Description
はコラーゲン成分をコラゲナーゼ酵素を用いて特異的に
分解して得られる、アナフィラキシー反応を誘発させな
い非抗原性安定化剤およびそれにより安定化された生理
活性物質に関する。
生理活性物質の医薬品製剤、殊に、蛋白質製剤、酵素製
剤、ワクチン製剤が開発されているが、これらの安定化
剤としてゼラチンやコラーゲンが使用されたり(特開昭
57-140715 号公報、特開平1-279843号公報、特開平6-23
4659号公報、特開昭51-16488号公報、特開昭60-260523
号公報、特開昭62-149628 号公報、特開平2-49734 号公
報参照) 、あるいはそれらを酸や加熱によって処理した
分解物が用いられている(特開昭49-109520 号公報、特
開昭54-140715 号公報、特開昭57-114527 号公報、特開
昭63-307827 号公報、特開平6-234659号公報、特開昭54
-143197 号公報参照) 。これらの用法としては、単独で
使用されたり、その他の一般的な安定化剤と併用された
りしている。ゼラチンやコラーゲンおよびこれらの分解
物は、アレルギー反応を引き起こしにくいという経験に
基づいて、各種生理活性物質の安定化剤として広く使用
されるようになった。
患の患者数は、近年欧米各国を始め、日本国内でも急激
に増加の一途をたどっており、現在では実に3人に1人
の割合で何らかのアレルギー疾患をもっているとさえ言
われている。このようなアレルギー患者数の増加を背景
にして、抗原性/アレルゲン性がほとんどないと考えら
れていたゼラチンやコラーゲンを安定化剤として含む各
種生理活性物質に対しても、アナフィラキシーなどの副
作用を引き起こす(ゼラチン特異IgE抗体をもった)
患者が最近少しずつ増えてきている為、社会問題化し始
めている。事実、これらに関する学術的な発表は199
0年代に入ってから初めて散見されるようになってきた
(Kelso, J. M. 等, J. Allergy Clin. Immunol., 91
巻, 867-872 頁およびSakaguchi, M. 等, 感染・炎症・
免疫, 26巻, 48-50 頁参照) 。本来、各種疾病を治療お
よび予防するために使用される生理活性物質の医薬品製
剤が原因で、アナフィラキシーのような副作用が引き起
こされることはあってはならないことであり、重要な問
題である。
鑑みて、抗原性やアレルゲン性を示さないゼラチンやコ
ラーゲンの誘導体について鋭意研究を重ねた。その結
果、コラゲナーゼ酵素を単独ないしは各種の担体に固定
化させた状態で、ゼラチン成分ないしはコラーゲン成分
を含む原料に直接作用させ、特異的な酵素分解を行わせ
ることにより、抗原性がなく、コラーゲン本来の特徴的
なアミノ酸配列である(Gly−X−Y)n を保持し
た、分子量範囲が1,000以下のペプチド組成物が得
られることを見出した(特開平7-82299 号公報参照) 。
54-80406号公報に示すように、従来、ゼラチンの分子量
が約10,000以下のものはウロキナーゼの安定化効果が殆
どないと考えられており、本発明者らによる特開平7-82
299 号公報に示すペプチド組成物に安定化剤としての用
途は考えられなかった。また、特開平7-82299 号公報に
示すペプチド組成物では、分子量範囲が狭いため、収率
の向上に限界があるという問題点があった。

の安定化効果を有する非抗原性安定化剤およびそれによ
り安定化された生理活性物質を提供することを目的とし
ている。
アレルゲン性を示さないゼラチンやコラーゲンの誘導体
について鋭意研究を重ねた結果、ゼラチン成分またはコ
ラーゲン成分をコラゲナーゼ酵素を用いて特異的に分解
して得られる、アミノ酸配列が(Gly−X−Y)n の
ペプチド組成物であって、分子量が20,000以下の
ものは、抗原性/アレルゲン性を示さないだけでなく、
各種生理活性物質を安定化させる作用があることを見出
した。

0,000以下でアミノ酸配列が(Gly−X−Y)n
のペプチドを主成分とすることを特徴とする。特に、本
発明に係る非抗原性安定化剤は、ゼラチン成分またはコ
ラーゲン成分をコラゲナーゼ酵素を用いて特異的に分解
して得られるペプチド組成物であって、分子量が20,
000以下であり、アミノ酸配列が(Gly−X−Y)
n のペプチドを70%以上含有するものから成ることが
好ましい。非抗原性を高めるため、特に、本発明に係る
非抗原性安定化剤は、前記ペプチドを85%以上、より
好適には95%以上含有することが好ましい。本発明に
係る非抗原性安定化剤は、ウロキナーゼおよびインター
フェロンに対して安定化効果を有する安定化剤である。

ある。本明細書中で、「生理活性物質」とは、ウイル
ス、ワクチン、各種サイトカイン、抗生物質のほか、蛋
白質、酵素、細菌、ホルモン、核酸および抗体などの成
分、あるいはそれらを有効成分とする治療や予防目的の
医薬製剤を含む概念である。本発明に係る生理活性物質
は、遺伝子工学的手法で調製された抗体または酵素であ
ってもよい。本発明に係る生理活性物質は、遺伝子工学
的手法で調製された蛋白質またはペプチドであってもよ
い。
インスリン、カリクレイン、アプロチニン、プロラクチ
ン、胎盤性性腺刺激ホルモン、黄体ホルモン、トランス
フォーミング因子類(TGFα、TGFβ、TGFγ
等)、血小板由来因子(PDGF)、繊維芽細胞成長因
子(FGF)、組織プラスミノーゲンアクチベータ(t
−PA)、その他刺激因子やサイトカイン類などのホル
モン/蛋白質/サイトカイン;ウロキナーゼ、チトクロ
ームC、リボヌクレアーゼ、パパイン、キモトリプシ
ン、ペプシン、トリプシンなどの酵素;プラスミン、ト
ロンビン、アンチトロンビンなどの凝固因子;アデノシ
ントリフォスフェート(ATP)、ヌクレオチド、核酸
などの核酸関連物質;セフェム系・ペニシリン系・テト
ラサイクリン系・クロラムフェニコール系・ポリペプチ
ド系などの抗生物質;その他プロスタグランジンなどの
生体成分などが挙げられる。
存中に経時的に変化して、その活性を著しく低下するこ
とが多く、実際の使用に際しては、前述のようにゼラチ
ンあるいはコラーゲン由来の成分を安定化剤として添加
する場合が多い。それら生理活性物質に本発明に係る非
抗原性安定化剤を安定化剤として用いることで、アナフ
ィラキシーなどの副作用を引き起こすことのない生理活
性物質などの製剤化が可能となる。
ゲナーゼ酵素を用いて特異的に分解して得られるペプチ
ドであって、アミノ酸配列が(Gly−X−Y)n のも
のであっても、分子量が20,000を越えるものでは
抗原性が現れるおそれがある。本発明に係る非抗原性安
定化剤は、分子量が20,000以下である。従って、
同じ原料から分子量が1,000以下のものより高収率
で製造することができる。本発明に係る非抗原性安定化
剤は、非抗原性を高めるため、望ましくは、分子量が1
0,000以下である。本発明に係る非抗原性安定化剤
の、分子量が20,000以下でアミノ酸配列が(Gl
y−X−Y)n のペプチドのうち、分子量が10,00
0以下のものの比率は、90%以上であることが好まし
い。

ことを特徴とする。本発明に係る生理活性物質は、好ま
しくは、非抗原性安定化剤を0.01〜10重量%含有
する。しかしながら、非抗原性安定化剤の生理活性物質
への添加量は、対象となる生理活性物質の種類および剤
型などにより異なるので、一様には定まらない。

ニトール、サッカロース、乳糖、グルタミン酸ソーダ、

る。

に、牛や豚を始めとした動物由来の新鮮な骨、皮、鍵あ
るいは軟骨などを原料として調製されたコラーゲンない
しはゼラチンを用いることができる。その際、コラーゲ
ンないしはゼラチンの精製度合いは高い方が望ましい
が、使用するコラーゲン酵素の純度や特異性が優れてい
たり、また、酵素処理後に純化のための工程を組み込む
ことができる場合には、それらの精製度合いは特に問わ
ない。むしろ、これら出発原料の選択基準は、製造され
る非抗原性安定化剤の用途との関係が重要であり、それ
に合わせて適宜選択することが望ましい。
際に酵素分解に用いるコラゲナーゼ酵素には、Clostrid
ium histolyticum, Streptomyces parvulus などの細
菌、放線菌あるいは真菌など由来で、コラーゲン特有の
アミノ酸配列:(Gly−X−Y)n のグリシンのアミ
ノ基側を特異的に切断する酵素を用いることができる。
また、このコラゲナーゼ酵素は、これらの酵素遺伝子を
遺伝子工学的に特定のベクターに組み込んで、乳酸菌や
酵母などの他の菌体あるいは動物に産生させて得られた
遺伝子組み替えによる酵素で、類似の基質特異性を有す
るコラゲナーゼ酵素であってもよい。
分解する前の素材の抗原性を一部なりとも保持している
というのが一般的な考え方である。事実、後述する実施
例で示すように、ゼラチンやコラーゲンを酸や加熱によ
って処理して調製した分子量500〜20,000のゼ
ラチン分解物の場合、ゼラチンとしての抗原性の1/1
5〜1/50程度が残存していた。また、酸や加熱によ
る分解方法ではなく、一般的なプロテアーゼ酵素など、
例えば、ペプシン、トリプシン、パパイン、キモトリプ
シン、パンクレアチンあるいはアクチナーゼなどを、単
独であるいは2種類以上混合して酵素分解処理したゼラ
チン分解物の場合も、ゼラチンとしての抗原性が1/5
0〜1/200程度残存していた。このようにゼラチン
の抗原性を失わせるには単純に分解を行って低分子化を
図れば良いということではなく、抗原性の消失とゼラチ
ンの分解方法との間には密接な関係があるといえる。
うえで重要な点は、使用するコラゲナーゼ酵素の純度で
ある。通常、各種菌体から調製されたコラゲナーゼ酵素
には、他の蛋白質分解酵素(プロテアーゼ)が混入して
いることがある。この不純酵素が多く含まれると、原材
料中のゼラチン成分あるいはコラーゲン成分以外の蛋白
質も分解されてしまったり、ゼラチンあるいはコラーゲ
ン成分自身もその不純酵素によって非特異的に分解され
てしまったりする。その場合、精製されてくる非抗原性
安定化剤の品質が低下してしまい、ひいてはアナフィラ
キシーを誘発させる原因となる可能性が高い。従って、
使用するコラゲナーゼ酵素の純度については、その基質
特異性とともに十分注意を払う必要がある。

して使用しても良い。また、コラゲナーゼ酵素による酵
素分解の方法として、(a)バッチ法、(b)カラム法
あるいは(c)これらを組み合わせた方法を用いること
ができる。これら(a)〜(c)の方法による製造ライ
ンと、使用するコラゲナーゼ酵素の形態とは、それぞれ
自由に組み合わせることができる。本発明に係る非抗原
性安定化剤の製造は、特開平7-82299 号に記載の方法に
準じて実施することができるが、他の方法でも可能であ
る。むしろ、原料となるゼラチン成分やコラーゲン成分
に応じた方法を選択したり、コラゲナーゼ酵素の特異性
や純度などを維持するのに適した方法を選択することが
好ましい。
化剤の製造方法の一例を挙げれば、以下の実施例で示す
ように、ゼラチン成分またはコラーゲン成分を出発原料
とし、酵素の回収率を良くするために固定化コラゲナー
ゼ酵素を用いたバッチ法やカラム法によるバイオリアク
ター方式で製造することができる。すなわち、コラゲナ
ーゼ酵素を物理的吸着法あるいは化学結合法によって各
種の担体、例えば、キトパールなどに結合させ、クロマ
トグラフィー用のカラムに充填し、一定温度下、可溶化
させたゼラチン溶液あるいは分解が生じない程度の温
度、望ましくは40〜45℃で変性させたコラーゲンを
カラムに通して酵素分解をさせる。この際のコラーゲン
原料の送液速度は、固定化酵素の活性および必要とされ
る分解度合いに応じて適宜選択される。

活性物質に使用する方法は、すでに公知となっている安
定化剤の生理活性物質への使用方法に準じて実施するこ
とができる。
50gを1,000 mlの20mM Tris-HCl 緩衝液(pH 7.4)/
0.1M NaCl に加温しながら溶解後、50℃に冷却した。酵
素分解用の固定化酵素は、100mg のコラゲナーゼ酵素
(ワシントン社製、type IVから精製した高純度品) を5
0g のキトパール (富士紡績社製) に2 架橋試薬を用い
て結合させて調製した。担体への結合量は、結合前後の
280nm における吸光度の変化を計測して算出したが、99
%以上の結合率であった。使用時、本固定化酵素をカラ
ムとカラムの間にpHセンサーを設置した2連式のカラム
式バイオリアクターに充填し、20mM Tris-HCl 緩衝液(p
H 7.4)/0.1M NaCl で良く洗浄・平衡化を行った。pHの
測定システムは、この2連式カラムのカラム間に設置し
たpHセンサーが変化を感知して、繋いであるチューブか
ら濃厚Tris緩衝液が流入するシステムとなっている。
上記工程で調製した縦型2連式のコラゲナーゼ酵素固定
化カラムにアプライし、カラム法による酵素分解を行っ
た。この間、流速は毎分50〜80mlに、また、カラムの温
度は39±1 ℃にコントロールした。最終 (2連目)のカ
ラムから出てきた酵素反応終了液を分取し、0.45 μm
のフィルターで濾過を行った。
で粉末化した後、ゲル濾過器 (商品名:スーパーデック
ス G-30、ファルマシア社製)を用いたゲル濾過法、あ
るいはODS カラム (ワイエムシー社製) を用いた逆相ク
ロマトグラフィー法によって、分子量1,000 以下あるい
は分子量約20,000以下のペプチド組成物をそれぞれ精製
した。この分子量1,000 以下あるいは分子量約20,000以
下のペプチド組成物は、本発明の実施例の非抗原性安定
化剤である。
クロマトグラフィー:HPLC (島津製作所社製LC-10A、カ
ラム:スーパーデックスpeptide)で測定した。各試料
は、HPLCカラムへインジェクションする前に、0.2 μm
のメンブレンフィルターで濾過を行った。各ペプチド組
成物、すなわち、分子量約20,000以下のペプチド画分お
よび分子量1,000 以下のペプチド画分について、平均分
子量、分子量範囲、分子量500 以下のペプチドの割合を
表1にまとめた。この結果から、分子量約20,000以下の
ペプチド画分を製造する場合、分子量1,000 以下のペプ
チド画分を製造するのに比べて、収率が向上することが
わかる。
0,000以下のペプチド組成物および分子量1,000 以下の
ペプチド組成物を凍結乾燥した後、それぞれのNH2 末端
アミノ酸およびNH2 末端から2番目のアミノ酸について
エドマン分解法によって検定した。その結果、両者のNH
2 末端側のアミノ酸はそれぞれ95.7%および97.5%がグ
リシンであることが判明し、本発明に係る非抗原性安定
化剤に特徴的なアミノ酸配列である(Gly−X−Y)
n 構造を保持していることが証明された。以下、実施例
で得られたペプチド組成物の抗原性/アレルゲン性の消
失に関する検定方法およびその結果について示す。
牛皮および豚皮由来のゼラチンをPBS に溶解して2mg /
mlとし、0.22μm のフィルターで濾過を行った溶液をフ
ロイント完全アジュバントと等量ずつ混和してエマルジ
ョンを調製し、3羽のウサギに1mlずつ注射した。その
3週間後、同じペプチド組成物の溶液を等量のフロイン
ト不完全アジュバントと共に混和してエマルジョンを作
成し、同様にウサギに注射した。この操作を3回繰り返
し、最終免疫後1週間目に抗血清を得た。
牛皮および豚皮由来のゼラチンをPBS に溶解して2mg /
mlとし、0.22μm のフィルターで濾過滅菌を行った溶液
に水酸化アルミニウム (Alum) で沈澱させ、洗浄したも
の100 μg /mlに調製し、3羽のモルモットの皮内に1
mlずつ注射した。その4週間後、同様にして追加免疫を
行った3〜5日後に抗血清を得た。
橋試薬で活性化されたアミノ化ポリスチレンボール(住
友ベークライト社製)の官能基(NH2 基) に牛皮あるい
は豚皮由来のゼラチンを固定化させ、牛血清アルブミン
あるいは界面活性剤などでプロッキング操作を行ったゼ
ラチン感作イムノボールを作成した。
ールに、実施例で得られた分子量約20,000以下のペプ
チド組成物(実施例の非抗原性安定化剤)、実施例で
得られた分子量1,000 以下のペプチド(実施例の非抗原
性安定化剤)、加熱分解して得られた分子量200 〜7,
000 の部分分解ゼラチン(比較例)、トリプシンとペ
プシンで酵素分解させた分子量500 〜12,000の酵素分解
ゼラチン(比較例)およびゼラチン(比較例)、を各
200 μl ずつ添加し、次いで、前記IgG タイプのウサギ
ゼラチン抗血清あるいはIgE タイプのモルモットゼラチ
ン抗血清のいずれかを200 μl 加えて37℃で30分間反応
させて、抗血清とゼラチン抗原との反応系における〜
各成分の競合反応を行わせた。次いで、洗浄し、山羊
抗ウサギIgG 抗体の西洋ワサビパーオキシダーゼ (HRP)
標識複合体 (コスモ・バイオ) あるいは山羊抗モルモッ
トIgE 抗体の西洋ワサビパーオキシダーゼ (HRP)標識複
合体 (コスモ・バイオ) を2次反応させた。37℃で1時
間反応後、イムノボールに結合して残っているHRP 標識
複合体の活性を測定した。
ルトフェニレンジアミン塩酸塩(OPD)及び0.02%過
酸化水素を含む活性測定用の「基質溶液」と、37℃で20
分間反応させた後、希硫酸液で反応を停止させ、492nm
の波長の吸光度を分光光度計( 日本分光社製、V-550)で
測定して行った。HRP 標識複合体の活性の測定によっ
て、〜各成分の抗原性およびアレルゲン性をその競
合反応の程度から検討した。その結果を表2に示す。表
2の結果から、およびの実施例の非抗原性安定化剤
は、阻害率が0%であり、〜の比較例のものが抗原
性を有するのに対して抗原性を有しないことがわかる。

の1/2希釈系列(1/10, 1/20, 1/40, 1/80, 1/160)を
作成し、各々の各希釈血清50μl を背毛を刈ったSD系
ラット(オス、8週齢)2匹ずつ(計4匹)の背部皮内
に注射した。24時間後、このうちの1匹に、実施例で
得られた分子量約20,000以下のペプチド組成物(実施例
の非抗原性安定化剤)の1mgを含む0.6 %エバンスブル
ー溶液1.0ml を尾静脈より注射した。また、ペプチド
組成物に対する陽性コントロールとして、2匹目のSD
系ラットにも同様に牛ゼラチンの1mgを含む0.6 %エバ
ンスブルー溶液1.0 mlを尾静脈より注射した。60分後、
4匹とも屠殺し、背部皮膚を剥いで紫斑を観察し、それ
らの大きさを測定した。判定は、紫斑径が10ml以上を
(++) または (+++) 、9mm〜5mmを (+) 、4mm
〜1mmを (±) とし、紫斑が生じない場合を (−) とし
た。その結果を表3に示す。表3の結果から、牛ゼラ
チンの場合、1/160 の希釈系列でも紫斑が生じるのに
対し、実施例で得られた分子量約20,000以下のペプチ
ド組成物の場合には、1/10の希釈系列でも紫斑が生じ
ず、抗原性を有しないことがわかる。
てアレルギー症状を呈している患者6名(表4中の患者
A〜F)から採血された血清(ゼラチン特異IgE を含
む) および実施例7に示したゼラチン感作イムノボール
を用いて、ゼラチン特異IgE 抗体を測定する蛍光酵素免
疫測定法を実施する際に、蛍光酵素免疫測定法で陽性と
なった患者の特異IgE 抗体の力価が実施例の非抗原性安
定化剤によってどの程度阻害されるかについて、標識酵
素により分解された蛍光基質の蛍光強度を測定すること
によって判定した。蛍光強度の測定は、蛍光光度計(日
本分光社製 FP777)を用い、励起波長495nm、蛍光波長5
20nm で行った。なお、ゼラチン特異IgE 抗体を検出す
るための第2抗体としてはマウス抗ヒトIgE 抗体のβガ
ラクトシダーゼ標識体を使用し、その酵素活性は蛍光基
質を用いて検定した。その結果を表4に示す。
分子量約20,000以下のペプチドおよび分子量約20,000以
下のペプチド(実施例の非抗原性安定化剤)は、阻害反
応による抗体力価 (蛍光強度) の低下が観察されず、ゼ
ラチン特異IgE 抗体とは全く反応性を有しないことが判
明した。これに対し、もとの原料であるゼラチン、加熱
分解ゼラチンあるいは非特異的なプロテアーゼで分解し
たゼラチンでは強い阻害が生じ、ゼラチン特異IgE 抗体
と良く反応することが示され、アナフィラキシーの原因
の一端が明らかになった。本実施例による試験を行うこ
とによって、I型アレルギーを誘導するゼラチン特異Ig
E と各ペプチド組成物が反応性をもっているか否かを知
ることができる。
分子量20,000以下のペプチド(実施例の非抗原性安定化
剤)あるいはマンニトールを下記処方に従い、ウロキナ
ーゼに添加後、凍結乾燥を行ってウロキナーゼ製剤を調
製した。〔処方〕ウロキナーゼ……160,000iu 、実施例
で得られたペプチド(実施例の非抗原性安定化剤)……
0gあるいは0.8g、マンニトール……0gあるいは0.2g。
これらは注射用蒸留水で全量100ml とする。
は、ウロキナーゼ製剤凍結乾燥品を生理食塩水で溶解
し、30℃で0時間、4時間、24時間および48時間放置し
た後、残存活性率をウロキナーゼ測定法二段法〔参照:
医薬品研究 5, 295, 1974, ウロキナーゼ製剤の力価測
定〕によって測定した。その結果を表5に示す。表5の
結果から、実施例で得られたペプチド(実施例の非抗原
性安定化剤)を添加した場合、添加しない場合やマンニ
トールのみを添加した場合に比べて、ウロキナーゼ製剤
の安定性を高めることがわかる。
ウロキナーゼ凍結乾燥品を45℃で保存し、一定期間 (1
カ月〜3カ月)後のウロキナーゼ残存活性を試験4と同
様に測定し、実施例で得られた分子量20,000以下のペプ
チド(実施例の非抗原性安定化剤)の安定化効果を検定
した。その結果を表6に示す。表6の結果から、実施例
で得られた分子量20,000以下のペプチド(実施例の非抗
原性安定化剤)を添加した場合、添加しない場合やマン
ニトールのみを添加した場合に比べて、ウロキナーゼ凍
結乾燥品の安定性を高めることがわかる。
れた分子量20,000以下のペプチドおよび対照となる他の
安定化剤を下記処方に従い、遺伝子工学で生産したイン
ターフェロン−α(コスモ・バイオ社製、比活性107U
/mg)に添加後、凍結乾燥を行ってインターフェロン−α
製剤を調製した。〔処方〕インターフェロン−α……1
06U、実施例で得られた分子量20,000以下のペプチド
(実施例の非抗原性安定化剤)……0%あるいは0.2
%、他の安定化剤……0%あるいは0.2 %。これらは注
射用蒸留水で全量200mlとする。
約1カ月保存後、生理食塩水で溶解し、30℃で30分間あ
るいは4時間放置した後、残存するインターフェロンの
活性を測定した。残存活性率を検定し、その結果を表7
に示す。表7の結果から、実施例で得られた分子量20,0
00以下のペプチド(実施例の非抗原性安定化剤)を添加
した場合、添加しない場合やヒト血清アルブミン以外の
他の安定化剤のみを添加した場合に比べて、インターフ
ェロン−αの安定性を高めることがわかる。
ば、抗原性を消失していると同時にゼラチンあるいはコ
ラーゲンのアミノ酸配列の特性を有していることから、
アナフィラキシー反応を誘発させず、治療用・予防用の
生理活性物質の安定化剤として有用である。また、本発
明に係る非抗原性安定化剤によれば、従来の非抗原性ペ
プチド組成物に比べて分子量範囲を広げることができ、
収率を向上させることができる。
Claims (6)
- 【請求項1】ゼラチン成分またはコラーゲン成分をコラ
ゲナーゼ酵素を用いて特異的に分解して得られる、分子
量が20,000以下でアミノ酸配列が(Gly−X−
Y)n のペプチドを主成分とすることを特徴とする非抗
原性安定化剤。 - 【請求項2】ゼラチン成分またはコラーゲン成分をコラ
ゲナーゼ酵素を用いて特異的に分解して得られるペプチ
ド組成物であって、分子量が20,000以下であり、
アミノ酸配列が(Gly−X−Y)n のペプチドを70
%以上含有するものから成ることを特徴とする非抗原性
安定化剤。 - 【請求項3】前記ペプチドを85%以上含有することを
特徴とする請求項2記載の非抗原性安定化剤。 - 【請求項4】請求項1,2または3記載の非抗原性安定
化剤を0.005〜15重量%含有することを特徴とす
る生理活性物質。 - 【請求項5】ウイルス、ワクチン、各種サイトカインま
たは抗生物質であることを特徴とする請求項4記載の生
理活性物質。 - 【請求項6】遺伝子工学的手法で調製された蛋白質また
はペプチドであることを特徴とする請求項4記載の生理
活性物質。
