Description
本発明は、sigG遺伝子から少なくとも150ヌクレオチドが欠失していることを特徴とする非胞子形成枯草菌株に関する。ここで記載する欠失は、議論の余地無く、胞子形成の防止におけるsigG欠失の効果を示すことができる初めてものである。さらに、遺伝子欠失において、いかにして欠失が生物体のその他の機能に影響を及ぼすか、あるいは、生物体全体としての生存能力に影響を及ぼすか、ということを予測することは困難である。しかし本発明は、sigG遺伝子の欠失は、欠失した細菌のその他の必須の機能、例えば、組換えポリペプチドの産生能に影響を与えなかったことを示す。

本発明の枯草菌はsigG遺伝子の有意な断片を欠失させることによって非胞子形成のものにされたものである。有意な断片の欠失は少なくともおよそ150、好ましくは少なくともおよそ300ヌクレオチドの欠失を意味する。sigG遺伝子は少なくとも2つの機能性の領域を有する。即ち、これら領域はタンパク質の活性のために重要であり、一方はおそらくRNAポリメラーゼのコア断片に結合するドメインをコードし、これは第67アミノ酸から開始して第80アミノ酸で終結する。このドメインは、その他の既知のシグマ因子配列と枯草菌細菌のシグマ−G因子のアミノ酸配列を比較することによって規定される。もう1つの重要な機能性の領域はH−T−H(ヘリックス−ターン−ヘリックス)モチーフであり、これはDNAへのタンパク質の結合部位に存在し、これもまた類似性に基づいて規定され、SigGタンパク質のアミノ酸229から248である。欠失させるべき領域は好ましくはSigGタンパク質のアミノ酸67および248をコードするヌクレオチドの間の領域である。より好ましくは、欠失は、sigG遺伝子の2つの機能性領域のいずれかの少なくとも一部を含むようし、該領域はsigGタンパク質のアミノ酸67から80またはアミノ酸229から248をコードする。特に、欠失はsigG遺伝子の該二つの機能性領域の両者の少なくとも一部を含む。あるいは、欠失は機能性領域の間にあってもよく、この場合機能性領域のいずれかの発現が阻害される、および/またはこれらの間の距離がネイティブなシグマ−G因子様分子を変化させてシグマ−G因子のRNAポリメラーゼおよびDNAへの結合が阻害されるようにする。1つの態様によると、少なくとも600、特に少なくとも670ヌクレオチドがsigG遺伝子から欠失される。例えば、ヌクレオチド39から715を欠失させるのが好ましい。sigG遺伝子全体および該遺伝子のプロモーター領域を欠失させることも可能である。
実施例において用いる細菌株とプラスミドの特性を表1に挙げる。宿主生物の枯草菌168sigG遺伝子とその隣接領域の配列は、データバンクから得た(Micado データベース、http://locus/jouy.inra.fr)。cat遺伝子をコードする領域はプラスミドpSJ2739E由来であって黄色ブドウ球菌(Staphylococcus aureus)のプラスミドpC194のものと同一である(GenBank 登録番号VO1277)。産生ベクターpRSH10は、バシラス・リチェニフォルミスのβ−ラクタマーゼpenPをコードするプラスミドである。該プラスミドにおける、penPは分泌ベクターpKTH141(Lundstrom、1985)のHindIIIクローニングサイトに導入される。ベクターのα−アミラーゼプロモーターとシグナル配列をコードする断片は、バシラス・アミロリクエファシエンス由来である。
細菌株をルリア・ブロス(LB)培地(Harwood and Cutting、1991)、5g/lイーストエクストラクト(Difco)、10g/lトリプトン(Oxoid)および10g/l NaCl(Merck)で、+37℃、250rpmにて撹拌培養した。タンパク質産生をモニタリングする場合、細胞を濃縮LB培地(10g/lイーストエクストラクト、20g/lトリプトンおよび10g/l NaCl)で培養した。クロラムフェニコール(Cm)5μg/mlおよびカナマイシン(Km)10μg/mlを細菌培養における選抜のために用いた。
常套の方法によるDNA技術を用いた(Sambrook et al.、1989)。染色体DNAをMarmur(1961)の指示に従って単離した。プラスミドDNAを細菌細胞からイオン交換カラム(QIAGEN Plasimid Midi Kit)を用いて製造業者の指示に従って単離した(Qiagen Plasimid Purification Handbook July/1999)。この指示から逸脱して、リゾチーム処理をプロトコールに追加してペプチドグルカン層を分解し、この処理において1mg/mlリゾチームをバッファーP1に添加し、細胞をこのバッファー中で37℃で30分間インキュベートした。精製したプラスミドを30から100μlの滅菌水に溶解した。プラスミド調製物のDNA含量は50から500ng/μlであった。
sigG欠失の胞子形成に対する効果を、胞子形成培地で細菌細胞を培養し、細胞を熱処理(+85℃、10分間)することによって試験した(Harwood and Cutting、1991)。用いた処理は栄養細胞を殺すものであるが、胞子を殺すには十分でない。処理後に生育したコロニー数は胞子の量を示す。
sigG遺伝子欠失をPCRとSOE技術を用いて構築した直鎖状組換えDNA断片によって野生株枯草菌168を形質転換することによって行った(Horton et al.、1989、および米国特許第5023171号)。行った欠失のフローチャートを図1に示す。PCR反応のプライマーは、PCR産物が相補的配列を用いた第2のPCR反応によって連結されるように設計した。表2にPCR反応に用いたプライマーを挙げる。この表において、プライマー中の相互に相補的な領域を太字と下線で示した。プライマーにおいて用いた相補的領域は、20ヌクレオチドの領域であって、sigG遺伝子のヌクレオチド19から開始する。プライマーBはsigG遺伝子の開始コドンからのこの領域をコードする。プライマーBとDおよびCとEはそれぞれ部分的に相同的である。
欠失領域の安定性を欠失領域とその隣接領域における細菌の染色体DNA配列決定により確認した。PCR産物を染色体DNAから調製し、配列決定した。凍結乾燥したプライマー(Amersham Pharmacia Biotech)を100μlの水に溶解し、3μM(3pmol/μl)の希釈液をPCR反応のためのプライマーから調製した。5μM(5pmol/μl)の希釈液を配列決定のためのプライマーから調製した。
株RS303とRS310の染色体DNAからの欠失領域の配列決定に必要であったPCR産物に用いたプライマーを以下に示す。配列におけるプライマーの位置は図2に示す。

胞子形成に対するsigG欠失の効果を、胞子形成培地にてsigG−欠失枯草菌株RS303を培養し、栄養細胞を+85℃で熱処理することによって試験した。欠失の安定性はRS303を選択圧下(クロラムフェニコール5μg/ml、SS培養液)および選抜無しで培養することによってモニターした。熱処理後、細胞をLBプレートに播いた。野生株168を対照株として用い、同様に処理した。野生株168の培養液には、1ミリリットルあたり2.1x108の胞子があった。RS303は全く胞子を産生しなかった(表3)。SS培養液中で選抜無しでRS303細胞を培養した場合も胞子は産生されなかった。このことから、sigG遺伝子の欠失が細菌の胞子形成を妨げ、そしてsigG欠失は安定であると結論づけられる。
外来タンパク質の増殖培地への分泌をバシラス・リチェニフォルミス749/C細菌のβ−ラクタマーゼ遺伝子を分泌ベクターpKTH141(Lundstrom、1985)に挿入することによって調査した。このベクターにおいては、外来タンパク質の分泌はα−アミラーゼプロモーターおよびバシラス・アミロリクエファシエンスのシグナル配列によって制御される。外来タンパク質をコードする領域の所望の断片を、シグナル配列を有する正しいリーディングフレームにてHindIIIクローニングサイトに挿入することができ、それによってタンパク質が増殖培地に分泌される。挿入は以下に簡単に説明するように行った:染色体DNA(Marmur、1961)をバシラス・リチェニフォルミス細菌749/Cから単離し、PCR反応におけるテンプレートとして使用した。817塩基対長のPCR産物がPEL−141F(配列番号7)およびPEN−141R(配列番号8)プライマーによってテンプレートから増幅され、産物はいわゆる小エキソペニシリナーゼ(small exopenicillinase)をコードする領域を含み、GenBank配列V00093のヌクレオチド129から921(およびSwiss-Prot配列P00808のアミノ酸43から307)に対応していた。100μlのPCR反応混合物は、10ngテンプレートDNA、0.3μMプライマー、0.2mM dNTP、10μl酵素製造業者のバッファー、1.5μlエキスパンド酵素(3.5U/μl、Boehringer Mannheim)を含むものであった。PCRプログラムは最初の94℃4分の加熱、ついで28回の、94℃40秒、50℃20秒、72℃1分、そして最後に72℃5分からなるものであった。得られたPCR産物を精製し、プライマーに付加されたHindIII制限部位によってpKTH141に挿入し、そして産生プラスミドpRSH10を得た。プラスミドpRSH10のβ−ラクタマーゼをコードする領域を配列決定したところ、その配列は遺伝子バンクの配列と相違しなかった。枯草菌168とRS303をpRSH10で形質転換し(Gryczan et al.、1978)、それぞれ細菌株RS201とRS310を得た。
産生ベクターpRSH10による非胞子形成コンピテント株RS303の形質転換によって産生株RS310が生じた。sigG遺伝子の欠失は細菌の能力(competence)に影響を与えなかった。新しい産生株の胞子形成とsigG遺伝子の安定性をクロラムフェニコールで選抜せずに胞子形成培地でRS310株を培養することによってモニターした。胞子形成の発達を、24時間培養物をSS培地に再播種することによってさらに数増殖周期にわたってモニターした。本明細書において増殖周期とは、新しい胞子形成培地に1:100にて播種した24時間の増殖を意味する。胞子形成を3増殖周期にわたってモニターした。細胞をIからIIIの増殖から取り出して熱処理した。熱処理後、細胞をLBプレートまたはLBクロラムフェニコールプレートに播いた。
新しい非胞子形成細菌株、RS310のβ−ラクタマーゼ(BLP)産生を、産生プラスミドpRSH10で形質転換した野生株168、即ち株RS201におけるβ−ラクタマーゼの産生レベルと比較した。β−ラクタマーゼ産生は100mlの撹拌培養においてモニターした。増殖培養液のβ−ラクタマーゼ活性はO’Callaghan ら(1972)によって開発された方法に従って色素生産性基質であるニトロセフィン(nitrocefin)を用いて分光測定によって測定した。486nmでの吸光度変化を活性単位BUに変換した。BU:30℃で1時間あたりベンジルペニシリン1μmol分解する単位。変換には、実験的に決定した係数26.3を用いた。この活性単位をついで、β−ラクタマーゼμg/ml単位に変換した。この変換には340BU/μgタンパク質のβ−ラクタマーゼ比活性を用いた(Simons et al.、1978)。選抜のためにカナマイシン10μg/mlを含む濃縮LB培地を増殖培地として用いた。0.1mlの対数増殖期の細胞を100mlの増殖培養液に播種し、細胞を2lの撹拌ビンで培養した(+37℃、250rpm)。増殖を25時間にわたって吸光度(OD600nm)を測定することによってモニターした。結果を図3に示す。細菌株RS310とRS201は対数増殖期において同様の増殖速度を有しており、最高のOD600nm値は11時間目に達成された。両方の株の増殖培養液はおよそ250μg/mlのβ−ラクタマーゼを含有していた。11時間後に、細菌密度は減少し始め、培養の最後にはRS310株のOD600nm値は7.6であり、RS201株については8.1であった。しかし増殖培養液中のβ−ラクタマーゼ含量は増加し、培養の最後には新しい非胞子形成産生株RS310については430μg/ml、そしてRS201株については380μg/mlであった。さらに増殖培養液に分泌されるタンパク質のプロフィールに対するsigG遺伝子欠失の効果をSDS−PAGEゲル泳動によって調べたところ、クーマシーブルー染色したゲル(15%アクリルアミド)において非胞子形成株と野生株との相違は検出されなかった。得られた結果に基づくと、sigG遺伝子欠失は細菌増殖または分泌タンパク質産生を妨げはしないようである。

Claims (14)
- sigG遺伝子から少なくとも150ヌクレオチドが欠失し、sigGタンパク質のアミノ酸67から80をコードするsigG遺伝子の機能性領域およびアミノ酸229から248をコードするsigG遺伝子の機能性領域のそれぞれの少なくとも一部を欠失していることを特徴とし、組換えβ−ラクタマーゼを産生するよう形質転換されている非胞子形成枯草菌株であって、該非胞子形成枯草菌株におけるβ−ラクタマーゼの産生が、野生株枯草菌株におけるβ−ラクタマーゼの産生よりも多い、非胞子形成枯草菌株。
- sigG遺伝子全体が欠失していることを特徴とする、請求項1に記載の枯草菌株。
- 選抜マーカー遺伝子が欠失部位に挿入されていることを特徴とする、請求項1から2のいずれかに記載の枯草菌株。
- 選抜マーカー遺伝子の下流の少なくとも20ヌクレオチド長のオーバーラップ領域と相同的な、少なくとも20ヌクレオチド長のオーバーラップ領域が選抜マーカー遺伝子の上流に存在することを特徴とする、請求項3に記載の枯草菌株。
- 配列番号9を含むことを特徴とする、請求項4に記載の枯草菌株。
- 枯草菌株のsigG遺伝子から少なくとも150ヌクレオチドを欠失させることによって、sigGタンパク質のアミノ酸67から80をコードするsigG遺伝子の機能性領域およびアミノ酸229から248をコードするsigG遺伝子の機能性領域のそれぞれの少なくとも一部をsigG遺伝子から欠失させること、および組換えβ−ラクタマーゼを産生するよう該枯草菌株を形質転換することを含むことを特徴とする、非胞子形成枯草菌株の調製方法であって、該非胞子形成枯草菌株におけるβ−ラクタマーゼの産生が、野生株枯草菌株におけるβ−ラクタマーゼの産生よりも多い、調製方法。
- 欠失させるべき遺伝子領域の隣接領域および選抜マーカー遺伝子を含むインサートをオーバーラップ伸長によるスプライシング(SOE)技術によって調製すること、欠失させるべき枯草菌株を該インサートで形質転換すること、そして形質転換体を選抜することによる方法であり、該インサートが所望の遺伝子領域が欠失するように細菌の染色体に組み込まれることを特徴とする、請求項6に記載の方法。
- 欠失させるべき遺伝子領域の隣接領域の間に選抜マーカー遺伝子を含む直鎖状DNAインサートをオーバーラップ伸長によるスプライシング(SOE)技術により調製すること、欠失させるべき枯草菌株を該インサートで形質転換すること、そして形質転換体をスクリーニングすることによる方法であり、インサートが相同的二重組換えの結果組み込まれることにより、所望の遺伝子領域が欠失することを特徴とする、請求項7に記載の方法。
- オーバーラップ伸長によるスプライシング(SOE)技術が、少なくとも20ヌクレオチド長のオーバーラップ領域を有するプライマーを利用することを特徴とする、請求項8に記載の方法。
- 選抜マーカー遺伝子の上流に、選抜マーカー遺伝子の下流において反復する少なくとも20ヌクレオチド長のオーバーラップ領域を有する、直鎖状DNAインサートを利用することを特徴とする、請求項9に記載の方法。
- 直鎖状DNAインサート中の、欠失させるべき領域の上流の隣接領域、選抜マーカー遺伝子、および欠失させるべき領域の下流の隣接領域がすべて実質的に同じ長さである直鎖状DNAインサートを利用することを特徴とする、請求項9または10に記載の方法。
- 3kb長の直鎖状DNAインサートを利用することを特徴とする、請求項11に記載の方法。
- 組換えβ−ラクタマーゼを産生するための産生生物としての請求項1から5のいずれかに記載の非胞子形成枯草菌株の使用。
- 生物学的に調製される産物の調製方法であって、該産物を、β−ラクタマーゼをコードするDNAによって形質転換され、該DNAがそのなかで発現する請求項1から5の何れかに記載の非胞子形成枯草菌株を用いて調製し、生成した組換えβ−ラクタマーゼを回収することを特徴とする、方法。
