4502はTNFaの用量依存阻害およびIL6放出の用量依存阻害をもたらす。これらの結果は、COG化合物が、細胞系、腹膜マクロファージ、および丸ごとの動物において有力な抗炎症性分子であることを強く示す(上記で詳述)。
炎症はクローン病および潰瘍性結腸炎の進行および症状の基礎にあり、場合によって、これは狭窄形成、瘻、閉塞、およびせん孔の合併症に進行しうる。IBDにおけるこの炎症性反応の一部が、一般に疾患活性の悪化と結合しているNO放出の刺激である。NOは基質としてアルギニンからのNO合成(NOS)によって生成され、この酵素の誘導性形態(iNOS)はIBDに関与する主要な担い手である。しかし、アルギニンは、ポリアミンを生成する別の経路によっても使用される。これは、制限アルギニン基質の2つの経路間での競合を誘発する。
アルギナーゼIおよびiNOSが両方とも結腸炎組織において多量に発現され、L−Argは完全に代謝されたため、われわれは、飲料水中1%L−Argの投与の効果を調査した。野生型(WT)マウスにおいて、C.ローデンティウム結腸炎は、感染後9日も開始した高レベルの死亡を誘発した(図31A)。L−Argで処置したWT動物において、死亡は、感染後12および14日にそれぞれ、水のみが与えられたマウスと比べ42%および62%阻害された(図31A)。Cox回帰分析によって、L−Argで処置したWTマウスは、水のみが与えられたマウスと比べ死亡の31%の危険のみを示した(p<0.0009)。iNOS−/−マウスにおいて、L−Argの有無に関係なく、死亡は確認されなかった(図31A)。WTマウスにおいて、L−Arg処置は体重喪失を軽減し、L−Argが与えられたiNOS欠乏マウスはさらに改善を示し、実際に感染の存在下に体重を増加した(図31B)。ここで留意すべきは、依然として生きている動物の体重のみが含まれたため、WTマウスの体重喪失が過小評価であることである。結腸重量は、C.ローデンティウム感染WTマウスでは2倍より多く大幅に増大し(図31C)、WT−L−ArgおよびiNOS−/−マウスでは、それぞれ、28%、および38%減少した。iNOS欠失およびL−Arg投与の追加の効果が認められ、結腸重量が68%減少した。iNOS欠失でもL−Arg処置でも非感染対照マウスにおける結腸重量に対する影響はなかった。重要なことに、これらの変化は結腸組織学における変化によって密接に対比された(図32)。
アルギナーゼおよびポリアミン形成の有利な効果をさらに明らかにするために、マウスに対し、アルギナーゼおよびODCの阻害剤である、S−(2−ボロノエチル)−L−システイン(BEC)または−ジフロロメチルオリニチン(DFMO)をそれぞれ、飲料水中で与えた。非感染対照マウスにおいて、BBCまたはDFMOは効果を示さなかった(表I)。しかし、BECまたはDFMOで処置したC.ローデンティウム感染マウスにおける有意な生存の喪失が認められた(表9)。実際に、実験は、感染後10日に、この時点での死亡および重篤な疾患のため早期に終了されなければならなかった。C.ローデンティウム−BECおよびC.ローデンティウム−DFMO群の結腸は、C.ローデンティウム−水群のものよりも重量および組織学的損傷の大幅な増大を示した(表9)。
このモデルは、結腸炎のマウスモデルとして一般的に受け入られており、単にDSSを飲料水に添加するだけのため、使用するのにきわめて実用的であるため選択された(モチュー(Moteau)Oら、2000年、ウィリアムズ(Williams)KLら、2001年、アンドレス(Andres)PGら、2000年、マーラー(Mahler)M.ら、1998年、テッサー(Tesser)TGら、1998年)。初期実験では、われわれは、2.5%〜5%の文献(モチュー(Moteau)Oら、2000年、ウィリアムズ(Williams)KLら、2001年、アンドレス(Andres)PGら、2000年、マーラー(Mahler)M.ら、1998年、テッサー(Tesser)TGら、1998年)からの報告された範囲でさまざまな用量を試験し、われわれが4%DSSで最も信頼できる反応を得たことがわかった。また、われわれは、十分に生存しているマウスが提供されれば、DSSを水中に6日間含め、次いでそれを除去すること、DSSが長い連続した日数の間、水中に放置された場合、死亡率は高くなりすぎるため、生存したものを容易に分析しうる方法を生存するマウスを十分に得られなかったことがわかった。
確立され次第、われわれは、本モデルにおける10日の時点でマウスにおけるアルギナーゼおよびiNOS発現を評価した。図35Aに示されているように、C.ローデンティウムモデルにおけるように、アルギナーゼIのアップレギュレーションが認められたが、アルギナーゼII mRNA発現については認められず、iNOSレベルの増大が認められた。われわれは、ウェスタンブロット法(図35B)および免疫組織化学(図36)によってタンパク質レベルでのアルギナーゼI発現を確認した。
われわれのC.ローデンティウムモデルにおける上記の結果と一致して、さらにiNOS欠失またはL−Arg投与のいずれかによるDSSモデルにおける結腸炎の臨床パラメータの軽減が認められ、L−ArgがiNOS−/−マウスに投与されると、さらに生存、体重、および結腸重量の改善が認められた(図37)。これらのデータは、結腸組織病理の改善によって対比された(図38)。
興味深いことに、ポリアミンレベルを評価したところ、臨床改善した群における一貫した増大が認められた(図40)。C.ローデンティウムモデルにおけるように、L−Argの添加はポリアミンレベルを増強した。しかし、C.ローデンティウムと対照的に、L−Argを含まないWTマウスにおけるポリアミの増大は認められず、われわれは、これは、われわれがこれらの組織において確認した酵素スペルミンオキシダーゼおよびスペルミジン/スペルミンN1−アセチルトランスフェラーゼ(SSAT)を代謝するポリアミンの誘発によるものであると推測する(データは示さず)。
DSSモデルにおけるアルキナーゼの保護的役割と一致して、アルギナーゼ阻害剤BECが投与されると、われわれは、WTおよびiNOS−/−マウスにおける結腸炎の臨床および組織学的パラメータの悪化が確認された(データは示さず)。最後に、潰瘍性結腸炎およびクローン病からのヒトIBD組織において、われわれは、アルギナーゼIおよびODC mRNAレベルの増大を確認した(図4)。マウスとは異なり、アルギナーゼIIにおける増大も認められ、ミトコンドリア酵素もこれらの組織において誘発される。
試験を開始し、IBDのモデルに対するアポE類似COGペプチドの考えうる関連性を検証するために、われわれは、それらをC.ローデンティウムで活性化したマウスRAW264.7マクロファージにおいて試験した。われわれは、細菌溶解物を使用し、固有層マクロファージが曝露される可能性ある細菌産物により密接に模倣する。実際に、われわれは、規則的に感染マウスにおける上皮下粘膜において細菌凝集体を確認した(図32、高倍率像を参照)。

前記ペプチドは、アンテナペディア由来のタンパク質導入ドメイン(PTD)にコンジュゲートし、且つ前記ペプチドはLDL結合活性を有する、組成物。 - 請求項1に記載の組成物であって、前記ペプチドコンジュゲートが、RQIKIWFQNRRMKWKK−LRVRLASHLRKLRKRLLからなる、組成物。
- 請求項1に記載の組成物であって、前記COG133ペプチドのアミノ末端又はカルボキシ末端に1から15のアミノ酸が結合している、組成物。
- 請求項1に記載の組成物であって、前記COG133ペプチドのアミノ末端又はカルボキシ末端に1アミノ酸が挿入されている、組成物。
- 請求項1に記載の組成物であって、前記アンテナペディア由来のPTDが、COG133を含むペプチドのアミノ末端にコンジュゲートしている、組成物。
- 請求項5に記載の組成物であって、前記アンテナペディア由来のPTDが、RQIKIWFQNRRMKWKK(配列番号51)である、組成物。
- 請求項1から6のいずれか一項に記載のペプチドコンジュゲートを含む、医薬組成物。
- 請求項1から6のいずれか一項に記載のペプチドコンジュゲートを含む、多発性硬化症又はギランバレー症候群の治療または予防のための医薬。
- 請求項1から6のいずれか一項に記載のペプチドコンジュゲートを含む、アルツハイマー病の治療または予防のための医薬。
- 請求項1から6のいずれか一項に記載のペプチドコンジュゲートを含む、炎症性腸疾患、クローン病、又は潰瘍性結腸炎の治療または予防のための医薬。
- 請求項1から6のいずれか一項に記載のペプチドコンジュゲートを含む、外傷性脳損傷の治療または予防のための医薬。
- 請求項11に記載の医薬であって、前記外傷性脳損傷が、CNS炎症又はCNS浮腫である、医薬。
- 請求項1から6のいずれか一項に記載のペプチドコンジュゲートを含む、放射線の少なくとも1つの作用から対象を保護する医薬。
- 請求項13に記載の医薬であって、前記放射線が全身照射である、医薬。
- 請求項13に記載の医薬であって、前記対象が移植を受けた、医薬。
- 請求項15に記載の医薬であって、前記移植が血液又は骨髄移植である、医薬。
- 請求項13に記載の医薬であって、前記放射線が放射線療法である、医薬。
- 請求項17に記載の医薬であって、前記放射線療法が癌の治療用である、医薬。
