Description
1)振動が横方向であるため、水中でのシステムのQファクターは非常に高い。したがって、オンラインかつ高い分解能の検出を実現することが可能である。2)Qファクターが十分に高いため、質量を実質的に減少させる必要はなく、振動周波数を比較的低く保つことができる。さらに、これらのシステムは電気機械的に作動し、電子読取り回路を用いて製作することができるために検出システムは光学的マーキングおよび読取りを必要としない。その結果、これらは独立型であり、非常に低い価格で製作される。3)標的を、装置が埋め込まれているチャネル内の付着表面に向け直すことができる。また、4本のビームの機械平衡を有するプルーフマスのおかげで、出力は量に関して直線的に変化するが、標的の付着位置に依存しない。したがって、高い濃度分解能および再現性を有する装置を得ることが可能である。4)既知の製作技法を使用して、これらの装置を微小流体チャネルおよび電子回路と一緒に生産することができる。さらに、製作に使用する独特な技法によってQファクターを高く保つことができる。
以下の表では、最近の文献における、カンチレバー型の重量測定検出器と提案したシステムとを比較する。
本発明は、横方向に振動し、微小流体チャネルの下に埋め込むことができる、生体細胞および分析物の検出を目的とする重量測定センサ装置である。この装置の主な特徴は以下のとおりである:
・高い分解能
・速い
・低費用で製作される
・低い運転費用
・より少ない試料消費
・CMOS適合性および独立型
・高い再現性
・独特であるが、既知の製作技術を用いて容易に生産することができる
・小さく、持ち運びが可能
・使い捨てまたは再利用可能
・複雑かつ高価な外部機器なしに作動が可能
・光学的マーキングおよび顕微鏡を必要としない
・細胞の検出および計数に使用できる
・分析物の検出および濃度の測定に使用できる
・フィードバック構造
・振動が横方向であるため、液体中でのQファクターは非常に高い。したがって、オンラインかつ高い分解能の検出を有することが可能である。
・Qファクターが高いため、質量を顕著に減少させる必要はなく、振動周波数を比較的低く保つことができる。低い周波数により、電子フィードバック制御および読取り回路内で作動することが可能となる。
・さらに、これらのシステムは電気機械的に作動し、電子読取り回路と一緒に製作することができるために検出システムは光学的マーキングおよび検出を必要としない。したがって、これらは単独で作動することができ、非常に低い費用で生産することができる。
・標的は、チャネル内に配置することができる付着表面に向けることができる。それに加えて、向け直しは自動的に誘導される。これにより、既存のカンチレバー型の重量測定検出器と比較して分解能が改善される。
・プルーフマスは4本のビームの機械平衡の構造を有するため、出力は直線的に変化し、また、付着位置に依存しない。したがって、高い濃度分解能および再現性を有する装置を得ることが可能である。
・既存のSOI(絶縁体上シリコン)プロセスを用いてMEMS生産技術に独特な変化を与えることによって得られる装置で使用する微小製作技法は、共振器を微小流体チャネルの下に配置し、外部の微小流体接続点を容易に統合することを可能にする。
・これは、微小流体チャネル内の分析物または細胞懸濁流体が櫛形フィンガーの間から漏れることを遮断することによって、より効率的に作動し、より高いQファクターで振動する。
・標的のより速い分析は、即時に結果を得ることを可能にし、既存の患者処置方法の成功を測定するために使用されている処置プロセスを加速する。既知の高価かつ限定的な診断および分析の方法は、医師が処置プロセス中にこれらの確認を実現することを妨げる。
・低費用の生産は診断および処置の費用を減少させ、したがって、個人、病院および研究室による製品の利用可能性を改善させる。同様に、低い運転費用は、固定費および変動費を減少させる。
・結果を得るためにわずかな試料しか必要としないこの検出装置のおかげで、外科的処置を最小限のレベルに維持することが可能となる。
・製作技法の高い再現性は、生産後の較正および品質管理のための時間および金銭に関して費用を減少させる。
・小さく、持ち運びが可能であり、使い捨てであり、また、高価かつ複雑な外部機器に依存せずに作動する能力のため、この装置を遠隔地の保健所または軍隊に統合することが非常に容易かつ実用的となる。
・これは光学的システムを必要としないため、この装置の作動に必要な固定費用がさらに減少される。

共振器構造体上に配置された微小流体チャネル(9)は、チャネルの基礎上に配置された共振器と比較して、より広い面積上に配置されている。チャネルの壁および上部は、透明なポリマーまたはPyrexから作製されている。その結果、装置の作動に補足的な技法が必要な場合に、光学的方法を適用することが可能である。光学的読取り方法および共振器を統合することによって、装置を他の応用に使用することさえも可能である。
最初に、プルーフマス(1)およびプルーフマス上に堆積させた金膜(7)を、既知の生物学的認識分子を金上に固定するプロトコルによって、標的認識のために活性化する。このプロセスによって、自身を標的の細胞または分析物に付着させる能力を有する生物剤を、金膜表面上に、選択的な様式で固定する。
細胞検出の応用では、プルーフマス(1)と静止電極(5)との間の電気的領域から生じた誘電泳動力は、プルーフマス(1)のコアに向けられ、その付着の確率を増加させることによって最小の検出可能な細胞数を減少させる。さらに、4面の横振動を実行するプルーフマス(1)および周波数の変化の量の、細胞付着の位置に対する依存性が最小限となる。

Claims (2)
- その表面上に付着した質量の変化を測定することによって分析物ならびに/または生体細胞および分析物を検出するための、微小流体チャネルの下に埋込み可能な横振動重量測定センサ装置であって、前記横振動重量測定センサ装置が、
微小流体チャネル内に配置することができる疎水性ポリマーでコーティングしたプルーフマス(1)であって、前記疎水性ポリマーがプルーフマスと液体との直接接触を妨げ、かつ、プルーフマスの電気的な動作および読取りを容易にする、プルーフマス(1)と、
前記プルーフマスの電気的な読取りおよび動作のために同じ疎水性ポリマーでコーティングした、プルーフマスに固定された櫛形フィンガー(2)であって、前記疎水性ポリマーが、振動のQファクターを液体環境内で高く保つために、伝導性の櫛形フィンガーと液体との直接接触および櫛形フィンガー間の液体の充填のどちらも防止する、櫛形フィンガー(2)と、
前記プルーフマスの電気的な読取りおよび動作のために前記疎水性ポリマーでコーティングした、静止電極(5)であって、前記疎水性ポリマーが、振動のQファクターを液体環境内で高く保つために、静止電極(5)と液体との直接接触および櫛形フィンガーと静止電極との間の液体の充填のどちらも防止する、静止電極(5)と、
前記プルーフマス(1)を機械的アース(4)に接続する折り畳まれたばねビーム(3)であって、この構造が、出力の周波数のシフトの、質量の付着位置に対する依存を最小限にする、折り畳まれたばねビーム(3)と、
