Description
図1は主要部が4つのコンポーネント、すなわち、レーザ走査型顕微鏡検査のための励起光を生成する光源モジュール2、励起光をコリメートして試料上の走査のため然るべき偏向を行なう走査モジュール3、走査モジュールによって用意された走査ビームを顕微鏡光路内で試料の方向に向ける顕微鏡モジュール4および試料からの光線を受け止め検出する検出モジュール5から成るレーザ走査型顕微鏡1の模式図である。その場合検出モジュール5は、図1に描かれているように、スペクトル別のマルチチャネル型に構成することができる。
光源モジュール2は、レーザ走査型顕微鏡検査に適した照明光、したがって特に蛍光を誘起し得るビームを生成する。適用法に対応させるため、光源モジュールは当目的用に複数の光源を有している。図示された実施態様では光源モジュール2に2つのレーザ6および7が配備されている。それらの後にはそれぞれ光バルブ8および減衰器9が接続されており、それらはビームを結合ポイント10を通じて光ファイバ11に連結させている。光バルブ8は、レーザユニット6または7のレーザ自体の作動を遮断しなくてもビームを遮断させることのできるビーム偏向器として機能する。光バルブ8は、例えば、ビーム遮断のためレーザビームを光ファイバ11へ連結する手前で、図示されていない光の落下方向に偏向させるAOTF(音響光学フィルタ)として形成されている。
光ファイバ11に通されたビームは、移動式のコリメーション光学系12および13によりビーム結合ミラー14、15を通じて合一化され、ビーム形成ユニット内でビームの特性が変更される。
図1に矢印で示されたモータによるズーム光学系41の自由移動度は、結像倍率、焦点位置、ひとみ位置の3つのパラメータの適合化に想定されている自由度の数値に精確に一致している。その出力側のひとみに固定式絞り42の配置されたズーム光学系41が特に好ましい。絞り42を実地で簡単に実現するには、スキャナ18の鏡面幅の制限によって構成することもできる。ズーム光学系41を持つ出力側絞り42により、ズーム倍率の設定如何に拘わらず常に一定したひとみ直径を走査対物レンズ19上に結像させることが可能になる。このように、ズーム光学系41を任意に設定した場合でも対物レンズのひとみは依然として完全に照らし出されたままである。スキャナ18の領域での願わしくない散乱光発生事態の阻止には独自型絞り42の使用が好ましい。
共鳴スキャナに関しては、例えばPawley著“Handbook of Biological Confocal Microscopy”Plenum Press社1994年刊、461ページ以降に記述されている。スキャナがフィールドを、光軸に対し、すなわち走査ミラーの定常位置に対し非対称に走査するように、スキャナの制御を行えば、ズーム作用との関連性から選定ROI領域の補正移動OFが得られる。既に触れたスキャナ18のでスキャン作用およびズーム光学系41の再通過により、検出器方向への検出光路における観察対象領域ROIの選択が改めて中断される。それにより、走査画像SF内のROI領域に対し任意の選択が可能になる。様々なROI領域の選択毎に画像を追加獲得することができ、それらを高分解性画像に合成することができる。
これは、スプリッタTにより蛍光励起のための光源LQ1と合一化させることができる。両光源は円筒型光学系ZLを通じラインとして試料上に結像する。その場合、LQ1は試料上/中に均一ラインを、LQ2は格子Gの作用により周期的に変調されたラインを生成する。
スキャナP2は試料PRに沿って照明光線を移動させるのに用いられる。そのほか、走査光学系SO、鏡筒レンズTL、対物レンズLが共通の光路に、ピンホール光学系PO、フィルタおよび検出器またはスリット絞りが検出光路に配置されている。
LQ1およびLQ2は、試料照射するLQ1、LQ2の同期化のための制御ユニットと結合している。
この干渉性フィールドを試料へ例えば線形で結像させれば、Z方向にタルボット構造(文献ではタルボット効果)が発生する。このタルボット効果はコヒーレント光が周期dの周期性平坦構造で回折した場合に出現する。元の構造を持つ画像が
Zn=2nd2/λ
の間隔で現われる。この場合、構造は奇数倍ごとにオリジナル構造の周期dの丁度半分ずつシフトする(文献 : Lexikon der Optik/Spektrum Akademie刊、1999年、ベルリン)。なお、λは波長およびnはタルボット平面の整数倍を表わしている。図6の部分図a)にはいわゆるタルボット平面(xに沿う方向)が図解されている。+/-1および0の回折次数の重畳により、タルボット長atの間隔にしてちょうどpi/2の構造シフトが生じる。
△z=πa2/2λから得られる。ただし、aは対物レンズの横方向の分解能である。格子Gの格子定数は、好ましくは、試料においてd=√πa/2の周期的構造が得られるように選択する。この場合タルボット平面の間隔は顕微鏡対物レンズの深度分解能に等しい。
光源LQ2は格子と組み合わせれば、蛍光過程の抑制に有用である
(文献 : S. W. HellおよびJ. Wichmann“Opt. Lett.”第19巻、780号、1994年)。
過疎分布のメカニズムとしては、例えば刺激による放出(文献 : A. Klar、M. DybaおよびS. W. Hell“Appl. Phys. Let.”第78巻第4号、393ページ、2001年)、基底準位における過疎分布化または色素の様々な放出状態/吸収状態への照準的切換が考えられる。LQ1のビームは蛍光励起に用いられる。
刺激放出の場合、これは黒塗りストライプ領域における刺激的放出を手段とする色素分子の照準的鎮静化によって行われる。そのようにして、比較的高い蛍光強度を持つ領域(非鎮静化領域)と低い蛍光強度を持つ領域(鎮静化領域)が生じる。強度の高い領域を検出することにより撮像分解能を高めることができる。そのためには、光源LQ1およびLQ2はパルス化するのが有利である。LQ1からのパルスによって先ず色素が励起される。蛍光の寿命(ナノ秒領域)が続いている間は、LQ2の光分配により蛍光分子の鎮静化が行われる。その後、励起された残留蛍光分子の自然放出により蛍光光子の検出が行われる。それに続き、改めてLQ1のパルスにより新たなサイクルで均一に励起することができる。

有機体の生育
記述の本発明は、なかでも1/10秒から時間レベルまでのダイナミックな変遷を特徴とする生育過程の研究に適している。ここでは細胞結合面および有機体全体への適用例について記述する :
・ Abdul-Karin、M. A.他は2003年“Microvasc. Res.”第66巻、113〜125ページに動物生体における血管の変化に関する長期分析結果を記録した。その場合、蛍光画像は数日間隔で撮影された。運動の定角軌道を模式的に描くために、3次元のデータ記録が適合アルゴリズムで評価されている。
・ Soll、D. R.他は2003年“Scientic World Journ.”第3巻827〜841ページに3次元空間全体における生体細胞の核および偽足に関する顕微鏡データのソフトウェアベースによる運動分析について記述している。
・ Grossmann, R他は2002年“Glia”第37巻229〜240ページにラットの微小神経膠細胞における運動の3次元分析について記述している。そのデータは10時間以上に亘って記録されたものである。神経膠細胞にはトラウマ性傷害の後に同時に迅速反応が発現するので、高いデータ収得率およびそれ相応のデータ量が得られる。
これに関しては特に次のことが重要なポイントである :
・ その隣接細胞がレーザ照明に敏感に反応するので3次元ROI照明から保護されねばならない3次元領域での生細胞の分析
・ 例えば、FRET実験などにおいて、3次元のレーザ照準照明下で退色するマーカーによる生細胞の3次元領域での分析
・ 例えば、3次元FRAP、FLIP実験などにおいて、レーザ照準照明下で退色する、同時にROI外の観察も必要なマーカーによる生細胞の3次元領域での分析
・ 例えば3次元伝達物質の活性化など、レーザ照明下での操作原因により変化するマーカーおよび薬剤による生細胞の3次元領域での照準分析
・ 例えば、paGFP、Kaedeなど、レーザ照明下での操作原因により変色するマーカーによる生細胞の3次元領域での照準分析
・ 例えば、コンフォーカル性と検出感度との最適バランスが要求される微弱マーカーによる生細胞の3次元領域での照準分析
・ 例えばCFP、GFP、YFP、DsRed、HcRedなど可変性多重マーキングのなされた3次元組織結合における生細胞
・ 機能に依存して変色する、例えば、Ca+マーカーなどでマーキングのなされた3次元組織結合における生細胞
・ 生育に起因して変色するマーキングのなされた3次元組織結合における生細胞、例えばGFPによる形質転換動物

記述の本発明は細胞内運搬過程の研究にはこの上なく適している。この場合では正しく非常に微小な運動構造体、例えば、タンパク質を高速度で(殆どが1/100秒の領域)描写しなければならないからである。複雑な運搬過程のダイナミックスを捕捉するためには、ROIブリーチングを伴うFRAPもしばしば適用される。そのような研究例として、ここでは以下のものを挙げておく :
・ Gimpl, G.他が2002年“Prog. Brain Res.”第139巻43〜55ページに、ROIブリーチングによる実験、運動性分析のための蛍光撮像およびGFPマーキングされたオキシトシン受容体の線維芽細胞内での分布について記述している。その場合、空間位置設定、分解能およびブリーチングと撮像との直接的な時間的連続性に関して高い要求が課されている。
・ Zhang他が2001年“Neuron”第31巻261〜275ページに、GFP変換された神経細胞における生細胞の撮像について記述している。その場合、顆粒の運動がブリーチングと蛍光撮像との組み合せにより分析された。神経細胞のダイナミックスに起因して、撮像速度には高い要求が課される。
記述の本発明は、特に分子間およびその他副細胞間の相互作用の描写に適している。これらの場合では非常に微小な構造が高速度(1/100秒レベル)で描出されねばならない。相互作用に必要な分子の空間ポジションの解明には、例えばROIブリーチングを伴うFRETなどの間接的な技術を使用することもできる。適用される例として、ここでは以下のものを挙げておく :
・ Petersen, M. A.およびDalley, M. E.が2004年“Glia”第46巻195〜206ページに、ラット海馬角の培養における2チャネル撮影について記述している。この場合は、マーカーとしてのレクチンとシトックスについて3次元空間において長時間に亘り2チャネルで記録される。
・ Yamamoto, N.他が2003年“Clin. Exp. Metastasis”第20巻633〜638ページに、ヒトの線維肉腫細胞の2色撮影について記述している。この場合では緑色と赤色の蛍光タンパク質(GFPおよびRFP)が同時にリアルタイムで観察された。
・ Bertera, S.他が2003年“Biotechniques”第35巻718〜722ページに、合成後色が緑から赤に変化するタイムレポータプロテインによってマーキングされた転換マウスのマルチカラー撮影について記述している。撮像は生体動物の組織内3次元空間で迅速シリーズとして行われる。
記述の本発明は、殆どが極端に迅速になる信号伝達過程の研究には他に抜きん出て適している。殆どが神経生理学に関するこの過程では経時的分解能に最大限の要求が課される。それは、イオンによって媒介される活動が1/100秒から1/1000秒以下の範囲で起こるからである。筋肉系または神経系の検査への適用例として、ここでは次のものを挙げておく :

・ Schmidt H他が2003年“J Physiol”第551巻13〜32ページに、転換マウスの神経細胞突起におけるCa+イオンの分析について記述している。変化するCa+に結合するタンパク質による、マウス内での迅速なCa+変化状況についての研究は、高分解能を持つ共焦点顕微鏡により初めて行うことができた。それは、神経細胞内におけるCa+活動の位置確認およびその精確な経時的動力学が重要な役割を果たしているからである。

Claims (21)
- 試料が、第1および第2の照明光によって照明され、その場合、第1照明光が試料の励起を誘起し、第2照明光が試料内で照射横方向および照射軸方向に空間的な周期性構造を有し、該第2照明光が試料内で少なくとも3つのコヒーレント・ビームの干渉によって生成される、ライン走査式のレーザ走査型顕微鏡。
- 第1および第2のレーザ光パターンが、試料で立体的に重なり合い、試料内第2照明光の領域内で励起状態の切換がなされる、請求項1に記載の顕微鏡。
- 第1および第2の照明光がレーザビームである、請求項1又は2に記載の顕微鏡。
- 第1および第2の照明光が共同で試料に沿って動かされる、請求項1乃至3のいずれか1項に記載の顕微鏡。
- 照明が、試料に対してライン状になされる、請求項1乃至4のいずれか1項に記載の顕微鏡。
- 第2照明光の構造が、複数の回折次数のコヒーレント重畳により干渉模様として生成される、請求項1乃至5のいずれか1項に記載の顕微鏡。
- 回折次数が、格子にて生成される、請求項1乃至6のいずれか1項に記載の顕微鏡。
- タルボット格子効果が生成される、請求項1乃至7のいずれか1項に記載の顕微鏡。
- 顕微鏡対物レンズの深度分解能が、タルボットラインの間隔領域内にある、請求項1乃至8のいずれか1項の1つに記載の顕微鏡。
- 少なくとも1つの照明光がパルスレーザである、請求項1乃至9のいずれか1項に記載の顕微鏡。
- 請求項1乃至10のいずれか1項に記載された顕微鏡の動作方法であって、試料内で過疎分布化及び励起が交互に行われるように該顕微鏡が動作される、方法。
- 過疎分布化の後に励起が行われる、請求項11に記載の方法。
- 励起後に過疎分布化が行われる、請求項11に記載の方法。
- 第1、第2照明光の生成のために2つのパルスレーザが同期化されて、試料内で励起と過疎分布化が交互に行われる、請求項11乃至13のいずれか1項に記載の方法。
- 光学分解能が周期的構造の周波数の変更によって調整できる、請求項11乃至14のいずれか1項に記載の方法。
- 顕微鏡対物レンズの交換時に、第2照明光の構造形成装置が交換される、請求項11乃至15のいずれか1項に記載の方法。
- 第2照明光内の格子が交換される、請求項11乃至16のいずれか1項に記載の方法。
- 生育過程の研究のための方法であって、請求項1乃至10のいずれか1項に記載された顕微鏡を用いて細胞群および有機体全体のレベルにおいて1/10秒から時間単位までのダイナミックなプロセスを研究することを含む方法。
- 細胞内運搬過程の研究のための方法であって、請求項1乃至10のいずれか1項に記載された顕微鏡を用いて微小な運動構造体を高速度で描写することを含む方法。
- 分子および細胞内の相互作用の描写のための方法であって、請求項1乃至10のいずれか1項に記載された顕微鏡を用いて非常に微小な構造を高速度で描写することを含む方法。
- 迅速な信号伝達過程の表現のための方法であって、請求項1乃至10のいずれか1項に記載された顕微鏡を用いて筋肉系または神経系における高い経時的分解能による神経生理学過程を表現することを含む方法。
