Abstract
Description

この比較例は、官能性一級-二級アルコールデヒドロゲナーゼ(secAdh)を有する微生物、及び二官能性アルデヒド-アルコールデヒドロゲナーゼ(adhE1+adhE2)遺伝子座位でのアセトン経路(thlA、ctfAB、adc)のノックインを説明する。一級-二級アルコールデヒドロゲナーゼ(secAdh)は、この株がアセトンではなくイソプロパノールを生産するように、アセトンをイソプロパノールに変換する。
この実施例は、budA座位での他の遺伝子又は経路の組み入れについて説明する。
Claims (22)
- アセトラクテートデカルボキシラーゼ遺伝子座位でのDNAのノックインを含む、遺伝子操作された微生物。
- 前記DNAが、前記アセトラクテートデカルボキシラーゼ遺伝子のコード領域を置き換える、請求項1に記載の微生物。
- 前記DNAが、アセトラクテートデカルボキシラーゼプロモータを置き換えない、請求項1に記載の微生物。
- 前記微生物が、2,3-ブタンジオールを生産しない、請求項1に記載の微生物。
- 前記DNAが、1つ以上の酵素をコードする、請求項1に記載の微生物。
- 1つ以上の酵素が、前記微生物に対して非天然である、請求項1に記載の微生物。
- 1つ以上の酵素が、前記微生物に対して天然である、請求項1に記載の微生物。
- 1つ以上の酵素が、アセトラクテートデカルボキシラーゼプロモータの制御下にある、請求項1に記載の微生物。
- 前記DNAが、プロモータを含む、請求項1に記載の微生物。
- 前記1つ以上の酵素が、アセトラクテートデカルボキシラーゼプロモータ及び少なくとも1つの他のプロモータの両方の制御下にある、請求項5に記載の微生物。
- 1つ以上の酵素が、チオラーゼ、CoAトランスフェラーゼ、及びアセトアセテートデカルボキシラーゼ又はアルファ-ケトイソバレレートデカルボキシラーゼから選択されるデカルボキシラーゼを含む、請求項1に記載の微生物。
- 前記微生物が、アセトン及びイソプロパノールのうちの1つ以上を生産する、請求項11に記載の微生物。
- 前記微生物が、一級-二級アルコールデヒドロゲナーゼ遺伝子に破壊的変異を更に含む、請求項11に記載の微生物。
- 前記1つ以上の酵素が、1-ブタノール、ブチレート、ブテン、ブタジエン、メチルエチルケトン、エチレン、アセトン、イソプロパノール、脂質、3-ヒドロキシプロピオネート、テルペン、イソプレン、脂肪酸、2-ブタノール、1,2-プロパンジオール、1-プロパノール、1-ヘキサノール、1-オクタノール、コリスマート由来の生産物、3-ヒドロキシブチレート、1,3-ブタンジオール、2-ヒドロキシイソブチレート若しくは2-ヒドロキシイソブタン酸、イソブチレン、アジピン酸、1,3-ヘキサンジオール、3-メチル-2-ブタノール、2-ブテン-1-オール、イソバレレート、イソアミルアルコール、又はモノエチレングリコールの生産を可能にする、請求項5に記載の微生物。
- 前記微生物が、C1固定微生物である、請求項1に記載の微生物。
- 前記微生物が、Wood-Ljungdahl微生物である、請求項1に記載の微生物。
- 前記微生物が、細菌である、請求項1に記載の微生物。
- 前記微生物が、Acetobacterium、Alkalibaculum、Blautia、Butyribacterium、Clostridium、Eubacterium、Moorella、Oxobacter、Sporomusa、及びThermoanaerobacterから選択される属のメンバーである、請求項1に記載の微生物。
- ガス状基質の存在下で請求項1に記載の微生物を培養することを含む、生産物を生産する方法。
- 前記ガス状基質が、CO、CO2、及び/又はH2を含むC1炭素源を含む、請求項19に記載の方法。
