[実施例1〜10]
実施例1〜10における反応は、他に特に明記していない限り、マグネチックスターラーを使用して窒素下で実施した。「ワークアップ(Work−up)」とは、有機溶媒を用いての抽出、飽和NaCl溶液を用いての有機相の洗浄、乾燥(MgSO4)、および蒸発(ロータリー・エバポレーター)を意味する。薄層クロマトグラフィーは、シリカゲル60°F―254が事前に塗布されたメルク(Merck)社製ガラスプレートを用いて実施し、ヨウ素蒸気によりスポットを可視化した。すべての質量スペクトルは、コロラド州所在のコロラド州立大学高分子資源(Macromolecular Resources)によって測定され、m/z、(相対強度)の大きさで報告された。元素分析および融点測定は、テネシー州ノックスビル所在のガルブレイス・ラボラトリーズ(Galbraith Laboratories)社によって実施された。実施例1〜実施例10は、図2に示したスキームを参照する。
無水ジクロロメタン(50mL)中の非多分散トリエチレングリコールモノメチルエーテル分子(4.00mL,4.19g,25.5mmol)およびトリエチルアミン(426mL,3.09g,30.6mmol)の溶液を氷浴中で冷却し、窒素雰囲気下に置いた。無水ジクロロメタン(20mL)中の塩化メタンスルホニル(2.37mL,3.51g,30.6mmol)の溶液を添加用漏斗から滴下法で加えた。塩化物添加の完了10分後、この反応混合液を氷浴から取り出し、室温になるのに任せた。この混合液をさらに1時間撹拌したが、その時点でTLC(溶出剤として15%のMeOHを含むCHCl3)でトリエチレングリコールモノメチルエーテルの残留は証明されなかった。
無水DMF(25.7mL)中の非多分散化合物11(35.7mmol)の撹拌溶液に、N2雰囲気下で鉱油中のNaHの60%分散液を少しずつ加え、そしてこの混合物を室温で1時間撹拌した。この塩12に無水DMF(4mL)中の非多分散メシレート9(23.36)の溶液を1部分で加え、この混合液を室温で3.5時間撹拌した。反応の進行は、TLC(12% CH3OH−CHCl3)によって監視した。この反応混合液を等量の1NのHClを用いて希釈し、そして酢酸エチル(2×20mL)を用いて抽出して廃棄した。水溶液の抽出およびワークアップにより、非多分散ポリマー10が得られた(収率82〜84%)。

非多分散化合物14は、9について記載した通りにアルコール13(m=4)および塩化メタンスルホニルから油として定量的収率で入手した;Rf 0.4(酢酸エチル:アセトニトリル=1:5);MS m/z C17H37O10に対する計算値433.21(M++l)、実測値433.469。
非単分散化合物15は、化合物10について上記に記載した方法を使用することによりジオールから調製した。

実施例11〜実施例18は、図3に示したスキームを参照する。
4mLの水に3.99g(100mmol)のNaOHを溶解させることで調製した水酸化ナトリウム水溶液を非多分散ヘキサエチレングリコール(28.175g,25mL,100mmol)へ緩徐に加えた。塩化ベンジル(3.9g,30.8mmol,3.54mL)を加え、この反応混合液を撹拌しながら100℃へ18時間加熱した。その後、この反応混合液を冷却し、食塩水(250mL)を用いて希釈し、そして塩化メチレンを用いて抽出した(200mL×2)。結合した有機層を食塩液により1回洗浄し、Na2SO4の上方を通して乾燥させ、そして真空中で暗褐色油へ濃縮した。粗生成物混合物をフラッシュクロマトグラフィー(シリカゲル、勾配溶離:酢酸エチレンから9/1酢酸エチル/メタノール)により精製すると、黄色油として8.099g(70%)の非単分散化合物16が産生した。
無水ジクロロメタン(75mL)中の非多分散エチル6−ヒドロキシヘキサノエート(50.76mL,50.41g,227mmol)の溶液を氷浴中で冷却し、窒素雰囲気下に置いた。トリエチルアミン(34.43mL,24.99g,247mmol)を加えた。無水ジクロロメタン(75mL)中の塩化メタンスルホニル(19.15mL,28.3g,247mmol)の溶液を添加用漏斗から滴下法で加えた。この混合液を3時間半撹拌し、氷浴が溶けるにしたがって緩徐に室温になるに任せた。この混合液をシリカゲルに通して濾過し、そして濾液を水、飽和NaHCO3、水および食塩液を用いて連続的に洗浄した。Na2SO4の上方を通して有機相を乾燥させ、濾過し、そして真空中で浅黄色油へ濃縮した。粗生成物の最終精製は、フラッシュクロマトグラフィー(シリカゲル、1/1のヘキサン/酢酸エチル)により達成し、透明な無色油として非単分散生成物(46.13g,85%)を得た。FAB MS:m/e 239(M+H),193(M−C2H5O)。
水素化ナトリウム(3.225gもしくは60%油分散液,80.6mmol)を80mLの無水トルエン中に懸濁させ、窒素雰囲気下に置き、そして氷浴中で冷却した。80mLの無水トルエン中の非多分散アルコール16(27.3g,73.3mmol)の溶液をNaH懸濁液に加えた。この混合液を0℃で30分間撹拌し、室温になるに任せ、さらに5時間撹拌したところ、その時間内に混合物は透明褐色溶液になった。80mLの無水トルエン中の非多分散メシレート17(19.21g,80.6mmol)をNaH/アルコール混合液に加え、そして結合した溶液を室温で3日間撹拌した。この反応混合液を50mLのメタノールを用いて急冷し、塩基性アルミナを通して濾過した。この濾液を真空中で濃縮し、フラッシュクロマトグラフィー(シリカゲル、勾配溶離:3/1酢酸エチレン/ヘキサンから酢酸エチル)により精製すると、浅黄色油として非単分散生成物(16.52g,44%)が産生した。FAB MS:m/e 515(M+H)。
非多分散ベンジルエーテル18(1.03g,2.0mmol)を25mLのエタノール中へ溶解させた。この溶液に270mgの10%のPd/Cを加え、この混合液を水素雰囲気下に置き、4時間攪拌したところ、その時点でTLCは出発原料の完全な消失を示した。この反応混合液をセライト(Celite)545に通して濾過して触媒を除去し、そして濾液を真空中で濃縮すると透明油として非多分散の標題化合物(0.67g,79%)が産生した。FAB MS:m/e 425(M+H),447(M+Na)。
非多分散アルコール19(0.835g,1.97mmol)を35mLの無水ジクロロメタン中に溶解させ、窒素雰囲気下に置いた。トリエチルアミン(0.301mL,0.219g,2.16mmol)を加え、この混合液を氷浴中で冷却した。2分間後、塩化メタンスルホニル(0.16mL,0.248g,2.16mmol)を加えた。この混合液を0℃で15分間、その後室温で2時間攪拌した。この反応混合液をシリカゲルに通して濾過して塩化トリエチルアンモニウムを除去し、そして濾液を水、飽和NaHCO3、水および食塩液を用いて連続的に洗浄した。有機相をNa2SO4の上方を通して乾燥させ、濾過し、真空中で濃縮した。残留物をカラムクロマトグラフィー(シリカゲル、9/1酢酸エチル/メタノール)により精製して、透明油として非多分散化合物20(0.819g,83%)を得た。FAB MS:m/e 503(M+H)。
NaH(88mgの60%油中分散液,2.2mmol)をN2雰囲気下で無水トルエン(3mL)中に懸濁させ、0℃へ冷却した。トルエンを用いた共沸蒸留を介して乾燥させた非多分散ジエチレングリコールモノメチルエーテル(0.26mL,026g,22mmol)を加えた。この反応混合液が室温に加温するに任せ、4時間攪拌したところ、その間に濁った灰色の懸濁液は透明な黄色になり、その後褐色に変色した。2.5mLの無水トルエン中のメシレート20(0.50g,1.0mmol)を加えた。室温で一晩攪拌した後、2mLのメタノールを添加することによりこの反応液を急冷し、結果として生じた溶液をシリカゲルに通して濾過した。濾液を真空中で濃縮した。FAB MS:m/e 499(M+H),521(M+Na)。予備クロマトグラフィー(シリカゲル、19/3クロロホルム/メタノール)による追加の精製で、透明黄色油として非多分散生成物(0.302g,57%)が得られた。FAB MS:m/e 527(M+H),549(M+Na)。
非多分散エステル21(0.25g,0.46mmol)を0.71mLの1NのNaOH中で18時間攪拌した。18時間後、この混合液を真空中で濃縮してアルコールを除去し、残留物をさらに10mLの水に溶解させた。2NのHClを用いてこの水溶液をpH2へ酸性化させ、その生成物をジクロロメタン中で抽出した(30mLx2)。その後、結合した有機相は食塩液を用いて洗浄し(25mL×2)、Na2SO4の上方を通して乾燥させ、濾過し、真空中で濃縮すると黄色油として非多分散の標題化合物(0.147g,62%)が産生した。FAB MS:m/e 499(M+H),521(M+Na)。
非多分散酸22(0.209g,0.42mmol)を4mLの無水ジクロロメタン中に溶解させ、既にNHS(N−ヒドロキシスクシンイミド)(57.8mg,0.502mmol)およびEDC(l−(3−ジメチルアミノプロピル)−3−エチルカルボジイミドヒドロクロリド)(98.0mg,0.502mmol)を含有する乾燥したフラスコにN2雰囲気下で加えた。この溶液を室温で一晩攪拌し、シリカゲルに通して濾過して過剰な試薬およびEDCから形成された尿素を除去した。この濾液を真空中で濃縮すると暗黄色油(0.235g,94%)として非多分散生成物が得られた。FAB MS:m/e 596(M+H),618(M+Na)。
氷浴中で0℃に冷却したCH2Cl2(100mL)の溶液に非多分散トリエチレングリコールモノメチルエーテル(25g,0.15mol)を加えた。その後、トリエチルアミン(29.5mL,0.22mol)を加え、この溶液を0℃で15分間攪拌し、この後に滴下法で塩化メタンスルホニル(13.8mL,0.18mol,20mLのCH2Cl2に溶解させた)を加えた。この反応混合液を0℃で30分間攪拌し、室温へ加温するに任せ、その後2時間攪拌した。この粗反応混合液をセライトに通して濾過し(〜200mLのCH2Cl2で洗浄した)、その後H2O(300mL)、5%のNaHCO3(300mL)、H2O(300mL)、飽和NaCl(300mL)を用いて洗浄し、MgSO4の上方を通して乾燥させ、乾燥するまで蒸発させた。この油を、その後乾燥を保証するために〜2時間、真空ライン上に置き、黄色油として非多分散の標題化合物(29.15g,収率80%)を得た。
THF(1L)中の非多分散テトラエチレングリコール(51.5g,0.27mol)の溶液へt−カリウムブトキシド(14.8g,0.13mol,30分間にわたり少しずつ)を加えた。この反応混合液を1時間攪拌し、その後THF(90mL)中に溶解させた24(29.15g,0.12mol)を滴下法で加え、この反応混合液を一晩攪拌した。粗反応混合液をセライトに通して濾過し(〜200mLのCH2Cl2で洗浄した)、乾燥するまで蒸発させた。この油をその後HCl(250mL,1N)中に溶解させ、過剰な24を除去するために酢酸エチル(250mL)を用いて洗浄した。残っている24を除去するために、酢酸エチル(125mL)による追加の洗浄が必要になることがある。25の大部分が水性相から除去されるまで、CH2Cl2(容量125mL)を用いて水性相を繰り返し洗浄した。初回抽出では24、25および二重結合副生成物を含むであろうから、HCl(125mL,1N)を用いて逆抽出しなければならない。有機層を結合し、乾燥するまで蒸発させた。生じた油をその後CH2Cl2(100mL)中に溶解させ、25が除去されるまでH2O(容量、50mL)を用いて繰り返し洗浄した。総容量が500mLになるように水性フラクションを結合し、この溶液が濁るまでNaClを加え、その後CH2Cl2(2×500mL)を用いて洗浄した。有機層を結合し、MgSO4で乾燥させ、乾燥するまで蒸発させると、油として非多分散の標題化合物(16.9g,収率41%)が得られた。高純度を保証するために1以上の精製ステップを繰り返すのが望ましいことがある。
エタノール(100mL)中の8−ブロモオクタン酸(5.0g,22mmol)の溶液にH2SO4(0.36mL,7.5mmol)を加え、この反応液を加熱して攪拌しながら3時間環流させた。この粗反応混合液を室温へ冷却し、H2O(100mL)、飽和NaHCO3(2×100mL)、H2O(100mL)を用いて洗浄し、MgSO4で乾燥させ、乾燥するまで蒸発させると、透明油(5.5g,収率98%)が得られた。
エーテル(90mL)中の非多分散化合物25(3.0g,8.8mmol)の溶液にt−カリウムブトキシド(1.2g,9.6mmol)を加え、この反応混合液を1時間攪拌した。その後、エーテル(10mL)中に溶解させた非多分散化合物26(2.4g,9.6mmol)を滴下法で加え、この反応混合液を一晩攪拌した。粗反応混合液をセライトに通して濾過し(〜200mLのCH2Cl2で洗浄した)、乾燥するまで蒸発させた。生じた油を酢酸エチル中に溶解させ、H2Oで洗浄し(2×200mL)、MgSO4で乾燥させ、乾燥するまで蒸発させた。カラムクロマトグラフィー(シリカ、酢酸エチル対酢酸エチル/メタノール、10:1)を実施して、透明油として非多分散の標題化合物(0.843g,収率19%)を得た。
非多分散化合物27の油(0.70g,1.4mmol)へ1NのNaOH(2.0mL)を加え、この反応混合液を4時間攪拌した。この粗反応混合液を濃縮し、酸性化し(pH〜2)、NaClを用いて飽和させ、そしてCH2Cl2(2×50mL)を用いて洗浄した。有機層を結合し、NaClを用いて飽和させ、MgSO4で乾燥させ、乾燥するまで蒸発させると、透明油として非多分散の標題化合物(0.35g,収率53%)が得られた。
非多分散mPEG7−C8酸(28)(0.3lg,0.64mmol)を3mLの無水塩化メチレン中に溶解させ、無水塩化メチレン中のN−ヒドロキシスクシンイミド(0.079g,0.69mmol)およびEDCI−HCl(135.6mg,0.71mmol)の溶液を加えた。この反応液を数時間攪拌し、その後1NのHCl、水を用いて洗浄し、MgSO4で乾燥させ、濾過して濃縮した。粗物質をカラムクロマトグラフィーにより精製し、濃縮すると透明油として非多分散の標題化合物が得られたので、これを真空により乾燥させた。
エタノール(100mL)中の非多分散10−ヒドロキシデカン酸(5.0g,26.5mmol)の溶液にH2SO4(0.43mL,8.8mmol)を加え、この反応液を加熱して攪拌しながら3時間還流させた。この粗反応混合液を室温へ冷却し、H2O(100mL)、飽和NaHCO3(2×100mL)、H2O(100mL)を用いて洗浄し、MgSO4で乾燥させ、乾燥するまで蒸発させると、透明油として非多分散の標題化合物(6.9g,収率98%)が得られた。
CH2Cl2(27mL)の溶液に非多分散10−ヒドロキシデカノエート30(5.6g,26mmol)を加え、氷浴中で0℃に冷却した。その後トリエチルアミン(5mL,37mmol)を加え、この反応混合液を0℃で15分間攪拌した。CH2Cl2(3mL)中に溶解させた塩化メタンスルホニル(2.7mL,24mmol)を加え、この反応混合液を0℃で30分間攪拌し、氷浴を取り除き、この反応液をさらに2時間、室温で攪拌した。この粗反応混合液をセライトに通して濾過(CH2Cl2、80mLを用いて洗浄)し、濾液をH2O(100mL)、5%のNaHCO3(2×100mL)、H2O(100mL)、飽和NaCl(100mL)を用いて洗浄し、MgSO4で乾燥させ、乾燥するまで蒸発させると黄色油として非多分散の標題化合物(7.42g,収率97%)が得られた。
テトラヒドロフラン(100mL)中の非多分散ヘプタエチレングリコールモノメチルエーテル25(2.5g,7.3mmol)の溶液に水素化ナトリウム(0.194g,8.1mmol)を加え、この反応混合液を1時間攪拌した。その後、テトラヒドロフラン(10mL)中に溶解させた非多分散10−ヒドロキシデカノエートのメシレート31(2.4g,8.1mmol)を滴下法で加え、この反応混合液を一晩攪拌した。粗反応混合液をセライトに通して濾過し(〜200mLのCH2Cl2で洗浄した)、乾燥するまで蒸発させた。生じた油を酢酸エチル中に溶解させ、H2O(2×200mL)を用いて洗浄し、MgSO4で乾燥させ、乾燥するまで蒸発させ、クロマトグラフィー(シリカ、酢酸エチル/メタノール、10:1)にかけ、そしてさらにクロマトグラフィー(シリカ、酢酸エチル)にかけると透明油として非多分散の標題化合物(0.570g,収率15%)が得られた。
非多分散のmPEG7−C10エステル32の油(0.570g,1.1mmol)へ1NのNaOH(1.6mL)を加え、この反応混合液を一晩攪拌した。この粗反応混合液を濃縮し、酸性化し(pH〜2)、NaClを用いて飽和させ、そしてCH2Cl2(2×50mL)を用いて洗浄した。有機層を結合し、NaCl(2×50mL)を用いて飽和させ、MgSO4で乾燥させ、乾燥するまで蒸発させると、透明油として非多分散の標題化合物(0.340g,収率62%)が得られた。
非多分散酸33は実施例24で記載した方法に類似する方法を使用して活性化した。
非多分散塩化ステアロイル(0.7g,2.31mmol)をベンゼン中のPEG6(5g,17.7mmol)およびピリジン(0.97g,12.4mmol)の混合液へ緩徐に加えた。この反応混合液を数時間(〜5)攪拌した。この反応の後に展開溶媒として酢酸エチル/メタノールを使用するTLCを実施した。その後、この反応液を水で洗浄し、MgSO4で乾燥させ、濃縮して真空により乾燥させた。精製した非多分散化合物36をFAB MSにより解析した。m/e 549/M+H。
非多分散C18(PEG6)の活性化は次の2段階で実施した。
1)非多分散ステアロイル−PEG6 36(0.8g,1.46mmol)をトルエン中に溶解させ、ホスゲン液(10mL,トルエン中で20%)に加え、これを氷浴を用いて冷却した。この反応混合液を0℃で1時間攪拌し、その後室温で3時間攪拌した。蒸留によりホスゲンおよびトルエンを除去し、残っている非多分散ステアロイルPEG6クロロホルメート37をP2O5で一晩乾燥させた。
非多分散テトラエチレングリコールの油(19.4g,0.10mol)にNaOH(4.0mL中の4.0g)の溶液を加え、この反応液を15分間攪拌した。塩化ベンジル(3.54mL,30.8mmol)を加え、この反応混合液を100℃へ加熱し、一晩攪拌した。この反応混合液を室温へ冷却させ、飽和NaCl(250mL)を用いて希釈し、そしてCH2Cl2(2×200mL)を用いて洗浄した。有機層を結合し、飽和NaClを用いて洗浄し、MgSO4で乾燥させ、クロマトグラフィー(シリカ、酢酸エチル)にかけると、黄色油として非多分散の標題化合物(6.21g,収率71%)が得られた。
CH2Cl2(20mL)の溶液に非多分散テトラエチレングリコールモノベンジルエーテル39(6.21g,22mmol)を加え、氷浴中で0℃に冷却した。その後トリエチルアミン(3.2mL,24mmol)を加え、この反応混合液を0℃で15分間攪拌した。その後CH2Cl2(2mL)中に溶解させた塩化メタンスルホニル(1.7mL,24mmol)を加え、この反応混合液を0℃で30分間攪拌し、氷浴を取り除き、この反応液をさらに2時間、室温で攪拌した。この粗反応混合液をセライトに通して濾過(80mLのCH2Cl2で洗浄)し、濾液を水(100mL)、5%のNaHCO3(2×100mL)、H2O(100mL)、飽和NaCl(100mL)を用いて洗浄し、MgSO4で乾燥させた。生じた黄色油を活性炭(10g)を含有するシリカのパッド上でクロマトグラフィーにかけると、透明油として非多分散の標題化合物(7.10g,収率89%)が得られた。
水素化ナトリウム(0.43g,18mmol)を含有するテトラヒドロフラン(140mL)の溶液に、テトラヒドロフラン(10mL)中の非多分散テトラエチレングリコール(3.5g,18mmol)の溶液を滴下法で加え、その反応混合液を1時間攪拌した。その後、テトラヒドロフラン(10mL)中に溶解させた非多分散テトラエチレングリコールモノベンジルエーテル40のメシレート(6.0g,16.5mmol)を滴下法で加え、この反応混合液を一晩攪拌した。粗反応混合液をセライトを通して濾過(250mLのCH2Cl2で洗浄)し、濾液をH2Oで洗浄し、MgSO4で乾燥させ、乾燥するまで蒸発させた。結果として生じた油はクロマトグラフィー(シリカ、酢酸エチル/メタノール、10:1)にかけ、さらにクロマトグラフィー(シリカ、クロロホルム/メタノール、25:1)を実施して、透明油として非多分散の標題化合物(2.62g,収率34%)を得た。
非多分散オクタエチレングリコールモノベンジルエーテル41(0.998g,2.07mmol)およびピリジン(163.9mg,2.07mmol)の攪拌冷却溶液にベンゼン中の非多分散塩化ステアロイル42(627.7mg,2.07mmol)を加えた。この反応混合液を一晩(18時間)攪拌した。翌日、この反応液を水で洗浄し、MgSO4で乾燥させ、濃縮して真空により乾燥させた。その後、粗生成物を、10%メタノール/90%クロロホルムを使用してフラッシュシリカゲルカラム上でクロマトグラフィーにかけた。生成物を含有するフラクションを結合し、濃縮し、真空で乾燥させると非多分散の標題化合物が得られた。
非多分散ステアリン酸−PEG8−Bzl 43(0.854g,1.138mmol)のメタノール液へPd/C(10%)(パラジウム、活性炭上で10重量%)を加えた。この反応混合液を水素下で一晩(18時間)攪拌した。その後、この溶液を濾過し、濃縮し、10%メタノール/90%クロロホルムを使用してフラッシュカラムクロマトグラフィーにより精製し、Rt=0.6のフラクションを収集し、濃縮し、乾燥させると非多分散酸44が得られた。
非多分散活性化C18(PEG8)オリゴマー45を得るために、非多分散ステアリン酸−PEG8オリゴマーの2段階活性化を上記実施例31におけるステアリン酸PEG6について記載した通りに実施した。
以下の説明は、図8に示したスキームを参照する。20%ホスゲンを含有するトルエン溶液(100mL,約18.7g,189mmolのホスゲン)をN2雰囲気下で0℃へ冷却した。非多分散mTEG(トリエチレングリコール、モノメチルエーテル、7.8g,47.5mol)を25mLの無水酢酸エチル中に溶解させ、これを冷却したホスゲン溶液に加えた。この反応混合液を6℃で1時間攪拌し、室温へ加温するに任せ、その後さらに2時間半攪拌した。残っているホスゲン、酢酸エチルおよびトルエンは真空蒸留を介して取り除くと、透明油性残留物として非多分散mTEGクロロホルメート46が残された。
以下の説明は、図9に示したスキームを参照する。非多分散パルミチン酸無水物(5g;10mmol)を無水THF(20mL)中に溶解させ、室温で攪拌した。この攪拌溶液に、3モル過剰のピリジンを加え、その後に非多分散トリエチレングリコール(1.4mL)を加えた。この反応混合液を1時間攪拌した(反応の進行はTLC;酢酸エチル−クロロホルム;3:7により監視した)。反応の終了時に、THFを除去し、生成物を10%のH2SO4と混合し、酢酸エチルにより抽出した(3×30mL)。結合した抽出物を水、食塩液を用いて連続的に洗浄し、MgSO4で乾燥させ、蒸発させて非多分散生成物48を得た。DMF(〜10mL)中のN−ジスクシンイミジルカルボネート(3mmol)の溶液を、10mLの無水DMF中の非多分散生成物48(1mmol)の溶液へ攪拌しながら加えた。水素化ナトリウム(3mmol)をこの反応混合液に緩徐に加えた。この反応混合液を数時間(例、5時間)攪拌した。活性化オリゴマーを沈降させるためにジエチルエーテルを加えた。このプロセスを3回繰り返し、生成物を最終的に乾燥させた。
以下の説明は、図10に示したスキームを参照する。非多分散活性化ヘキサエチレングリコールモノメチルエーテルは上記の実施例39における非多分散トリエチレングリコールと同様に調製した。トルエン中の20%ホスゲン溶液(35mL,6.66g,67.4mmolホスゲン)をN2雰囲気下の氷/塩水浴中で冷却した。非多分散ヘキサエチレングリコール50(1.85mL,2.0g,6.74mmol)を5mLの無水EtOAc中に溶解させ、これをシリンジによりホスゲン溶液に加えた。この反応混合液を1時間、氷浴中で攪拌し続け、取り出し、さらに室温で2.5時間攪拌した。ホスゲン、EtOAc、およびトルエンを真空蒸留を介して取り除くと、透明油性残留物として非多分散化合物51が残された。
サケカルシトニン−オリゴマー抱合体の混合物についての分散係数は次のように決定する。サケカルシトニン−オリゴマー抱合体の混合物は、例えば実施例41に記載したように用意する。この混合物の第1サンプルは、サンプル中の様々なサケカルシトニン−オリゴマー抱合体を分離し、単離するためにHPLCにより精製する。各単離フラクションが抱合体の純粋な単分散混合物を含有すると仮定すると、「n」は収集されたフラクションの数に等しい。この混合物は、コンジュゲートの位置、抱合体の程度の順に記載されている次の抱合体の1以上を含む可能性がある。Lys11モノ抱合体;Lys18モノ抱合体;N−末端モノ抱合体;Lys11,18ジ抱合体;Lys11,N−末端ジ抱合体;Lys18,N−末端ジ抱合体;および/またはLys11,18,N−末端トリ抱合体。フラクションの質量を決定するために、この混合物の各単離フラクションは、各単離フラクションをモノ抱合体、ジ抱合体もしくはトリ抱合体として分類することができ、サンプル中の各抱合体に対して可変の「Mi」に対する数値を提供する質量分析法により解析する。
T−47D細胞(乳腺管癌細胞系、アメリカンタイプカルチャーコレクションから入手)をランニング緩衝液(カリフォルニア州サニーベール所在のMolecular Devicesから入手した低緩衝、血清フリー、重炭酸塩フリーRPMI 1640培地)中の1×107cell/mLの密度で懸濁させた。およそ100,000cellを10μLの液滴中のアガロース細胞捕捉培地中に固定化し、サイトセンサー・カプセルカップ内の2枚の厚さ3μmのポリカーボネート膜の間に挟んだ。Cytosensor(登録商標)マイクロフィジオメーター(微小生理機能測定器)上のセンサーチャンバー内に入れたサイトセンサー・カプセルカップをpH感受性検出器の極めて近くに保持した。流動が停止する30秒間の間隔を除いて、ランニング緩衝液を100μL/分の速度で全細胞に行き渡るようにポンプで送り込み、センサーチャンバー内のランニング緩衝液の酸性化を測定した。酸性化率は2分間毎に測定した。センサーチャンバー内の温度は37℃であった。細胞は、実験開始前2〜3時間はセンサーチャンバー内で平衡化させ、その間に基底酸性化率を監視した。その後、細胞を様々なnM濃度のランニング緩衝液中で希釈した試験化合物(サケカルシトニンまたはオクチル−ジ−カルシトニン)に曝露させた。試験化合物への細胞の曝露は、合計20分間の反復パターンで行った各2分間のポンプサイクルの最初の40秒間に行った。これは細胞代謝におけるレセプター媒介性反応を引き出すための細胞の試験化合物への十分な曝露を許容し、その後に化合物を含有しないランニング緩衝液を約50秒間流動させた。この方法は酸性化率を測定する前にセンサーチャンバー内から試験液(ランニング緩衝液単独よりわずかに低いpHを有していた)を洗い流した。したがって、酸性化率は完全に細胞活性の尺度であった。類似の方法を使用して、PEG7−オクチル−sCT、モノ抱合体(オクチル−モノ);PEG7−デシル−sCT、モノ抱合体(デシル−モノ);PEG7−デシル−sCT、ジ抱合体(デシル−ジ);ステアリン酸−PEG6−sCT、モノ抱合体(PEG6 St.モノ);およびステアリン酸−PEG8−sCT、モノ抱合体(PEGS St.モノ)についてのデータを入手した。データは、各サイトセンサーチャンバーの酸性化率のグラフについて曲線下面積(AUC)を計算し、同一実験条件下で実施した複数の実験から入手した平均AUC測定値を示している図14に示した棒グラフとしてプロットすることにすることにより化合物の相対活性について解析した。
凍結乾燥粉末として供給された化合物をpH7.4の10mMのリン酸緩衝液中に再懸濁させ、その後HPLCによる濃度測定のために提出した。リン酸緩衝液を使用して、各特定の腸酵素の活性のために最適なpHを備える溶液を作製した。こうして調製した化合物のアリコートを1.7mL微量高速遠心管へ移し、37℃水浴中で15分間攪拌して化合物をその温度へ平衡化させた。15分後、適切に濃縮した腸酵素2μLを各遠心管へ加えて所望の最終濃度を達成した。キモトリプシンおよびトリプシンを1mMのHCl中へ再懸濁させた。同様に、コントロールとしての化合物も2μLの1mMのHClを用いて処理した。添加の直後に、コントロール遠心管から100μLのサンプルを取り出し、25μLのキモトリプシン/トリプシン急冷液(1:1 1%TFA:イソプロパノール)のどちらかを用いて急冷した。このサンプルはT=0minとする。使用した腸酵素に依存する様々な時間間隔でサンプリング方法を繰り返す。キモトリプシンは15分間、30分間および60分間のサンプルを有する。トリプシンは30分間、60分間、120分間および180分間のサンプルを有する。全時点が獲得されたら、観察された分解が温度もしくは緩衝液関連性ではないことを確認するために最終サンプルをコントロール遠心管から取り出す。キモトリプシンおよびトリプシンサンプルは、HPLCバイアル内へ直接に採取してよい。RP−HPLC(アセトニトリル勾配)を使用して各サンプルについてAUCを決定し、T=0minコントロールに基づいて分解率(%)を計算する。結果は以下の表1〜4に示した。
体重20〜25gの雄性CF−1マウス(チャールスリバー(Charles River)、ローリー、NC)をノベックス(Nobex)社動物飼養場内の照明(12:12の明暗周期、午前6時からの照明)、温度(21〜23℃)、そして湿度(40〜60%の相対湿度)を調節した飼養室に収容した。動物には、実験動物用飼料(PMI Nutrition社製)および水道水を自由に摂取させた。マウスは、実験当日前の45〜72時間に収容条件へ馴化させた。
Claims (17)
- 抱合体の単分散混合物であって、該抱合体の100%が同一の分子量を有する混合物、
抱合体の実質的に純粋な単分散混合物であって、該抱合体の少なくとも95%が同一の分子量を有し、同一の分子構造を有する混合物、または
抱合体の純粋な単分散混合物であって、該抱合体の100%が同一の分子量を有し、そして同一の分子構造を有する混合物
であって、
前記抱合体が、第1オリゴマーと、第2オリゴマーとを含み、各オリゴマーがポリエチレングリコール部分を含み、各オリゴマーがカルシトニンに結合しており、該第1オリゴマーが該カルシトニンのLys 11 のアミノ基に共有結合的に結合しており、該第2オリゴマーが該カルシトニンのLys 18 のアミノ基に共有結合的に結合している、混合物。 - 前記ポリエチレングリコール部分が少なくとも2個のポリエチレングリコールサブユニットを有する請求項1に記載の混合物。
- 前記ポリエチレングリコール部分の付加が、(a)血清中カルシウム濃度を少なくとも5%低下させる能力、(b)オリゴマーに結合されていないカルシトニンのキモトリプシンまたはトリプシンによる分解に対する抵抗性と比較した、キモトリプシンまたはトリプシンによる分解に対する高い抵抗性、(c)オリゴマーに結合されていないカルシトニンの生体有効性より高い生体有効性、及びこれらの組み合わせから選択される性質を提供する請求項1に記載の混合物。
- 前記カルシトニンが前記オリゴマーに、加水分解可能な結合、加水分解可能でない結合、またはそれらの両方により共有結合されている請求項1に記載の混合物。
- 前記カルシトニン薬剤が前記オリゴマーのポリエチレングリコール部分に共有結合されている請求項1に記載の混合物。
- 前記オリゴマーが親油性部分をさらに含む、及び/又は各オリゴマーが同一の分子構造を有する請求項1に記載の混合物。
- 前記カルシトニン及び/又はポリエチレングリコール部分が親油性部分に共有結合され
ている請求項6記載の混合物。 - 請求項1に記載の混合物と
医薬的に許容可能な担体と
を含む医薬組成物。 - 前記第1オリゴマーと、前記第2オリゴマーとが、カルボン酸含有化合物をさらに含み、該化合物のカルボン酸末端が、前記Lys 11 、及びLys 18 にアミド結合として結合され、該カルボン酸含有化合物の他方の末端が、少なくとも7個のポリエチレングリコールサブユニットを有するメチル末端ポリエチレングリコール部分に共有結合的に結合されている、請求項1に記載の抱合体の実質的に単分散の混合物。
- 前記カルボン酸含有化合物が、オクタン酸である請求項9に記載の混合物。
- 過度の破骨細胞性骨吸収または低カルシウム血性血清作用を特徴とする骨疾患の治療のための医薬の調製のための、請求項1〜10のいずれかに記載の抱合体の混合物の使用。
- 前記骨疾患が骨粗鬆症、ぺージェット病、または低カルシウム血症である請求項11に記載の使用。
- 各抱合体が少なくとも4個のポリエチレングリコールサブユニットを有するポリエチレングリコール部分を含むオリゴマーに結合されたカルシトニンを含む、抱合体の混合物であって、約22ダルトン未満の標準偏差を伴う分子量分布を有する、請求項1に記載の混合物。
- 前記分散係数が100,000より大きい請求項14に記載の抱合体の混合物。
- 各抱合体が同一の分子量を有し、各抱合体が次の式:
Bは、結合部分であって、共有結合、エステル部分、カルボネート部分、アミド部分、及び第二級アミン部分から選択される結合部分であり、
Lはリンカー部分であって、アルキル部分及び脂肪酸部分から選択されるリンカー部分であり、
G、G’およびG”は独立して選択されたスペーサー部分であって、糖コレステロール部分及びグリセリン部分から選択されるスペーサー部分であり、
Rは親油性部分およびR’はポリアルキレングリコール部分である、またはR’は親油性部分およびRはポリアルキレングリコール部分であり、
Tは末端部分であって、アルキル部分またはアルコキシ部分から選択される末端部分であり、
j、k、mおよびnは独立して0または1であり、
pは、1からカルシトニン薬剤上の求核性残基数までの整数である)を有する、請求項1に記載の抱合体の混合物であって、
該カルシトニン薬剤が、サケカルシトニンである、混合物。 - 各抱合体がポリエチレングリコール部分を含むオリゴマーに結合されたカルシトニンを含む、請求項1に記載の抱合体の実質的に単分散の混合物を合成するプロセスであって、式I:
式IIIのポリマーを含む実質的に単分散の混合物を活性化させて、カルシトニンと反応することのできる活性化されたポリマーの実質的に単分散の混合物を得るステップであって、該実質的に単分散の混合物を活性化するステップが、前記式IIIのポリマーの実質的に単分散の混合物を、N−ヒドロキシスクシンイミドと反応させるステップを含む、ステップと、
m+n個のサブユニットを備えるポリエチレングリコール部分を含むオリゴマーにカルシトニンのLys 11 及びLys 18 のアミノ官能基で結合されたカルシトニンを各々が含んでいる抱合体の実質的に単分散の混合物を得るのに十分な条件下で、該活性化されたポリマーの実質的に単分散の混合物と、カルシトニンとを反応させるステップと
を含むプロセス。
