Abstract

Claims (22)
- 第1の外因性CoAトランスフェラーゼと、少なくとも1つの追加の外因性CoAトランスフェラーゼと、を含む、遺伝子操作されたWood-Ljungdahl微生物。
- 前記第1の外因性CoAトランスフェラーゼが、アセトラクテートデカルボキシラーゼ遺伝子のコード領域を置き換える、請求項1に記載の微生物。
- 前記少なくとも1つの追加の外因性CoAトランスフェラーゼが、アルデヒド-アルコールデヒドロゲナーゼ遺伝子のコード領域を置き換える、請求項1に記載の微生物。
- 外因性チオラーゼ、及びアセトアセテートデカルボキシラーゼ若しくはアルファ-ケトイソバレレートデカルボキシラーゼから選択される外因性デカルボキシラーゼ、又はこれらの任意の組み合わせを更に含む、請求項1に記載の微生物。
- 前記外因性チオラーゼ、及びアセトアセテートデカルボキシラーゼ若しくはアルファ-ケトイソバレレートデカルボキシラーゼから選択される前記外因性デカルボキシラーゼ、又はこれらの任意の組み合わせが、前記第1の外因性CoAトランスフェラーゼとともに機能するか、又は前記最小の1つの追加の外因性CoAトランスフェラーゼとともに機能する、請求項4に記載の微生物。
- 前記外因性CoAトランスフェラーゼが、Clostridium acetobutylicum CtfA及びCtfB、又はClostridium beijerinckii CtfA及びCtfBである、請求項1に記載の微生物。
- 前記外因性CoAトランスフェラーゼが、前記微生物に対して非天然である、請求項6に記載の微生物。
- 前記外因性CoAトランスフェラーゼが、前記微生物に対して天然である、請求項6に記載の微生物。
- 前記外因性CoAトランスフェラーゼが、同じである、請求項6に記載の微生物。
- 前記外因性CoAトランスフェラーゼが、異なる、請求項6に記載の微生物。
- 前記外因性CoAトランスフェラーゼが、アセトラクテートデカルボキシラーゼプロモータ及び少なくとも1つの他のプロモータの両方の制御下にある、請求項6に記載の微生物。
- 前記微生物が、2,3-ブタンジオールを生成しない、請求項1に記載の微生物。
- 前記微生物が、アセトン及びイソプロパノールのうちの1つ以上を生成する、請求項1に記載の微生物。
- 一級-二級アルコールデヒドロゲナーゼ遺伝子、チオエステラーゼ遺伝子、3-ヒドロキシブチリルcoAデヒドロゲナーゼ遺伝子、又はこれらの任意の組み合わせにおける破壊的変異を更に含む、請求項1に記載の微生物。

- 前記微生物が、C1固定微生物である、請求項1に記載の微生物。
- 前記微生物が、細菌である、請求項1に記載の微生物。
- 前記微生物が、Acetobacterium、Alkalibaculum、Blautia、Butyribacterium、Clostridium、Eubacterium、Moorella、Oxobacter、Sporomusa、又はThermoanaerobacterから選択される属のメンバーである、請求項1に記載の微生物。
- ガス状基質の存在下で、請求項1に記載の微生物を培養することを含む、生成物の生成を増加させるための方法。
