【従来の技術】
介護老人においては、特有の日和見感染微生物、例えばCandida albicans、Pseudomonas aeruginosa等が増殖し、誤嚥性肺炎の原因となる可能性が高い。その対策として介護老人施設では、ポビドンヨードを主体とした各種抗菌剤、殺菌剤を用いた口腔内清掃が行なわれている。

しかしながら、ガーゼによる口腔内清掃では、口腔内微生物を除去するだけであり清掃後すぐに増殖が開始され、清掃頻度を高くする必要がある。また、ポビドンヨード等の殺菌剤は、殺菌効果は優れているものの、人体に対して安全であるとは言いがたい。また、ポビドンヨード等の殺菌剤は不快な味覚を与えるため介護者が殺菌剤による口腔洗浄を嫌うあるいは拒否する場合もあり、そうした場合有効な殺菌効果が得られない。

従って、本発明の目的は、介護老人の口腔に特有な微生物の増殖抑制効果が高く、安全性が高く、且つ呈味性が改良された口腔用組成物を提供することである。

本発明者は、ポリリジン又はその塩、糖類を親水基とする界面活性剤、並びに糖アルコール及び/又はスクラロースを組合わせることにより、特有の渋みを有するポリリジンの有効濃度を低減でき、呈味性良く、しかも安全に、固相に付着したCandida albicans(以下、C.albicansと言う)やPseudomonas aeruginosa(以下、P.aeruginosaと言う)等の日和見感染口腔微生物の増殖を強力に抑制できることを見出した。

即ち本発明は、(A)ポリリジン及び/又はその塩、(B)糖アルコール及び/又はスクラロース、並びに(C)糖類を親水基とする界面活性剤を含有する日和見感染を予防するための口腔用組成物を提供するものである。

【発明の実施の形態】
本発明で、ポリリジンとはポリ−D−リジン、ポリ−DL−リジン又はポリ−L−リジンであり、塩としては塩酸、硫酸、及びリン酸などの無機酸の塩、及び酢酸、プロピオン酸、フマル酸、リンゴ酸、クエン酸、マレイン酸、アジピン酸、グルコン酸、及び乳酸などの有機酸の塩を用いることができる。本発明の組成物において、ポリリジン又はその塩は上記のいずれか一種又は二種以上を組合わせて使用できる。ポリリジンの含有量は、微生物増殖抑制効果を得る観点から、口腔用組成物の0.0001重量%以上が好ましく、呈味性に悪影響が生じない観点から、口腔用組成物の0.1重量%以下が好ましい。即ち、ポリリジンの含有量は、口腔用組成物の0.0001〜0.1重量%が好ましく、0.001〜0.06重量%がより好ましく、0.01〜0.06重量%が特に好ましい。

糖アルコールとしては、グリセロール、エリスリトール、キシリトール、リビトール、アラビトール、ガラクチトール、ソルビトール、イジトール、マンニトール等の単糖アルコール類、パラチニット、マルチトール、ラクチトール、マルトトリイトール、イソマルトトリイトール、マルトテトライトール、イソマルトテトライトール、その他透過度別還元水あめ糖類のオリゴ糖・多糖アルコール類などを用いることができ、これらの中でもキシリトール、エリスリトール及びマンニトールが好適に使用される。本発明の組成物においては、上記のいずれかの糖アルコール又はスクラロースを一種又は二種以上組合わせて使用できる。糖アルコール及び/又はスクラロースの含有量は、微生物増殖抑制効果を十分発揮する観点から、口腔用組成物の0.005重量%以上が好ましく、香味への影響を生じない観点から、口腔用組成物の40重量%以下が好ましい。即ち、糖アルコール及び/又はスクラロースの含有量は、口腔用組成物の0.005〜40重量%が好ましく、0.01〜30重量%がより好ましく、0.02〜20重量%が特に好ましい。

糖鎖を親水基とする界面活性剤としては、サポニン、シュガー脂肪酸エステルが挙げられる。サポニンとしては、茶、大豆、キラヤ、ユッカ、アルファルファ、人参、オンジ、ジギタリス、モクツウ、キキョウ、サイコ、甘草、タイゾウ、オウギ、ヤマノイモ、トラガント等の植物から抽出あるいは精製して得られるものを一種あるいは二種以上組合わせて用いることができる。シュガー脂肪酸エステルとしては、蔗糖と脂肪酸がエステル結合したものであればいずれの脂肪酸エステルでも良い。かかる界面活性剤の含有量は、微生物増殖抑制効果を十分発揮する観点から、口腔用組成物の0.0005重量%以上が好ましく、香味への影響を生じない観点から、口腔用組成物の1重量%以下が好ましい。即ち、界面活性剤の含有量は、口腔用組成物の0.0005〜1重量%であるのが好ましく、0.0015〜0.4重量%がより好ましく、0.004〜0.2重量%が特に好ましい。

本発明の組成物のpHは唾液pHに合せる観点から、pH5〜8の弱酸性から弱アルカリ性域が好ましく、pH6.2〜7.7の範囲がより好ましい。pH調整剤としては、各種アミノ酸又はその塩、有機酸又はその塩が使用できる。

本発明の口腔用組成物は、洗口液、マウスフォッシュ、塗布剤、人口唾液などとして調製又は適用し得、上記成分(A)〜(C)に加えて組成物の種類等に応じた適宜な成分を配合し得る。たとえば粘稠剤、成分(C)以外の界面活性剤、成分(B)以外の甘味剤、香料、着色剤、成分(A)〜(C)以外の有効成分などを配合し得、これらの成分を水と混合して製造することができる。

本発明の口腔用組成物は、呈味性が良く安全であり、口腔内のC. albicans及びP. aeruginosa等の増殖を効果的に抑制できるので、これらの微生物が原因で起こる日和見感染の予防に有効に利用することができる。

実施例1〜15及び比較例1〜11
表1〜表3に示す組成の口腔用組成物を調製し、日和見感染原因菌であるC. albicansあるいはP. aeruginosaの増殖抑制の評価を以下の方法で行なった。

評価方法
ソイビーン−カゼイン ダイジェスト ブロスに寒天1.8%を添加し、滅菌後φ85mmのシャーレに、15mLを流し入れて寒天培地を作成した。前記寒天培地に生理食塩水に懸濁した約1×106個/mLのC. albicans IFO1061あるいはP. aeruginosa IFO13275を0.1mL塗布して乾燥させた後、表1及び表2に示す口腔組成物を50μL滴下し、35℃、24時間好気的に培養して増殖抑制効果を評価した。即ち、増殖抑制効果の高い組成物を滴下した部分には菌体の増殖が認められず、ハローを形成する。このハローの明瞭度で増殖抑制能の強さを判断した。抑制効果の基準は以下の通りである。
