Description
チロシン→ドーパ→ドーパキノン→ドーパクロム→メラニン。
a) 試験物質を提供し、
b) Myo−Xを発現する細胞を提供し、
c) ミオシン−X(Myo−X)の発現または活性化を調節する試験物質の能力を測定する
段階を含んでなる、方法を提供する。
細胞中または細胞上のMyo−Xタンパク質の量を測定し、
Myo−Xタンパク質の合成における前駆体の量を測定し、または
細胞中のメラノソーム移動レベルを測定する
ことを包含する。
請求項1に記載の方法の段階a)〜c)によりメラニンの移動を調節する前記物質の能力を評価し、
Myo−Xの発現または活性化を増加させることができる物質を選択する
ことを含んでなる、皮膚または毛髪の色素沈着を高めることができる活性物質を選択する方法、および
請求項1に記載の方法の段階a)〜c)によりメラニンの移動を調節する前記物質の能力を評価し、
Myo−Xの発現または活性化を減少させることができる物質を選択する
ことを含んでなる、皮膚または毛髪の色素沈着を減少させることができる物質を選択する方法を提供する。
下記に報告される結果に対して、本発明によって用いられる植物抽出物を、それぞれ独自のコード名によって下記の対応で同定する。
L−18:Artocarpus heterophyllus種子抽出物、
L−19:Cyathea cumingii葉抽出物(極性溶媒抽出)、
L−20:Secale cereale種子抽出物(極性溶媒、特に水で抽出後、酵素加水分解)、
L−21:Thalassiosira pseudonanaの分泌物(海水中培養により入手)、
L−22:大豆種皮抽出物(水に懸濁後、酵素加水分解)、
L−23:Buddleja axillaris葉抽出物(抽出/濃縮後、凍結乾燥/滅菌)。
本発明の植物抽出物を用いたメラニン細胞および黒色腫細胞の刺激
細胞培養:
下記の報告において、用いた出発材料は、
L−18については、粉末(水性母溶液に加えた後、指示濃度に調整した)であり、
L−19については、乾燥抽出物30%での水/グリセロール混合物の溶液であり、
L−20については、乾燥抽出物5、6%での水溶液であり、
L−21については、乾燥抽出物6%での水溶液であり、
L−22については、乾燥抽出物15%での水溶液であり、
L−23については、粉末(水性母溶液に加えた後、指示濃度に調整)である。
植物抽出物L−18〜23の用量選択:
細胞を、植物抽出物および色素沈着モジュレーター(IBMXおよびナイアシンアミド)をある範囲の濃度で用いてインキュベーションした。幾つかの用量では、細胞死または細胞形態の異常変化(例えば、空胞形成)を生じた。本発明者は、最適なL−18〜23活性用量は、変更された細胞増殖または形態と関連しないことを示した。従って、これらの用量を用いて、残りの検討を行った。
正常なヒト表皮メラニン細胞(F52;p5)およびヒト黒色腫(FM55)を、L−18(0.001%)、L−19(1%)、L−20(0.5%)、L−21(10μg/ml)、LVMH−22(1%)およびL−23(0.05μg/ml)で72時間処理した。さらに、IBMXおよびNH4C1(ポジティブメラニン細胞モジュレーター)およびナイアシンアミド(ネガティブモジュレーター)を、コントロールとして用いた。目視変化は、正常なメラニン細胞では、それらの基底メラニンレベルが既に高いため特に明確ではなかった(図1)。対照的に、黒色腫細胞では、基底メラニンレベルが低いため、目視変化は明らかであった(図2)。L−19、L−20、L−21およびL−23では、正常なメラニン細胞およびFM55黒色腫細胞の両方における基底レベルと比較してメラニン含量が有意に減少した。しかしながら、L−22は、基底レベルと比較してメラニン含量の増加と関連していた。L−18は、これらの細胞では有意な変化を誘発しなかった。結果は、表1および図1および2にまとめてある。

gp100免疫染色は、ケラチン形成細胞へのメラニン移動の包括的評価およびメラニン細胞表現型モジュレーターの評価のための強力な追跡方法を提供する(Singh et al, 2008)。全L−18〜23、ホスホジエステラーゼ阻害剤IBMX(すなわち、cAMPインデューサー)およびナイアシンアミドで処理した後のEM:KC共培養の二重免疫標識(NKi/betebおよび抗細胞骨格抗体)では、すべて、ケラチン形成細胞における蛍光緑のgp100ポジティブメラニン顆粒の数が明らかに変化した(図6)。
メラニン細胞−ケラチン形成細胞の適合した共培養物(女性−39)を、既知のメラニン細胞モジュレーター(IBMX(100μM)およびナイアシンアミド(10μM))と平行してL−18(0.001%)、L−19(1%)、L−20(0.5%)、L−21(10μg/ml)、L−22(1%)およびL−23(0.05μg/ml)で24時間処理した。抗−gp100抗体(緑色)および抗−アクチン抗体(赤色)による二重免疫標識では、ケラチン形成細胞へ移動した蛍光スポットの数に明らかな変化が見られた。異なる処理群のそれぞれについて、5つの無作為に選択した顕微鏡視野(視野当たり全部で20個の細胞)から採取した移動したメラノソームの定量。値は、未刺激コントロールレベルと比較してケラチン形成細胞当たりのgp100ポジティブスポットの数の増加率として表した。平均値は、4回の独立した実験の±SEMである。**P<0.01、***P<0.001。
Myo−X(データは示さず)およびgp100に対する抗体による二重免疫標識では、EM細胞周辺および樹状突起における顕著なMyo−X発現が明らかになった。Myo−Xは、EM糸状仮足でも検出された。本発明者らは、IBMXがMyo−Xの発現および(EM樹状突起を越える黄色スポットの全数によって示唆されるように)糸状仮足の全数を増加させたことを見出した。IBMXは、未処理細胞と比較してEM糸状仮足におけるMyo−X局在化も増加した。対照的に、Myo−X発現はEMではL−18、L−19、L−20、L−21およびL−23によって有意に下方制御されたが、L−22単独によって上方制御された。EM糸状仮足中のMyo−Xの局在化は、これらの植物抽出物によっても有意に減少した。
正常なヒト表皮メラニン細胞培養物(F39−EM)を、L−18(0.001%)、L−19(1%)、L−20(0.5%)、L−21(10μg/ml)、L−22(1%)およびL−23(0.05μg/ml)で12時間処理した。コントロール:IBMX(100μM)およびナイアシンアミド(10μM)。

Claims (7)
- メラニン細胞からケラチン形成細胞へのメラニンの移動を調節する物質の能力を評価する方法であって、
a)試験物質を提供し、
b)Myo−Xを発現する細胞を提供し、
c)ミオシン−X(Myo−X)の発現または活性化を調節する試験物質の能力を測定する
段階を含んでなる、方法。 - Myo−Xを発現する細胞がメラニン細胞またはケラチン形成細胞である、請求項1に記載の方法。
- 試験物質で処理した細胞でのMyo−Xの発現または活性化を対照細胞でのMyo−Xの発現または活性化と比較することを含んでなる、請求項1または2に記載の方法。
- Myo−Xの発現または活性を調節する試験物質の能力を測定する段階が、
細胞中または細胞上のMyo−Xタンパク質の量を測定し、
Myo−Xタンパク質の合成における前駆体の量を測定し、または
細胞中のメラノソーム移動レベルを測定する
ことを包含する、請求項1〜3のいずれか一項に記載の方法。 - 請求項1に記載の方法の段階a)〜c)によりメラニンの移動を調節する前記物質の能力を評価し、
Myo−Xの発現または活性化を増加させることができる物質を選択する
ことを含んでなる、皮膚または毛髪の色素沈着を高めることができる活性物質を選択する方法。 - 請求項1に記載の方法の段階a)〜c)によりメラニンの移動を調節する前記物質の能力を評価し、
Myo−Xの発現または活性化を減少させることができる物質を選択する
ことを含んでなる、皮膚または毛髪の色素沈着を減少させることができる物質を選択する方法。 - 評価される物質が植物抽出物である、請求項1〜6のいずれか一項に記載の方法。
