Description
本発明に従って調整される試験片のための水溶液及び全血の両方の中のグルコースに対する用量応答は、小孔径及び大孔径の両方の試験片膜について決定された。使用された多孔質膜は、0.2ミクロン及び5.0ミクロンの等方性孔径をもつ、Pall Corporationから購入したSuporブランドのポリエーテルスルフォン膜であった。各々の多孔質膜を、表1に記されている組成物中に膜の平滑な反応面を浸漬することによって試薬で処理した。
この例は、上述の例1で使用された流入形技術ではなく図1Cに関連して例示され論述された側方流毛細管方式を用いて標本が適用される浸漬式試薬膜技術のフィージビリティを実証している。Pall Corporationから商品名「Presense」で販売されている孔径0.8ミクロンのポリエーテルスルフォン膜の標本を、例1に記述されている通りに処理した。処理済み膜のリボンを、図1Cに例示されている通り毛細管フロー試験片上に取付けた。各試験片には、40%ヘマトクリットの全血中のグルコース又はグルコース水溶液のいずれかの600〜800ナノリットルの標本を塗布した。水性試験標本中のグルコースの濃度は、0、50、100、250及び500mg/dLであった。全血標本中のグルコース濃度は、血漿値に変換されて、0、50、100、200、400及び600mg/dLであった。
Pall CorporationによりPresenseの商品名で販売されている孔径0.8ミクロンのポリエーテルスルフォン多孔質膜材料を用いて、試験片を調製した。使用された試薬組成物は、表1で記述されたものであった。浸漬ではなくむしろ多孔質膜の反応面上へのストライピングにより、試薬組成物を塗布した。次に試験片に、Arcare7843感圧接着剤により固定された透明ポリエステルウインドウを備え、これを、全て以上の例1に記述された通りに試薬リボンと共に積層させた。試験片は、反応面を下にして方向づけした。その後、500ナノリットルのグルコースでスパイクされた全血の試験標本を、各試験片の粗い標本面に塗布した。グルコース全血標本は、Yellow Springs Instrument検出器を用いて血漿グルコースとして測定された場合、0mg/dL、50mg/dL、100mg/dL、200mg/dL、400mg/dL及び600mg/dLであった。各標本試験片上で各標本溶液について2つの複製を試験させた。
この例は、光学ベースのPQQ−GDH還元系における〔特許文献4〕で報告されたメディエータの有用性を実証している。試薬は、以下のように調製された:
この例は、本発明の状況下でNAD依存性系において〔特許文献5〕及び〔特許文献6〕に開示されているもののようなテトラゾリウム塩指示薬を用いることのフィージビリティを実証している。WST−4テトラゾリウム塩の代りに40mMの2−(4−ジフルオロメチル−5−クロロチアゾール−2−イル)−3−〔2−(3−トリメチルアンモニオプロプロキシ)フェニル〕−5−(3.4−メチレンジオキシフェニル)テトラゾリウムジメチルスルフォネートテトラゾリウム塩指示薬を用いた。0.8ミクロンのポリエーテルスルフォンPresense膜の平滑な面をコーティングするのに試薬ミックスを使用し、次に毛細管充填方式に組立てた。およそ30〜10パーセントの反射率の範囲で0〜600mg.dLの全血グルコースに対する用量応答を実証した。

Claims (9)
- 全血標本中のグルコースの濃度を決定するための方法において:
(a)標本面及び反応面をもち、標本面上に標本を受容しそれを反応面に向けて通過させるのに適合されている多孔質膜を含み、標本中のグルコースの存在を表示するように色を変える変色指示薬系を含む試験用試薬を含む試験片であって、さらに試験片の少なくとも一領域の不透明度を増大させる重合体を少なくとも反応面上に有している試験片を用意する段階;
(b)試験片の標本面に標本を塗布する段階;及び
(c)標本中のグルコースの濃度を決定するように試験面の色の変化を測定する段階、
を含み、
前記重合体が、アクリル系ラテックス重合体、ポリスチレン硫酸ナトリウム重合体及びそれらの組合せから成るグループの中から選択される、方法。 - 前記重合体が、ポリスチレン硫酸ナトリウム重合体である、請求項1に記載の方法。
- 試験用試薬が、PQQ−GDH系、NAD−依存性GDH系、ヘキソキナーゼ還元系及びグルコースオキシダーゼ酸化系から成るグループの中から選択された化学薬品系を含んで成る、請求項1に記載の方法。
- 試験片が0.2ミクロンから5.0ミクロンの範囲内の孔径をもつ、請求項1に記載の方法。
- 前記試験用試薬が、グルコースデヒドロゲナーゼ(GDH)系を含む、請求項1に記載の方法。
- 全血標本中のグルコースの濃度を測定するための試験片において:
(a)標本面及び反応面をもち、標本面上に標本を受入れそれを反応面に向かって通過させるのに適合されている多孔質膜;
(b)膜によって支持され、標本中のグルコースの存在を指示するように色を変える変色指示薬系を含んで成る試験用試薬;及び
(c)試験片の少なくとも一領域における不透明性を増大させる、多孔質膜の反応面の少なくとも一領域上にある重合体、
を含み、
前記重合体が、アクリル系ラテックス重合体、ポリスチレン硫酸ナトリウム重合体及びそれらの組合せから成るグループの中から選択される、試験片。 - 前記重合体が、ポリスチレン硫酸ナトリウム重合体である、請求項6に記載の試験片。
- 前記多孔質膜が、ポリエーテルスルフォン、変成ポリエーテルスルフォンまたはそれらの組合せを含んで成る、請求項6に記載の試験片。
- 試薬が、置換済み又は未置換3−フェニルイミノ−3H−フェノチアジンまたは3−フェニルイミノ−3H−フェノキサジンを含むメディエータ化合物を含んで成る、請求項6に記載の試験片。
