〔関連出願の相互参照〕
結腸直腸腫瘍の進行におけるEGF経路内の下流側分子の免疫組織化学染色
癌腫の病理学的病期分類と相関関係をもつバイオマーカープロファイルを結腸直腸癌について同定できるか否かを判定するために、個々の症例からの組織試料及び市販の組織アレイを用いて、結腸直腸癌内のEGFRの発現にリンクされるバイオマーカーの発現レベルを検討した。バイオマーカーは免疫組織化学(「IHC」)を用いて評価された。
インサイチュハイブリダイゼーションを用いたEGFR遺伝子コピー数と結腸直腸癌腫瘍病期分類の相関性
EGFRについての螢光インサイチュハイブリダイゼーション(FISH)を、個々の症例からの単一スライド切片及び多重組織アレイについて実施し、結腸直腸癌の進行における遺伝子の状態を評価した。
腫瘍内部のEGFR経路分子の発現レベルの不均質性
EGFR経路分子の発現レベルは、単一の腫瘍内のこれらの分子の発現レベルの考えられる不均質性を評価するべく、結腸直腸癌の事例研究において評価された。
Claims (17)
- 個体内での結腸直腸癌の進行を評価するための方法であって、2つ以上の生物学的マーカーの発現、リン酸化又は発現およびリン酸化の両方のパターンを検出するために前記個体から得た組織又は細胞試料を検定する工程を含み、
前記2つ以上の生物学的マーカーは、EGFRとPTENを含み、
前記2つ以上の生物学的マーカーの前記発現、リン酸化又は発現およびリン酸化の両方のパターンが、結腸直腸の進行病期が立証された、異なる個体由来の組織もしくは細胞試料由来の、前記2つ以上の生物学的マーカーの前記発現、リン酸化又は発現およびリン酸化の両方のパターンと比較した時に実質的に類似している、又は同じ個体由来の非結腸直腸癌組織もしくは細胞試料由来の、前記2つの生物学的マーカーの前記発現、リン酸化又は発現およびリン酸化の両方のパターンと異なる、方法。 - 前記2つ以上の生物学的マーカーがpAKT、pMEK、pERK、Ki67およびpHER1からなる群より選択される少なくとも1種をさらに含む、請求項1に記載の方法。
- 標識された核酸ベースのプロープを用いてEGFRをコードするゲノムDNAの増幅、欠失又は再配列を測定する工程をさらに含む、請求項1又は2に記載の方法。
- 小腺腫期での個体内の結腸直腸癌の進行を評価する、請求項1〜3のいずれか1項に記載の方法。
- 2つ以上のマーカーがpMEKをさらに含み、前記個体から得た前記組織又は細胞試料由来の前記2つ以上の生物学的マーカーの前記発現、リン酸化又は発現およびリン酸化の両方のパターンが、非腫瘍組織又は細胞試料内で発現、リン酸化又は発現およびリン酸化されるものよりも高いEGFRの発現、リン酸化又は発現およびリン酸化の両方、それよりも高いPTENの発現、リン酸化又は発現およびリン酸化の両方、それよりも低いpMEKの発現、リン酸化又は発現およびリン酸化の両方を含む、請求項4に記載の方法。
- 標識された核酸ベースのプローブを用いてEGFRをコードするゲノムDNAの増幅、欠失又は再配列を測定する前記工程をさらに含み、前記個体から得た前記組織又は細胞試料内の前記増幅、欠失又は再配列の度合が安定な二染色体性である、請求項4又は5に記載の方法。
- 腺癌期での個体内の結腸直腸癌の進行を評価する、請求項1〜3のいずれか1項に記載の方法。
- 前記個体から得た前記組織又は細胞試料由来の前記1つ以上の生物学的マーカーの発現、リン酸化又は発現およびリン酸化の両方のパターンが、小腺腫組織又は細胞試料内で発現、リン酸化又は発現かつリン酸化されるものよりも低いPTENの発現、リン酸化又は発現およびリン酸化の両方、又はそれよりも大きいpMEKの発現、リン酸化又は発現およびリン酸化の両方が含まれる、請求項7に記載の方法。
- 標識された核酸ベースのプローブを用いてEGFRをコードするゲノムDNAの増幅、欠失又は再配列を測定する前記工程をさらに含み、前記個体から得た前記組織又は細胞試料内の前記増幅、欠失又は再配列の度合が安定な二染色体性および安定な三染色体性である、請求項7又は8に記載の方法。
- 悪性腺癌期での個体内の結腸直腸癌の進行を評価する、請求項1〜3のいずれか1項に記載の方法。
- 前記個体から得た前記組織又は細胞試料由来の前記2つ以上の生物学的マーカーの前記発現、リン酸化又は発現およびリン酸化の両方のパターンが、小腺腫組織又は細胞試料内で発現、リン酸化又は発現かつリン酸化されるものよりも低いPTENの発現、リン酸化又は発現およびリン酸化の両方、又はそれよりも大きいpMEKの発現、リン酸化又は発現およびリン酸化の両方が含まれ、かつ標識された核酸ベースのプローブを用いてEGFRをコードするゲノムDNAの増幅、欠失又は再配列を測定する前記工程をさらに含み、前記個体から得た前記組織又は細胞試料の遺伝子形態が安定な二染色体性および安定な多染色体性である、請求項10に記載の方法。
- 標識された核酸ベースのプローブを用いてEGFRをコードするゲノムDNAの増幅、欠失又は再配列を測定する前記工程をさらに含み、前記個体から得た前記組織又は細胞試料の前記増幅の度合が安定な二染色体性および安定な多染色体性である、請求項10又は11に記載の方法。
- 前記検定する工程が、標識された特異的結合試薬を用いた染色後の組織切片のコンピューター援用画像解析を含む、請求項1〜12のいずれか1項に記載の方法。
- 2つ以上の生物学的マーカーの発現、リン酸化又は発現およびリン酸化の両方を検出するための少なくとも3つの試薬を含み、
前記2つ以上の生物学的マーカーは、EGFRとPTENを含む、個体内の結腸直腸癌の進行を評価するためのキット。 - 前記2つ以上の生物学的マーカーがpAKT、pHER1、pMEK、pERKおよびKi67からなる群より選択される少なくとも1種をさらに含む、請求項14に記載のキット。
- 前記2つ以上の生物学的マーカーがpMEKをさらに含む、請求項14に記載のキット。
- EGFR遺伝子増幅、欠失又は再配列の検出のための試薬をさらに含む、請求項14〜16のいずれか1項に記載のキット。
