Description

本文に用いられる用語「C1-C20 アルキル」は、非置換の、または1以上の置換基により置換されてよい1〜20個の炭素原子を含む直鎖または分岐鎖飽和炭化水素を表す。C1-C20アルキル基の例には、メチル、エチル、プロピル、2-プロピル、n-ブチル、イソブチル、tert-ブチル、ペンチル等が含まれる。同様に、用語「C1-C20エーテル」及び「C1-C20エステル」は、非置換の、または1以上の置換基により置換されてよい1〜20個の炭素原子を含む化合物を表す。
図1を参照されたい、図1は、本発明においてフラボノイド化合物を製造する全合成方法を示すフローチャートである。
この態様では、次の化合物を合成するための半合成方法を説明する:化合物8a:プロトフラボノン(IIUPAC名称:2-(1-ヒドロキシ-4-オキソシクロへキサ-2,5-ジエニル)-4H-クロメン-4-オン)と、化合物10a:5-ヒドロキシプロトフラボン(IIPAC名称:2-(1-ヒドロキシ-4-オキソシクロへキサ-2,5-ジエニル)-5-ヒドロキシ-4H-クロメン-4-オン)と、化合物15a:β-ナフトフラボノン(IIPAC名称:3-(1-ヒドロキシ-4-オキソシクロへキサ-2,5-ジエニル)-1H-ベンゾ[f]クロメン-1-オン)と、化合物18a:5-ヒドロキシ-7-メトキシプロトフラボノン(IIPAC名称:5-ヒドロキシ-2-(1-ヒドロキシ-4-オキソシクロへキサ-2,5-ジエニル)-7-メトキシ-4H-クロメン-4-オン)。
細胞組織の培養
ヒト肝癌細胞(HepG2、Hep3B)、乳癌細胞(MCF-7、MDA-MB-231)及び肺癌細胞(A549)を10%(v/v) FBS、100units/mL ペニシリン及び100μL/mL ストレプトマイシンを補充したRPMI-1640 培地中で、37℃で5%二酸化炭素及び95%空気の加湿した雰囲気下で培養した。3日ごとに培地を交換しコンフルエントになるまで培養し、細胞がコンフルエントになるまで継代培養し、実験に用いた。実験の期間は目的に応じて決定される。
この実験では、MTT染色法を利用して細胞障害性を測定した。細胞を5×103から1×104細胞/ウェルの密度で96 ウェル組織培養プレート中に播種した。2日目に、細胞をさらに72時間試験化合物を用いて試験した。薬物処理の後、付着細胞をMTT (0.5 mg/mL) で1時間インキュベートし、続いてDMSOで可溶化した。ついで550 nmでの吸光度を、マイクロプレートリーダーで測定する。
結果:表1に示すとおりである。

- 前記フラボノイド化合物が、全合成又は半合成方式のいずれか1である化学的方法により得られる請求項1記載の医薬組成物。
- 哺乳動物の疾病の治療に用いられる請求項1記載の医薬組成物。
- 前記疾病が癌である請求項3記載の医薬組成物。
- 前記哺乳動物がヒトである請求項4記載の医薬組成物。
