Abstract
Description
i)Tim-3タンパク質結合性の上昇
ii)Th1及びTh17細胞分化の抑制
iii)ヘルパーT細胞(T helper cell)の抑制
iv)線維芽細胞様滑膜細胞(FLS;Fibroblast-like synoviocytes)の細胞死
v)破骨細胞(osteoclast)分化及び骨吸収の抑制
配列番号1のアミノ酸配列を有する組換え安定化ガレクチン9タンパク質をコードする遺伝子を含む発現ベクターを作製し、前記発現ベクターをheat shock法により大腸菌(E.coli)に導入した。大腸菌を50μg/ml kanamycin含有LB培地に培養し、600nmでの吸光度が0.7に達した時点でアラビノースを添加し、組換えタンパク質の発現を誘導することにより、組換えタンパク質を発現させた。次に、組換えタンパク質の発現を誘導した細胞を溶解及び濾過し、標的タンパク質を陽イオン交換法やaffinityカラムなどを用いて捕獲し、高純度の組換え安定化ガレクチン9タンパク質を高収率で得た。
濃度が0.5μg/mlの組換えヒトTim-3(R&D systems,Cat.No.10241-TI-050)を96ウェルELISAプレート(Nunc,Cat.No.44-2404-21)の各ウェルに100μlずつ入れ、常温で一晩反応させ、その後0.05%のツイーン20(Sigma Cat.No.P9416)を含むDPBS(Dulbecco's phosphate-buffered saline;Welgene,Cat.No.LB011-02)で3回洗浄した。洗浄後に、1%のウシ血清アルブミン(bovine serum albumin;BSA)を含むDPBSを各ウェルに200μlずつ入れ、常温で1時間培養し、次いで洗浄緩衝液(wash buffer)で3回洗浄した。3回の洗浄後に、<製造例1>で得られた組換え安定化ガレクチン9タンパク質(sGal-9)を各濃度(0、5、10及び20μg/ml)に希釈して各ウェルに100μlずつ入れ、常温で2時間培養し、次いで洗浄緩衝液で3回洗浄した。
PBS(Welgene,Cat.No.LB 001-02)で希釈した濃度が2μg/mlの抗CD3(eBioscience,Cat.No.16-0031-82)を96ウェルプレートに100μlずつまたは24ウェルプレートに500μlずつ分注し、4℃で一晩反応し、その後PBSで1回洗浄して細胞培養に用いた。CD4、CD62L及びT細胞でコーティングした96ウェルプレートまたは24ウェルプレートに分注し、37℃、5%CO2の条件で4日間培養し、その後Th1細胞への分化を誘導するために、抗IL-4、抗IL-2及び抗IL-12サイトカインを添加し、Th17細胞への分化を誘導するために、抗IFN-γ、抗IL-4、抗IL-2及び抗IL-6サイトカインを添加した。サイトカイン添加後に、濃度が1μg/mlのsGal-9を添加し、37℃、5%CO2の条件で4日間培養した。
FLS細胞(cell passage:RA_FLS 2-92#P7)を48ウェルプレート(SPL,Cat.No.30048)の各ウェルに5×104cells/mlの細胞密度で200μlずつ分注し、24時間後に各濃度(2.5、5及び10μl/ml)のsGal-9を各ウェルに100μlずつ入れた。16時間後に、上清を回収し、超低温冷凍庫(deep freezer)に保管した。
破骨細胞(osteoclast)を12ウェルプレート(Corning,Cat.No.3513)の各ウェルに2.5×105cells/mlの細胞密度で1mlずつ継代し、24時間後に各濃度(0.1、0.5、1、2、4及び10μg/ml)の実験群としてのsGal-9及び対照群としてのPBSを各ウェルに1mlずつ入れた。
破骨細胞(osteoclast)を生体内(in vivo)の骨構造に類似する無機3次元結晶(inorganic 3-dimensional crystalline)をコーティングしたOsteo Assay Surfaceマルチウェルプレート(Corning,Cat.No.3987)の各ウェルに2.5×105cells/mlの細胞密度で1mlずつ継代し、24時間後に各濃度(0.1、0.5、1、2、4及び10μg/ml)の実験群としてのsGal-9及び対照群としてのPBSを各ウェルに1mlずつ入れた。分化が終了するまで、実験群(sGal-9)、対照群(PBS)及び細胞培養液(MEM-alpha,gibco,Cat.No.12561-056)を毎日交換した。
Claims (18)
- 配列番号1で表されるアミノ酸配列を有する組換え安定化ガレクチン9タンパク質。
- 前記組換え安定化ガレクチン9タンパク質は、野生型ガレクチン9の糖鎖認識活性を保持することを特徴とする、請求項1に記載の組換え安定化ガレクチン9タンパク質。
- 請求項1に記載のタンパク質をコードする遺伝子を含む組換えベクター。
- 請求項3に記載の組換えベクターが挿入された形質転換体。
- 配列番号1で表されるアミノ酸配列を有する組換え安定化ガレクチン9タンパク質またはそれをコードするポリヌクレオチドを有効成分として含む骨疾患予防または治療用薬学的組成物。
- 前記組換え安定化ガレクチン9タンパク質は、次の少なくとも1つの特性を有することを特徴とする、請求項5に記載の骨疾患予防または治療用薬学的組成物。
i)Tim-3タンパク質結合性の上昇
ii)Th1及びTh17細胞分化の抑制
iii)ヘルパーT細胞(T helper cell)の抑制
iv)線維芽細胞様滑膜細胞(FLS;Fibroblast-like synoviocytes)の細胞死
v)破骨細胞(osteoclast)分化及び骨吸収の抑制 - 前記骨疾患は、関節炎、骨粗鬆症、人工関節周囲骨溶解(periprosthetic osteolysis)、インプラントの摩耗粉による骨溶解、パジェット病(Paget's disease)、骨軟化症、くる病、骨減少症、カルシウム調節異常、転移性骨癌(metastatic bone cancer)、炎症性骨喪失、内分泌疾患または薬物による二次性骨喪失(secondary bone loss)、及び歯槽骨の破壊を伴う歯周疾患からなる群から選択される少なくとも1つであることを特徴とする、請求項5に記載の骨疾患予防または治療用薬学的組成物。
- 前記関節炎は、骨関節炎、変形性関節症、関節リウマチ、剥離性骨軟骨炎、関節靭帯損傷、半月板損傷、関節の不整列、無血性壊死及び小児特発性関節炎からなる群から選択されることを特徴とする、請求項7に記載の骨疾患予防または治療用薬学的組成物。
- 前記骨疾患は、関節リウマチまたは骨粗鬆症であることを特徴とする、請求項5に記載の骨疾患予防または治療用薬学的組成物。
- 前記薬学的組成物は、薬学的に許容される担体をさらに含むことを特徴とする、請求項5に記載の骨疾患予防または治療用薬学的組成物。
- 前記薬学的組成物は、経口投与用、筋肉内投与用、静脈内投与用、腹腔内投与用、皮下投与用、皮内投与用または局所投与用に製剤化されることを特徴とする、請求項5に記載の骨疾患予防または治療用薬学的組成物。
- 配列番号1で表されるアミノ酸配列を有する組換え安定化ガレクチン9タンパク質を有効成分として含む骨疾患予防または改善用保健機能食品。
- 配列番号1で表されるアミノ酸配列を有する組換え安定化ガレクチン9タンパク質またはそれをコードするポリヌクレオチドを薬学的に有効な量で個体に投与するステップを含む骨疾患治療方法。
- 配列番号1で表されるアミノ酸配列を有する組換え安定化ガレクチン9タンパク質またはそれをコードするポリヌクレオチドを薬学的に有効な量で個体に投与するステップを含む骨疾患予防または改善方法。
- 骨疾患の予防または治療に用いるための配列番号1で表されるアミノ酸配列を有する組換え安定化ガレクチン9タンパク質またはそれをコードするポリヌクレオチドを含む薬学的組成物。
- 骨疾患の予防または改善に用いるための配列番号1で表されるアミノ酸配列を有する組換え安定化ガレクチン9タンパク質またはそれをコードするポリヌクレオチドを含む保健機能食品。
