2)腐植抽出液の殺菌は、121℃、15分の熱殺菌とした。
(1)腐植土を含水率50〜80%に保って明反応させ、
(2)得られる腐植に、水および酢酸マグネシウムカルシウム水和物を混合し、発酵および熟成させることからなる。その詳細を以下に説明する。
〔実施例1〕
(工程1)
エンザイム株式会社長崎工場敷地内で約8000年間地下に埋蔵されていた腐植土を掘削し、太陽光の下、常温で約12ヶ月間、明反応を行った。この明反応により、酵素および補酵素の含有量を高めた腐植の物性を表3に示す。
工程1で得られた腐植と水とを重量比30:70の割合で混合した(腐植の含水率は50%となる)。その混合物に栄養剤として市販のスキンミルク100ppm、生理活性化剤としてCMA100ppm、微生物増殖用支持体として軽石0.1重量%を添加して、常温で3〜12ヶ月間発酵を熟成した。熟成から2〜11ヶ月間は曝気と槽内攪拌を続け、そして残る1ヶ月間は静止沈降させた。上澄液をろ紙またはカセットフィルターでろ過して、腐植抽出液を得た。腐植抽出液の物性を表4に示す。
比較(ポジティブコントロール)のために、エンザイム株式会社長崎工場敷地内で得た腐植土を用いて特開2006−273734に記載の製造方法に従って腐植抽出液を調製した。具体的には、実施例1の工程(2)において、スキンミルク、CMA、および軽石を混合しなかった以外は、実施例1と同様の手順で、腐植抽出液を調製した。この腐植抽出液の成分を表5に示す。表5の有機物、無機物ともに含有量は少量であるが、腐植抽出液は、腐植成分、ミネラル、微生物、酵素、補酵素が含まれた活性化された液といえる。
実施例1および比較例1の腐植抽出液に、大腸菌、緑膿菌、サルモネラ菌または黄色ブドウ球菌の菌液を接種後、25℃で保存し、60秒、6時間、および、24時間後、それぞれ、試験液中の生菌数を測定した。その結果を表6に示す。また、比較(ネガティブコントロール)として、試験液を精製水に変えた試験を追加した。
2)開始時:菌液接種直後の対照の生菌数を測定し、開始時とした。
この試験に先立ち予備試験を行ない、ウイルス感染価の測定法について検討した。実施例1の腐植抽出液の原液では十分な不活性化が予測できたので、注射用水を用いて腐植抽出液の5倍液を調製して、本試験を行なうことにした。さらに、細胞維持培地で作用液を100倍に希釈して感染価を測定した。また、比較(ネガティブコントロール)として、試験液を精製水に変えた試験を追加した。

Claims (3)
- 以下の工程:
(1)腐植土を含水率50〜80%に保って太陽光のもとで発酵と熟成をさせる明反応を行わせ、
(2)得られる腐植を、水および酢酸マグネシウムカルシウム水和物と混合し、発酵および熟成させる
ことからなる、腐植抽出液の製造方法。 - 工程(2)で、前記腐植に、さらにスキムミルクを混合することを特徴とする、請求項1に記載の腐植抽出液の製造方法。
- 工程(2)で、前記腐植に、さらに軽石を混合することを特徴とする、請求項1または2に記載の腐植抽出液の製造方法。
