Description
本出願は、1996年11月29日出願の米国仮特許出願第60/032,325号に対する優先権を主張している1997年11月26日出願の米国特許出願第08/978,756号の一部継続である2000年8月4日出願の米国特許出願第09/631,906号の一部継続である2002年7月22日出願の米国特許出願第10/200,682号に対する優先権を主張し、その各々は、参照として、その全体が本明細書で援用される。
本発明は一般に、支持流動性二重層およびそれらを選択された領域に限定する方法に関する。より具体的に、本発明は、独立して処理できる支持流動性二重層膜のミクロ組立てアレイ、それらの使用および製造法に関する。

この数年来、数百万とまでは行かなくても、数千種の化合物の所望の1つの活性または複数の活性に関するスクリーニングを促進するための多量原料スクリーニング法が多数開発されている。このような方法は、典型的にはレセプターに対する潜在的に有効な化合物の結合検出に基づいている。これらの結合アッセイは、所望の活性を有し得る化合物の領域を制限する点では有効であるが、この活性をいくらかでも詳細に評価するためには、一般に十分に好適というわけではない。
一態様において、本発明は、表面検出器アレイデバイスを含む。前記デバイスは、1つ以上の二重層バリア領域によって分離された複数の異なった二重層適合性面領域を規定している表面を有する基体、前記基体表面を覆っている大量の水相、各二重層適合性面領域上に維持された脂質二重層空間、および各二重層適合性面領域と、対応する脂質二重層空間との間に挟まれた水性薄膜を含む。一般的な好ましい実施形態において、二重層適合性面領域と二重層バリア面領域とは、異なる材料から形成される。
・ 複数の脂質二重層適合性領域を有し、各適合性領域が1つ以上の二重層バリア領域によって包囲されてバイオセンサーアレイを提供する工程、
・ 2つ以上の異なる、脂質二重層組成物を担持し、表面上にスポットを形成するためのスポット形成装置と流体連絡しているグラジエント形成装置を提供工程、
・ バイオセンサーアレイ製品を保持し、移動するための多軸移動テーブルを提供する工程、
・ 1つ以上のバリア領域に包囲された複数の二重層適合性領域を有するバイオセンサーアレイ製品を配置する工程、および
・ グラジエントを形成しているグラジエント形成装置から生じる混合脂質二重層組成物のスポットを形成し、そのような組成物の混合物がそれによって形成される際に、それを分配しながら、少なくとも1本の軸で前記テーブルを移動し、異なった連続的な位置に各脂質二重層組成物の異なった比率で分配する工程、
を含んでなるバイオセンサー領域のアレイを形成する方法を提供し、各領域は異なった公知の脂質二重層組成物を有する。
I. 定義
全ての用語は、下記で特に定義されない限り、当業者によって理解されている通常の意味を有することが意図されている。請求された質量と用量は、本発明の実施に適合する言明された量における変量を包含することが意図されている。このような変量は、例えば、言明された量の約±10〜20パーセントの範囲内にあると考えられる。本節に含まれる特定の定義と当業者によって理解されている通常の意味との間に矛盾がある場合は、下記に供された定義が規定するものとする。
図1は、本発明による表面検出器アレイデバイス(SDAD)20の一部の斜視図である。前記デバイスは、酸化シリコン、または溶融シリカウェーハなどの基体22から組立てられる。前記基体の寸法は、典型的には、一面当たり約0.1cmから約10cmの間であり、厚さが約0.01mmから約1cmである。
本発明の表面検出器デバイスは、ミクロ組立てと、例えば実施例1に記載されるような脂質ベシクル技術との組合わせを用いて簡便に製造できる。
1つ以上の二重層バリア領域によって分離された複数の異なった二重層適合性面領域を有する基体表面を製造するための基体のパターンづけは、本明細書の利益を受けるミクロ組立て技術における熟練者によって理解される多数の異なった方法で実施できる。例えば、当業界で周知のミクロ機械作製法としては、スパッタリング、スピンコーティングおよび化学蒸着などのフィルム蒸着法、レーザー組立てまたはフォトリソグラフィ法または、湿式化学的方法またはプラズマ法のいずれかによって実施し得るエッチング法が挙げられる。これら、および他のミクロ機械作製法は、例えば、参照として本明細書に組み込んであるPetersen(1982)に要約されている。当業界で公知の一般的なシリコン処理法は、例えば、参照として本明細書に組み込んでいるWolfおよびTauber(1986)に記載されている。
パターンづけした支持格子の作製後、表面に隣接する脂質二重層の形成を妨げる恐れのある基体表面上に存在する不純物または汚染物を剥がし、削り取るためにそれを洗浄し、および/または処理する。洗浄法は、二重層バリア領域の機能性を実質的に損なわないように選択される。例えば、前記バリア領域がホトレジストから作製されている実施形態では、酸が二重層バリア領域を剥ぎ取る恐れがあるため、伝統的なピランハ(piranha)溶液酸洗浄液(3:1 H2SO4:H2O2)を用いて洗浄すべきではない。ホトレジストを損傷させない例示的洗浄/処理法では、アルゴンまたは酸素のプラズマに対するパターンづけした格子の数分間曝露が用いられる。前記プラズマは、ホトレジストをいくらかは削り取るが、それは、ホトレジスト層を実質的に損傷させる前に基体(例えば、SiO2基体)の表面層から汚染物質を剥ぎ取る。
このような洗浄/削り取り/処理工程の後に、前記格子をチャンバーに入れ、選択された脂質から形成され、また(場合によっては)選択されたタンパク質または他の生体分子を含有するベシクルまたはリポソームの懸濁液を、各二重層適合性面領域に接触させる。懸濁液中のベシクルは一般に、二重層適合性面領域と1分以下で融合して、支持二重層膜を形成する(Xiaら、1996年;Grovesら、1996年)。二重層が形成し、前記格子が大量の水性物質で満たされる前に、実質的な蒸発を可能にするほど脂質懸濁液の小滴容量が、十分に小さい(例えば、<約5μl)適用においては、加湿チャンバーを用いることが好ましい。
A. バイオセンサー
一態様において、本発明は、表面検出アレイデバイスを有するバイオセンサーを含む。前記検出アレイデバイスは、(i)1つ以上の二重層バリア領域によって分離された複数の異なった二重層適合性面領域を規定している表面を有する基体、(ii)前記基体表面を覆っている大量の水相、(iii)各二重層適合性面領域上に維持された脂質二重層空間、および(iv)各二重層適合性面領域と、対応する脂質二重層空間との間に挟まれた水性薄膜を含んでなり、各二重層空間はレセプターまたは生体分子の一種を含有し、異なった二重層空間は、レセプターまたは生体分子の異なった種類を含有する。前記レセプターまたは生体分子は、各脂質二重層空間へまたは、その中に固定されている。脂質空間中のレセプターに対する特定のリガンドの特異的結合は、光学的または電気的検出などの種々の公知のバイオセンサー検出機構のいずれかによって検出される。電気的検出を採用しているバイオセンサーにおいては、支持格子は、伝導性の電極および前記デバイスの各アレイ要素に対する電導線を含有することが好ましい。前記導線は、典型的には、前記デバイスからの延長線または「ピン」として終わっており、これは、プロセッサーに導く接続ケーブルまたはリボンによりインターフェース接続ができる。前記電極は、二重層適合性面の少なくとも一部を形成することが好ましく、絶縁物質の細片によって、互いに分離されている。それらは、容量性過渡電流ならびに伝導性電流(持続的または過渡的であり得る)の検出に使用できる。一実施形態において、前記電極は、二重層適合性面の一部を形成する。他の実施形態において、その構成は実施例5に詳述されているが、前記電極は、二重層適合性面のすぐ下に位置する。すなわち、電極表面は、低温成熟酸化物(例えば、SiO2)などの材料の薄層で被覆され、それが二重層適合性面を形成する。この層が絶縁性材料である実施形態において、イオン輸送レセプターに対するリガンド結合により生じた容量性過渡電流の検出を可能にするために、その厚さは、約1μm未満であることが好ましい。バイオセンサーと共に使用する上で好適な表面検出アレイデバイスの構造部分の一実施形態は、上記のとおり、図4に示されている。
表面検出アレイデバイスを(i)二重層空間を作製するために使用されるベシクルが、典型的には、所望のレセプターまたは生体分子を含有すること(このレセプターまたは生体分子は二重層が形成された後にも導入され得るが)および(ii)異なったアレイ要素は、典型的には、異なったベシクル懸濁液によって作製されることを除いては、本質的に上記のとおりに製造される。
上記のバイオセンサー適用に関する一実施形態において、本発明のデバイスを、化合物の生物活性スクリーンにおいて使用されるレセプターのアレイを保持する基体として採用できる。特に、多数の化合物ライブラリーの高スループットスクリーンは、典型的には引き続く生物活性試験に関して、候補となる化合物を迅速に同定するために、スピードと効率に関して最適化される。このような生物活性試験が、例えば、イオンチャンネルアゴニスト、またはアンタゴニスト活性に関するアッセイに関係している場合、試験は、標的イオンチャンネルまたはレセプターを発現している個々の細胞の電気生理学的測定(例えば、パッチクランピング;例えば、Hamillら、1981年を参照)を用いて、一時期に1つの化合物が1人の科学者によって行われることが多い。このタイプの分析は、各化合物に関して、詳細で高品質のデータを提供するが、多数の化合物を生物活性に関してアッセイすべき時は、時間がかかり非効率的である。本発明の装置は、高スループットスクリーンにおいて同定された化合物の生物活性を評価する二次スクリーンにおいて使用でき、科学者が、真に関心対象の化合物に集中することを可能にする。このデバイスは、本質的にはバイオセンサーに関して上に記載されるとおりに作製される。同じタイプの結合検出体系を採用し得るが、イオノトロフィックレセプターまたはイオンチャンネルについての生物活性に関して化合物をアッセイする場合は、典型的には光学的検出よりも電気的検出が好ましい。
本発明は、非常に高密度の膜タンパク質の領域により支持二重層を形成する方法も含む。上述のように、タンパク質含有ベシクルまたはプロテオリポソームは、典型的には、約1:500以下のタンパク質:脂質のモル比でのみ形成することができ、より高いモル比を有するベシクルは、均一な平面二重層を一貫して形成することはない。したがって、支持二重層を形成するために、単にタンパク質含有ベシクルを表面と融合させることによっては、脂質二重層におけるタンパク質の高密度アレイは形成できない。
本発明の他の態様は、前記支持二重層内に統合されたまたは結合した生体分子に係る分類装置に関する。前記分類装置は、前記二重層バリア面領域を、異なった区画へと表面を区画分けするために使用するのではなく、前記装置の一端から他端までの、徐々に小さくなる「開口」を有する2次元のふるいとして作用させるために使用する。本発明のこの態様の構造部分の一実施形態を図5の平面図に示している。ここで、装置70の構造部分は、全側面が二重層バリア領域76に接している二重層適合性領域を規定している基体表面74を有するウェーハ72から形成される。前記二重層適合性領域はまた、二重層バリア面領域を規定している複数の実質的に平行な破線78によって中断されている。前記の線における隙間は分子の大きさであり、この装置の一端80から反対の端82に進むにつれて徐々に小さくなっていく。電極84、86は、この装置の端近くに位置しており、ワイヤ90、92によって、電圧源88に接続されている。
他に指定されない限り、化学物質は、Sigma(ミズーリ州セントルイス)または米国Biochemical(オハイオ州クリーブランド)から購入した。

B. 脂質および標識
卵由来のL−αホスファチジルコリン(卵−PC)を、Avanti Polar Lipids(アラバマ州アラバスター)より入手した。蛍光プローブN(Texas Redスルホニル)−1,2−ジヘキサデカノイル−sn−グリセロ−3ホスホエタノールアミン、トリエチルアンモニウム塩(Texas Red DHPE)は、Molecular Probes(オレゴン州ユージーン)から入手した。
小型単層ベシクル(SUV)類は、卵L−αホスファチジルコリン(Avanti)を用いて、Barenholzら(1977)に概説されたプルトコルに従って調製した。ホスファチジルコリンを、HPLCグレードのクロロホルム(SigmaAldrich)中、1モル%のTexas red DHPEと混合し、真空デシケータ中、一晩乾燥した。乾燥した脂質をSibataフィルタユニットを用いてRainin Nylon−550.45μmフィルタを通してろ過した標準緩衝液中、約6mg/mlに再懸濁した。前記懸濁液を、Branson超音波処理器を用いて、氷上アルゴン流下、1分間の冷却時間をはさんで、3分間、音波処理して透明にした。(Martin、1990)。
電気泳動試験のため、PBS中の支持膜を、1mM総イオン強度に希釈した。次にこれを緩衝液下、他のカバースリップによって、サンドイッチ状に組み立てた。電気泳動セルは、Teflon槽の溶液を満たしたウェル中0.01インチ直径の2本の白金ワイヤ電極から構成した。前記カバースリップサンドイッチを、2つの電極ウェル間に橋を形成するように調整した。電気的接続は、前記カバースリップサンドイッチ内の溶液を介して達成した。標準的電気供給により、60V/cmまでの電場をかけた。Keithleyピコ電流計(オハイオ州クリーブランド)を用いて電流をモニターすると、15V/cmにおいて、18mm正方形のカバースリップサンドイッチ1個に関して、典型的には凡そ3μAであった。これは、総電力消費、9x10−5Wに相当し、生成されるジュール熱は無視し得るはずである。
標準的な方法(WolfおよびTauber、1986年)を用いてパターンづけした支持格子をミクロ組立てした。100nm直径のシリコン1−0−0ウェーハを、Silrec社(カリフォルニア州サンジョセ)から入手した。前記ウェーハを酸化炉(Tylan社、カリフォルニア州サンディエゴ)において、1000℃で蒸気中に維持し、熱酸化物の約1μm厚さの層を生成した。標準的なポジホトレジスト(S1800:Shipley社、マサチューセッツ州マールボロー)をトラックコーター(Silicon Valleyグループ、カリフォルニア州サンジョセ)により1ミクロンの厚さでウェーハ上にスピンした。
二重層適合性領域または囲い内の長範囲流動性は、光退色後の蛍光回復(FRAP)によって観察された。この実験は、25の二重層適合性面領域または囲いを含有し、これらの各表面領域上に維持された、対応する脂質二重層空間を有する表面検出器アレイデバイスの略図を示す図2Aおよび2Bに関して記載されている。前記デバイスは、1辺が100μmの寸法の囲いを生成させるために、ホトレジストによってパターンづけした酸化シリコンウェーハから作製される。幅10μmのホトレジスト(二重層バリア領域)は、図2Aおよび2Bにおいて、25個の囲いを分離している黒い境界線として表されている。Texas Red DHPE脂質プローブ(Molecular Probes、オレゴン州ユージーン)を、標識として働くように二重層膜の中に組み込んだ。
二重層適合性面領域上の支持二重層の流動性は、荷電した膜成分の電気泳動再分配によっても評価された。この方法は、前記脂質二重層の流動性、および異なった組成の二重層区画を異なった、独立してアドレス指定できる二重層適合性面領域へ局限することの双方を示す。
二重層バリア領域が、隣接二重層空間を分離するのは、機械的分離によるのか、それとも二重層バリア表面領域を作製している材料の固有な性質によるのかを判断するために実験を行った。上記のとおり、パターンづけされていないSiO2基体(すなわち、単一の二重層適合性面領域)上に、二重層膜を被覆した。しかし、前記二重層適合性面領域のトポグラフィは、上記のホトレジストパターンづけSiO2基体のものと同じであった。
この実施例では、二酸化ケイ素の薄層で覆われたシリコン電極の作製を記載する。ワイヤをシリコン電極に接続するためにボンドパッドを使用できる。
酸化物上シリコンウェーハを、Ibis Technology社マサチューセッツ州デンヴァースから購入した。前記ウェーハは、直径100mmで、厚さが凡そ500μmである。製造元により供給されたウェーハは、ウェーハ上面を形成する約0.2μmの厚さの純粋シリコン層の下に埋められた約0.4μmの厚さの二酸化シリコン層を有する。
前記ウェーハを、従来のRCA洗浄操作(KernおよびPuotinen、1970年;WolfおよびTauber、1986年、p.516)によって洗浄し、150℃で30分焼付けし、Silicon Valleyグループ(SVG)トラックコーターシステムにより、従来のスピンコーティングを用いて、1μmのホトレジスト(Shipley S−1813)によって被覆した。
前記マスクパターンを石英基体上のクロムパターンから構成されている電子ビームマスターマスクを用いて、Karl Suss MA−4接触マスク整列器における8秒曝露を用いて曝露させた。次に前記ウェーハを、クロロベンゼン中に15分間浸漬してから、Silicon Valleyグループ(SVG)トラック現像器システムを用いて、標準的なTMAH(水酸化テトラメチルアンモニウム)基剤の現像剤(Shipley)により現像した。
選択的にシリコンを食刻するが、二酸化ケイ素は、食刻しない従来のフッ素基剤のプラズマ食刻(WolfおよびTauber、1986年)を用いて、シリコン層上面に電極パターンを食刻した。プラズマ食刻に用いられる気体は、SF6、O2およびCHF3である。1秒間のRCA洗浄後に、薄オキシドを蒸気オーブン中、1000℃で0.1μmの厚さにした。
再度RCA洗浄を行い、新たなレジスト層を上記のとおり被覆した。先の工程において厚化させたオキシド層の開口を規定する新たなパターンを上記のとおり、ホトリソグラフィにより、レジストに移し、曝露したレジストを上記のとおり現像した。
前記ウェーハを、Applied Materials(カリフォルニア州サンタクララ)の反応性イオン食刻剤中で食刻し、ボンドパッド用に基礎シリコン層への接触が実施できるように、オキシド層上面に穴を開けた。
先の工程のレジストを除去する前に、0.3μm層の金を、前記ウェーハ上に蒸着させた。次に、この金をアセトンで取り去ると、先の工程で食刻された穴の中に金のボンドパッドが配置された。
本発明の表面検出器アレイデバイスを、Proteomic System社により現像した。MembraneChipsTMとして公知のこれらのデバイスは、試験薬物標的に会合した膜の試験にとって重要な道具である。これらのMembraneChipsTMについてのごく初期の研究は、生体膜研究のための手動による流動性膜アレイの構成に関係していた。その後の研究により、アレイ表面にわたって膜組成を連続的に変化させられるグラジエントアレイの構成が可能になった。これらの結果は、表面検出器アレイデバイスに関するコンセプトの証拠を明らかに示したが、これらのアプローチは、実験的なものであり、したがって、工業的製造にはコスト効率がなく、小型サイズ特性の利点を維持しつつ、アレイ表面上で個別の予め決定された膜組成物を扱う能力に関して、特に制限を課していた。
分配タイプ:分配するための2つの異なる方法に関するソフトウェアプログラムの書き込みを行った。それらは、「直接」および「タッチ−オフ」と呼ばれる。両分配タイプとも、下記のパラメータを用いて、好結果を伴って実行された。
分配時におけるチップの高さ:

MembraneChipsTMは、以下に要約した工程後にマイクロアレイヤーを用いて組立てられた。
2. チップを洗浄し、真空乾燥する(3回)
3. システム圧を除去するために通気する
4. セラミックチップ(直径3mm、オリフィスサイズ190ミクロン)を供給源ウェル(96/384ウェルプレート)に移動する
5. 供給源96ウェル(20〜300μlの範囲の作業容量)プレートまたは384ウェルプレート(10〜100μlの範囲の作業容量)から脂質/プロテオベシクルを吸引する(典型的に約50μL)
6. システム圧を除去するために通気する
7. バルブを開けずに実際の分配容量を1回分配することにより前加圧してシステム内を陽圧にする
8. 上記のとおり前分配する
9. 実際の分配容量:10〜90nL−−用意したMembraneChipsTMパターンづけ支持格子に分配する
10. 脱イオン水でチップホルダーを満たす
11. 脱イオン水で掃流する
12. チップを洗浄し、真空乾燥する
プログラムのパラメータを、精密に最適化し、アレイヤーが、機能的で流動性の膜二重層を創製することを確保する。マイクロアレイヤーが、チップ上にプリント膜を仕上げた直後にMembraneChipsTMを水に浸した(上記工程9)。より大きなオリフィスサイズチップ(508ミクロン)を用いて、チップホルダーを脱イオン水で満たした。膜形成後、空気への短時間曝露によっても、直ちにそれらの構造が破壊され、膜の流動性欠如のみならず、生体膜中に埋め込まれているまたはそれらと会合しているタンパク質の破壊が生じる。したがって、膜アレイは、組立て後、常時水面下に維持しなければならない。
光退色後の蛍光回復(FRAP)実験は、マイクロアレイヤーにより組立てられたMembraneChipsTMにおけるNBD標識膜(図10)およびテキサスレッド標識膜(データは示していない)から代表的な囲いに対して実施された。囲いの1つにおいて、凡そ100ミクロン/ミリメートル直径スポットは、開口絞りを通して方向付けられた100W水銀アークランプ(Ushio社、USH−102DH、日本国東京)で2分の照射により光退色した(図10a)。光退色後、広範な拡散により、囲い内の蛍光回復が生じ(照射10分後に撮影された図10bを参照)、マイクロアレイヤーにより整列されたMembraneChipsTMにおける膜の流動性を証明している。
MembraneChipTMの表面を、前記アレイの全ての特性点(すなわち、囲い)が同時に単独試剤に曝露するようにデザインする。この性質を立証するために、MembraneChipTM実験を実施して、整列された膜が、試験物質の結合をアッセイするためも特異的な標的として使用できることを示した(図11)。MembraneChipTMは、実施例6に記載された自動化方法に従って構築された。500平方ミクロンの特性点(すなわち、囲い)を有する4×4のアレイにおいて、1つを除いて全ての囲いを、1モルパーセントNBD−ホスファチジルグリセロールを有する99モルパーセント卵ホスファチジルコリンを含有する溶液で整列させた。最後の囲い110(第3欄、第3列、上部左コーナーが原点)は、98モルパーセント卵ホスファチジルコリン、1モルパーセントNBD−ホスファチジルグリセロールおよび1モルパーセント非標識GM1により整列させた。適切なFITCフィルタセット(Nikon Instruments社、96106807B−2A、ニューヨーク州メルビレ)を装備した蛍光顕微鏡(Nikon Instruments社、Nikon Eclipse E400、ニューヨーク州メルビレ)下で観察すると、チップは、均一にグリーンが表れた(データは示していない)。
序論
大多数の薬物標的は、G−蛋白共役レセプター(GPCR)類などの膜タンパク質である。例えば、全処方医薬品の60%がGPCR類を標的にする。これらのタンパク質は、GPCR類に結合するホルモン類および神経伝達物質など、リガンドにより開始される大多数の細胞作用を媒介するので、このような注目に値する。GPCR類に結合するリガンドは、代謝、細胞増殖、および神経伝達に関連する広範囲の生理的機能を媒介する。
MembraneChipTMは、並行フォーマットにおいて任意の数のGPCR類をディスプレイするために使用できる。GPCR膜製剤は、Stratagene(カリフォルニア州ラホーヤ)またはPackard BioScience(コネチカット州メリデン)より購入した。材料と方法に記載された方法を用いて調製された、膜製剤対卵ホスファチジルコリン(Avanti Polar Lipids、アラバマ州アラバスター)ベシクル製剤の1:1比を、37℃で一晩インキュベートし、拡散溶媒として用いた。次に、GPCR類を、実施例6に記載されたマイクロアレイヤーを用いてチップ上にデザインされた囲い内に整列した。チップ上の各GPCRの座標を追跡した。GPCR類が埋め込まれている脂質二重層は生物学的に活性である。すなわち、GPCR類は、機能的であり、天然または天然様膜環境内に適応されていた。
MembraneChipTMの重要な特徴の1つは、今日薬物開発に使用されている他の伝統的なアッセイ(フィルタ結合およびシンチレーション近接アッセイ(SPA))と比較して各データポイントに必要なGPCRが実質的により少ないことであることである。表1に、典型的なフィルタ結合アッセイおよびSPAに必要な膜と特定のGPCR、AR−β1の量が算出され、MembraneChipTMに対するアッセイに必要なものと比較されている。これは特に、GPCR類の量獲得がしばしば時間がかかり、困難であることから重要である。さらに、GPCR類は、薬物スクリーニングおよび最適アッセイの開発において全コストのうち最も大きな割合になることが多い。表1に例示されるように、MembraneChipTMは、データポイント1つ当たりに必要とされるGPCR量において約300倍以上の減少を提供する。

4SPAに概要されるG−タンパク質共役レセプターアッセイ(Amersham Biosciences、RPNQ0210、ニュージャージー州ピスカタウェイ)
競合結合アッセイは、2種のGPCR、β−アドレナリンサブタイプ1レセプター(AR−β1、Packard BioScienceコネチカット州メリデン)(図12および表2)およびムスカリンレセプターサブタイプM1(M1、Packard BioScienceコネチカット州メリデン)(表2)を用いて開発された。このタイプの実験は、薬物最適化に使用され得る。脂質のみのコントロール(ネガティブコントロール)で整列されたMembraneChipTMの表面囲いを、AR−β選択的蛍光アンタゴニスト類縁体、BODIPY TMR CGP12177(Molecular Probes、オレゴン州ユージーン)の100nM溶液により探査した。表面の蛍光は、著しく低かった(図12および表2の「ネガティブ」)。他のGPCR類に対する結合特異性を評価するために、CXCR4(CXCケモカインレセプター4)を含有するMembraneChipsTMは、同一濃度のBODIPY TMR CGP12177(図12および表2「CXCR4」)により探査された。ネガティブコントロールよりも僅かに高いシグナルが得られ、このCXCR4細胞膜調製物は、低内在性濃度のAR−β1を含有することから予想されたことであった。AR−β1を過剰発現している細胞から細胞膜製剤を含有するMembraneChipTMが、100nM BODIPY TMR CGP12177で探査された場合、大きなシグナルが観察された(図12および表2の「ポジティブ」)。重要なことに、5mMの非標識AR−β選択的アンタゴニスト、プロプラノール(Sigma、ミズーリ州セントルイス)が、ヒット薬物の模擬物である、100nM BODIPY TMR CGP12177と共に含まれた場合(図12および表2の「競合」)、ポジティブシグナルの約95%は、競合で追い出された。
6独立して実施された実験に関して。
MembraneChipTMは、複合GPCRアッセイのためのMembraneChipTMシステムの有用性を立証するために開発された。複数の標的が、単一の基体に存在し、同時に試験物質に曝露されている複合アッセイにより、基体上の種々の標的に対する試験物質の相対的親和性を極めて精確に判定することが可能である。GPCR類、CXCR1、CXCR3、CXCR4、M1およびAR−β1を含有するMembraneChipTMは、実施例6に要約された方法に従って組み立てられ、実施例9に要約された方法に従ってAR−β1特異的リガンドCGP12177により探査された。AR−β1に結合する20nM BODIPY TMR CGP12177からの蛍光シグナルは、純粋脂質(図14、「ネガティブ」)CXCR1、CXCR3、CXCR4およびM1に対する蛍光リガンドの非特異的結合からの平均蛍光の22倍である(図14、「BODIPY CGP」バー)。蛍光標識CGP12177のAR−β1に対する結合は、予想されるとおり、未標識5mMプロプラノロールの添加により競合した(図14、AR−β1欄に関する「CGP+プロプラノロール」のバー)。AR−β1にではなく、M1レセプターに高親和性を有する試剤、15mMスコポラミンの添加時には蛍光減少が殆ど見られなかった(図14、AR−β1欄に関する「CGP+スコポラミン」のバー)。
ケモカインレセプター、CXCR1、CXCR3およびCXCR4(Stratagene、カリフォルニア州ラジョラ)を、上記のとおり、プロテオリポソームベシクルに組み込み、実施例6に記載された方法を用いてMembraneChipsTMの表面上に整列させた。次にそれらは、各CXCR類を認識する蛍光標識抗体と共にMembraneChipsTMをインキュベートし(図17)、MembraneChipsTMを洗浄し、上記のとおり蛍光顕微鏡写真画像をとらえ、処理することにより検出された。GPCR類が置かれている位置のバックグランド(それぞれ図17中の「+」および「−」)と比較した相対的蛍光単位(RFU)が統計的に有意に増加していることは、MembraneChipsTM表面上に整列後も、レセプター上の抗原部位構造が保存されたことを証明している。この結果は、MembraneChipsTMが、抗原に結合する抗体を特徴付けるために使用でき、抗体結合が、MembraneChipsTMの表面に存在する標的を検出するために使用できることを説明している。
高感度膜アッセイをMembraneChipsTMに適合させた。前記アッセイは、簡潔で、試剤ロボット操作システムに適合性があり、複合フォーマットにおけるレセプターの個別の膜アレイに対する新規のアンタゴニストおよびアゴニストのスクリーニングを可能にする。
Claims (33)
- 表面検出器アレイデバイスを組立てする方法であって、該方法は、
1つ以上の二重層バリア領域によって分離された複数の異なった二重層適合性面領域を規定している表面を有する基体であって、前記二重層適合性面領域と前記二重層バリア領域とが異なった材料から形成される基体を提供する工程と;
第1の組成物を有する脂質二重層ベシクルの第1の懸濁液を提供する工程と;
第2の組成物を有する脂質二重層ベシクルの第2の懸濁液を提供する工程であって、前記第1の組成物が、前記第2の組成物とは異なる、工程と;
前記第1の懸濁液を100nl未満自動操作によって、前記複数の異なった二重層適合性面領域のうち第1の領域へ移動させる工程と;
前記第2の懸濁液を100nl未満自動操作によって、前記複数の異なった二重層適合性面領域のうち第2の領域へ移動させる工程と;
前記第1の異なった二重層適合性面領域上に安定に局在化した第1の脂質二重層空間および前記第2の異なった二重層適合性面領域上に安定に局在化した第2の脂質二重層空間を形成するために第1および第2の懸濁液と共に前記基体をインキュベートする工程であって、前記二重層空間の各々が、隔離されたままで他の前記二重層空間と混合せず、前記各脂質二重層空間と、前記各二重層適合性面領域との間で共有結合することなく、前記各空間が、前記各表面領域上に局在化しており、前記二重層適合性面領域と、対応する前記脂質二重層空間との間に挟まれた水性膜によって前記各表面領域から分離されているものとする前記工程と;
前記脂質二重層空間上に大量の水相を確立する工程と、
を含んでなる方法。 - 前記基体が、約10個から約100個の間の異なった二重層適合性面領域を含んでなる請求項1に記載の方法。
- 前記基体が、少なくとも約2500個の異なった二重層適合性面領域を含んでなる請求項2に記載の方法。
- 前記基体が、少なくとも約25,000個の異なった二重層適合性面領域を含んでなる請求項3に記載の方法。
- 前記基体が、少なくとも約250万個の異なった二重層適合性面領域を含んでなる請求項4に記載の方法。
- 前記二重層適合性面領域が、約1μmから約10μmの間の幅の二重層バリア領域によって互いに分離されている請求項1に記載の方法。
- 前記第1の組成物が、さらに生体分子を含んでなる請求項1に記載の方法。
- 前記生体分子が、膜貫通型レセプターおよびイオンチャンネルよりなる群から選択される請求項7に記載の方法。
- 前記生体分子が、脂質分子に共有結合している請求項8に記載の方法。
- 前記移動工程が、前記第1および前記第2の懸濁液の小滴を生成させること、および前記小滴を前記第1および前記第2の二重層適合性面領域各々に接触させることを含んでなる請求項1に記載の方法。

- 前記各小滴が、15nl未満を含んでなる請求項11に記載の方法。
- 前記移動工程が、前記第1および前記第2の懸濁液のアリコートを、空隙を通って前記第1および第2の二重層適合性面領域各々の上に射出することを含んでなる請求項1に記載の方法。
- 前記射出された各アリコートが、90nl未満を含んでなる請求項13に記載の方法。
- 前記射出された各アリコートが、15nl未満を含んでなる請求項14に記載の方法。
- 請求項1から15のいずれかに記載の方法によって製造された表面検出器アレイ装置。
- 表面検出器アレイデバイスを組立てする装置であって、
1つ以上の二重層バリア領域によって分離された複数の異なった二重層適合性面領域を規定している面を有する基体を受ける台であって、前記二重層適合性面領域と前記二重層バリア領域とが、異なった材料から形成される、台;
第1の組成物を有する脂質含有ベシクルまたはリポソームの第1の懸濁液の供給源;
第2の組成物を有する脂質含有ベシクルまたはリポソームの第2の懸濁液の供給源であって、前記第1の組成物が、前記第2の組成物とは異なる、供給源;
前記第1の懸濁液を100nl未満、前記複数の異なった二重層適合性面領域のうちの第1の領域へ移動させ、および前記第2の懸濁液を100nl未満、前記複数の異なった二重層適合性面領域のうちの第2の領域へ移動させる自動手段であって、前記アリコートを隔離されたままで互いに混合しないように移動させる、自動手段;および
前記の組立てられた表面検出器アレイデバイスを覆うために使用される大量の水相の供給源、
を含んでなる装置。 - 前記基体表面の近傍における1つ以上の環境パラメータを制御するためのチャンバーをさらに含んでなる請求項17に記載の装置。
- 前記パラメータが、温度、相対湿度、圧力および照明よりなる群から選択される請求項18に記載の装置。
- 前記台と前記移動手段との間の相対的運動を制御するために、操作可能にプログラム化されたマイクロプロセッサーをさらに含んでなる請求項17に記載の装置。

第1の組成物を100nl未満有し、前記複数の異なった二重層適合性面領域のうち第1の領域の上に安定に局在化されている第1の脂質二重層空間、
前記第1の組成物とは異なる第2の組成物を100nl未満有し、前記複数の異なった二重層適合性面領域のうち第2の領域の上に安定に局在化されている第2の脂質二重層空間、
を含んでなるものとし、
前記第1の組成物が、前記第2の組成物とは異なり;
前記二重層空間の各々が、隔離されたままで他の前記二重層空間と混合せず、前記各脂質二重層空間と、前記各二重層適合性面領域との間で共有結合することなく、前記各空間が、前記各表面領域上に局在化しており、前記二重層適合性面領域と、対応する前記脂質二重層空間との間に挟まれた水性薄膜によって前記の各表面領域から分離されているものとする、工程と;
前記デバイスを、試験物質を含んでなる大量の水相に接触させる工程と;
前記試験物質と前記第1の組成物との相互作用および前記試験物質と前記第2の組成物との相互作用をアッセイする工程と、
を含んでなる複合アッセイ。 - 前記第1の組成物が、第1の生体分子を含んでなり、前記第2の組成物が、第2の生体分子を含んでなる請求項21に記載の方法。
- 前記生体分子が、膜貫通型レセプターおよびイオンチャンネルよりなる群から選択される請求項22に記載の方法。
- 前記第1と第2の生体分子が、レセプタータンパク質ファミリーの異なったメンバーである請求項22に記載の方法。
- 前記第1の組成物が、第1の脂質を含んでなり、前記第2の組成物が、第2の脂質を含んでなる請求項21に記載の方法。
- 前記第1の脂質二重層空間と前記第2の脂質二重層空間の各々が、5μg未満を含んでなる請求項21に記載の方法。
