Description
Claims (10)
- 液体試料中の1つ又は複数の標的分析物抗体に対して血清学的凝集アッセイを実施するための方法であって、
a)対向する平面状の表面であって、そのうちの少なくとも一方が透明である表面を有し、高さは20μm未満であり、且つ垂直方向の寸法と水平方向の寸法との差を1,000倍よりも大きく設定して薄く形成した平面状チャンバを提供するステップと、
b)前記平面状チャンバに有効量の検出可能な第1の粒子を入れるステップであって、前記第1の粒子が、その表面上に、標的分析物抗体が特異性を有する抗原を含む、ステップと、
c)前記第1の粒子と粒径及び形状が同様の第2の対照粒子を前記チャンバに入れるステップであって、前記対照粒子が、色又は蛍光又は他の識別可能な特徴について前記第1の粒子と異なり、及び前記対照粒子が前記抗原を有しない、ステップと、
d)前記検出可能な粒子と前記試料との互いの混和を可能にする、又は生じさせるステップであって、それにより前記標的分析物抗体が、それが存在する場合には、前記第1の粒子の凝集を生じさせる、ステップと、
e)前記混和液を電子的に撮像又は走査して、検出された粒子の凝集によって生じる画素強度のパターンを分析することにより個々の粒子凝集を検出するステップと、
f)任意の凝集した第1の粒子の信号を、任意の凝集した対照粒子の信号と比較することにより、有意な粒子凝集の有無を決定するステップと、
を含む、方法。 - 前記粒子凝集の存在を検出するステップが、凝集した第1の粒子の画素面積を、任意の凝集した対照粒子を含む画素面積と共に計測することにより行われる、請求項1に記載の方法。
- 前記粒子凝集を検出するステップが、凝集した、又は凝集塊を形成した任意の所与のタイプの粒子の割合を決定することにより行われる、請求項1に記載の方法。
- 個別に識別することのできる検出可能な第1の粒子に2つ以上のクラスがあり、各クラスが異なる抗原を含むため、複数のアッセイが同じ試料チャンバで同時に行われ得る、請求項1に記載の方法。
- 前記第1の粒子と前記対照粒子とが、互いを識別することができるように異なる形で標識される、請求項1に記載の方法。
- 液体試料中の1つ又は複数の標的分析物抗体に対して血清学的凝集アッセイを実施するための方法であって、
a)対向する平面状の表面であって、そのうちの少なくとも一方が透明である表面を有し、高さは20μm未満であり、且つ垂直方向の寸法と水平方向の寸法との差を1,000倍よりも大きく設定して薄く形成した平面状チャンバを提供するステップと、
b)前記平面状チャンバに有効量の検出可能な第1の粒子を入れるステップであって、前記第1の粒子が、その表面上に、前記試料中に存在し得る標的抗原に対するリガンドを含む、ステップと、
c)前記第1の粒子と粒径及び形状が同様の第2の対照粒子を前記チャンバに入れるステップであって、前記対照粒子が、色又は蛍光又は他の識別可能な特徴について前記第1の粒子と異なり、及び前記対照粒子が前記リガンドを有しない、ステップと、
d)前記検出可能な粒子と前記試料との互いの混和を可能にする、又は生じさせるステップであって、それにより前記標的分析物抗体が、それが存在する場合には、前記第1の粒子の凝集を生じさせる、ステップと、
e)前記混和液を電子的に撮像又は走査して、検出された粒子の凝集によって生じる画素強度のパターンを分析することにより個々の粒子凝集を検出するステップと、
f)任意の凝集した第1の粒子の信号を、任意の凝集した対照粒子の信号と比較することにより、有意な粒子凝集の有無を決定するステップと、
を含む、方法。 - 前記粒子凝集の存在を検出するステップが、凝集した第1の粒子を含む画素面積を、任意の凝集した対照粒子を含む画素面積と共に計測することにより行われる、請求項6に記載の方法。
- 前記粒子凝集を検出するステップが、凝集した、又は凝集塊を形成した任意の所与のタイプの粒子の割合を決定することにより行われる、請求項6に記載の方法。
- 個別に識別することのできる検出可能な粒子に2つ以上のクラスがあり、各クラスが異なる抗原を含むため、複数のアッセイが同じ試料チャンバで同時に行われ得る、請求項6に記載の方法。
- 前記第1の粒子と前記対照粒子とが、互いを識別することができるように異なる形で標識される、請求項6に記載の方法。
