Description
1.血液凝固分析器10(一般に使用されているもの)は、最も強力な血小板アクチベーターのトロンビンにより刺激された血小板機能(MA又はMAP)を測定する。血小板機能の小さい阻害に対してMA又はMAPを敏感にするためには、血小板機能は、トロンビンより強力でない血小板アクチベーター例えばADPによって活性化されるべきである。それ故、この例で血液試料を血液凝固分析器10中を流す場合には、トロンビンの形成を、例えば、クエン酸ナトリウムにより阻止し、代わりにADPを用いて血小板機能を活性化する。
1.血液凝固分析器10の捩り針金の変形:剪断力に対する様々な感度のための種々の強度の捩り針金を、適当に測定するために、作成することにより、様々な潜在能力の抗血小板剤の効果を測定することができる。捩り針金の感度は、一般に、その太さに関係する。約150〜1000dyn/cm2の範囲の血餅検知感度の太さを有する捩り針金が、上記の米国特許出願第09/255,066号に記載された血液凝固分析器に適合させるのに適当である。
Claims (10)
- 抗血小板療法の効果を測定する方法であって、下記のステップを含む当該方法:
血液凝固分析器を用意し、該血液凝固分析器は、血餅強度を測定することができ;
該血液凝固分析器を用いて、抗血小板療法の非存在下で得た第一の血液試料の第一の血液凝固パラメーターを測定し;
該血液凝固分析器を用いて、抗血小板療法の存在下で得た第二の血液試料の第二の血液凝固パラメーターを測定し;
ここに、第一の血液凝固パラメーター及び第二の血液凝固パラメーターの少なくとも一つが、それぞれの血液試料の試料として採ったままの部分の特性並びにトロンビン活性化を阻止し且つフィブリノーゲン及び血小板の活性化を保護するためにイン・ビトロで処理したそれぞれの血液試料の一部分の特性に基づくことを特徴とし;そして
第一及び第二の血液凝固パラメーターに基づいて、抗血小板療法の効力を測定する。 - 血液凝固分析器が、100〜1000dyn/cm2の範囲の血餅強度を測定するために操作可能な、請求項1に記載の方法。
- 血液凝固分析器が、第一及び第二の血液試料の実質的に同時の試験を提供する、請求項1に記載の方法。
- 血栓症の危険の評価と共に行なう、請求項1に記載の方法。
- 過剰凝固性状態及び低凝固性状態の評価の一つと共に行なう、請求項1に記載の方法。
- 抗血小板剤の投与量を決定するステップを更に含む、請求項1に記載の方法。
- 血液凝固分析器が、処理能力を含み、効力の測定のステップが該血液凝固分析器により行なわれる、請求項1に記載の方法。
- 第一及び第二の血液凝固パラメーターが、次の一つに基づく、請求項1に記載の方法:血餅形成までの潜伏期、血餅形成速度、最大血餅強度及び血餅溶解速度。
- 第一及び第二の血液凝固パラメーターが、フィブリン−血小板相互作用の評価に対応する、請求項1に記載の方法。
- 第一の血液凝固パラメーターが、第一の最大幅測定値によって表され、第二の凝固パラメーターが、第二の最大幅測定値によって表され、効力の測定のステップが、第一の最大幅を第二の最大幅と比較することを含む、請求項1に記載の方法。
