Description
2.フォトビオチン、AAF,水銀、ズルフオン族の様に直接的な化学結合による標識。
1.酵素若しくは有機合成法を必要としない、直接−殆んど瞬時での−巨大分子の標識
2.簡単な常套の手法による標識分子の一段階精製
3.殆んど全ての周知の(顕微鏡的)手法による直接的あるいは間接的な標識分子の検出にある。
標識目的のためのPtFの合成
まず100mgのフルオレセン−N=C=Sを100mlの水に溶けた1mlのCH3NH3と反応させ、フルオレセン−NH(CS)NHCH3を作成する。反応は室温、暗所で連続撹拌しながら約1時間行なった。塩酸(1モル/リッター{M})でpHを2〜3に酸性化し、得られた反応産物、フルオレセン−NH(CS)−NHCH3を溶液内で沈澱させる。沈澱物を水で洗浄し回収する。
PtFによる核酸標識
乾燥したPtF化合物を蒸留水に1mg/mlの濃度に溶解し、NaOHでpHを9〜10に上げた。
PtM標識核酸の使用例:
(実施例III)
ヒト乳頭腫ウイルスは培養できないが、ある種の亜型(HPV16/18)は殊に頸管および陰茎の悪性腫瘍の発生に確実に関連している。
a)ヒト乳頭腫ウイルスは培養できないが、ある種の亜型(HPV16/18)は高い頻度で頸管あるいは陰茎における悪性腫瘍の進行に確実に関連している。且つ、殊にDNAウイルス(ワクシニア、単純ヘルペス(HSV1/2)、エプスタインバール、アデノウイルス)あるいはRNAウイルス(ロタウイルス、インフルエンザA、コツクサキーB)の検出のためにプローブが開発された。B型肝炎ウイルスはヒト細胞中では培養できないので、今まではこの急性感染の診断がチンパンジーへの接種(!)によってのみ可能であった。
細菌診断
DNAプローブを用いた細菌核酸の検出もまた近年可能となったことは明らかであろう。これにより細菌毒素の遺伝子が明らかになる。しかしながらこれらの遺伝子が発現しているか否かを常に識別することは不可能である。染色体およびプラスミドにコードされた病原性因子(殊にリステリアモノサイトジーン、ガス菌腸毒素、コレラ腸毒素、大腸菌腸毒素そしてインバシビティー、シゲラ及びエルシニアエンテロコルチチカ、エンテロインバシビチィー)の迅速な検出は、食中毒及び食品産業での品質管理(最終産物管理)の診断において重要である。
寄生虫感染の診断
世界中で二百万人の人々がマラリヤで死んでいる。原理的には、時をえた正確な診断によってこれは防ぐ事ができる。現在の(常例の)顕微鏡的手法は、第三世界の国々には殆んど全てが複雑すぎる。西洋においても困難な顕微鏡的手法は、PtMプローブを用いた常例の試料に対するインサイチュウハイブリダイゼーションに置換されよう。これによりマラリヤ種の鑑別診断が相当に簡素化されまた最小限の訓練を受けた人によって行うことができた。第三世界ではPtMに基づいてディップスティック試験が迅速で簡単な診断のための適当な方法となる。
遺伝子異常の検出
PtMプローブを用いたハイブリダイゼーションの手法は、例えば羊水穿刺物や絨毛膜バイオプシーによる先天性異常の胎仔期診断の可能性を提供する。HLA関連症診断のためのHLA型の検出の増進と供に、異常(例えば悪性腫瘍)の出生後の検出もまた可能である。
遺伝子発現の検出
免疫組織化学の手法を用いた細胞内抗原の存在の可視化によっては、その時点で関連した遺伝子が発現していることは証明されない。またその可視化された産物が細胞内に起因するのか細胞外に起因するものであるかも示されない。細胞内でのmRNAの検出が遺伝子発現の直接の証拠を提供する。この情報は、細胞の機能性に関するデーターを与えるばかりでなく、診断の手助けともなろう。
Claims (4)
- 検出する固有序列を補足する核酸序列でなるハイブリダイゼーションによる固有序列検出用の核酸プローブであって、該核酸プローブが、下記の構造式(1)または(2)で表わされ、化学式{Pt(II)(w)(x)(y)(z)}または{Pt(IV)(u)(v)(w)(x)(y)(z)}を持つ、Pt含有化合物の少なくとも一つで標識されており、
- 検出可能なマーカー基がフルオレセンまたはテトラメチルローダミンであることを特徴とする請求項1に記載の核酸プローブ。
- 検出可能なマーカー基がビオチン、ジゴキシゲニン、またはAMCAであることを特徴とする請求項1に記載の核酸プローブ。
- Pt含有化合物が{Pt(エチレンジアミン)(Me2SO)(フルオレセン−NH(CS)−NHCH3}であることを特徴とする請求項1に記載の核酸プローブ。
