Description
Claims (10)
- 生体物質をカプセル封入したヒドロゲル組成物の調製方法であって、
(a)イソシアナート官能性ヒドロゲルプレポリマー、生体物質、および前記イソシアナート官能性ヒドロゲルプレポリマーと反応する少なくとも2個のチオール基を有する架橋剤を用意するステップ、
(b)前記イソシアナート官能性ヒドロゲルプレポリマー、前記生体物質、および前記架橋剤を水溶液中で混合するステップ、および
(c)前記生体物質を重合ヒドロゲル内にカプセル封入して、前記生体物質が生物活性を維持するようにステップ(b)の混合物を6.5と7.5の間のpHに維持するステップ
を含むことを特徴とする方法。 - 前記水溶液が、2種の異なるイソシアナート反応性求核基を有する水溶性二座架橋剤も含有していることを特徴とする請求項1に記載の方法。
- 前記チオール官能性架橋剤が、ポリエチレングリコール、ポリプロピレングリコール、またはそのコポリマーを含み、分子量が2,000と6,000の間にあることを特徴とする請求項1または2に記載の方法。
- 前記ヒドロゲルプレポリマーを、基板の予め特定した多数の領域上に繰り返し分配し、アレイとして配列した、個別のスポットを形成し、既知の位置にある前記スポットが、少なくとも2種の異なる生体物質を含有し、そして、前記生体物質が、重合するヒドロゲルに固定化されるためのチオール反応性基を含有することを特徴とする請求項1から3のいずれか1項に記載の方法。
- 生体物質をカプセル封入したヒドロゲル組成物であって、
イソシアナート官能性ヒドロゲルプレポリマー、生体物質、および少なくとも2個のチオール基を有する架橋剤を含み、
前記イソシアナート官能性ヒドロゲルプレポリマーが、生理的条件下の水溶液中での前記架橋剤との反応によって、前記生体物質の存在下で架橋されており、前記架橋の結果、前記生体物質が生物活性を維持するように6.5と7.5の間のpHに維持された、安定な重合ヒドロゲル内にカプセル封入されていることを特徴とするヒドロゲル組成物。 - 前記プレポリマーは、ポリエチレングリコール、ポリプロピレングリコール、またはそのコポリマーを含み、かつ、イソシアネート末端基を有し、前記生体物質が、タンパク質、核酸、生細胞、および分子量が100と2000の間にある有機分子からなる群から選択され、そして、前記架橋剤が、ポリエチレングリコール、ポリプロピレングリコール、またはそのコポリマーの主鎖を含み、かつ分子量が2,000と6,000の間にあることを特徴とする請求項5に記載のヒドロゲル組成物。
- 前記安定な重合ヒドロゲルが、光学的に透明であり、かつ、前記生体物質が、タンパク質、核酸、生細胞、および分子量が100と2000の間にある有機分子からなる群から選択されることを特徴とする請求項5または6に記載のヒドロゲル組成物。
- 1個のチオール基およびもう1個の異なるイソシアナート反応性基を有する二座架橋剤を更に含むことを特徴とする請求項5、6、または7のいずれか1項に記載のヒドロゲル組成物。
- 請求項5のヒドロゲル組成物が、基体に結合した複数の個別のスポットとして形成され、前記スポットが空間的に配列されてアレイを形成し、かつ複数のスポットが、前記アレイ内の既知の位置に少なくとも2種の異なる生体物質を含有するいくつかのスポットを含むことを特徴とするバイオチップ。
- (a)請求項9によるバイオチップを提供するステップ、
(b)バイオチップを分析物溶液に接触させるステップ、および
(c)バイオチップと分析物溶液の相互作用を検出するステップ
を含むことを特徴とするバイオアッセイ方法。
