Description
本発明は、組織の封止剤(sealant)および止血剤として用いることができる血漿濃縮物を調製する装置および方法に関する。血漿濃縮物は、細胞から遊離していることが好ましい。
血液は分画することができ、血液の異なるフラクションは、それぞれ異なる医学的用途に使用することができる。重力または遠心力の影響下においては、血液は自然に三層に分離する。平衡状態では、最上層の低密度層は、血漿と呼ばれる淡黄色の透明な液体である。血漿は、塩、代謝産物、ペプチド、および小さな蛋白質(インスリン)から非常に大きな蛋白質(補体成分)までの多くの蛋白質の水溶液である。
本発明の使い捨て可能な装置は、細胞フリーの血漿画分から非常に貴重な天然の(autologous)血漿濃縮物を調製するのに適している。
用語「分離重力」は、上記濃縮ゲルビーズの表面から血漿濃縮物を分離し、分離された結晶濃縮物に上記フィルターを通過させるのに十分な遠心力をいう。
導水管18が減圧されたときに、濃縮物出口16まで液体濃縮物(図示せず)の大部分を引き込むために、陥凹部42の中に伸長している。
Claims (11)
- 濃縮チャンバーと、上記濃縮チャンバー内の多数の濃縮ゲルビーズと、フィルターと、攪拌機とを備える血漿濃縮装置であって、
上記攪拌機は、上方攪拌端部および下方攪拌端部を備える攪拌軸と、上記下方攪拌端部から外側に広がる攪拌刃と、上記濃縮チャンバー内に配置され、上記攪拌機の中心軸の周りの回転運動と、上記中心軸に沿った往復運動とが可能なように支持された上記下方攪拌端部と、を備え、
上記濃縮装置は、上記攪拌軸の上端が通過する上方開口部を持つ天井と、上記フィルターが配置される下方開口部とを備えることを特徴とする血漿濃縮装置。 - 上記濃縮チャンバーは、円筒形の内壁と、攪拌刃とを有しており、当該攪拌刃は、上記内壁に近接する外刃とを有しており、当該外刃と上記内壁との間の間隔は、上記ゲルビーズの直径以下の間隔であることを特徴とする請求項1に記載の血漿濃縮装置。
- 上記濃縮装置の上記上方開口部が、上記濃縮装置の天井から上記濃縮チャンバー内に伸びるストップスリーブを備えており、上記ストップスリーブは、下方接合部表面を有し、上記攪拌軸は、上記ストップスリーブの直径よりも大きい径を持つ停止突出部を有し、上記停止突出部の上方の表面には、上記ストップスリーブの上記下方接合部表面と接触したときに、上記攪拌機の軸上方への動きを止めるように接合部表面が配置されて構成されていることを特徴とする請求項1に記載の血漿濃縮装置。
- 上記フィルターが、上方表面を有し、上記攪拌刃が、下方部を有している血漿濃縮装置であって、
上記下方部は、上記フィルターの上記上方表面と接触し、上記攪拌機が回転している間は、上記上方表面を清掃し、上記攪拌機が上記攪拌機の中心軸に沿って下方へ動いている間は、上記上方表面に押し付けられるように配置され、上記攪拌機が往復運動している間は、上記攪拌刃の下方への動きが、上記フィルターの上記上方表面が備える接合部によって抑制されることを特徴とする請求項3に記載の血漿濃縮装置。 - 上記フィルターが、周囲の重力条件下において血漿の流れを効果的にブロックし、遠心力として分離重力をかけた場合に、自由に血漿濃縮物を通すことができることを特徴とする、請求項1に記載の血漿濃縮装置。
- 上記分離重力が10g以上であることを特徴とする請求項5に記載の血漿濃縮装置。
- 血漿濃縮物の出口となる導水管と、上記フィルターとつながっている上方開口部を備え、上記フィルターを通過した血漿濃縮物を受けるように配置される血漿濃縮物貯蔵部とを持つ血漿濃縮装置であって、傾斜した床を有し、当該床の最も低い端部に貯留槽を有し、上記導水管の一端が上記貯留槽と連結されていることを特徴とする請求項1に記載の血漿濃縮装置。
- 血漿中のフィブリノゲンを有意に変性させることなく水を除去することによって血漿を濃縮する方法であって、以下のステップを含むことを特徴とする方法。
a)上記血漿を多数の脱水濃縮ゲルビーズおよび攪拌機を備えた濃縮チャンバー内に導入するステップ。
b)下記のi)およびii)を行う間に、水を、上記血漿の蛋白質濃度が未処理の血漿の蛋白質濃度の1.5倍以上になるまで、上記血漿から除去するステップ。
i)上記ビーズが上記血漿から水を吸収している間、血漿の局在化を低減するために、上記ビーズを攪拌するように上記攪拌機を動かすこと。
ii)上記攪拌中に形成されるビーズの凝集を解くために上記攪拌機を動かすこと。
d)上記ビーズから上記血漿濃縮物の本質的部分を分離するのに十分な遠心力を、濃縮された血漿にかけるステップ。 - 上記濃縮チャンバーが、攪拌機の下方端部から外側に広がる攪拌刃を有する攪拌機を備えており、上記攪拌機は、上記攪拌機の中心軸の周囲を回転でき、上記攪拌機の中心軸に沿って往復運動できるように支持されており、
