Description
1)第1の層には、前処理として、蒸留水で洗浄を行い、乾燥させた後、UVオゾン処理を行う。
2)第1の層と第2の層を、第2の層がチップの上層となるように積層する。
3)第1の層と第2の層を積層して形成した第1の流路の狭小区間の底面部分に、分析対象物質を特異的に吸着するためのプローブを固定する。その後、非特異的吸着を抑制するためのブロッキング処理、固定されたプローブの活性を維持するためのイムノスタビライザー処理を施す。
4)第3〜第5の層にはそれぞれブロッキング処理を施す。
5)第2の層の上に、さらに第3〜第5の層を順次積層する。

Claims (6)
- 第1の流路において分析対象物質を特異的に吸着する吸着反応を行う反応方法であって、
前記第1の流路には、該第1の流路に接続する第2の流路との接続部から続く区間であって、その断面積aが前記第2の流路の断面積Aよりも小さい狭小区間が設けられており、
前記第2の流路に前記分析対象物質及び該分析対象物質に結合する標識物質を含む検体液を流し、該検体液を前記第1の流路に送液する工程と、
前記第1の流路の内圧の変化に基づいて前記検体液の後端が前記第1の流路に流入したことを検出し、該検体液の送液を停止させる工程と、
前記第1の流路に接続する前記第2の流路の接続部に合流した第3の流路に洗浄液を流し、前記第1の流路に停止する前記検体液の後端に該洗浄液を合流させる工程と、
前記検体液の後端に前記洗浄液が合流した後に該洗浄液を前記第1の流路に送液する工程と、
を備える反応方法。 - 請求項1記載の反応方法であって、
前記狭小区間の断面積aは、前記第2の流路の断面積Aの2/5〜1/300である反応方法。 - 請求項1又は請求項2記載の反応方法であって、
前記第2の流路に接続する前記第1の流路の接続部の開口が、該第2の流路の一面にあって該面のエッジから離れた位置にある反応方法。 - 第1〜第3の流路、及びこれら第1〜第3の流路の基端部にそれぞれ設けられた第1〜第3のポートを含むマイクロ流体チップと、
前記第1〜第3のポートにそれぞれ圧力を作用させ、前記第1〜第3の流路に送液する送液手段と、
前記送液手段を駆動する制御手段と、
前記第1のポートに作用する圧力を測定する圧力測定手段と、
を備え、
前記第1の流路及び前記第2の流路は、それらの先端部において互いに接続し、前記第3の流路は、前記第1の流路に接続する前記第2の流路の接続部に合流しており、
前記第1の流路は、分析対象物質を特異的に吸着する吸着反応を行い、前記第2の流路との接続部から続く区間であって、その断面積aが該第2の流路の断面積Aよりも小さい狭小区間を有しており、
前記制御手段は、前記分析対象物質及び該分析対象物質に結合する標識物質を含み前記第2の流路に流れる検体液を前記第1の流路に送液し、前記圧力測定手段から送出される測定信号に基づいて該検体液の後端が該第1の流路に流入したことを検出して、該検体液の送液を停止させると共に前記第3の流路に流れる洗浄液を該第1の流路に停止する該検体液の後端に合流させ、該検体液の後端に該洗浄液が合流した後に該洗浄液を該第1の流路に送液する反応装置。 - 請求項4記載の反応装置であって、
前記狭小区間の断面積aは、前記第2の流路の断面積Aの2/5〜1/300である反応装置。 - 請求項4又は請求項5記載の反応装置であって、
前記第2の流路に接続する前記第1の流路の接続部の開口が、該第2の流路の一面にあって該面のエッジから離れた位置にある反応装置。
