Description
生産性酵母と酵母一倍体との戻し交雑により得られたFOH生産性酵母を使用する。すなわち、FOH生産性酵母と酵母(協会7号系清酒酵母)の一倍体株と交雑させ、胞子形成させた後、子嚢解剖を行い得られた一倍体株で、高濃度のFOHを生産する酵母とする。
すなわち、各菌株をYPD培地で、30℃で2日間振盪培養後、培養液中のFOHをジエチルエーテルで抽出し、極性キャピラリーカラムと水素炎イオン化検出器(FID)を装着したガスクロマトグラフを用いて分析した。なお、2-ドデカノールを内部標準物質として使用した。
HL296株の菌学的性質を以下に示す

(3)接合性
接合型:a型
接合能:協会7号酵母の型の一倍体と接合する。また、実験室酵母サッカロマイセス・セレビシエX2180−1Bと接合する。
(4)胞子形成
HL296株を、胞子形成用培地にて30℃で4日間培養し、胞子の有無を顕微鏡により観察した。HL296株は一倍体のため胞子形成しないが、X2180−1Bとの交雑株は良好に胞子形成した。
HL296株をYPD培地1000 mlで30℃で一晩培養し、水で2回洗浄した。その後、強力粉280g、酵母8.6g(wet)、上白糖21g、スキムミルク7.2g、食塩4.6g、無塩バター20g、水180mlを用い、ホームベーカリーにてパンを作製した。作製したパン1gをジエチルエーテル抽出後、ガスクロマトグラフによりFOH生産性の測定を行った。対照は市販のドライイースト3gを用いた。

- スクアレン合成酵素をコードするERG9遺伝子が、その塩基配列の714位のgをaに置換された配列番号1で表わされるerg9-2遺伝子に変異したサッカロミセス・セレビシエ(Saccharomyces cerevisiae)と、清酒酵母一倍体との戻し交雑により得られたファルネソール生産性酵母。
- ファルネソール生産性酵母が、サッカロミセス・セレビシエ(Saccharomyces cerevisiae)HL296株[FERM P−20132菌株]である請求項1に記載のファルネソール生産性酵母。
- 請求項1または2に記載のファルネソール生産性酵母で発酵させて製造されたアルコール飲料。
- 請求項1または2に記載のファルネソール生産性酵母を用いて発酵させて製造されたファルネソール含有食品。
