Description

本発明は、眼の潤滑性の調節に関連するものである。詳細には、本発明は、角膜および結膜表面の潤滑性の欠乏により起こる疾患を治療するための製薬学的組成物、およびその使用方法に関するものである。
プロテオグリカン4(proteoglycan 4(prg4))遺伝子は、巨核球刺激因子(Megakaryocyte Stimulating Factor)(MSF)、ルブリシン(lubricin)、及び、スーパーフィシャル・ゾーン・プロテイン(Superficial Zone Protein)(SZP)を容易にコード化する(1)。ルブリシンは、最初に滑液から分離されて、軟骨とガラスの界面で滑液に類似してイン・ビトロ(in vitro)での潤滑能力が示された(2)。ルブリシンは、後で滑膜線維芽細胞(3)の製品と確認されて、そのうえJayら(3−9)によって、ラテックス・ガラスの界面で、境界潤滑能力を有することが示された。940のアミノ酸(10)(エクソン6によってコード化される)の大きなムチンのような領域の中のOにリンクされたβ(1−3)Gal−GalNAcオリゴ糖は、部分的に、境界潤滑能力を考慮することがその後示された(8)。SZPは、最初に表面的な地帯から外植体軟骨の表面で局所化されて、馴化培地から単離された(11)。SZPも、軟骨ガラスの界面で潤滑能力を示した(12)。これらの分子は、集合的に、PRG4と呼ばれている。PRG4は、関節滑膜(58)、腱(13)と半月板(14)の表面に存在することも示された。そのうえ、生理的で病態生理学的濃度において、並べられた関節軟骨面の境界潤滑に貢献することが、PRG4について示された(59)。

Claims (13)
- 個人における眼の潤滑性の欠乏若しくはそれにより引き起こされる症状の治療において使用するための製薬学的組成物であって、
眼科治療に許容される平衡塩類溶液にけん濁されるPRG4の治療有効濃度を含む、製薬学的組成物。 - 請求項1の製薬学的組成物であって、眼科治療に許容される粘滑剤、眼科治療に許容される賦形剤、眼科治療に許容される収斂剤、眼科治療に許容される血管収縮剤、眼科治療に許容される電解質、眼科治療に許容されるヒアルロン酸ナトリウム又はヒアルロン酸、眼科治療に許容される界面活性リン脂質、及び、眼科治療に許容される軟化剤からなる群より選択される眼科治療に許容される薬剤の1又は2以上を含むことを特徴とする製薬学的組成物。
- 請求項1又は2の製薬学的組成物であって、前記製薬学的組成物は、10−10,000μg/mLの治療有効濃度のPRG4を含むことを特徴とする製薬学的組成物。
- 請求項1又は2の製薬学的組成物であって、前記製薬学的組成物は、50−500μg/mLの治療有効濃度のPRG4を含むことを特徴とする製薬学的組成物。
- 請求項1から3の何れかに記載の製薬学的組成物であって、治療有効濃度のヒアルロン酸ナトリウム又はヒアルロン酸を更に含むことを特徴とする製薬学的組成物。
- 請求項5の製薬学的組成物であって、前記製薬学的組成物は、10−100,000μg/mLの治療有効濃度で、ヒアルロン酸ナトリウム又はヒアルロン酸を含むことを特徴とする製薬学的組成物。
- 請求項5の製薬学的組成物であって、前記製薬学的組成物は、500−5,000μg/mLの治療有効濃度で、ヒアルロン酸ナトリウム又はヒアルロン酸を含むことを特徴とする製薬学的組成物。
- 請求項1から7の何れかに記載の製薬学的組成物であって、L−α−ジパルミトイルフォスファジルコリン、ホスファチジルコリン、ホスファチジルエタノールアミン、及び、スフィンゴミエリンからなる群より選択される治療有効濃度の表面活性リン脂質を更に含むことを特徴とする製薬学的組成物。
- 請求項8の製薬学的組成物であって、前記製薬学的組成物は、10−10,000μg/mLの治療有効濃度の表面活性リン脂質を含むことを特徴とする製薬学的組成物。
- 請求項1から9の何れかに記載の製薬学的な組成物であって、前記眼科治療に許容される平衡塩類溶液が少なくとも3つの異なる電解質を含み、該電解質が、リン酸ナトリウム、塩化ナトリウム、塩化カリウム、炭酸水素ナトリウム、炭酸水素カリウム、塩化カルシウム、塩化マグネシウム、酢酸ナトリウム、クエン酸ナトリウム、塩酸、及び、水酸化ナトリウムからなる群より選択されることを特徴とする製薬学的組成物。
- 請求項1から9の何れかに記載の製薬学的な組成物であって、前記PRG4が、潤滑性を有する潤滑フラグメント、又は、それのマルチマーを含む、精製された天然発生の若しくは遺伝子組換えのPRG4タンパク質であり、平均モル質量が50kDa及び400kDaの間であることを特徴とする製薬学的組成物。
- 請求項1から11のいずれかに記載の製薬学的な組成物であって、必要のある個人は、眼の境界潤滑における欠乏を持ち、それは、涙液欠乏性若しくは蒸発性ドライアイ症状、シェーグレン症候群、乾性角結膜炎、アンドロゲン欠乏症、マイボーム腺病、エストロゲン補充療法、コンタクトレンズ装着、屈折矯正手術、アレルギー、涙膜層破壊時間の減少、易感染涙膜、眼表面障害、涙膜内及び眼の表面でのプロテアーゼ・レベルの上昇、慢性炎症、高浸透度、加齢、又は、それらの組合せであることを特徴とする製薬学的組成物。
- 請求項1から11のいずれかに記載の製薬学的な組成物であって、眼の潤滑欠乏に関連し若しくは引き起こす前記状態は、涙液欠乏性若しくは蒸発性ドライアイ症状、シェーグレン症候群、乾性角結膜炎、アンドロゲン欠乏症、マイボーム腺病、エストロゲン補充療法、コンタクトレンズ装着、屈折矯正手術、アレルギー、涙膜層破壊時間の減少、易感染涙膜、眼表面障害、涙膜内及び眼の表面でのプロテアーゼ・レベルの上昇、慢性炎症、高浸透度、加齢、又は、それらの組合せからなる群より選択されるものであることを特徴とする製薬学的組成物。
