哺乳動物体液としてヒト末梢血、細胞分離材としてHES、顆粒球吸着材として部分鹸化酢酸セルロースビーズを用いて以下の操作を行った。
酢酸セルロースをジメチルスルホキシドとプロピレングリコールの混合溶剤に溶解し、この溶液を特開昭63−117039に記載された方法(振動法)により液滴化し、凝固させて酢酸セルロースの球形の粒子を得た。得られた球形粒子の粒径は、約300μmであった。この粒子を沈降体積比で、0.1規定水酸化ナトリウム水溶液と1:1(V/V)の割合で混合し、30分間接触させることにより、部分加水分解反応を行った。吸着体沈降体積の約200倍量の蒸留水にて洗浄後、生理食塩水液(最終濃度で5IU/mlのヘパリンを含む)にて置換を行い、部分鹸化酢酸セルロースビーズを得た。このときの部分鹸化酢酸セルロースビーズの鹸化度は、47.5%(置換率は52.5%)であった。
健常人ボランティアから末梢血を採取した。このものは、1mlあたり赤血球を4.3×109個、白血球を4.6×106個(そのうち単核球が2.1×106個、顆粒球が約2.3×106個)が含まれるものであった。
(1)で調製した沈降体積1mlの部分鹸化酢酸セルロースビーズに(2)で調製した末梢血を2ml加え、37℃、1時間、20rpmで撹拌しながらインキュベートした。この操作を吸着材処理とした。吸着材処理後、細胞分離剤として400μl(全量に対してHES濃度が1%となる量)のHES40(NIPRO社製、6%HES含有0.9%食塩液)を加え、50gで5分間遠心して、上清を回収し、これを処理区1とした。また、吸着材処理後、1500μl(全量に対してHES濃度が2.57%となる量)のHES40を加え、50gで5分間遠心して、上清を回収し、これを処理区2とした。さらに、吸着材処理後、RPMI1640培養液(Gibco)を2ml加え、800μl(全量に対してHES濃度が1%となる量)のHES40を加え、50gで5分間遠心して、上清を回収し、これを処理区3とした。
対照区2〜4においては、赤血球除去率は全て98%以上と優れていたが、顆粒球除去率は最も高い対照区2で64%に留まり、対照区3,4では30%程度であった。
哺乳動物体液としてブタ骨髄液、細胞分離剤としてHES、顆粒球吸着材として部分鹸化酢酸セルロースビーズを用いて以下の操作を行った。
酢酸セルロースをジメチルスルホキシドとプロピレングリコールの混合溶剤に溶解し、この溶液を特開昭63−117039に記載された方法(振動法)により液滴化し、凝固させて酢酸セルロースの球形の粒子を得た。得られた球形粒子の粒径は、約300μmであった。この粒子を沈降体積比で、0.1規定水酸化ナトリウム水溶液と1:1(V/V)の割合で混合し、30分間接触させることにより、部分加水分解反応を行った。吸着体沈降体積の約200倍量の蒸留水にて洗浄後、生理食塩水液(最終濃度で5IU/mlのヘパリンを含む)にて置換を行い、部分鹸化酢酸セルロースビーズを得た。このときの部分鹸化酢酸セルロースビーズの鹸化度は、47.5%(置換率は52.5%)であった。
ミニブタ(クラウン、30kg)の腸骨より骨髄液を採取した。このものは、1mlあたり赤血球が5.0×109個、白血球が1.0×107個(そのうち単核球画分細胞が3.0×106個、顆粒球が5.0×106個)含まれるものであった。
(1)で調製した沈降体積1mlの部分鹸化酢酸セルロースビーズに(2)で調製したブタ骨髄液を2ml加え、37℃、1時間、20rpmで撹拌しながらインキュベートした。この操作を吸着材処理とした。吸着材処理後、400ul(全量に対してHES濃度が1%となる量)のHES40(NIPRO社製、6%ヒドロキシエチルデンプン加0.9%食塩液)を加え、50gで5分間遠心して、上清を回収し、これを処理区4とした。また、吸着材処理後、1500ul(全量に対してHES濃度が2.57%となる量)のHES40を加え、50gで5分間遠心して、上清を回収し、これを処理区5とした。さらに、吸着材処理後、RIMI1640培養液(Gibco)を2ml加え、800ul(全量に対してHES濃度が1%となる量)のHES40を加え、50gで5分間遠心して、上清を回収し、これを処理区6とした。
対照区6〜8では、赤血球は除去できるものの、顆粒球画分除去は3〜20%程度であったのに対し、処理区4〜6では赤血球、顆粒球共に80%以上であり、処理区5、6では顆粒球画分除去率が90%以上であった。
Claims (6)
- 単核球、顆粒球および赤血球を含有する哺乳動物の体液からの、単核球を含む細胞画分の調製法であって、
前記体液をアセチル基によって置換されている水酸基の割合が全水酸基の35〜53%である酢酸セルロースである顆粒球吸着材と接触させる工程Aと、
工程Aによる処理中または処理後の試料への、単核球と赤血球とを分離することができるHES(ヒドロキシエチル澱粉)である細胞分離剤の添加を行う工程Bとを含む、単核球を含む細胞画分の調製法、あるいは
前記体液への、単核球と赤血球とを分離することができるHES(ヒドロキシエチル澱粉)である細胞分離剤の添加を行う工程Cと、
工程Cによる処理後の試料をアセチル基によって置換されている水酸基の割合が全水酸基の35〜53%である酢酸セルロースである顆粒球吸着材と接触させる工程Dとを含む、単核球を含む細胞画分の調製法。 - 工程Aまたは工程Dが、顆粒球吸着材が充填されたカラムに前記体液または工程Cによる処理後の試料を通過させるか、顆粒球吸着材と前記体液または工程Cによる処理後の試料とを同一容器内で接触させることにより行われる請求項1記載の方法。
- 工程Aと工程B、あるいは工程Cと工程Dとが同一容器内で行われる、請求項1記載の方法。
- 単核球、顆粒球および赤血球を含有する哺乳動物の体液から、単核球を含む細胞画分を調製するための装置であって、
アセチル基によって置換されている水酸基の割合が全水酸基の35〜53%である酢酸セルロースである顆粒球吸着材を収容してなる第1手段と、
単核球と赤血球とを分離することができるHES(ヒドロキシエチル澱粉)である細胞分離剤を収容してなる第2手段とを備える、前記装置。 - 第1手段と第2手段とが液体連通している、請求項4記載の装置。
- 単核球、顆粒球および赤血球を含有する哺乳動物の体液から、単核球を含む細胞画分を調製するための装置であって、
アセチル基によって置換されている水酸基の割合が全水酸基の35〜53%である酢酸セルロースである顆粒球吸着材と、単核球と赤血球とを分離することができるHES(ヒドロキシエチル澱粉)である細胞分離剤とを収容してなる手段を備える、前記装置。
