る病気、即ち、遺伝子疾患に対しては、これまで様々な
治療法が考えられてきた。その一つとして、欠陥遺伝子
そのものを正常な遺伝子と入れ換えたり、正常な遺伝子
を補ったりすることにより、遺伝子疾患を根本的に解決
しようとするのが、遺伝子治療である。この遺伝子治療
に当たって重要なことは、正常な遺伝子を標的細胞に正
確に導入すると共に、導入した遺伝子を正確に発現させ
ることである。正常な遺伝子を標的細胞に導入するため
の遺伝子のベクターとしては、これまで、レトロウイル
スベクター、アデノウイルスベクター、アデノ随伴ウイ
ルスベクター等のウイルスベクターや、リポソーム等に
代表される非ウイルスベクターが用いられてきた。しか
し、いずれも標的細胞への遺伝子の導入効率が低い等の
欠点が存在した。また、導入された遺伝子の発現効率が
悪い等の欠点もあったため治療に不十分な場合が多かっ
た。この場合でも、アデノシンデアミナーゼ(ADA)欠
損症のように、正常ADA遺伝子が導入された細胞が生存
優位性(survival advantage)あるいは増殖優位性(gr
owth advantage)を獲得し、体内で次第に選択され優位
になっていくものと期待されるものであれば、たとえ遺
伝子導入効率が低くても次第に治療効果が現れるように
なっていく可能性がある。しかし、このような体内での
選択性が期待できないタイプの治療用遺伝子を導入する
必要のある場合も多く、遺伝子を導入した細胞を選択的
に増幅させるシステムの確立が望まれていた。
サイトカイン(造血因子)として従来から考えられてき
たが、近年、G−CSFを投与すると、好中球の増加がみ
られるばかりでなく、造血幹細胞/前駆細胞の体内プー
ルの増加がみられることが報告された(臨床血液.,35,1
080(1994))。また、G−CSFが機能する機構として、
G−CSF刺激によりG−CSF受容体が活性化されるときに
は、G−CSF受容体の二量体化がみられること(Proc Gr
owth Factor Res.,3(2),131−141(1991))や、G
−CSF受容体には、増殖誘導ドメインと分化誘導ドメイ
ンが存在することが報告された(Cell.,74,1079−1087
(1993))。さらに、G−CSF受容体同様、二量体化に
より活性化する受容体としては、エストロゲン受容体が
知らており(J Biol Chem.,264,2397−2400(198
9))、細胞内でエストロゲン受容体とc−Ablチロシン
キナーゼの融合タンパク質を発現させることによりc−
Ablチロシンキナーゼの活性化が起こることも報告され
た(The EMBO Journal.,12,2809−2819(1993))。
内あるいは体外で選択的に増幅させることにより、遺伝
子導入効率が低いという問題点を克服することを狙った
もので、造血幹細胞等を標的とした遺伝子治療の基盤技
術を提供することを目的とする。
伝子の導入効率及び導入された遺伝子の発現効率の面で
克服すべき課題が多い。従って、遺伝子導入がなされた
標的細胞のみを選択的に増殖させるシステムが確立され
れば、大きな飛躍がもたらされることは、明らかであ
る。特に、赤血球、白血球など多くの血液細胞の基とな
る細胞であり、遺伝子治療の標的細胞として最も好まし
いとされる造血幹細胞に対し、かかるシステムが確立さ
れれば、遺伝子治療の分野における貢献度は、非常に大
きい。
イン(造血因子)として従来から考えられてきたG−CS
Fが、造血幹細胞の増殖をも引き起こし、このG−CSF受
容体が活性化される際に、G−CSF受容体の二量体化が
みられることに鑑み、遺伝子工学的手法を用いて工夫し
たG−CSF受容体の二量体化により、造血幹細胞を増殖
させるシステムを想到した。また、エストロゲン刺激に
よりエストロゲン受容体が二量体化するということに鑑
み、G−CSF受容体遺伝子とエストロゲン受容体遺伝子
のキメラ遺伝子を作製し、そのキメラ遺伝子を導入した
細胞に対して外部からエストロゲン刺激を与えることに
より、強制的にキメラ遺伝子産物中のG−CSF受容体部
分を二量体化させることを想到した。
より、キメラ遺伝子産物中のG−CSF受容体部分の活性
化を引き起こし、遺伝子を導入した造血幹細胞を選択的
に増殖させるシステムを新規に開発し、これを遺伝子治
療の分野に応用すべく本発明を完成した。

融合タンパク質をコードする遺伝子を含むベクター、該
ベクターを含む細胞及びステロイドホルモンを作用させ
ることにより該細胞を体内ないし体外で選択的に増殖さ
せる方法等に関する。また、本発明は、該ベクターが外
来遺伝子を含む場合は、該外来遺伝子が導入された細胞
を選択的に増殖させる方法等に関する。

タンパク質の「リガンドが結合するドメイン」に作用す
るリガンドを作用させて、(6)記載の細胞を選択的に
増殖させる方法、 (8)(a)リガンドが結合する領域、(b)(a)の
領域にリガンドが結合すると会合する領域、及び(c)
会合すると細胞に増殖活性を付与する領域、とを含む融
合タンパク質をコードする遺伝子と所望の外来遺伝子、
を含むベクター、 (9) 「会合すると細胞に増殖活性を付与する領域」
がサイトカイン受容体に由来する、(8)記載のベクタ
ー、 (10) サイトカイン受容体がG−CSF受容体である
(9)記載のベクター、 (11) 「リガンドが結合する領域」がステロイドホル
モン受容体に由来する、(8)記載のベクター、 (12) ステロイドホルモン受容体がエストロゲン受容
体である(11)記載のベクター、 (13) 「融合タンパク質をコードする遺伝子」と「外
来遺伝子」とが同一の分子上に位置している(8)記載
のベクター、 (14) 「融合タンパク質をコードする遺伝子」と「外
来遺伝子」とが別々の分子上に位置している(8)記載
のベクター、 (15) (8)〜(14)のいずれかに記載のベクターを
保持する細胞、 (16) (15)記載の細胞に対して、(8)記載のベク
ターに含まれる遺伝子がコードする融合タンパク質の
「リガンドが結合するドメイン」に作用するリガンドを
作用させて、(15)に記載の細胞を選択的に増殖させる
方法、 (17) (a)(5)または(8)に記載のベクター、
及び(b)該ベクターに含まれる遺伝子がコードする融
合タンパク質の「リガンドが結合するドメイン」に作用
するリガンド、を含むキット、 に関する。
作用することにより該タンパク質を会合せしめるもので
あれば、特に制限はないが、ステロイドホルモンが好適
である。ステロイドホルモンとしては、例えば、エスト
ロゲン、アンドロゲン、プロゲステロン、グルココルチ
コイド、ミネラルコルチコイドなどが挙げられ、それぞ
れの受容体タンパク質との組み合わせで用いられる。ま
た、本発明に用いられるサイトカイン受容体は、会合す
ることによって細胞に増殖活性を付与するものであれば
よく、例えば、G−CSFなどサイトカイン受容体ファミ
リーに属するものや、c−kitやflk2/flt3などのように
チロシンキナーゼ受容体ファミリーに属するもの等があ
る。
付与する領域」としては、細胞内増殖シグナルを伝える
分子、例えば、サイトカイン受容体分子全体を用いるこ
とが可能であるが、分子中の細胞に増殖活性を付与する
領域のみを用いることも可能である。この場合、融合タ
ンパク質遺伝子が導入された細胞の分化を起こさず増殖
のみを起こすので、該細胞をそのままの形で増殖させる
際に有利である。さらに、本発明において用いられるベ
クターには、融合タンパク質をコードする遺伝子を含む
単一種分子のベクター、融合タンパク質と外来遺伝子の
双方を含む単一種分子のベクターの他、融合タンパク質
をコードする遺伝子を含むベクターと外来遺伝子を含む
ベクターの組み合わせからなる複数種の分子からなるベ
クター系、例えば、バイナリーベクター系も含まれる。
この複数種の分子からなるベクター系は通常、共形質転
換(co−transformation)によって細胞に導入される。
子とを同一のベクターに挿入する場合には、IRES(inte
rnal ribosome entry site)(特表平6−509713号公
報)を含むジシストロニックの形にすることができる。
例えば、「5'−プロモーター−外来遺伝子−IRES−融合
タンパク質をコードする遺伝子−3'」の構造を有するベ
クター、又は、「5'−プロモーター−融合タンパク質を
コードする遺伝子−IRES−外来遺伝子−3'」の構成を有
するベクターを用いることができる。該融合タンパク質
遺伝子を発現している細胞のほぼ全てが外来遺伝子を発
現しているようにするには、前者の形が一般に用いられ
る。
しては、造血幹細胞、リンパ系細胞、これら血球系以外
の細胞などが挙げられる。特に、本発明は、自己複製能
を有する造血幹細胞に好適に適用される。なお、本発明
において細胞に導入される外来遺伝子については、特に
制限はないが、遺伝子治療の分野においては、欠陥遺伝
子に対応する正常な遺伝子を用いるのが有効である。

/エストロゲン受容体キメラ遺伝子の作製 G−CSF受容体全体とエストロゲン受容体のリガンド
(エストロゲン)結合領域のキメラタンパク質(以下、
単に「GCRER」と称する)が産生されるように、それぞ
れのタンパク質をコードするcDNAの融合遺伝子を作製し
た(図1(A))。次いで、G−CSFに対する反応性を
欠いたキメラタンパク質を産生するために、「GCRER」
に対応する融合遺伝子のうちG−CSF受容体の細胞外ド
メインの5番目のGluから195番目のLeuまでの部分を除
いた変異誘導体(以下、単に「GCRΔ(5−195)/ER」
と称する)を作製した(図1(B))。さらに該変異誘
導体から、G−CSF受容体の分化誘導ドメイン(725〜75
6)を含んだ部分を除いた変異誘導体(以下、単に「GCR
Δ(5−195、725−756)/ER」と称する)を作製した
(図1(C))。
/エストロゲン受容体キメラ遺伝子が導入されたBa/F3
細胞の単離 実施例1で作製した3種類の選択的増幅遺伝子をプラ
スミド「pCMX」(Cancer Res.56:4164(1996))に組み
込んで作成したプラスミド10μgを、Sca Iにて直線化
したブラストサイジン耐性遺伝子を有する「pSV2bsr」
(科研製薬社製)1μgとともに、IL−3依存性細胞株
であるBa/F3細胞に電気穿孔法にて導入した。次いで、
電気穿孔施行後の細胞を5×105個ずつ24ウェルプレー
トに分配し、ブラストサイジン10μg/mlを含む培地で培
養した。この結果、「pCMX−GCRER」を導入した場合
は、17ウェル中11ウェルで、「pCMX−GCRΔ(5−195)
/ER」の場合は、29ウェル中3ウェルで、「pCMX−GCRΔ
(5−195、725−756)/ER」では、52ウェル中52ウェル
で、ブラストサイジン耐性の細胞増殖がみられた。次い
で、これらブラストサイジン耐性の細胞をウェルごとに
IL−3で増殖させた後、IL−3の代わりにエストラジオ
ール10-7Mを加えて培養を行ったところ、「pCMX−GCRE
R」を導入した場合は、11ウェル中7ウェルで、「pCMX
−GCRΔ(5−195)/ER」の場合は、3ウェル中3ウェ
ルで、「pCMX−GCRΔ(5−195、725−756)/ER」の場
合は、16ウェル中13ウェルで、IL−3非依存性・エスト
ロゲン依存性の細胞増殖がみられた。なお、「pCMX−GC
RER」の代わりにレトロウイルスベクターである「pMX」
(Exp.Hematol.24:324(1996))に「GCRER」を挿入し
たもの(以下、単に「pMX−GCRER」と称する)(図2)
を用いて同様の実験を行ったところ、各1細胞を含む24
ウェル中2ウェルで、IL−3非依存性・エストロゲン依
存性の細胞増殖がみられた。また、「pCMX−GCRER」を
導入した細胞に対し、エストラジオールに代えて、1nM
のG−CSFを加えたところ、G−CSFで増殖のあるウェル
は、エストラジオールで増殖のあるウェルと一致した。
さらに、プラスミド未導入のBa/F3細胞をコントロール
として用いたところ、G−CSF及びエストラジオールに
よる増殖は認められなかった。なお、目的の融合タンパ
ク質が細胞内で生産されていることは、抗G−CSF受容
体抗体又は抗エストロゲン受容体抗体を用いたウエスタ
ンブロッティング法により確認した。
トラジオールに対する反応性の良いものを選択し、以下
の実験(XTTアッセイ)に用いた。
を行った。この結果、G−CSF及びエストラジオール刺
激にて、IL−3非依存性の細胞増殖が認められた(図
3)。さらに、エストラジオール濃度を10-14〜10-7Mの
間で変化させて同様の実験を行ったところ、10-9〜10-7
Mの間で細胞増殖が認められた(図4)。これにより、1
0-9〜10-7Mの間の濃度によるエストラジオール刺激によ
り、細胞増殖シグナルが伝達されることが示唆された。
a/F3細胞にて検討をおこなった。この結果、G−CSF刺
激による細胞増殖はブロックされ、エストラジオール刺
激によってのみ細胞増殖が認められた(図5)。
導入したBa/F3細胞でも、エストロゲン刺激で細胞増殖
が起こり、G−CSFに対する反応性は認められなかっ
た。
ー断片(「断片1」)を回収した。また、「pCMX−GCRE
R」および「pCMX−GCRΔ(5−195)/ER」それぞれか
ら、Hind IIIとKpn I断片(それぞれ「断片2」:1672b
p、「断片3」:1099bp)、EcoR IとKpn I断片(それぞ
れ「断片4」:1888bp、「断片5」:1792bp)を回収し
た。pBCEC(pBluescriptIIKSにEMCV由来のIRESとCD24を
連結したもの,Migita,M.,Proc.Natl.Acad.Sci.USA 92:1
2075(1995))をApo Iにて消化し、IRES−CD24を含む
断片(「断片6」:950bp)を回収した。「断片1」、
「断片2」、「断片4」、および「断片6」をライゲー
ションし「pCMXGCRER−IRES−CD24」(図6)を、「断
片1」、「断片3」、「断片4」、および「断片6」を
ライゲーションし「pCMX−GCRΔ(5−195)/ER−IRES
−CD24」(図7)を、「断片1」、「断片3」、「断片
5」、「断片6」をライゲーションし「pCMX−GCRΔ
(5−195、725−756)/ER−IRES−CD24」(図8)を構
築した。
製)にて1回洗浄した107細胞を0.2mlの「OPTI−MEMI」
に懸濁し、「pCMX−GCRER−IRES−CD24」、「pCMX−GCR
Δ(5−195)/ER−IRES−CD24」、「pCMX−GCRΔ(5
−195、725−756)/ER−IRES−CD24」をそれぞれ10mgず
つ加え、「Gene Pulser」(BioRad社製)を用いて290
V、960mFにて導入した。導入後、2日間10%FCS,10U/ml
mIL−3(R & D SYSTEMS社製)を含むRPMI培地にて培
養した。106細胞を5%FCS/PBSにて洗浄後、1mg/ml抗CD
24抗体(Pharmingen社製)を室温20分反応させ、5%FC
S/PBSにて2回洗浄後、1:20希釈したPE標識抗マウス抗
体(DAKO社製)を室温20分反応させ、5%FCS/PBSにて
2回洗浄した。5mg/mlヨウ化プロピジウム(propidium
iodide)/PBS 1mlに懸濁し、フローサイトメトリー(Be
cton Dickinson)にて585nmのディテクターを用いてCD2
4の発現の解析を行った。この結果、「pCMX−GCRΔ(5
−195)/ER−IRES−CD24」が導入された細胞では、多く
の細胞でCD24の発現が検出された。なお、pCMX−GCRΔ
(5−195)/ER−IRES−CD24」が導入された細胞の対照
としては「pCMX−GCRΔ(5−195)/ER」が導入された
細胞を用いた。結果を図9および表1に示す。なお、死
細胞検出のために用いたヨウ化プロピディウムからのシ
グナルもデーター値に含まれている。
U:和光純薬社製)生理食塩水溶液(10mg/ml)を300ml匹
静脈注射を行った。投与後2日目に、大腿骨より骨髄を
採取し、「Lympholyte−M」(Cederlane社製)上にて
遠心(1500rpm、25℃、22分)により単核球を単離し
た。20%FCS、100U/ml IL6、100mg/mlラットSCFを添加
したイスコフ変法ダルベッコ基本培地(IMDM;Gibco社
製)にて2日間培養した。CH296(宝酒造社製、Hanenbe
rg,H.et al.Nature Med.2:876(1996))をコートした
プレート(1146:ファルコン社製)上にて、IL6とSCFに
て前刺激した骨髄細胞106を、エコトロピックパッケー
ジング細胞株「GP+E−86」(J.Virol.62:1120(198
8))に「pMX−GCRER」を組み込み培養上清に得られた
レトロウィルス「vMXGCRER」、またはエコトロピックパ
ッケージング細胞株「GP+E−86」に「pMXGCRΔ(5−
195)/ER」を組み込み培養上清に得られたレトロウィル
ス「vMXGCRΔ(5−195)/ER」を108含む培養上清で懸
濁し、IL6とSCFを添加し培養した。2,24,26,36,38時間
後にウィルス上清を交換した。6回目のウィルス上清交
換の24時間後、細胞を104/wellになるようにメチルセル
ロースを含む培地(IMDM,1.2%メチルセルロース1500c
p;Wako,20%FCS,1%脱イオン化BSA,10mM 2−メルカプト
エタノール,10-7M b−エストラジオール)にて培養し
た。10日培養後にコロニーを顕微鏡観察した後、塗抹標
本を作成しライトギムザ染色後、構成細胞の同定を行っ
た。
が感染した骨髄細胞では、エストラジオール刺激で分化
した顆粒球−マクロファージ系コロニー、赤芽球系コロ
ニーが観察された。エストラジオール刺激によって「vM
XGCRER」感染骨髄細胞より形成された顆粒球−マクロフ
ァージ系コロニーを図10に示す。エストラジオール刺激
によって「vMXGCRΔ(5−195)/ER」感染骨髄細胞によ
り形成された赤芽球系コロニーを図11に示す。これらの
コロニーの構成細胞を塗抹標本にしてライト−ギムザ染
色したところ、分化した血球細胞像が得られた。「vMXG
CRER」感染骨髄細胞より形成された顆粒球−マクロファ
ージ系コロニーの塗抹標本で観察されたマクロファージ
のにしてライト−ギムザ染色像を図12に、「vMXGCRΔ
(5−195)/ER」感染骨髄細胞より形成された赤芽球系
コロニーの塗抹標本で観察された赤芽球のライト−ギム
ザ染色像を図13に示す。
刺激により選択的に増幅させることが可能となり、標的
細胞への遺伝子の導入効率などが低い場合であっても、
有効な遺伝子治療が行えるようになった。また、本発明
における細胞の選択的増幅システムは、様々な血液細胞
に適用が可能であるため、遺伝子治療の対象となる細胞
の範囲の拡大が図られた。従って、本発明によって、特
に遺伝子治療の分野において重要な基盤技術が提供され
た。
Claims (14)
- 【請求項1】(a)ステロイドホルモン受容体に由来
し、リガンドが結合する領域、(b)(a)の領域にリ
ガンドが結合すると会合する領域、及び(c)サイトカ
イン受容体又はその一部からなり、会合すると細胞に増
殖活性を付与する領域、を含む融合タンパク質。 - 【請求項2】ステロイドホルモン受容体がエストロゲン
受容体である、請求項1記載の融合タンパク質。 - 【請求項3】請求項1記載の融合タンパク質をコードす
る遺伝子。 - 【請求項4】請求項3記載の遺伝子を含むベクター。
- 【請求項5】請求項4記載のベクターを保持する細胞。
- 【請求項6】請求項5記載の細胞に対して、請求項1記
載の融合タンパク質の「ステロイドホルモン受容体に由
来し、リガンドが結合する領域」に作用するリガンドを
作用させて、請求項5記載の細胞を選択的に増殖させる
方法。 - 【請求項7】ステロイドホルモン受容体がエストロゲン
受容体である請求項6記載の方法。 - 【請求項8】(a)ステロイドホルモン受容体に由来
し、リガンドが結合する領域、(b)(a)の領域にリ
ガンドが結合すると会合する領域、及び(c)会合する
と細胞に増殖活性を付与する領域、とを含む融合タンパ
ク質をコードする遺伝子と所望の外来遺伝子、を含むベ
クター。 - 【請求項9】ステロイドホルモン受容体がエストロゲン
受容体である請求項8記載のベクター。 - 【請求項10】「融合タンパク質をコードする遺伝子」
と「外来遺伝子」とが同一の分子上に位置している請求
項8記載のベクター。 - 【請求項11】「融合タンパク質をコードする遺伝子」
と「外来遺伝子」とが別々の分子上に位置している請求

含む、請求項13記載の方法に用いるキット。
