Description
本発明は、腫瘍に固有の抗原マーカーとその抗体およびそれらの抗原マーカーに基づいた免疫アッセイ法に関する。また、本発明はワクチン、腫瘍の画像化、および腫瘍に特異的な試薬を用いた免疫療法に関する。
異物が体内に侵入すると、免疫系は抗体として知られるタンパク質を産生し、抗体は異物に特異的に結合する。抗体生産を促し、抗体が結合する標的となる物質がいわゆる抗原である。
細胞の表面に存在する抗原は、細胞表面のマーカーとして用いられる。マーカーに特異的な抗体によって、このマーカーを検出できるからである。したがって、抗体を用いれば、細胞の有無を検出することができることになる。細胞はその表面にあるマーカーで特徴づけることができ、抗体を用いれば、細胞型を決定できる。
細胞表面マーカーとなるものの多くはグリコシル化されたタンパク質(糖タンパク質)である。なかでもムチン型糖タンパク質が際立って多い。ムチン型糖タンパク質は高分子で、糖鎖の大部分は、N−アセチルガラクトサミンを介してタンパク質コアのセリン及び/又はスレオニンとO−結合型で連結する。
抗原の報告は数多くあるが、その中には、同じ宿主中で、腫瘍細胞の表面には現れるが、正常細胞の表面には現れない抗原として、ムチン型糖タンパク質を報告したものもある。また、このような腫瘍特異的抗原とその抗体の使用法に関する発表も数多くある。
例えば、エピグリカニン(epiglicanin)として知られるシアロムチン(sialomucin)に関する研究を報告した論文はかなり多い。エピグリカニンはマウスの癌細胞系統TA3−Haの細胞表面に存在することが分かっている。エピグリカニンは1972年に「Codington et al.,(1972)Biochemistry 11:2559-2564」によって初めて報告された。TA3−Haの腹水生細胞をTPCK−トリプシンで穏やかにタンパク質分解した後、ゲル濾過クロマトグラフィーにより遊離ペプチドを分画すると、高分子量の糖ペプチド分画がピークを示したため、この物質を対象として、以下に示すような数多くの研究がなされてきた(Codington et al.,(1973)J.Nat’l Cancer Inst. 51:585-591 ; Slayter et al.,(1973)J.Biol.Chem. 248:3405-3410 ; Codington et al.,(1975)Biochemistry 14:855-859;Codington et al.,(1979)J.Nat’l Cancer Instit. 63:153-162;Watkins et al.,(1990)Carbohydr.Res. 213:185-200;Henningson et al.,(1987)Cancer Immunol. Immunother. 25:231-241)。エピグリカニンは通常、分子量500,000で、その約80%は糖からなり、そのほとんどはGalβ(1→3)GalNAcの側鎖で、およそ1300個のアミノ酸からなるポリペプチド鎖のセリンまたはスレオニン残基に結合している。「Van den Eijnden et al.,(1979)J.Biol.Chem. 254:12153-12159」参照のこと。これらの論文の総説は「Codington et al.,(1992)Glycobiology 2:173-180;Haavik et al.,(1992)Glycobiology 2:217-224」参照のこと。
エピグリカニンに対するポリクローナル抗体は「Codington et al.,(1984)J.Nat’l Cancer Instit. 73:1029-1038」で報告されている。エピグリカニンに対するいくつかのモノクローナル抗体もまた報告されている(Codington,U.S. Patent 4,837,171;Haavik et al.,(1992)Glycobiology 2(3):217-224)。これらのポリクローナル抗体は腹膜液または胸膜液中に含まれる物質および転移性癌患者の血清中の物質と反応することが知られている(Codington et al.,(1984)J.Nat’l Cancer Inst. 73:1029-1038)。
他の腫瘍特異的マーカーも報告されている。「Samuel et al.,米国特許公報第5,110,911号」は腺癌に由来し、ヒト腫瘍細胞が産生する抗原を報告している。この糖タンパク質はトムセンーフリーデンライヒ(TF)抗原と呼ばれ、分子量1,000,000以上で、非潜在性のGalβ(1→3)GalNAcエピトープをもつのが特徴である。
「Kortright,米国特許公報第4,708,930号及び第4,743,543号」はヒトの癌細胞や組織の表面または細胞質内にある抗原決定基に特異的なネズミのモノクローナル抗体を報告している。この抗原決定基は”KC−4抗原”と名付けられ、2つの形態を有するといわれている。分子量が480,000−510,000のものと390,000−450,000のものである。KC−4抗原はヒトの前立腺癌から取り出された。
「Salem et al.,米国特許公報第4,921,789号」はヒトの結腸癌に対する、分子量約160,000の抗原マーカーを報告している。この抗原は、他の一定の抗原に対する抗体とは反応しないとされている。
「O’Brien,米国特許公報第4,921,790号」は卵巣嚢胞腺癌に関連する抗原であるCA125抗原のサブユニットを報告している。CA125抗原の分子量は約40,000である。
「Kufe,米国特許公報第4,963,484号及び第5,053,489号」はヒトの乳癌から得たDF3抗原の抗原決定基を(組換え法を用いて)同定したと報告している。「Chu et al.,米国特許公報第4,939,240号」が報告するモノクローナル抗体は、マウスに移植したヒトの乳癌の腫瘍を小さくしたとされる。
「Hellstrom et al.,米国特許公報第4,918,164号」はヒトの黒色腫のp97抗原に対する抗体の抗原結合部位に関する結合部位を有する抗イディオタイプモノクローナル抗体を報告している。
「Toth et a1.,米国特許公報第4,914,021号」は癌またはオロソムコイド関連抗原(CORA)という抗原に特異的なモノクローナル抗体を報告している。CORAは、分子量が約46,000−50,000で、等電点3.0−3.5、糖の含量は(重量比で)30%とされる。
「Yoshida,米国特許公報第4,892,935号」はヒトの肺癌抗原に対する抗体を報告している。この抗体は鱗状細胞の肺癌とは反応するが、小細胞の肺癌とは反応しない。
「Kjeldsen et al,(1988)Cancer Res.48:2214-2220」はTAG−72と命名された乳癌の糖タンパク質を報告している。
「Springer et a1.,(1988)Carbohydr.Res.178:271-292」はヒトの乳癌の糖タンパク質のTハプテンとTnハプテンを報告している。
「Tjandra et a1.,(1988)Br. J. Surg. 75:811-817」はMSAという乳癌の糖タンパク質を報告している。
「Isida et al.,(1989)Tumor Bio1. 10:12-22」はMFGMという乳癌抗原を報告している。
「Lan et a1.,(1985)Cancer res. 45:305-310」はDU−PAN−2という膵臓癌抗原を報告している。
「Hanisch et a1.,(1988)Carbohydr. Res. 178:29-47」はCA125という卵巣癌抗原を報告している。
「Hinoda et a1.,(1988)Cancer J. 42:653-658」はYH206という肺癌抗原を報告している。
発明の概要
本発明者らは、いずれの上皮組織で癌が発生するかに関わらず、一般的にヒトの癌腫に特徴的な糖タンパク質抗原を発見した。本発明者らは、この抗原をヒト癌腫抗原(HCA)と名付けた。HCAは通常、正常細胞中には存在しない。本発明の第一の観点は、最も広く示すなら、HCAとHCAの抗原決定基を含む断片とを、HCAが自然に存在する成分の中から、実質的に分離したことである。ここで、抗原決定基とは、単一抗原に特異的な抗HCAモノクローナル抗体が、HCAと結合するのに十分なHCAの部位、あるいは、(単独で、あるいはハプテン結合体として)哺乳動物の体内で、HCA特異的な免疫反応を惹き起こすのに十分なHCAの部位を意味するものとする。本発明者らは、分子量が50,000以下で、一般的には適当な抗原決定性を提示しないHCAの断片を見い出した。ある物質を十分に精製、単離して、後述する本発明に関わるさまざまな方法、例えば、抗HCA抗体を得るための免疫原として、あるいは、競合免疫アッセイ法における競合抗原として用いるとき、その物質はHCAが自然に生成している環境要素から実質的に分離されたという。例えば、HCAとHCA結合抗体からなるHCA免疫複合体[この複合体は非放射性標識を用いる免疫アッセイ法(競合法やELISA法)を行なって得ることができる]を洗浄して得たHCAは、HCAが自然に生成している環境要素から分離されたものとみなせる。
HCAは通常、以下の特徴を有する。
a)750,000を超える分子量を有する。
b)糖成分は、ムチン型糖タンパク質の特徴をもち、シアル酸とガラクトース、N−アセチルガラクトサミン残基の含有率が比較的高い(即ち、これら3つの残基の重量を合わせると糖含量の大部分(60%以上)を占める)。
c)等電点はpH3.0より低い。
d)一般的にヒト癌腫細胞上に存在する。
e)一般的に癌化されていないヒト細胞には存在しない。
f)抗ネズミ−エピグリカニン抗体AE3(ハイブリドーマHAE−3から産生された。HAE−3は「American Tissue Type Culture Collection(ATCC)」に「寄託番号HB-9467」として寄託されている。)と特異的に反応する抗原決定基を1つ以上もつ。
g)pH2.0以下の水溶液(例えばリン酸溶液やHCl溶液)中では一般に不溶である。
さらに典型的には、HCAの分子量は100万以上で等電点はpH2.5以下である。HCAは通常、腫瘍細胞上に見られる。例えば、結腸、肺、膵臓、乳房、前立腺などの癌細胞上あるいは、上皮由来の卵巣癌にも見られる。
HCAは抗マウス−エピグリカニン抗体と親和性を持つのが、一般的な特徴であろう。このような抗体への親和性は、O−グルカナーゼか過ヨウ素酸あるいはその両者に対して感受性を有する(即ち、これらの物質によって、免疫反応性が実質的に失われてしまうような)抗原決定基が存在するためであろう。また、このような抗体への親和性は、HCAのアシアロ化によって促進されよう。HCAは、糖の含有量が重量比にして50%以上あるのも特徴である。
また、HCAは単糖類の構成においても特徴がある。HCAはフコース、N−アセチルガラクトサミン、N−アセチルグルコサミン、ガラクトース、マンノース、N−アセチルノイラミン酸を、好ましくは以下の比率(±10%〜40%)で含む。以下の比率は、HCAをトリフルオロ酢酸で加水分解したのち、HPAEC−PADにより単糖類の構成を決定して求めた:フコース、3.6%;N−アセチルガラクトサミン、15.2%;N−アセチルグルコサミン、11.2%;ガラクトース、27.8%;マンノース、25.6%;N−アセチルノイラミン酸、16.6%。
HCAはアミノ酸の構成においても特徴がある。HCAは以下のアミノ酸残基を含む:セリン、スレオニン、グルタミン、アスパラギン、ロイシン、アラニン、グリシン、バリン、プロリン、リシン、イソロイシン、アルギニン、フェニルアラニン、チロシン、ヒスチジン、システイン。プロリン、フェニルアラニン、チロシン、システインはそれぞれ、HCAに含まれる全てのアミノ酸残基のなかで0−5%を占める。真空内で6M HClを用いてHCAを加水分解した後、アミノ酸解析により決定すると、アミノ酸残基は、主に以下の量、存在する:セリン、10.2%;スレオニン、3.6%;グルタミン、14.6%;アスパラギン、8.2%;ロイシン、14.5%;アラニン、6.6%;グリシン、23.4%;バリン、3.7%;プロリン、<1.0%;リシン、4.8%;イソロイシン、3.7%;アルギニン、4.3%;フェニルアラニン、<1.0%;チロシン、<1.0%;ヒスチジン、2.4%;システイン、<1.0%。
HCAは浮遊密度1.3−1.45g/ml、好ましくは1.34−1.41g/mlという特徴を有する。この数値は、47%セシウム−トリフルオロ酢酸を用いて、実施例19に述べる方法に従って遠心分離して決定したものである。エピグリカニン特異的抗体、例えばAE3やAD7、BF11などへの結合能力によっても、HCAを特徴づけることができる。HCAは、抗体AE3への結合能力において、抗体49H.8との親和性の100倍以上の親和性をもつという特徴がある。
本発明者らがHCAを発見したことにより、以下に述べる本発明のその他の観点が可能になった。
本発明の第二の観点は、一般的にヒト癌腫抗原に特異的な抗体が得られたことを特徴とする。中でも、当該抗体は、マウスのエピグリカニンよりもHCAに強く反応する。好ましい態様において、抗体は、HCAまたはその抗原決定基を含む断片を動物に抗原として投与することによって得るものとする。また、HCAに結合する抗体は、後述する抗−イディオタイプ抗体を動物に抗原として投与しても得られる。当該抗体が結合するには、Galβ(1→3)GalNAc鎖があるのが望ましい。また、本発明で用いられる抗体は、モノクローナル抗体であることが好ましい。
本発明の第三の観点は、HCAと結合する抗体、好ましくはモノクローナル抗体AE3をサンプルと反応させて、生物学的サンプル中のHCAの存在を検出するための免疫アッセイ法を提示することを特徴とする。この反応は、抗体とHCAが免疫複合体を形成するのに必要な時間と条件の下で行われる。反応後、この免疫複合体の定量が行われる。本発明の第三の観点の第一の局面において使用される抗体は、HCAまたはHCAの免疫原となる断片、または抗HCA抗体に結合するイディオタイプ抗体のいずれかを免疫原として哺乳動物(例えば、ウサギ、ヤギ、マウス、ブタなど)に投与して得た抗体である。本発明の本観点の説明の他の場合には、一般的にHCAに結合する抗体を(それらが上記した免疫原に対して得られたものであるか否かを問わないで)使用する。例えば、第二の局面においては、免疫複合体の形成は競合免疫アッセイ法によって測定されるという方法を特徴とする。即ち、HCA結合抗体を、試料および競合抗原に反応させる。例えば、標識したトレーサー抗原または固定した競合抗原に反応させる。三の局面においては、HCAに結合する別の抗体を用い、2種の抗体の一方を固定し、他方を標識して用いるサンドイッチ式の免疫アッセイ法を行うことを特徴とする。
本発明の第三の観点の第一の説明の好ましい態様は、HCAとHCAに結合する抗体とが形成する固定化した複合体を、この抗体に結合する別の標識された2次抗体を用いて検出するという方法である。これに代わる方法としては、HCAに結合する2次抗体を用いるサンドイッチ方式がある。サンドイッチ式の免疫アッセイ法においては、HCA結合抗体は不溶性物質に結合する捕獲抗体となり、HCAに結合する2次抗体が標識抗体となる。
上述の競合免疫アッセイ法またはサンドイッチ式免疫アッセイ法についても、本発明の第三の観点の第一の局面において述べられた抗体が用いられうる。本発明の第三の観点に関して、競合法に用いられる適当な抗原とは以下のものである。HCA、抗原決定基を含むHCA断片、エピグリカニン、抗原決定基を含むエピグリカニン断片。好ましい競合抗原の一つは、ピーナッツレクチンに結合しないエピグリカニンの画分である。競合抗原は標識されるか固定化される。
本発明の第三の観点のアッセイ法はサンプル中のHCAを測定するために用いられる。サンプルには、尿、血漿、血清、腹水、リンパ液など、また、乳癌腫などの固形の生体組織が含まれる。本発明のこの観点は、また、ヒト癌腫抗原を検出するためのキットを提示している。キットには、HCA結合抗体、およびこの抗体の、生体サンプル中のHCAへの結合を測定する手段が含まれる。好ましい態様においては、キットには上記の2次抗体あるいは競合抗原の一つが含まれる。
本発明の第四の観点は、腫瘍を(好ましくは生体内で)画像化する方法を提示する。画像化は、放射性同位元素などの画像化物質に結合した(一対になった)HCA結合抗体を投与して行われる。
本発明の第五の観点は、ヒトの腫瘍組織を選択的に標識する薬剤ないし方法を提示する。標識には、酵素標識や蛍光標識、染色、放射性同位元素などの検出用標識を結合(連結)したHCA結合抗体が用いられる。
本発明の第六の観点は、細胞毒素にHCA結合抗体を結合(一対になるように)させたものを投与することによって、乳癌腫などのヒト腫瘍を治療することを提示する。細胞毒素とはケモトキシン(例、リシン)や細胞毒性の放射性同位体などである。
本発明の第七の観点は、HCAに結合する抗イディオタイプ抗体を提示したことである。抗イディオタイプ抗体は以下のように利用できる。a)癌腫を予防するためのワクチンとして;b)免疫療法で、癌患者の免疫機構から抗癌腫免疫反応を誘起するために;c)前述の競合免疫アッセイ法における競合抗原として;d)HCAに結合する基質と結合しない基質とを分離するための精製試薬として。
本発明の第八の観点は、HCAをヒトに投与するのに適した組成物に処方して提示することにある。例えば、予防ワクチンとして、あるいは免疫療法で、癌患者の免疫機構から抗癌腫免疫反応を誘起するために用いるなど。

好ましい態様の記述
I.HCAの供給源
前述したように、HCAはヒトの癌腫抗原で、ヒト癌腫の樹立細胞系の培養によって、また、癌患者、特に乳癌と卵巣癌の患者の腹水から一般的に得ることができる。適当な細胞系を、後述する免疫アッセイ法により、HCAの産生についてスクリーニングすることができる。好ましい細胞系とは、このアッセイ法で測定して、比較的高濃度のHCAを産生すると認められる細胞系である。HCAの供給源となる培養細胞系の一つは、ヒト子宮内膜腺癌細胞系KLE(ATCC受託番号CRL1622号)で「Richardson et al. Gynecologic Oncology 17:213-230(1984)」に記載されている。当業者は、他にも多くの培養細胞系が供給源となりうると認識しているだろう。例えば、ATCCは上に述べたKLEを含む、様々なタイプの癌腫から得た癌細胞系を持っている。
HCAはまた、乳癌や結腸、前立腺、卵巣、肺などの癌に罹った患者の体液(胸膜水や腹水、血清など)からも分離することが出来る。
II.HCAの精製
一般に、癌細胞系を培養して(例えば、リチャードソン(Richardson)らによって記述されているように)、培養後の培地を濃縮する。例えば、100Kメンブレンを付けたフィルトロン-オメガ-ウルトラセット(Filtron Omega Ultrasette filtration system)濾過システムなどを用いて、高分子画分を単離する。次に、後述する抗HCA抗体を用いるか、前述し「Codington,米国特許公報第4,837,171号」に報告されているAE−3などの抗エピグリカニン抗体を用いて、HCAの免疫反応性を調べる。HCAの免疫反応性のほとんどの部分は高分子画分に保持されている。
高分子画分をさらに、セファロースCL−2Bなどのクロマトグラフィーにかける。再び、免疫反応性を測ると、その大部分は最も高い分子量の範囲(分子量100万−200万)に存在するはずである。
さらに、この高分子画分をクロマトグラフィー(例えば、モノPカラムを連結したファルマシアのFPLCシステム)にかける。次いでHCAの活性を、酵素連結型競合的結合アッセイ法などの免疫アッセイ法によって測る。HCAはモノPカラムにかけると沈澱しやすくなり、この不溶沈澱はNaClの濃度勾配にかけても除去できない。この沈澱は尿素やジチオスレイトール、塩酸グアニジン、過塩素酸などの水溶液中でもほとんど不溶である。
III.抗原決定基を含むHCA断片の採取
HCA本体は、分子量が過大で、不溶性でもあるため、抗原決定基を含むHCA断片を実験に用いる方が、通常便利である。そのような断片はタンパク質の部分加水分解によって作製できる。(例えば、TPCKトリプシンやプロナーゼTM、またはペプシンなどの酵素を用いる)。分解産物は、ゲル濾過などのクロマトグラフィーによって精製され、HCAの免疫反応を示す画分を採集する。
IV.HCAに対する抗体
上記の方法に従って回収したHCA、またはその断片を、免疫原として、マウスやウサギ、ラット、ヤギなどの哺乳類に投与する。これに関しては、さまざまなプロトコールが用いられよう。抗エピグリカニン抗体(後述される)を産生させるときは、抗HCAモノクローナル抗体を作成するときのプロトコールを用いるのが望ましい。好ましくは、動物には相当の期間(例えば、5カ月)にわたって抗原投与する。一般的には、「Haavik et al.,(1992)Glycobiology 2:217-224」を参照。マウスのモノクローナル抗体を採取するときは、抗原投与された動物の脾臓細胞が、標準の融合細胞によって不死化される。例えば、「Kohler and Milstein,(1977)Nature 256:495-497」参照。得られた抗HCAモノクローナル抗体やポリクローナル抗体は後に述べるさまざまな免疫学的処置に用いられよう。
V.HCAに対する免疫アッセイ法
HCAに対する免疫アッセイ法は上述の抗HCA抗体や、抗エピグリカニン抗体などのHCA結合抗体を用いて行うことができる。一般的には、適当な免疫アッセイ法であれば、いずれの方式も用いることができる。例えば、固定化したHCA/抗HCA抗体複合体を(放射性標識した免疫沈澱により)形成させ、これを抗HCA抗体に特異的な抗体を用いて検出することもできる。従って、もしHCA結合抗体がマウス由来の抗体であれば、免疫複合体は、標識されたヤギの抗マウス抗体で検出すればよい。本発明者らは、標識という語を一般的にあらゆるタイプの公知の標識を表すものとする。そのような標識には、酵素(例えば、ホースラディッシュパーオキシダーゼ)やその他、放射性標識、蛍光標識、発色団などが含まれる。ビオチン/アビジン標識システムも使用できよう。
競合方式(実施例18で述べられる)においては、標識されるかあるいは固定化された競合抗原を用いることが可能である。競合方式においては適切な競合抗原を用いることが特に重要である。好ましい競合抗原の一つにエピグリカニンAとして知られるエピグリカニンの分画成分がある。エピグリカニンAの取得については後述する(実施例17)。
反応用のウエルにまず血清を加え、次にモノクローナル抗体を加えると、最も良い結果が得られる。この二つの成分を、ウエルに加える前に混合してしまうと、十分な結果は得られない。また、短時間の内(約30−50分)に、血清と抗体をウエルに加えることも重要である。
この競合アッセイ法で使用するPBSはpH7.5−7.6で、リン酸は通常の免疫アッセイ法で使用される濃度の3倍を入れる。こうすればpHの変化によって起こる測定結果のばらつきを最小限に抑えることができる。
至適条件下で行われれば、競合的結合アッセイ法は癌患者一人一人の血液(血清または血漿)中にあるHCAの濃度を定量的に測定することもできる。好ましい手順の態様は、以下の順序である。
96穴微量滴定用イムノプレートのウエルにPBSに溶解したエピグリカニンA溶液(7.5ng/ml)100μlを加え、4℃で15−20時間放置してウエルをコーティングする。またはコーティング溶液が乾くまで放置する。このプレートは乾いたままならば、4℃で数週間保存が可能である。コーティング溶液を捨てた後、スーパーブロック(ピアス化学(株)、275μl/ウエル)で、4℃で60分間ウエルをブロックする。そして、このウエルをトゥイーン20(0.05%)を含むPBS溶液で洗浄する。この溶液はすべての場合に洗浄液として用いられる。次に、測定すべきサンプル(即ち、血清原液、HCAスタンダード、他のHCAを含む試料またはコントロール)50μlを加えた後、速やかに抗体50μlを加える。HCAの至適濃度は約1−200ng/mlの範囲である。モノクローナル抗体は200ng/ml(10ng/ウエル)の濃度で使用する。希釈はすべてスーパーブロック溶液で行われ、アッセイは各サンプルにつき3回反復する。
プレートは14−20時間定温放置した後、上に述べたように洗浄する。ホースラディッシュパーオキシダーゼに結合したヤギの抗マウス抗体IgM(PBS中で1:3000)を100μl加えた後、プレートを4℃で2.0時間インキュベートしてから洗浄する。HCAの濃度は、結合する抗体の量に比例する。抗体の濃度は基質[クエン酸緩衝液中1mg/mlの濃度の2,2’−アジノビス(3-エチルベンゾチアゾリン-6-スルホン酸)(ABTS)]と、1μl/mlの30%H2O2を加えて測定する。20−25℃で発色するので、分光光度計で405nmの波長の吸光度を読む。
これらのアッセイ法は以下の場合に利用されうる。a)癌腫のでき始めを検出する;b)癌の病期を判定して、将来の治療(全身療法か局所療法か)の方針を決める;c)癌腫治療の経過を追跡する;d)癌腫の再発を判定する。
VI.抗HCA抗体を用いた生体内での画像化
抗HCA抗体は、生体内で、腫瘍を特異的に画像化するのに用いることができる。例えば、抗体は、111Inや99mTcのような、生体内での画像化に適切な放射性同位元素に共有結合させることができる。131Iはあまり好ましくない。放射性同位元素を発生させ、放射性同位元素を抗体に結合させるための公知の技術が数多くあることは、当業者であればよく知っていることである。例えば、「Bartorelli,米国特許第4,732,862号」を参照。これによって、抗体の放射性標識に関する文献が具体化された。また、「Goldberg et al.,(1978)New England J. Med. 298:1384;Seragini et al.,(1989)Clin. Nucl. Med. 14:580-587」を参照。また、放射性標識抗体の調製に関する「Surwit,(1990)Antibody, Immunoconjugates,and Radiopharmaceuticals 3:48-49」を参照のこと。放射性標識用のキットを入手することもできる。例えば、アマシャム、メディフィジィクス、シィンコア、マリンクロットなどの商業的な供給者(市販業者)から入手できる。
上述の画像化物質を投与(注入)するのに、特異的な部位に注入したり、あるいは静脈から注入する方法など、さまざまな公知の技術があることも、専門的には、よく知られている。例えば、「Treves, U.S. Patent 4,729,380」を参照。読み取りは、コンピューターに接続したガンマ線カメラを用いる、全身スキャニング・シンチグラムなどの、標準的技術を用いて行われる。
VII.HCA結合抗体を用いた組織染色
抗HCA抗体またはHCA結合抗体を、上述の放射性画像化物質や蛍光画像化物質、可視的画像化物質(染料)に結合することができる。例えば、既に参照した「Kortright U.S. Patent 4,708,930」参照。組織切片を調製したり、適当な標識に結合したHCA結合抗体でそれらの切片を標識するのには、標準的な技術を用いる。電子顕微鏡の(電子密度の濃い)画像化物質(例えば、金[粒子])を用いる別の技術もある。
VIII.細胞選択的免疫療法
腫瘍細胞を破壊するために、HCA結合抗体を細胞毒性物質に連結させる。細胞毒素には、リシンA鎖、アブリンA鎖、モデクシンA鎖、ゲロニン、メルファラン、ブレオマイシン、アドリアマイシン、ダウノマイシン、ポークウィード抗ウイルス蛋白質(PAP、PAPII,PAP−S)などがある。公知の技術をもって、腫瘍特異的抗体に結合させることのできる、多くの放射性標識物質や化学細胞毒性物質があり、腫瘍組織に運ばれて、これを特異的に破壊することが専門的には知られている。「Blattler et al.,米国特許公報第4,542,225号」参照。
IX.抗イディオタイプ抗体
抗イディオタイプ抗体を得るための既知の技術があることは、専門的に知られている。例えば、HCA結合抗体に結合するイディオタイプ抗体など。HCAに結合する抗エピグリカニン抗体や抗HCA抗体を免疫原として、前述した免疫化方式に従って、哺乳動物に抗原投与する。前に引用した「Hellstrom et al.,米国特許公報4,918,164号」参照。この結果得た抗イディオタイプ抗体は以下の用途に用いられる。a)免疫反応を誘起して、癌の成長を阻害するためのワクチンとして;b)癌に対する、癌患者の免疫反応を誘起する免疫療法に;c)前述の競合免疫アッセイ法における競合抗原として;d)HCAに結合する基質を、HCAに結合しない基質から分離するための精製試薬として。
生体への応用に関しては、ヒトの治療により適した、マウスのモノクローナル抗体を作製するために、さまざまな既知の技術がある。例えば、「Winter et al.,(1991)Nature 349:293-299;Sahagan et al.,(1986)J. Immunol. 137:1066-1074;Boss et al.,米国特許公報4,816,397号」を参照。
X.HCAに基づくワクチンおよび免疫治療法
前述の方法で精製したHCA(あるいは、その抗原決定基を含む断片)は、発熱物質を除去し、緩衝剤で処理した食塩水などの標準的な賦形剤を用いてワクチンとして調製することができる。ワクチンは標準的な手法(例えば静脈から)で注入され、抗癌免疫反応を誘起するために一連の適当な処置が施される。このような反応は予防的なものであるか、(すでに癌にかかっていると診断された患者にとっては)治癒的なものである。
XI.実施例
以下に実施された実験は、本発明を説明するためのものであって、本発明の範囲を限定するものではない。
実施例1:癌腫細胞系の培養後培地の濾過
先に引用された、KLE子宮内膜癌細胞系は、リチャードソン(Richardson)ら(1984)によって述べられた通常の方法で培養される。通常、細胞はHam F12とダルベッコの修正イーグル培地を等量混ぜ、これに15%のウシ胎児血清(ギブコ社)、60mU/mLのインシュリン、20U/mLのペニシリン、20μg/mLのストレプトマイシン、0.5μg/mlのアムホテリシンを加えた培地で培養した。細胞が集密状態になった培養液の培地のサンプルを回収して、遠心分離した。この培養後培地を100Kメンブレン(4℃)を備えたフィルトロン・オメガ・ウルトラセットで濃縮し、後述する酵素競合的結合アッセイ法により、高分子画分のHC抗原含量を測定した。
実施例2:セファロースCL−2Bカラムによるゲル濾過クロマトグラフィー
実施例1で述べた濃縮培地のサンプルは、セファロースCL−2Bカラム(5X180cm)にロードしてpH7.5のPBS緩衝液で溶出するゲル濾過クロマトグラフィーにかけた。各分画を採集して、糖含量を、通常の、「Dubois et al.(1956)Anal.Chem. 28:350-356」によるフェノール−硫酸法で測定した。490nmでの吸光度を測った。集められた画分について、後述する酵素競合的結合アッセイ法を用いて、HC抗原含量を測定した。クロマトグラフィーを10回(1回につき3日かかる)かけた後、画分毎にまとめた。画分Aをオメガ100Kメンブレン付フィルトロン・ウルトラセットで濃縮して、さらに精製、特性試験するまで−20℃で保存した。
図1は、セファロースCL−2Bカラムを2本(1本あたり5X90cm)連ねたクロマトグラフィーにかけた結果を示す。各画分について、HCA結合抗体に対する反応性、タンパク質および糖含量について、後に述べる方法で測定した。矢印は次の分子量マーカーの溶出量を示す。1:デキストランT2000(2,000,000);2:サイログロブリン(670,000);3:ウシ血清アルブミン(BSA)(67,000);4:K2Cr2O7(294)。図に示された分画A,B,C,Dはそれぞれ一つにまとめた。
実施例3:モノPカラムによるFPLCイオン交換クロマトグラフィー
セファロースCL−2Bカラム(実施例2)から採取した画分Aサンプルを、ファルマシア・FPLCシステムに連結したモノPカラム(ファルマシア)(10X300mm)で分画した。カラム中のサンプルは15mMリン酸緩衝液(pH7.20)中の塩化ナトリウムの濃度勾配により溶出された。流出液の280nmでの吸光度が測定され、採集された画分は、後述するように、酵素競合的結合アッセイ法により、HCA結合抗体への結合活性を測定した。
図2に流出液の吸光度を示す。
実施例4:HCAの再溶解
画分A(実施例3)の沈澱を、2M尿素、4M尿素、2%ジチオスレイトール、2%EDTA、4Mグアニジン塩酸のいずれかで懸濁し、室温で16時間定温放置した。そして、各サンプルについて、後述するように、酵素競合的結合アッセイ法により、HCA結合抗体への結合活性を検定した。溶液になると、HCAの活性は殆ど失われた。
実施例5:HCAの断片化
画分I(実施例3)の懸濁液を、20μg/mL TPCK−トリプシンと、0.1Mリン酸ナトリウム緩衝液(pH6.8)中で、16時間、37℃に定温放置した。そして、この混合液を次にスーパーデックス200(10X600mm)カラムに入れ、0.15MのNaClを含む0.1Mリン酸ナトリウム緩衝液によって、1.0mL/分の流速で溶出した。280nmでの吸光度が測定され、採集された画分は、後述するように、HCA結合抗体への結合活性を検定した。図3は、流出液の280nmでの吸光度を示したものである。図には、分子量マーカーの溶出量が示されている。1:デキストランT500(500,000);2:サイログロブリン(670,000);3:アルドラーゼ(158,000);4:ウシ血清アルブミン(BSA)(67,000);5:リボヌクレアーゼA(13,700)
実施例6:ピーナッツ・アグルチニン・セファロース・カラムによるアフィニティークロマトグラフィー
スーパーデックス200カラム(実施例5)から回収した画分1のサンプルを、4%アガロースビーズで不溶化したピーナッツ(Arachis hypogea)・レクチンのカラム(10X50mm)(シグマ)に入れた。カラムは、0.1mL/分の流速でa)0.2M NaClを含む0.1Mのリン酸ナトリウム緩衝液(pH6.8);b)0.2M NaClを含む0.1Mのリン酸ナトリウム緩衝液(pH6.8)中の0〜2Mのメチル−β−D−ガラクトピラノシドの濃度勾配によって溶出した。280nmでの吸光度が測定され、採集された画分は、後述するように、酵素・ビオチン二重アッセイ法により、HCA結合抗体への結合活性を検定した。図4に流出液の280nmでの吸光度を示す。
実施例7:HCAエピトープの評価
KLE培養後培地のセファロースCL−2Bカラム流出画分A(実施例1)のサンプルに、次の4つの反応をそれぞれ行った。過ヨウ素酸酸化(pH4.0の10mM NaIO4により、20℃で30分間反応)、TPCK−トリプシン、ノイラミニダーゼ(II型コレラ菌(Vibrio cholera))、またはエンド−α−N−アセチル−D−ガラクトサミニダーゼ(O−グリカナーゼ)を加えて37℃で16時間定温放置する。その後、サンプルを希釈し、酵素競合的結合アッセイ法によって、HC活性を測定した。上記の試薬によって、HCA結合抗体の結合活性は、次のように変化した。ノイラミニダーゼ→19%活性あり;エンド−N−アセチル−ガラクトサミニダーゼ→0.1%活性あり;過ヨウ素酸(10mM,20℃,1h)→10%活性あり;TPCK−トリプシン→137%活性あり。
実施例8:非血清サンプルに対する酵素競合的結合アッセイ法
ヌンク社製マキシソープ96穴プレートの各ウエルに、PBS(pH7.50)に溶解したエピグリカニン(50ng/mL)を100μl加え、4℃で16時間定温放置した。0.5%のBSAを含むPBS(pH7.50)(PBS−BSA)を加え、45分間定温放置して、ウエルをブロックした。それに、次の2種類の溶液を加えた。a)エピグリカニンの標準溶液(1−200ng/ml)またはサンプルを50μL、b)精製した抗エピグリカニンIgMモノクローナル抗体(希釈1:OD280×10,000)を50μL。抗体も抗原もPBS−BSAで希釈した。混合液は振盪機に載せ、20℃で16時間定温放置した。ウエルを0.05%トウィーン20を含むPBSで3回洗浄(スカトロン微量プレート用洗浄機)してから、アルカリフォスファターゼ標識の抗マウスのヤギIgMをPBS−BSAで溶解して加え、20℃で2時間定温放置した。ウエルを3回洗った後、0.1Mエタノールアミン(pH10.0)に1mg/mLのリン酸p−ニトロフェニルを溶かした、基質溶液100μLを加えた。マッキントッシュ社のSEコンピューターをつないだバイオ・ラド社の微量プレート読み取り機で、マイクロプレート・マネイジャー・TMプログラムを用いて、405nmにおける吸光度を測定した。
実施例9:酵素・ビオチン・サンドイッチアッセイ法
ヌンク社製マキシソープU(Nunc Maxisorp U)96穴プレートの各ウエルに、PBS(pH7.5)に希釈した(2μg/mL)モノクローナル抗エピグリカニン抗体を100μL加え、4℃で16時間定温放置した。このコーティング溶液を除いた後、0.5%BSA入りPBS(pH7.5)を200μL加えて、4℃で45分間ブロックした。そして、このブロッキング液を除いた後、エピグリカニンの標準液またはサンプルを各ウエルに100μLずつ分注した。20℃で2時間定温放置した後、ウエル中の溶液を除去し、0.05%トウィーン20を含むPBS(pH7.5)(PBS/TWeen)200μLで2回洗った(スカトロンプレート洗浄機)。ビオチニル化した抗エピグリカニン抗体をPBS−BSAで希釈して(1μg/mL)、100μLずつ各ウエルに分注して、20℃で2時間定温放置した。その後、ウエル中の溶液を除き、200μLのPBS/TWeenで2回洗った。PBS−BSAで1:5000に希釈したストレプタビジン(streptavidin)−パーオキシダーゼ溶液100μLを各ウエルに加え、混合液を20℃で2時間定温放置した後、ウエル中の溶液を除き、200μLのPBS/Tweenで3回洗った。そして、クエン酸−リン酸緩衝液(pH5.9)に、1mg/mLのO−フェニレンジアミンと0.5μL/mLのH2O2(30%)を使用直前に混和して、混合液100μlを各ウエルに加えた。このプレートを発色するまで20℃に置き、0.3Mのクエン酸を50μL加えて反応を止めた。バイオ・ラド社の微量プレート読み取り機で、マイクロプレート・マネイジャー・TMプログラムを用いて、490nmにおける吸光度を測定して、濃度を計算した。
実施例10:等電点電気泳動
卵巣癌患者の腹水から分離したHCAの等電点電気泳動を以下の方法で行った。泳動は、RototoforTMセル(バイオ・ラド社)中、バイオライト3−10両性電解質担体を用いて12ワットで4時間行った。
子宮内膜癌の培養細胞系から分離したHCAの等電点電気泳動もRototoforTMセル(バイオ・ラド社)中、バイオライト3−10両性電解質担体を用いて12ワットで4時間行った。図5は、この実験結果を示したものである。
実施例11:エピグリカニンのモノクローナル抗体
モノクローナル抗エピグリカニン抗体を、前述した方法で調製した(Codington,米国特許公報4,837,171号;Haavik et al., 1992 Glycobiology)。C57BL/Jマウスに106の生きたTA3−Ha腹水細胞を皮下注射して免疫し、精製したエピグリカニンで二次免疫した。免疫されたマウスの脾臓細胞をBALB/c(NS1)マウスミエローマ細胞と融合させた。方法については、他の文献に述べられている。エピグリカニンに対する抗体を産生するハイブリドーマを、精製したエピグリカニンでコートした、微量滴定用プレートを用いた、抗体捕捉アッセイ法によりスクリーニングした。得られた抗体は、以前述べた方法にしたがって精製された(Haavik et al.,1992,Glycobiology)。特殊な精製法としては、エイビッドA1アフィニティーカラムTM(9X20mm、バイオプローブ社)にかけて溶出し、続いて、ABxカラム(7.75 X 100mm、J.T.Baker)とプロテインパックDEAE 5PWカラム(7.5 X 75mm、ウォーターズ)にかけて、HPLCで分画するというものがある。
実施例12:腹水中のHCA
癌患者の腹水を標準的な方法で採取し、2種の抗マウスエピグリカニン・モノクローナル抗体を用いて、後述する競合免疫アッセイ法で測定した結果を図6に示す。2つの抗体をそれぞれ、塗りつぶしたバーと点描したバーで示す。図6の患者の病名は以下の通りである。I=乳癌、2=卵巣癌、3=乳癌、4=卵巣癌。
実施例13:腹水由来のHCA
実施例12の患者4の腹水と子宮内膜癌細胞系の培養後培地のHCA活性を、フィルトロン・ダイアフローメンブレンに通して、分子量100,000で分画して濃縮した。抗エピグリカニンモノクローナル抗体を用い、競合的結合アッセイ法で、HCA活性を測定した。結果を図7に示す。HCA活性のほとんどは、メンブレン上に濃縮された画分に現れた。メンブレンを通過した画分のHCA活性は低すぎて、図7に表すことができなかった。
実施例14:精製と断片化の促進
濃縮された腹水はセファロースCL−2Bでゲル濾過し、フィルトロンウルトラセット(フィトロン技術会社、ノースボロ、マサチューセッツ州)で、限界分子量を100,000ダルトンに設定して濃縮した。さらに、この腹水濃縮液をセファロースCL−2Bカラム(5X180cm)(予め、リン酸緩衝食塩水(PBS)(pH7.50)で平衡させておいた)にかけて、1ml/分の流速で流出させた。各画分を採集して、糖含量をフェノール−硫酸法(Dubois et al.,1956)で測定した。また、490nmでの吸光度を測った。クロマトグラフィーによる分析で、蛋白質のおおよその濃度を測定するために、島津製作所のUV−可視光記録分光光度計、UV−160A型で、280nmでの吸光度を測った。吸光度を測るときに、PBSバッファーの吸光度を対照にした。採集した分画は酵素競合的結合アッセイ法によって、4種の抗エピグリカニンモノクローナル抗体を用いて、HCA抗原を定量した。各画分は、13回分画を繰り返したものを、それぞれ一つに集めた。画分Aを、オメガの100Kメンブレン付きフィルトロンウルトラセットで濃縮して、次の精製に進むまで、−20℃に保存した。
図8に結果を示した。分子量マーカーの溶出量が示されている。1:デキストランT2000(2,000,000);2:サイログロブリン(670,000);3:ウシ血清アルブミン(BSA)(67,000);4:K2Cr2O7(294)。各画分は図に示すようにまとめられた。
実施例15:断片化とカラムクロマトグラフィーの進行
HCAを分解するために、HCAの懸濁液に、等量の、TPCK−トリプシン(40μg/ml)を含む、0.1Mリン酸ナトリウム緩衝液(pH6.8)を加えて、37℃で16時間定温放置した。
濃縮され、トリプシン分解されて、セファロースCL−2Bカラムから溶出した画分Aは、HPLC−システムに連結したスーパーデックス200HR16/60カラム(10X600mm)にかけた。カラムは予め、0.2M NaClを含む0.1Mリン酸ナトリウム緩衝液(pH6.8)で平衡し、1.0ml/分の流速で溶出させ、1.0mlずつ分画した。流出液の280nmでの吸光度が測定され、採集された画分は、前述したように、酵素競合的結合アッセイ法により、HC抗原の含量を測定した。
図9に結果を示した。分子量マーカーの溶出量が示されている。1:デキストランT500(500,000);2:サイログロブリン(670,000);3:アルドラーゼ(158,000);4:ウシ血清アルブミン(67,000);5:リボヌクレアーゼA(13,700)。各画分は、それぞれ図に示すようにまとめられた。
実施例16:ヒト腹水由来HCAのpH依存的沈澱
図10-12は、腹水から採ったHCA(実施例12)の懸濁液の濁度を示したものであるが、この腹水には約100U/mlのHCAが含まれていた(これは、100ng/mlのエピグリカニンに対する免疫反応性に等しい)。溶液のpHはリン酸を滴下法で加えて調整した。図10は、リン酸を添加してから1分後毎に、500nmにおける吸光度を測ったものである。HCAはおよそpH1.1で自然に沈澱する。図11では、溶液にリン酸を加えてpHを1.0に調整しておき、調整後5−30分後の吸光度を測定したものである。また、溶液を攪拌しながらNaOHでpHを10.0に調整した。この溶液の一部を採って吸光度を測定した(図12)。
実施例17:エピグリカニンA
競合的HCA免疫アッセイ法において、好ましい競合物質はエピグリカニンAと命名されたエピグリカニンの分画成分である。エピグリカニンAはTA3−MM/1腹水細胞をもつマウスの腹水を(固定化されたピーナッツレクチンを用いた)アフィニティークロマトグラフィーにかけて得られた。レクチンに結合する画分をエピグリカニンBと名付け、カラムからそのまま流出する画分をエピグリカニンAと名付けた。
実施例18:競合的結合アッセイ法
血清標品に対して特に有用な、競合的結合アッセイ法(CBA)がヒトの血清中のHCA濃度を測定するために開発された。次の順序で行われる。
1.コーティング 25ng/mlのエピグリカニンAを含む、pH7.6のリン酸緩衝食塩水(PBS)100μlをウエルに分注し、4℃で8−16時間定温放置する。
2.定温放置した後、ウエルをPBSで洗う。
3.ブロッキング ヒト血清アルブミン(HSA)(0.6%)を含むPBS溶液をウエルに加えて2時間放置する。
4.血清は、予め、安定化溶液(0.010Mホウ酸ナトリウム溶液)でサンプルを1:5に希釈して、4℃に4−12時間定温放置する。その後、この溶液を0.1%HSAで、さらに1:5に希釈する。
5.モノクローナル抗体を、0.1%HSAでおよそ1:(O.D.280×10,000)に希釈する。
6.PBSでウエルを洗った後、まず血清(1:25 上記4)を50μl、次に希釈した抗体(上記5)を50μl加える。
7.プラスチックシートでプレートを覆って、4℃で12-18時間、ゆっくり振とうしながら定温放置する。
8.PBSでプレートを洗った後、フォスファターゼで標識した、ヤギの抗マウスIgM(μ鎖特異的)(1:4,000-10,000)を加え、4℃で3-4時間、ゆっくり振とうしながら定温放置する。
9.プレートを洗い、基質(炭酸−重炭酸緩衝液(pH10.3)12mlに、リン酸パラニトロフェニル15mgを溶解する)を100μl加える。
10.室温(20-22℃)に放置して、発色するのを待つ。ダイナテック自動プレート読み取り機で、405nmにおける発色強度を測る。
実施例19:HCAの浮遊密度
腹水とKLE−1細胞系から抽出したHCAの浮遊密度を、技術的には既に知られているトリフルオロ酢酸セシウムの密度勾配遠心法によって測定した。HCAのサンプルを、47%トリフルオロ酢酸セシウムに入れ、ベックマンのT−1250ローターを130,000×gで72時間回転させて遠心した。1mlずつ分画して採取し、それぞれのサンプルの密度を決めてから、サンプルの一部を使って、先述した酵素競合的結合アッセイ法でHCA活性を検定した。卵巣癌患者の腹水由来のHCAと細胞系KLE−1の培養後培地由来のHCAを用いて行った結合アッセイ法の結果をそれぞれ図13と図14に示す。
図13では、ヒトの腹水から精製されたHCA(図8のセファロースCL−2Bカラムから採取した画分A)を、47%トリフルオロ酢酸セシウム中、130,000×gで72時間遠心した(○は濃度を表す)。HCA活性(■)を、抗EPGNG−1抗体を用いて、酵素競合的結合アッセイ法によって調べた。
図14では、子宮内膜癌細胞系(KLE)の培養後培地から精製したHCAを、47%トリフルオロ酢酸セシウム中、130,000×gで72時間遠心した。(○は濃度を表す)。HCA活性(■)を、抗EPGN B−4抗体を用いて、酵素競合的結合アッセイ法によって調べた。
実施例20:固定化されたモノクローナル抗エピグリカニン抗体カラムを用いたアフィニティークロマトグラフィー
HCAを精製するために、KLE−1細胞系の培養後培地とヒトの腹水から採取したサンプルを、抗エピグリカニンAE−3を共有結合させたカーボリンクTM・カップリングゲル(Carbolink Coupling Gel)(ピアス化学、ロックフォード、イリノイ州)のカラム(1×5cm)にかけた。サンプルをカラムに載せた後、PBS(pH7.5)で溶出し、その後さらに、4Mグアニジン塩酸を含むPBS(pH7.5)で溶出した。2mlずつ分画し、各画分の希釈した一部を用いて、酵素競合的結合アッセイ法によってHCA活性を測った。KLE−1細胞系の培養後培地の、HCAに関する典型的な溶出グラフを図15に示す。
図15はヒト子宮内膜癌細胞系KLEの培養後培地を抗エピグリカニン抗体AE−3のアフィニティーカラムにかけたクロマトグラフィー分析の結果である。4Mグアニジン塩酸による溶出は矢印で示した所から始まっている。「−」で示した、ピークの画分を集めて、透析し、さらに分析するためにSDS−PAGEを行ない、PVDFメンブレンにブロットした。
実施例21:SDS−PAGEとブロッティングによるHCAの可視化
アフィニティーカラムにかけて、4Mグアニジン塩酸で流出し、ピークを形成した画分を集めて、これをさらに蒸留水で透析し、凍結乾燥し、メルカプトエタノールを含むSDS−PAGE用サンプルブッファーに再溶解して、沸騰水の中で5分間加熱した後、8%ゲルを用いてSDS−PAGEを行なった(Laemmli,(1970)Nature 227:680-685)。電気泳動によって分離された成分を、電気ブロティング法により、イモビロンTM(Immobilon)PVDFトランスファー用メンブレン(ミリポア)に転移させた。ブロットされたメンブレンはクマシー・ブリリアントブルーR−250によりタンパク質を染色し、さらに、抗エピグリカニン抗体AE−3でインキュベートした後、アフィニティー精製したヤギの抗マウスIgMをアルカリホスファターゼに結合した2次抗体(ベーリンガー・マンハイム)とともに定温放置することによりHCAを検出した。ナフトールAS−MXリン酸とO−ジアニシジンを含む基質混合液で定温放置して、0.1Mのトリス塩酸緩衝液(pH10.0)中でテトラゾ化(ファーストブルーBソルト)し、適当な濃さに発色したところで蒸留水で洗浄すると、タンパク質のバンドを可視化することができる(図16B参照)。
図16Aと16Bはそれぞれ、SDS−PAGEゲル(8%ポリアクリルアミド)をクマシー・ブリリアントブルーで染色したものとKLE細胞から精製されたHCAをPVDFメンブレンにブロットしたものである。

実施例22:HCAのアミノ酸と単糖構成の決定
クマシー・ブリリアントブルーで染色した後、PVDFメンブレンからHCAのバンドを切り出した。バンドの一部は真空状態で6M塩酸を用いて加水分解した後、この加水分解物をアミノ酸分析した(表1)。他の一部は2Mトリフルオロ酢酸で加水分解した後、HPAEC−PADにより単糖構成を測定した(Weitzhandler,et al.(1993)J.Biol.Chem.268:5121-5130)、(表2)。
表に示したHCAのアミノ酸構成は、マウスの乳癌細胞系TA3−Haのエピグリカニン(Codington and Haavik(1992)Glycobiology 2:173-180)や、MUC1(エピシアリン)マウス腫瘍関連ムチン(Spicer,et al.(1991)J.Biol.Chem.266:15099-15109)、13762ラット乳腺癌細胞系のシアロムチンASGP−1(Carraway and Spielman(1986)Mol. Cell. Biochem.72:109-120)、ヒト結腸癌異種移植細胞LS−174Tに由来するTAG−72(Sheer,et al.(1988)Cancer Res.48:6811-6818)、ヒトの喉頭癌から分離されたエピテクチン(Ca抗原)(Bhavanandan, et al.(1988)Ind.J.Biochem.Biophys. 25:36-42)、またヒト膵臓癌細胞系HPAF由来の膵臓腫瘍ムチン(Lan,et al.(1990)J.Biol.Chem. 265:15294-15299)などのアミノ酸構成とはかなり異なっていた。(表3)
図17は、HCAとMUC1のアミノ酸構成を比較したものであるが、これをみると両者の違いは明らかである。同様に、図18はHCAとTF抗原(Samuel,et al. U.S. Patent 5,110,911)のアミノ酸構成を比較したものであるが、この2つの抗原がアミノ酸構成において、非常に異なっていることがわかる。
実施例23:抗エピグリカニン抗体AE−3の特異性の調査
抗エピグリカニンAE−3の結合特異性を決定するために、微量滴定用プレートのウエルをエピグリカニンでコートして、このウエルに、AE−3抗体と未処理のHCAあるいは、AE−3抗体と、下記のいずれかの処理を予め施したHCAとの混合液を加えて、定温放置した。即ち、10mM NaIO4を加えて30分間20℃で定温放置、ノイラミニダーゼ(コレラ菌(Vibrio cholerae))処理、エンド−α−N−アセチルガラクトサミニダーゼ(F)処理、トリプシン処理、またはプロナーゼ処理。これらの実験結果から、AE−3抗体は、糖質を含むエピトープあるいは抗体との結合構造を維持するために一定の糖の構造体を必要とするエピトープに結合することが示された。
図19は、エピグリカニンとモノクローナル抗エピグリカニン抗体AE−3との結合を、ヒトの腹水から分離したままのHCAまたは変性させたHCAによって阻害させる競合アッセイの結果を示す。エピグリカニンでコートされたウエルの中にAE−3抗体と未処理のHCA(×)、あるいはAE−3抗体と次のいずれかの処理を予め施した加えたHCAとの混合液を加えて定温放置した。即ち、10mM NaIO4を加えて30分間20℃で定温放置したHCA(●)、ノイラミニダーゼ(コレラ菌)処理したHCA(▲)、エンド−α−N−アセチルガラクトサミニダーゼ(F)処理したHCA(◆)トリプシン処理したHCA(◇)、プロナーゼ処理したHCA(○)。
単離された糖タンパク質またはエピグリカニンをTPCK−トリプシン、プロナーゼ、ノイラミニダーゼ、O−グルカナーゼ、または過ヨウ素酸で処理して、抗原活性を調べた結果から、抗エピグリカニン抗体は、糖タンパク質上のGalβ(1→3)GalNAcを含むエピトープに結合することが示された。
実施例24:抗エピグリカニン抗体と他の腫瘍特異的抗体の特異性の比較
図20に示した実験では、抗エピグリカニン抗体G−1とAE−3の間で起こる共発現と交差反応を、他のTF特異的抗体HH8(Clausen,et al.(1988)Mol. Immunol.25:199-204)、シアロシル−Tn−特異的抗体、TKH2(Kjeldsen, et al.(1988)Cancer Res.48:2214-2220)、Tn−特異的抗体、TKH6(Clausen and Hakomori(1989)Vox.Sang.56:1-20)、A型血液抗原特異的抗体、AH16(Clausen and Hakomori(1989,supra))などと比較した。
エピグリカニン(4−61)でコートした微量滴定用プレートに、希釈したマウス・モノクローナル抗体を加えて定温放置した後、アルカリフォスファターゼを結合した、ヤギの抗マウスIgを加えて定温放置した。
図20から、抗エピグリカニン抗体G−1、AE−3と他の抗体HH8、TKH2、TKH6、AH16の間にほとんど交差反応性がないことがわかる。このことは、抗エピグリカニン抗体が、HH8、TKH2、TKH6、AH16のいずれとも異なったエピトープに結合することを示している。
図21に示す実験では、エピグリカニンと二糖Galβ(1→3)GalNAc(TF二糖)の阻害活性を、AE−3とエピグリカニンの結合を阻害する効果に関して比較した。エピグリカニン(4−61)でコートした微量滴定用プレートをBSAでブロックしてから、抗エピグリカニンAE−3と阻害物質の混合液を加えて定温放置した。その後プレートにヤギ抗マウスIgM−アルカリフォスファターゼを加えて定温放置した。TF二糖は、非常に高い濃度で、エピグリカニンとAE−3の結合を阻害することがわかった。しかし、エピグリカニンの阻害活性の方が、二糖の阻害活性に較べて108倍高かったため、エピグリカニンとTF二糖は交差阻害しないといえる。
実施例25:ヒト癌腫の診断
本発明のイムノアッセイ法を用いれば、癌腫を検出し、その病期を決定することができる。既に述べた競合的結合アッセイ法において、抗エピグリカニン・モノクローナル抗体を用いれば、癌が進行した患者の血清からHCAを同定するこたができる。
本発明の競合的結合イムノアッセイ法は、HCA特異的モノクローナル抗体、AE−3を用いて行われる。23人の第4期癌患者と70人の正常な患者の血清を対象に行なった研究では、第4期患者の血清中のHCAの量は、正常な患者の血清中のHCAの量に較べて上昇していた。感度[異常な(上昇した)測定値を示した、第4期患者の割合]は88%で、このとき特異性[正常な測定値を示した正常患者の割合]は95%であった。この結果は、本発明のAE−3に基づくイムノアッセイ法で、転移性癌をもつ患者と持たない患者とを分類できることを示している。
検定法間の変動係数(標準偏差を平均値で割ったもの)を70人の正常患者の測定値を用いて算定した。本アッセイ法の再現性を判定するために、対象者たちの、日による変動係数を算定した。正常患者すべてを含む集団で、この変動係数は、4.0%であった。次に、対象者集団内の変動をみるために、一定の日の対象者間の変動係数を計算すると、これは0.6%だった。これらの値は、このアッセイ法にかなりの程度の再現性があることを示している。

Claims (37)
- HCAが天然に存在する環境の成分から実質的に単離された、ヒト癌腫抗原(HCA)またはその抗原決定基を含むプロテアーゼ処理断片において、HCAが以
下の特徴を有するものであるHCAまたはその断片:
a)分子量が750,000を超える;
b)糖質成分がムチン型糖タンパク質の特徴を示し、シアル酸、ガラクトース、N-アセチルガラクトサミン残基の総重量が糖質の全体量に比例して比較的高
い割合を占める;
c)等電点はpH3.0以下;
d)ヒト癌腫細胞に通常存在する;
e)非トランスフォーメーションヒト細胞には、一般的に存在しない;
f)抗ネズミ・エピグリカニン・モノクローナル抗体AE3(ハイブリドーマHAE-3(ATCC寄託番号HB-9467)より産生)と特異的に反応する
抗原決定基を少なくとも一つ有する;
g)pH2.0以下の水溶液には一般的に不溶性である。 - HCAが100万以上の分子量を有することを特徴とする、請求項1記載のHCAまたはその抗原決定基を含むプロテアーゼ処理断片。
- 抗原決定基が抗マウス・エピグリカニン・モノクローナル抗体に結合し、O-グルカナーゼもしくは過ヨウ素酸、またはその両者に感受性であ
ることを特徴とする、請求項1記載のHCAまたはその抗原決定基を含むプロテアーゼ処理断片。 - HCAの糖質含量の少なくとも50重量%が、シアル酸、ガラクトース、N-アセチルガラクトサミン、N-アセチルグルコサミン、マンノー
ス、およびフコース残基の総重量で占められている、請求項1記載のHCAまたはその抗原決定基を含むプロテアーゼ処理断片。 - 抗マウス・エピグリカニン抗体よりも抗HCA抗体に対して強い親和性を有する、請求項1記載のHCAまたはその抗原決定基を含むプロテ
アーゼ処理断片。 - HCAの少なくとも50重量%が糖質である、請求項1記載のHCAまたはその抗原決定基を含むプロテアーゼ処理断片。
- フコース、ガラクトース/N-アセチルガラクトサミン、N-アセチルグルコサミン、ガラクトース、マンノース、およびN-アセチルノイラ
ミン酸の各単糖類を含む、請求項1記載のHCAまたはその抗原決定基を含むプロテアーゼ処理断片。 - トリフルオロ酢酸を用いてHCAを加水分解した後、パルス電流滴定装置付き高速陰イオン交換クロマトグラフィー(HPAEC-PAD)に
より決定した以下の組成の単糖類を含む、請求項1記載のHCAまたはその抗原決定基を含むプロテアーゼ処理断片。

フコース 3.6%
N-アセチルガラクトサミン 15.2%
N-アセチルグルコサミン 11.2%
ガラクトース 27.8%
マンノース 25.6%
N-アセチルノイラミン酸 16.6% - トリフルオロ酢酸を用いてHCAを加水分解した後、パルス電流滴定装置付き高速陰イオン交換クロマトグラフィー(HPAEC-PAD)に
より決定した以下の組成の単糖類を含む、請求項1記載のHCAまたはその抗原決定基を含むプロテアーゼ処理断片。

ルギニン、フェニルアラニン、チロシン、ヒスチジン、システインのそれぞれのアミノ酸残基を含み、該タンパク質のアミノ酸残基全体の0〜5%がプロリン、
フェニルアラニン、チロシン、またはシステインである、請求項1記載のHCAまたはその抗原決定基を含むプロテアーゼ処理断片。 - 真空内で6M
HClを用いてHCAを加水分解した後アミノ酸分析して決定したアミノ酸組成が以下のものである、請求項1記載のHCAまたはその抗原決定基を含むプロテ

原決定基を含むプロテアーゼ処理断片。 - 生物サンプルにおける請求項1記載のヒト癌腫抗原(HCA)の存在を測定するためのアッセイ法において、次の工程を含む方法:
当該サンプルを準備する工程;
しばらくの間当該サンプルを、前記HCAに結合する抗体と、当該抗体と前記HCAとが免疫複合体を形成することができる条件下で反応させる工程;
当該免疫複合体形成を測定する工程。 - 前記抗体と前記サンプルとを反応させる過程、および該抗体を免疫学的にHCAと交差反応する競合抗原と反応させる過程を含む、競合的免
疫アッセイ法によって免疫複合体の形成を判定する、請求項13記載のアッセイ法。 - HCAに結合する抗体は1次抗体で、免疫複合体は、該1次抗体または該HCAに結合し標識された2次抗体との反応によって測定される、
請求項13記載のアッセイ法。 - 生物サンプルにおける請求項1記載のヒト癌腫抗原(HCA)の存在を測定するためのアッセイ法において、次の工程を含むアッセイ法:
当該サンプルを準備する工程;
しばらくの間当該サンプルを、前記HCAに結合するモノクローナル抗体と、当該モノクローナル抗体と前記HCAとが免疫複合体を形成することができる条
件下で反応させる工程;および
該モノクローナル抗体と該サンプルを反応させる過程と、該モノクローナル抗体を、免疫学的に前記HCAと交差反応する競合抗原と反応させる過程とを含
む、競合的免疫アッセイ法によって、免疫複合体の形成を判定する工程。 - 当該モノクローナル抗体がAE3(ハイブリドーマHAE-3(ATCC寄託番号HB-9467)より産生)である、請求項16記載の
アッセイ法。 - 競合抗原が、請求項1記載のHCA、該HCAの抗原決定基を含むプロテアーゼ処理断片、エピグリカニン、およびエピグリ
カニンの抗原決定基を含む断片からなる群より選択される、請求項14、16または17記載のアッセイ法。 - 競合抗原が固体の表面上に固定される、請求項18記載のアッセイ法。
- 競合抗原が検出可能な標識で標識される、請求項18記載のアッセイ法。
- 競合抗原がエピグリカニンのピーナッツレクチンに結合しない分画を含む、請求項18記載のアッセイ法。
- 生物サンプルにおける請求項1記載のヒト癌腫抗原(HCA)の存在を測定するためのアッセイ法において、次の工程を含むアッセイ法:
該サンプルを準備する工程;
しばらくの間、該サンプルを、前記HCAに結合する抗体と、該抗体と前記HCAとが免疫複合体を形成することができる条件下で反応させる工程;および
サンドイッチ式免疫アッセイ法によって該免疫複合体の形成の判定をする工程であって、前記HCAに結合する抗体が1次抗体で、該免疫複合体が前記HCA
に結合し標識された2次抗体との反応によって測定される、工程。 - サンプルが、尿、血漿、血清、およびリンパ液からなる群より選択される体液である、請求項13、16または22記載のアッセイ法。
- 抗体が、請求項1記載のHCA、およびHCAの抗原決定基を含むプロテアーゼ処理断片からなる群より選択される免疫原
