Description
本出願は、2007年10月31日出願の米国仮特許出願第60/984,046号、題名「クエン酸安定化中性メタロプロテアーゼの使用と生産」に基づく優先権を主張する。
本発明の要約
しかしながら、本発明の理解を促すために、多くの用語が以下に定義されている。
定義
中性メタロエンドペプチダーゼ(つまり、中性メタロプロテアーゼ)(EC 3.4.24.4)は、触媒活性のために亜鉛イオンを絶対的に必要とするプロテアーゼのクラスに属する。これらの酵素は中性pHで最も活性であり、30から40kDaの範囲にある。中性メタロプロテアーゼはこのタンパク質の構造上の安定性に寄与する2個から4個のカルシウムイオンと結合している。メタロプロテアーゼの活性部位にある結合金属イオンは水分子の活性化のために欠くことのできない特徴である。この水分子は、次に求核剤として働き、ペプチド結合のカルボニル基を開裂する。
ここでNは固有の形態であり、Iは部分的に折りたたみの解けた中間体を表し、Apは自己分解されたプロテアーゼである。NprEについては、クエン酸イオンが、カルシウムイオンを取り去り、部分的に折りたたみの解かれた中間体(I)を生じさせ、ループと表面の残基が自己分解を受けやすくすることによってタンパク質構造を最も不安定化させる可能性が最も高い。自己分解されたタンパク質断片の生成は迅速でありかつ、高次である。これらの領域またはその近傍での安定化を高めるための置換は、自己分解に対し安定性が高くなったNrpE変異種を生じさせた。この変異種のモデルは、自己分解されたタンパク質(Ap)の生成がかなり遅いことを除き、野生型に類似する可能性が非常に高い。クエン酸存在下又は非存在下における熱的な折りたたみの解けは、クエン酸存在下で活性で完全な変異種が折りたたまれた構造を取ることを強調する重要事実であった。このデータは、見出された変異のいずれも直接にカルシウムとの結合に係っていないことを考えると、クエン酸に対し安定化する置換はカルシウム-ストレスを加えられたタンパク質の安定性を増大させている可能性が最も高い。従って、本明細書で記載の通り、カルシウムの喪失によるNprEのクエン酸による不安定化はカルシウム-ストレスを加えられたタンパク質の安定性を改善することにより、間接的に補うことができる。
別に記載されている場合を除き、本明細書に記載された全ての成分または組成物の量はその成分または組成物の活性な量であり、市販品に含まれている可能性のある不純物、例えば、残留溶媒または副生物は含まない。酵素成分の重量は総タンパク質量に基づく。全てのパーセンテージと比率は、別に示されている場合を除き、重量により計算される。全てのパーセンテージと比率は別に示されている場合を除き、全組成物に基づき計算される。
中性メタロプロテアーゼを含む洗浄剤組成物
いくつかの好ましい実施態様では、本発明の組成物はいずれかの適した形態へ調合され、調合者により選択されたいずれかの方法により調製される。(非限定的例として例えば、U.S.5,879,584、U.S. 5,691,297、U.S.5,574,005、U.S.5,569,645、U.S.5,565,422、U.S.5,516,448、U.S.5,489,392及びU.S.5,486,303を参照)低pH洗浄組成物が望まれるいくつかの実施態様では、そのような組成物のpHはHClのような酸性物質の添加により調整される。

好ましい実施態様では、本発明の洗浄剤組成物は、表面及び/または布地の洗浄に使用される。いくつかの実施態様では、その表面及び/または布地の少なくとも一部は、原液とまたは洗浄液の希釈液で、本発明の洗浄剤組成物の少なくとも一実施態様と接触し、次にその表面及び/または布地は任意に洗浄及び/または濯ぎを受ける。本発明の目的のため、「洗浄」はこすることと機械的にかき混ぜることを非限定的に含む。いくつかの実施態様では、この布地は、普通の消費者の使用条件で洗濯され得るいずれの布地も含む。好ましい実施態様では、本発明の洗浄剤組成物は、溶液中で約500ppmから約15,000ppmの濃度で使用される。洗浄溶液が水であるいくつかの実施態様では、この水温は通常約5℃から約90℃の範囲にある。布地洗浄に関していくつかの好ましい実施態様では、水と布地の質量比は通常約1:1から約30:1である。

定量
以下の定量が下記の実施例で行われた。下記のプロトコールからのいかなる逸脱も実施例に示している。これらの実験では、分光光度計は反応の終了後、生成した産生物の吸光度を測定するために使用された。布見本の反射率を測定するため反射率計が使用された。
1.96ウェルマイクロタイタープレート(MTPs)でのタンパク質含量測定のためのBCA(ビシンコニン酸)定量
これら定量では、BCA(Pierce)定量は、MTP上でプロテアーゼサンプル中のタンパク質濃度を決定するために使用された。この定量系では、使用された化学物質及び試液は:BCAタンパク質定量試薬、及びPierce Dilution緩衝液(50mM MES,pH6.5, 2mM CaCl2、0.005%TWEEN(登録商標)-80)。使用された装置はSpectraMAX(340型)MTPリーダー。このMTPはCostar(9017型)から得られた。
これらの定量では、ブラッドフォード染色試薬(Quick Start)定量がMTPでプロテアーゼサンプルのタンパク質濃度を決定するために使用された。この定量系で使用された化学物質と試液は、:Quick Start Bradford Dye Reagent(BIO-RAD Catalog No. 500-0205)、稀釈緩衝液(10mM NaCl、0.1mM CaCl2、0.005%TWEEN(登録商標)-80)である。使用した装置はBiomek FX Robot (Beckman)とSpectra MAX (type 340)MTPリーダーであった。このMTPはCostar(9017型)のものであった。
クエン酸に対する安定性はmMのクエン酸存在下で、野生型NprEと変異種を保温した後に測定された。この初期及び残留活性はDMC 加水分解定量法を用いて決定された。この定量系で、使用された化学物質と試液は、以下のものであった。

クエン酸安定性定量法で好ましい稀釈率を得るため、各プレートの野生型NprE対照サンプルのプロテアーゼ濃度はTCA定量により決定された。この方法では、25μlのろ過を受けた液体培地が200μlの16.875%(w/v)TCAに加えられた。周囲温度で10から15分間保温後、405nmにおける光散乱/吸光度が決定された。このタンパク質濃度は、精製NprEで作成した検量線を使用して決定された。
ろ過した培養液は稀釈緩衝液2で稀釈された。このMTPはテープでカバーされ、数秒振とうした後、25℃、60分間、200rpmで恒温器に入れられた。保温終了後、各ウェルからこの混合物の20μlを採取して、180μl 1%DMCの予熱された基質溶液(この基質は25℃に予熱されていた)を含む新しいMTPへ移された。このMTPは直接に恒温器/振とう器に入れられ200rpmの撹拌で30分間25℃で保温された。残存プロテアーゼ活性はジメチルカゼイン加水分解定量を用いて、下記のように決定された。
ろ過を受けた液体培地が稀釈緩衝液1により稀釈された。直ぐに、20μlの混合物が各ウェルから採取され、180μlの予熱された1%DMC基質溶液(基質溶液は25℃で予熱された。)を含む、新しいMTPへ移された。このMTPは直接に恒温器/振とう器に入れられ、200rpmの撹拌で、30分間25℃で保温された。ジメチルカゼイン加水分解定量を用いて、TNBSにより決定された初期プロテアーゼ活性は、下記に述べられている。

PIPES 緩衝液(遊離酸)Sigma P-1851; 15.1gが約960mlの水に溶解される。;4N NaOHによりpHは6.0に調整され、1mlの5%TWEEN(登録商標)-80が加えられ、容量が1000mlにされた。PIPESとTWEEN(登録商標)-80の最終濃度は、それぞれ50mMと0.005%である。
ピクリルスルホン酸(TNBS)Sigma P-2297(5%水溶液)
試薬A 45.4g Na2B4O7 10H2O(Merck 6308)と15mlの4N NaOHが共に溶解され最終容量は1000mlである(必要時は加熱する)
試薬B 35.2g NaH2PO4 1H2O(Merck6346)と0.6gのNa2SO3(Merck 6657)が共に溶解され最終容量は1000mlである。
基質を合成するために、4gのジメチルカゼインが400mlのPIPES緩衝液に溶解された。
このろ過を受けた培養液の上澄み液はPIPES緩衝液で稀釈された。次に、10μlの各希釈上澄液がMTPのウェルの200μlの基質へ加えられた。このMTPはテープでカバーされ、数秒間振とうされ、30分間25℃で撹拌せずオーブンに入れられた。このオーブンから第1のプレートを出す前、約15分に、50mlの試薬Aについて1mlTNBS溶液を混合し、TNBS試薬が調製された。MTPには、各ウェルについて60μlのTNBS・試薬Aが加えられた。恒温器に入れられたプレートは数秒間、振とうされ、その後、10μlが、TNBS試薬を入れたMTPに移された。このプレートはテープでカバーしベンチ振とう器(BMG Thermostar)で20分間、室温、500rpmで振とうされた。最後に、200μlの試薬Bがウェルへ加えられ、振とう器で1分間混合し、405nmでの吸光度がMTPリーダーを用いて測定された。
下記に与えられている方法は時と場所に依存しない再現性のあるプロテアーゼ定量データを取る、ある程度の技術的詳細を与える。定量は所与の試験条件に適合させることが出来るが、変更した手順により得られたいずれのデータも元の方法によって得られた結果と照合されなければならない。中性メタロプロテアーゼは、2-アミノベンゾイル-L-アラニルグリシル-L-ロイシル-L-アラニノ-4-ニトロベンジルアミド(Abz-AGLA-Nba)のグリシンとロイシンの間のペプチド結合を開裂する。溶液中の遊離2-アミノベンゾイル-L-アラニルグリシン(Abz-AG)は、340nmに極大のある励起光により415nmに極大のあるケイ光発光をもつ。Abz-AGのケイ光は、開裂前のAbz-AGLA-Nba分子中のニトロベンジルアミドにより消光される。
52.6mM MES/NaOH、2.6mM CaCl2, pH6.5-MES 緩衝液
MES 酸(10.28g)と292mgの無水CaCl2は約900mLの精製水に溶解された。この溶液はNaOHによりpH6.5に調整された(25℃で、又は温度調整pHプローブにより)。このpH-調整された緩衝液は総容量1Lにされた。最終溶液は0.22μm無菌フィルターでろ過され、室温に保たれた。
約28mgのAbz-AGLA-Nbaが小さい試験管に加えられた。これはDMFに溶解され(容量は加えたAbz-AGLA-Nbaの量により変わる。)、数分ボルテックスでかき混ぜた。この溶液は遮光して室温で保存された。
1mLのAbz-AGLA-Nba 原液が19mLのMES緩衝液に加えられ、ボルテックスに掛けられた。この溶液は遮光して室温で保存された。
この緩衝液は95mLのMES緩衝液に5mLの精製水を加えることにより調製された。
5mLの純DMFが95mLのMES緩衝液に加えられた。この緩衝液は速度論的パラメータを決定するために使用された。
酵素原液は酵素稀釈緩衝液で約1ppm(1μg/mL)の濃度にまで稀釈された。MULTIFECT(登録商標)中性プロテアーゼ(野生型NprE)は、6ppm(6ug/mL)未満の濃度にまで稀釈された。連続的希釈が望ましい。これらの溶液は室温で1時間安定であった、しかしより長い貯蔵では、これらの溶液は氷上で保存された。
最初に全ての緩衝液、原液及び試液が調製された。各酵素希釈液は、別に記載のない場合は、3回定量された。全てに溶液を入れない場合、酵素試液の貯留マイクロプレートはプレートの左から縦の列を満たすようにされた(プレートリーダーで測定するため)。対応する定量用プレートも同様に準備された。マイクロプレート分光ケイ光光度計は先に述べたように準備された。
B.スブチリスにおけるNprEプロテアーゼ産生
この実施例では、B.スブチリスにNprEを産生させるために行われた実験が述べられる。特に、B.スブチリスをプラスミドpUBnprEで形質転換するときに使用される方法が提供される。形質転換は本技術分野で知られているように実施された(例えば、WO2002/014490とWO2007/044993参照。両者は引用により本明細書に組み入れられる。)下記のDNA配列(B.アミロリケファシエンス由来のnprEリーダー、nprE 前置及びnprE成熟DNA配列)は、NprE前駆体タンパク質をコードしている。

このpUBnprEベクターは、上記のnprE発現カセットを含み、鋳型DNAとして役立つ。このベクターは特有のBglII制限部位を含み、これが部位評価ライブラリー構築に利用された。簡単にいうと、nprE部位評価ライブラリーを構築するために、3回のPCR反応が行われた。これは、成熟nprE DNA配列中に目的の変異コドンを導入するための2回の変異導入PCRと成熟nprE配列中に目的とする変異コドンを含むpUBnprE発現ベクターを構築するためのこの2つの変異PCR 断片を融合するために使用された3回目のPCRを含む。

使用 B.スブチリス細胞(ΔaprE,ΔnprE、oppA、ΔspoIIE、degUHy32、Δamy E::(xylR,pxylA,pxylA-comK)株
1変異を含むpUBnprEプラスミド(実施例3及びWO2007/04493で先に述べられたnprESELのスクリーニングにより同定される。WO2007/04493は引用により組み入れられる)を得るために、目的とする各バチルス株の単一コロニーが5ml LB+10ppm ネオマイシンを入れた試験管(例.スターター培養)に接種するために使用された。この培地は6時間、225rpmで振とうしながら37℃で培養された。次に、100mlの新鮮なLB+10ppmネオマイシンが1mlのスターター培地で接種された。この培地は225rpmで振とうしながら37℃で培養された。この培養に続き、収集に十分な遠心分離により細胞の粒が集められた。この粒は10mlの緩衝液P1(Quiagen Plasmid Midi Kit)中で再懸濁された。次に、10μlのReady-Lyse-Lysozymeがこの再懸濁された細胞粒に加えられ、30分間、37℃で保温された。10mlのBuffer P2とP3を培養液に加えて容量を増やし、Quiagen Plasmid Midi Kitのプロトコールを続けた。単一のnprE変異を含む各pUBnprEプラスミドをバチルスから単離した後、各プラスミドの濃度が決定された。次に、これらのプラスミドはメーカーの指図書に従い、damメチラーゼキット(New England Biolabs)を用いてdamメチル化され、試験管当たり約2μgの各pUBnprEプラスミドをメチル化した。Zymoclean Gel DNA回収キットがdam-メチル化されたpUBnprEプラスミドを精製、濃縮するために使用された。このdam-メチル化pUBnprEプラスミドは、次に、定量され、それぞれ50ng/μlの試験濃度に稀釈された。混合した部位指定変異プライマーが各反応ごとに別に調製された。例えば、pUBnprE T14Rプラスミドを鋳型として使用して、混合した部位指定変異プライマーの試験管は10μlのnprE-S23R、10μl nprE-G24R、10μl nprE-N46K、及び10μl nprE-T54R(全てのプライマーは各10μM)を含むであろう。QuikChange Multi Site-Directed Mutagenesis Kit(Stratagene)を使用するPCR反応がメーカーの指図書に従い行われた(例.1μlの1変異を含むdam メチル化pUBnprEプラスミド(50ng/μl)、2μl nprE 部位指定変異誘導プライマー(10μM)、2.5μl 10xQuikChange Multi Reaction 緩衝液、1μl dNTP Mix、1μl QuikChange Multi enzyme blend(2.5U/μl)、及び17.5μlの蒸留、加圧滅菌水を用い、総計25μlの反応混合物を与える)。このnprE変異種ライブラリーは以下の条件を使用して増幅された。:95℃、1分(第1サイクルのみ)、続いて95℃、1分間、55℃、1分間、65℃、13.5分間、及び29サイクル繰り返す。反応生成物は4℃で一夜保存された。次に、この反応混合物は、メーカーのプロトコール(つまり、1.5μlのDpnI制限酵素が各試験管に加えられ、37℃で3時間保温される。)を用いてDpnI切断処理(QUIKCHANG(登録商標)Multi Site-Directed Mutagenesis Kitにより提供される)を受け親pUB-nprEプラスミドを切断した。2μlのDpnI-切断されたPCR反応物は次に1.2%E-ゲル上で分析され、PCR反応が進行し、親の鋳型が切断されたことを確認した。TempliPhiローリングサークル(rolling circle)増幅がメーカーのプロトコール(つまり、〜11μlの総反応量に対して、1μlのDpnI処理を受けたDNA
のQuikChange MultiSite-Directed Mutagenesis PCR, 5μl TempliPhi Sample 緩衝液、5μl TempliPhi Reaction 緩衝液、及び0.2μl TempliPhi Enzyme Mix、30℃、3時間で保温された。)を用いて、このnprE多数変異含有種のライブラリーのサイズを増やすために多量のDNAを作成するために使用された。このTempliPhi 反応物は200μlの蒸留、加圧滅菌水を加えることにより稀釈され、短くボルテックスが掛けられた。次に1.5μlの稀釈TempliPhi物はB・スブチリス細胞を形質転換し、nprE多数変異含有種は、LA+10ppm ネオマイシン+1.6%スキムミルクプレートを用いて選別された。コロニーが採取され、異なるnprE変異ライブラリーの組合せを決定するために配列決定された。
変異プロテアーゼの発現、発酵及び精製
本実施例は先の実施例の形質転換されたB・スブチリスのプロテアーゼの発現、発酵及び精製に使用される方法を述べる。
中性メタロプロテアーゼ
実施例5
本実施例では、野生型及び組換え変異NprEのクエン酸-誘導自己分解の評価に使用される方法が述べられる。これらの実験では、B・アミロリケファシエンス(B・スブチリスで発現された天然及び組換え変異種)由来の中性メタロプロテアーゼの自己分解がクエン酸ナトリウム(Sigma)を用いて誘導された。この自己分解過程は(i)25mM MES,pH6.5 中4℃又は(ii)5mM HEPES pH8.0 で、室温で反応を行うことにより制御された。これらの実験では、0.4mg/ml NprEの自己分解は、(a)10mMクエン酸において保温時間(0-120分)を変えるか;又は(b) 100分でクエン酸濃度(10-100mM)を変えることにより最適化された。緩衝液のみ(つまり、クエン酸を含有しない)で稀釈された中性メタロプロテアーゼの対照サンプルは同様の条件で保温された。残存プロテアーゼ活性は、合成ペプチド(Abz-AGLA-Nba)を用いて測定され、クエン酸のないその対照サンプルと比較して計算された。
クエン酸誘導自己分解に耐性のあるNprE 変異種の産生
本実施例はカルシウムキレート剤の存在下、安定性を改善したNprE変異種のスクリーニングを述べる。第1の切断部位、M138、L198、D220、A221及びG222とこれらの部位の近辺のアミノ酸及び/または酵素表面にあるアミノ酸は実施例3とWO2007/044993(引用により全てを本明細書に組み入れる)で述べられた遺伝子作成及び配列決定法を用いて変異誘導の目標とされた。S129、F130、M138、V190及びD220の部位評価ライブラリー(天然のアミノ酸残基が19個の別のアミノ酸残基の一つによりそれぞれ置き換えられている)が調製された。同様に、2重(M138L-D220P; F130L-D220P; S129I-D220P; V190I-D220P; S129I-V190I, S129V-V190I, S129V-D220P)、3重(M138L-V190I-D220P, S129I-F130L-D220P)及び4重(S129I-F130L-M138L-V190I-D220P)アミノ酸置換変異種が作成された。
)、3重(S129I-F130L-D220P、及びM138L-V190I-D220P)及び5重(S129I-F130L-M138L- V190I-D220P)NprE変異種のクエン酸に対する安定性は、合成ペプチドに対するプロテアーゼ活性の測定、及びSDS-PAGE分析により評価された。クエン酸スクリーニングデータはLeuによるM138、Pro又はGluによるD220、IleまたはLeuによるS129、LeuによるF130及びLeuまたはIleによるV190の置換により、クエン酸自己分解に対する感度が低くなったプロテアーゼが生じたことを示した。
示差走査熱量測定(DSC)
過剰熱容量曲線は超感度走査高効率ミクロ熱量測定計、VP-Cap DSC(MicroCal. Inc., Northampton, MA)を使用して、測定された。約500 Lの200から400ppmのタンパク質が必要であった。通常、400ppmのNprEとその変異メタロプロテアーゼ(クエン酸非存在下、130mMのクエン酸の存在下)は、20-100℃の温度範囲にわたり走査された。同一のサンプルは、この方法の可逆性を点検するため再走査された。中性プロテアーゼについては、熱による折れたたみの解ける過程は非可逆的であった。使用された緩衝液は5mM HEPES,pH8.0であった。NprEの融点の走査速度に依存するデータは、25から200℃/時間の走査速度で評価された。200℃/時間の走査速度が、凝集または自己分解から生じうる分解物を最小にするため、最終的に選ばれた。DSC曲線の融点(Tm)は、熱安定性の標識として使用された。440ppm野生型中性プロテアーゼは69.2±0.5℃の融点(Tm)を示した。野生型とNprE変異種の融点が図6に示され、この変異種はクエン酸がない条件では、融点に最小限の影響しか与えないことを示している。
NprEの相同モデリング
この実施例はB.セレウスNprEの既知の構造に基づくB・アミロリケファシエンスNprEの構造のモデリングを述べる。B・アミロリケファシエンスNprE の構造は、B.セレウスNprE に45%同一である。加えて、ミクロPIXES分析は、NprEの亜鉛とカルシウムの結合の化学量論を確認するため使用された。このモデルは溶媒に晒される可能性のあるNprEのアミノ酸を同定するために使用された。
最終モデルのRamachandronプロットは3つの外れた残基を明らかにした。これは、S23、N61及びS191である。表面に出ている残基の分析により、この表面に8個の脂肪族又は芳香族残基を明らかにした。これらは、Y49、L55、I117、F130、V190、L216、V260及びL282である。これらの残基は表面に出ているループにあり、一般的に相同モデリング法を用いて、最も正確にモデル化ができない領域であり、このモデル化が比較的信頼できない領域となっている可能性がある。
pH依存活性と安定性の評価
A. 野生型NprEと変異NprEのpH依存活性
異なるpHの緩衝液がMES1水和物、トリズマ(Trizma)塩基及び酢酸ナトリウム(すべてSigma)を用いて調製された。緩衝液の各成分の最終濃度は25mM酢酸塩、50mM MESと25mMトリスであった。この緩衝液のpHは水酸化ナトリウム又は塩酸のいずれかを使用して最終pH9.40、8.69、8.36、7.70、7.45、6.52、6.00、5.45、4.78及び4.27に調整された。AGLA基質(American Peptide Company)が、4.8mMの濃度でDMFに溶解された。各緩衝液に異なる濃度のAGLA基質が調製され、最終AGLA濃度は0.096mM、0.048mM、0.024mM、0.012mM及び0.006mMであった。
異なるpHでの野生型NprEと5重NprE変異種(S129I-F130L-M138L-V190I-D220P=「5重変異」)の安定性が塩化カルシウムのない条件又は2mM塩化カルシウム存在下で試験された。異なるpHの同一の組の緩衝液が、先に述べたように使用された。但し、96ウェルのプレートにタンパク質が結合することを避けるため0.005%Tween80も各緩衝液に加えられた。1組の緩衝液に、2mM塩化カルシウムが加えられた。精製されたタンパク質が最終タンパク質濃度が10μg/mlになるように加えられた。総量は96ウェルのプレートで200μl/ウェルであった。このプレートは室温(例.23℃)で、3時間保温された。異なるpHでの各タンパク質の活性がAGLA定量を用いて測定された。野生型NprEは、pH6.5で最も安定であり、6.5より低い又は高いpHでは安定性が低下した。pH4.3では、NprEは100%活性を失う。5重変異種は、pH5.5 からpH9.4の広いpHの範囲で非常に安定である。4.7未満のpHでは、5重変異種の安定性は低下する。しかし、2mMのカルシウムを加えると、低pHと高pHの両方でNprEと5重変異種の両方が安定化する。

- 金属キレート剤の存在下の野生型バチルス中性メタロプロテアーゼの抵抗性と比べて、前記金属キレート剤の存在下で抵抗性の向上した単離されたバチルス中性メタロプロテアーゼ変異種であり、前記変異種はSEQ ID NO:3として表されたアミノ酸配列の129、130、138、190及び220の位置と同等の位置からなる群から選択された3又は5箇所での置換を含むアミノ酸配列を有し、前記置換はM138L/V190I/D220P及びS129I/F130L/M138L/ V190I/D220Pから選択された複数の変異を含むバチルス中性メタロプロテアーゼ変異種。
- 請求項1の単離されたバチルス中性メタロプロテアーゼ変異種であって、野生型バチルス中性メタロプロテアーゼの抵抗性に比べクエン酸-誘導自己分解に対し抵抗性が改善しているバチルス中性メタロプロテアーゼ変異種。
- 請求項1の単離されたバチルス中性メタロプロテアーゼ変異種であって、前記バチルス属は、B・アミロリケファシエンスであるバチルス中性メタロプロテアーゼ変異種。
