Description
すなわち本発明は、以下の(1)〜(6)である。
本発明は、イモ焼酎粕が抗アレルギー活性及び/又は抗アトピー活性を有する物質を含有するという知見に基づいている。従って、本発明に係る抗アレルギー剤及び抗アトピー性炎症剤は、イモ焼酎粕又はイモ焼酎粕由来の抗アレルギー活性及び抗アトピー活性を有する画分若しくは物質から調製することができる。
本実施例においては、サツマイモ焼酎粕サンプルの前処理を行った。簡単に説明すると、サツマイモ焼酎粕(株式会社薩摩酒造より提供)をろ紙を通してろ過し、得られたろ液を10000rpm、4℃で15分間遠心した。上清(イモ焼酎粕原液)又はエバポレーターによる3倍濃縮液を−20℃にて保存した。
実施例1で調製したイモ焼酎粕サンプルを用いて、ダイヤイオンHP20を担体として用いたゲルろ過クロマトグラフィーを実施した。
本実施例においては、イモ焼酎粕サンプルのアレルギー抑制機能を調べるために、細胞実験においてラットRBL-2H3細胞(ラット肥満細胞株)を用いて脱顆粒反応の抑制効果を検討した。脱顆粒反応として、β-ヘキソサミニダーゼ分泌の抑制法を用いた。
サンプルとして、実施例2で得られた画分H-1及びH-2の5μl、10μl及び20μlを準備した。RBL-2H3細胞(東北大学加齢医学研究所より供与)は、10%FBS(牛胎児血清)を添加したEMEM培地(日水製薬製)で培養した。48ウエルプレートに2.0×105個/ウエル/200μlでRBL-2H3細胞をまき、37℃で24時間インキュベートした。続いて、終濃度0.5μg/mlとなるようにマウスモノクローナル抗DNP-IgE抗体(Sigma社製)を添加したEMEMで培地交換し、37℃で12時間インキュベートした後、300μlのSiraganian buffer(-)(500ml中、3.477gのNaCl、0.186gのKCl、0.0406gのMgCl2、3.779gのPIPES、0.7998gのNaOH、pH7.2)で3回洗浄し、100μlのSiraganian buffer(+)(500ml中、100mlのSiraganian buffer(-)、0.1008gのグルコース、0.01109gのCaCl2、0.1gのBSA、pH7.2)を加えて37℃で10分間予備加温した。その後、サンプル(5〜20μl)を加えて37℃で30分間インキュベートした。Siragnian buffer(-)で希釈したDNP-BSA(Calbiochem 社製)10μlを各ウエルに加え、37℃で30分間インキュベートした後、10分間氷冷して反応を止めた。
抗アレルギー測定法として、β-ヘキソサミニダーゼ分泌抑制法を開発した。簡単に説明すると、細胞として、肥満細胞(マスト細胞)株細胞であるRBL-2H3細胞(東北大学加齢医学研究所より供与)を用いて、放出及び細胞内残存β-ヘキソサミニダーゼの測定量に基づき、β-ヘキソサミニダーゼの分泌の抑制を調べた。
実施例3で調製した溶出液(H-1画分)を用いて、Sephadex G25を担体として用いたゲルろ過クロマトグラフィーを実施した。
実施例3のβ-ヘキソサミニダーゼ分泌抑制法を、実施例4で得られた画分S-1及びS-2をサンプルとして用いて同様に実施した。
実施例5で調製したS-2画分を用いて、Bio-Gel P-2を担体として用いたゲルろ過クロマトグラフィーを実施した。
実施例3のβ-ヘキソサミニダーゼ分泌抑制法を、実施例6で得られた画分P-1、P-2、P-3及びP-4をサンプルとして用いて同様に実施した。
抗アレルギー活性又は抗アトピー活性の測定に関して、イモ焼酎粕サンプル及びSephadex G25により分画した画分I〜Vを用いて、in vitroにおけるβ-ヘキソサミニダーゼ分泌の抑制と、in vivoにおけるIgE抗体の生合成抑制について検討した。
担体としてSephadex G25(BIO-RAD)を使用し、カラムサイズ:直径約7.3cm、ゲル長:48cm、ゲル体積:約2000cm3、フラクション体積:20ml、流速:20ml/32秒の条件で、バッファーとして0.02M HCOONH4又は蒸留水を使用して、実施例1で調製したイモ焼酎粕原液サンプルのゲルろ過クロマトグラフィーを行った。実施例2に記載の方法と同様にして、得られた画分の280nm及び490nmにおける吸光度を測定した。
実施例3のβ-ヘキソサミニダーゼ分泌抑制法を、実施例1で調製したイモ焼酎粕原液と、上記(1)で得られた画分I〜Vを10μlのサンプルとして用いて同様に実施した。なお、画分IIIについては1μl、5μl、10μl及び20μlを準備した。
イモ焼酎粕サンプルのアレルギー抑制機能を調べるために、動物実験としてマウスを用いてIgE抗体の生合成抑制効果を検討した。
Claims (6)
- イモ焼酎粕に由来する、抗アレルギー活性を有する糖を含む親水性画分若しくは物質を有効成分として含むことを特徴とする抗アレルギー剤。
- 抗アレルギー活性が、肥満細胞の脱顆粒の抑制、β-ヘキソサミニダーゼ分泌の抑制、IgE抗体産生の抑制、アレルギー性症状の軽減、ヘルパーT細胞のTh1/Th2比の増大、及びヒスタミン分泌の抑制からなる群より選択される少なくとも1つの活性である、請求項1記載の抗アレルギー剤。
- 抗アレルギー活性が抗アトピー活性である、請求項1記載の抗アレルギー剤。
- 抗アトピー活性がアトピー性症状の軽減である、請求項3記載の抗アレルギー剤。
- 前記糖を含む親水性画分若しくは物質が、イモ焼酎粕を1以上のゲルろ過クロマトグラフィー及び/又は吸着クロマトグラフィーに供して採取される、請求項1〜4のいずれか1項に記載の抗アレルギー剤。
- 前記糖を含む親水性画分若しくは物質が、ダイヤイオン(登録商標)HP20、Sephadex(登録商標)G25、及び/又はBio-Gel(登録商標)P-2を担体として用いるゲルろ過クロマトグラフィー及び/又は吸着クロマトグラフィーに供して採取される、請求項5記載の抗アレルギー剤。
