【発明の属する技術分野】
本発明は、血清アルブミン、特にヒトの血清アルブミンの精製に関する

【従来の技術】
ヒト血清アルブミン(HSA) は血漿の主要タンパク質成分である。血漿中のアルブミンの主な機能は血管中のコロイド浸透圧の維持である。さらに、該タンパク質は幾つかのリガンド、例えばビリルビン及び脂肪酸類等のキャリアーとして作用する。(F.ロスステイン、V.M.ローゼノア及びW.L.ヒュージズ(F.Rothstein,V.M.Rosenoerand W.L.Hughes) による概説:アルブミン・ストラクチュアル・アンド・ファンクショナル・ユージィズ(Albumin Struct.Funct.Uses) 、1977年、第7-25頁;U.クラッグ−ハンセン(U.Kragh-Hansen)、ファーマコロジカル・レビュー(Pharmacol.Rev.)、1981年、33巻、第17-53 頁;T.ピータース・ジュニア(T.Peters Jr.)、アドバンシィズ・オブ・プロテイン・ケミストリィ(Adv.Prot.Chem.)、1985年、37巻、第161-245頁、を参照のこと。)
精製血清アルブミンは、重篤な低アルブミン状態にある血液量減少ショックの予防及び治療用に、又、血液透析及び心肺バイパス処置における補助的手段として、さらに、新生児高ビリルビン血症の治療において交換輸血と共に用いることが示されている。
治療には大量の血清アルブミンが必要であり血清アルブミン(血漿)の材料は限られているために、HSAを多量に製造する他の技術が探求されてきた。組み換えDNA 技術により作られた形質転換された微生物又は細胞株を用いた醗酵によるHSA の製造において、成功が報告されている。例として、EP-A-0073646参照のこと。
しかしながら、形質転換細胞を用いた醗酵により製造された血清アルブミンの精製における重大な問題の一つは、精製された均質の血清アルブミンを得るために除去しなければならない、培地(醗酵ブロス)又は細胞ライセートからの混入成分の存在である。
EP-A-0361991において、形質転換酵母で製造されたHSAの、当技術分野において周知の技術を用いた精製は、99%より優れた純度の製品を得ている。医薬用製剤としては、より高い純度が望ましい。
最近、3ステップの処理を基本とする血清アルブミン類の精製方法がEP-A-0319067に開示された。アルカリ沈澱ステップで始まり、次に陰イオン交換クロマトグラフィーを行い、最後にアフィニティークロマトグラフィーを行うこの製法は、良好な収率及び純度の製品を得る。EP-A-0319067には、ウィッチマン及びアンダーソン(Wichman and Andersson)(1974 年)により述べられた方法により製造されたBrCN活性化セファロース4B支持体が言及されている。産業上の使用において、BrCN活性化セファロースを基礎とするアフィニティーマトリックスは二つの重大な欠点を有する。
第一に、第一アルキルアミンとの反応の結果できるイソウレア結合(スペーサー)(M.ウィルチェク、T.ミロン及びJ.コーン(M.Wilchek, T.Miron and J.Kohn):メソッド・イン・エンザイモロジィ(Methods in Enzymology) 、1984年、104巻、第3頁、W.B.ジャコレイ(W.B.Jakoley)編、アカデミック・プレス、ロンドン)が、生理的条件下ではプラスの電荷を持つことである。この荷電スペーサーは、生物特異的吸着を妨げるらしい陰イオン交換様特性を示す。第二の欠点は、医薬用製品の製造に必要な殺菌消毒条件(0.1-2.0 M NaOH)下でのこのマトリックスの使用を可能にする微アルカリ条件下では該イソウレア結合の安定性が制限されることである(C.M.ヤング及びG.T.ツァオ(C.M. Yang and G.T.Tsao):アドバンシィズ・オブ・バイオケミカル・エンジニアリング(Ad.Biochem.Engin.)、1982年、25巻、第19頁、A.フィーヒター(A.Fiechter)編、スプリンゲル・フェルラーク、ベルリン−ハイデルベルグ)。
従って、高い収率及び非常に高い純度でヒト血清アルブミンを大量精製するための実用的な製法が強く求められている。

【発明が解決しようとする課題】
本発明の目的は、高い収率及び非常に高い純度で血清アルブミン、特にヒト血清アルブミンを大量精製するための実用的な製法、および、それによって得られる高純度の血清アルブミンを提供することである。

【課題を解決するための手段】
形質転換された宿主細胞により製造されたヒト血清アルブミンをイオン交換クロマトグラフィーにより精製した後、2-メルカプト或いは2-ヒドロキシC4-C14アルカン酸、又はその塩或いはエステルに結合した担体を含む親油性表面固定相を用いたアフィニティークロマトグラフィーにより精製する。成熟無傷アルブミンに非常に似ている醗酵の終了時に存在する血清アルブミン分解産物は、脂肪酸アフィニティークロマトグラフィーの適用により選択的に除去される。本発明により高い収率で非常に高い純度のヒト血清アルブミンが得られる。

【発明の実施の形態】
組み換えDNA技術により作られた、例えば微生物又は細胞株等の形質転換された宿主細胞を用いた醗酵により調製されたヒト血清アルブミンが、当該発明によって、医薬用製品としての使用に向けて高い収率及び高い純度で精製される。該精製法に先立ち、醗酵培地の遠心及び限外濾過を行う。このように得られた上清又は溶解(lysis)後の細胞に該方法を適用することができる。それは一般的に以下のステップを含む:
−微酸性条件下における血清アルブミンの陰イオン交換樹脂への結合;該アルブミンは溶離剤のpHを下げることにより樹脂から脱着される;
−次に、場合により、媒質(medium)の限外濾過;
−最後に、親水性マトリックス、スペーサー及びリガンドとしての脂肪酸誘導体を含む親油性固定相、並びに溶離剤中に脱着剤としての親油性陰イオンを用いるアフィニティークロマトグラフィーから成る本質的なクロマトグラフィーステップ。

該血清アルブミンはその後、脱塩及び濃縮により回収することができる。当該製法により製造された血清アルブミンは実質的に均質でモノマーであり、すなわち、純度は99.5%よりも高い。
該方法は、微生物又は細胞株を用いた組み換えDNA技術により調製された血清アルブミン、特にヒト血清アルブミンの製造に用途が見出される。該微生物は原核生物又は真核生物でもよく、特に真核生物でもよく、大腸菌(E. coli)、バチルス・ズブチリス(B. subtilis)、バチルス・リヘニフォルミス(B.licheni-formis)、スプレプトミセス(Spreptomyces) 、シュードモナス(Pseudomonas)等のバクテリア類を含む。
真核生物には、サッカロミセス(Saccharomyces)、シゾサッカロミセス(Schizosaccharomyces)、クルイベロミセス(Kluyveromyces)、カンジダ(Candida)等のイースト類、繊維状真菌類、ニューロスポラ(Neurospora)、アスペルギルス(Aspergillus)等がある。

該血清アルブミン発現の結果、該産物は分泌されてもよく、該微生物内に保持されてもよい。培地は醗酵槽からバッチごとに、又は連続的に除去してもよい。分泌される場合、セルフリーの上清を精製工程に用いる。セルフリーの上清は、該培地を遠心すること且つ/又は、タンパク質生成物を濃縮するための限外濾過を用いて濾過することによれば簡便に、該醗酵培地を透明化することにより得られる。該フィルターは通常 500-100,000 Dのカットオフ値を持ち、より一般には最低約10,000 Dである。
該産物又はその一部が細胞の細胞質内に保持される場合には、該細胞を採収する。ライセートは、ライセート製造のために該細胞を機械的又は化学的粉砕することによる、いずれの便利な技術により製造してもよい。細胞の破片は遠心によって除去してもよい。
セルフリーの該上清又は該ライセートのpHを約5ないし9、より好ましくは約6ないし 6.5に調整する。該pHは、水酸化ナトリウム又は他の都合の良い塩基の、通常約0.1 規定度ないし濃縮液までの範囲の液を加えること等の都合の良いいずれの手段で加減してもよい。

次のステップは陰イオン交換クロマトグラフィーである。約20ないし200 mMのバッファー濃度のセルフリーの該上清を予め条件を整えたカラムに乗せる。陰イオンゲルの充填量はゲル1リットル当たり5から50gのタンパク質、より好ましくはゲル1リットル当たり15ないし25gのタンパク質になる。
次に、該ゲルを平衡化バッファーで洗浄することができる。該洗浄液の量は重要ではないが、一般的には該ゲル量に対して0.5-10倍に当たる量である。該バッファーは一般に0.1-100mS/cm 、好ましくは約1ないし10mS/cmの間の導電率を有する。該アルブミンは、若干低いpHの普通に希釈されたバッファーで溶出することにより該ゲルから脱着される。該バッファー濃度は10ないし100 mMである。望ましくは該バッファーは約pH 4.0-6.0の範囲のpHであり、より望ましいのはpH 4.25 及び 4.75 の間である。
該イオン交換ステップにより、核酸類、エンドトキシン類、及び混入非アルブミン様タンパク質類の大部分が除去される。市販の担体に結合したQAE又はDEAE等の陰イオン交換吸着剤を使用する。

次の必須のステップは、親油性固定相を用いるアフィニティークロマトグラフィーである。親油性分子のゲルへの結合は、該親油性分子の活性化及び結合の両方が可能な二官能価試薬を用いて行われる。そのような試薬の例には、例えば、エピクロロヒドリン等のエピハロヒドリン、又はアルカンジオールエーテル、例えば1,4-ブタンジオールジグリシドオキシエーテルようなビスオキシラン類等のエポキシ化合物類がある。該ゲルマトリックスとの反応から、2-メルカプト又は2-ヒドロキシC4-C14アルカン酸類等の求核性リガンドと反応することができる親水性反応性エポキシドを有する誘導体が得られる。より詳しくは以下を参照のこと:アフィニティークロマトグラフィー:実践的アプローチ;ディーン,P.D.G.、ジョンソン,W.S.及びミドル,F.A.(Dean, P.D.G.,Johnson, W.S. and Middle, F.A.)編、IRL プレス、オックスフォード、イングランド、ISBN-0-904147-71-1。
これらのアフィニティークロマトグラフィー用吸着剤は非常に安定であり、スペーサー中に電荷を持たない。
本発明において、親油性固定相は、好ましくはエポキシド化合物により活性化されている。ある実施態様では、前記担体はアガロース担体であって、下記の構造を有するポキシド化合物により活性化されている:

該イオン交換ステップの後、該媒質(medium)のpHを生理的条件、通常約6.5から8.0、より一般には約7から7.5の範囲に上げなければならない。バッファー濃度は一般に50から250mMの範囲である。該媒質をカラムに乗せた後、該ゲルを平衡化バッファーで洗浄する。該洗浄液の量は重要ではないが、一般的には該ゲル量に対して0.25-5倍容である。該バッファーは一般に1.0-20mS/cm、好ましくは約10ないし15mS/cmの間の導電率を有するである。該アフィニティーゲルの充填量はゲル1リットル当たり5から70gのタンパク質、より好ましくはゲル1リットル当たり15ないし30gのタンパク質になる。アルブミンは、例えばカプリル酸ナトリウム等の脂肪酸を25-250mM含むバッファーを使用することにより溶出される。

成熟ヒト血清アルブミンの他に、特定の分解産物が該醗酵培地の上清中に存在する。これらの分解産物は40-50kDの分子量を持ち、三つのドメインを含む成熟タンパク質のうちのドメインI及びIIから成る。当該40-50 kD断片中に存在しないドメインIIIが、アルブミンの原(principle)脂肪酸結合部位を含む(T. ピータース(T.Peters)、上記)。驚くべきことに、当該40-50kD断片は、親油性アフィニティマトリックスに対して成熟ヒト血清アルブミンよりも高い親和性を示し、このことが完全な該タンパク質からの該40-50kD断片の分離を可能にする。種々の多様な支持体及び吸着剤を該固形担体又は支持体として使用してよい。そのような固形担体には、ガラス及びシリカゲル等の無機担体類及び、例えば、アガロース、セルロース、デキストラン、ポリアミド、ポリアクリルアミド類、二官能価アクリル酸のビニルコポリマー類及び種々の水酸化モノマー類等の合成又は天然の有機担体類などが含まれる。市販の担体類は、セファデックスR、セファロースR、トリサクリルR、ウルトロゲルR、ダイノスファーズR、マクロソーブR、XAD樹脂、及びその他の名称で売られている。

種々のステップにおける条件は、通常約−10℃から+30℃の範囲の都合の良い温度、より一般にはおよそ室温の、変成しない条件が用いられるであろう。該クロマトグラフィーステップは、都合が良いように、バッチごと又は連続的に行ってもよい。遠心、濾過、デカント等の都合の良いいずれの分離方法を用いてもよい。このようにして、99.9%より高い純度、とりわけ99.95%より高い純度という驚くほど高い純度の単離血清アルブミン製剤を得ることができる。
以下の非限定的実施例により本発明はさらに明らかにされる。

実施例1.アフィニティーマトリックス類の合成
E-C10-C10:
−エポキシ活性化セファロース 6FF用の E鎖;
−10個の炭素原子のスペーサー長をもたらす第一のC10鎖;
−カルボン酸リガンド中の炭素原子数を決める第二のC10鎖。
該アフィニティーマトリックス類の合成には周知の方法を使用した(例えば、ディーンら(Dean, et al.);上記、を参照のこと)。
C3及びC10 スペーサーの製造には、エピクロロヒドリン及び1,4-ブタンジオールグリシドオキシエーテルをそれぞれ使用した。

E-C10-C10 マトリックスの合成:
1リットルのセファロース6FFを蒸留水で入念に洗浄した。過剰の水はガラス濾過器により除去した。該ゲルを0.6Lの1,4-ブタンジオールグリシドオキシエーテルに懸濁し、およそ30分間攪拌した。次に 0.6Lの0.5N NaOH溶液を加え、24時間攪拌を続けた。該エポキシ活性化ゲルを蒸留水で入念に洗浄し、0℃で保存した。0.7kgの湿った生成物が得られた(E-C10)。

遊離のカルボン酸の数を単純滴定により測定し、湿ったゲルマトリックス 1 g当たり19.3μ当量であることがわかった。

実施例2.透明化された醗酵培地からの組み換え HSAのQ-セファロース及びE-C10-C10-アフィニティークロマトグラフィーによる精製
醗酵:
HSAに対する遺伝子を含むプラスミドで形質転換されたクルイベロミセス・ラクティス(Kluyveromyces lactis)株CBS 2360 (菌培養中央局、バールン、オランダ)を、0.5 %(w/v) 酵母抽出物、1.4 %(w/v) グルコース、1.0 %(w/v)カゼイン加水分解物、ビタミン類及び金属塩類を含む培地中で、30℃、90時間生育させた。醗酵中にグルコースが供給された。

濾過及び限外濾過:
該醗酵培地を遠心した(4000 rpm、5分)。上清を EKSフィルターで濾過し、30,000 Dのカットオフ値を持つフィルターを用いた限外濾過により11回濃縮した。限外濾過後のタンパク質濃度は28 mg/mlであり、その内、65%はモノマーで完全な組み換え HSAであった。0.5 M 酢酸ナトリウム、pH 5.5、を加えて pH を 6.4に調整した。

Q- セファロースクロマトグラフィー:
該HSA溶液を、50mM 酢酸ナトリウム、pH 5.5、で平衡化してあるQ-セファロースFF カラムに結合させた(充填:セファロース1L当たりタンパク質25g)。該ゲルを平衡化バッファーで洗浄した後、RHAを50mM酢酸ナトリウム、pH4.6、で溶出した。1Mリン酸バッファー、pH6.6、(比率;溶出液:バッファー(v/v)=10:1)及び4M NaOH を加えて、該溶出液のpHを pH7.4 に上げた。タンパク質濃度は10 mg/mlであり、その内、78%はモノマーで完全な組み換えHSAであった。

アフィニティークロマトグラフィー:
HSAを含むQ-セファロース溶出液を、例1に述べたようにジグリシジルエーテルによってエポキシ活性化セファロース6FFに結合された2-メルカプトデカン酸(E-C10-C10-セファロース)に接触させた。該アフィニティー吸着剤を100mMリン酸ナトリウムバッファー、pH7.4、で平衡化した。該組み換え HSAを吸着剤に結合させた後(充填:ゲル1リットル当たりタンパク質22g)、該カラムを平衡化バッファーで洗浄した。該 HSAを75mMカプリル酸ナトリウムを含む100mMリン酸ナトリウム、pH7.4、で溶出した。該HSAを脱塩し、30,000 Dのカットオフ値を持つフィルターを用いた限外濾過により濃縮した。該精製HSAはタンパク質濃度が50mg/mlであり、その内、99.9%より多くが単量体成熟HSAであった。

この例ではHSAの回収率は50%であった。SDS-PAGE(図1)、HPLC-SEC(図2,3)、HPLC-IEC(図4,5)及びIEF(図6)では、僅かに痕跡量の分解産物が検出されたのみで、混入物は検出されなかった。HPLC-SECによる純度は、99.3%と見なされた。24.77 の塩関連ピークで補正すると>99.91%の純度が得られる。実際に、イムノブロッティング法を用いると該精製HSAの純度は少なくとも99.95%と評価された。この技術ではポリクローナル抗体類がタンパク質類に対して増加したが、それらはこの例に述べた方法によりブランクのK.ラクティス(K. lactis)醗酵培地から精製された。エンドトキシンの濃度は5%(w/v) 組み換えHSA 濃縮液1ml当たり1エンドトキシンユニットよりも低かった。エンドトキシンユニットは、いわゆるEC-5として、FDA交付の参考基準(RSE)に関連付けられている。エンドトキシンはQ-セファロースカラムステップの間に主に除去された;表1参照のこと。

実施例3A.透明化された醗酵培地からの組換えHSAのQ-セファロース及び E-C10-C8-アフィニティークロマトグラフィーによる精製
醗酵、濾過及び限外濾過:例2に述べてあるように行った。
Q- セファロースクロマトグラフィー:EP-A-0319067に述べてあるように行った。
採取した上清を30,000 Dのカットオフ値を持つフィルターを用いた限外濾過により濃縮した。該残留液のpHをpH7.4 に調整した。モノマー性HSAの濃度は14mg/mlであった。

E-C10-C8- マトリックスによるアフィニティークロマトグラフィー:
濃縮したQ-セファロース溶出液をジグリシジルエーテルによってエポキシ活性化セファロース6FFに結合されたC8(2-メルカプトオクタン酸)に加えた。該アフィニティー吸着剤を100mMリン酸ナトリウムバッファー、pH7.4、で平衡化し、吸着後洗浄した。該単量体HSA及び分解生成物類は100mMカプリル酸ナトリウムにより溶出することができた。無傷のHSA及び分解生成物類は 100mMリン酸ナトリウムバッファー、pH7.4、中の0-100 mMカプリル酸ナトリウム勾配を用いることにより分離することができた。HSA及び分解生成物類のE-C10-C8-マトリックスへの結合は、例2に述べたE-C10-C10-マトリックスへの結合ほど強くなかった。

実施例3B.透明化された醗酵培地からの組換えHSAのQ-セファロース及び E-C3-C12-アフィニティークロマトグラフィーによる精製
醗酵、濾過、限外濾過及び Q- セファロースクロマトグラフィー:実施例3Aに述べてあるように行った。
E-C3-C12- マトリックスによるアフィニティークロマトグラフィー:
HSAの精製は、2-メルカプトドデカオクタン酸がC3-エーテルによってエポキシ活性化セファロース6FFに結合されていることを除けば、例3Aに述べたと同様に行った。結合した単量体成熟 HSA、HSAの分解生成物類及び他の混入物類は、100mMリン酸ナトリウムバッファー、pH7.4、中の100 mMカプリル酸ナトリウムでは溶出できなかった。しかしながら、溶出は1% SDSにより可能であった。無傷のHSA及び他のタンパク質類の分離は不可能であった。

Claims (3)
- 以下の工程を含む、アルブミン分解産物の含量が 0.1 %未満である組換え血清アルブミンを製造する方法:
(a)組換え血清アルブミンを含む透明化された発酵培養液を含む生成物培地又は細胞ライセートを生理的 pH において陰イオン交換樹脂とインキュベーションする工程;
(b)前記陰イオン交換樹脂を酸性pHのバッファー溶液で溶出して、前記組換え血清アルブミンを含む第1の溶出画分を回収する工程;
(c)生理的pHにおいて前記第1の溶出画分を親油性固定相および水性移動相を含むアフィニティークロマトグラフィーにかける工程であって、前記固定相が、ジグリシジルエーテルによって活性化され、2-メルカプトまたは2-ヒドロキシC4-C14アルカン酸、またはその塩若しくはエステルに結合した担体を含む、前記工程;
(d)脱着剤として親油性陰イオンを前記水性移動相に加え、前記組換え血清アルブミンを含む第2の溶出画分を回収することにより、組換え血清アルブミンを溶出する工程;
(e)アルブミン分解産物の含量が 0.1 %未満である精製組換え血清アルブミンを前記第2の溶出画分から単離する工程。 - 精製組換え血清アルブミンをさらに透析する工程を含む、請求項1記載の方法。
- 血清アルブミンがヒト血清アルブミンである請求項1または2記載の方法。
