Abstract

本開示は全般に、HIVの予防および治療のための改善された抗HIV抗体に関する。本開示は全般に、抗HIV抗体のヒスチジン変異にも関する。特に、本開示は、HIVの予防および治療のためのヒスチジン変異抗HIV抗体に関する。
HIV-1の侵入は、ウイルスエンベロープ(Env)糖タンパク質gp120とT細胞受容体CD4のドメイン1(D1)との相互作用によって引き起こされる。イバリズマブ(iMab)は、広範にHIV-1を中和する強力なAbであり(Jacobson et al., Antimicrob. Agents Chemother. 53:450-457, 2009; Kuritzkes et al., J. Infect. Dis. 189:286-291, 2004)、宿主T細胞上のCD4受容体の、主としてドメイン2(D2)に結合し、それによってHIVがT細胞内への侵入を達成し感染を成立させるためにCD4受容体を使用する能力を阻害することによって、HIVを中和する(Burkly et al., J. Immunol. 149:1779-178, 1992)。最近試験された初代分離株の大きなパネル(118のEnvシュードタイプ化ウイルス)において、イバリズマブは、50%感染抑制で全ウイルスの92%を中和し、90%感染抑制でウイルスの47.4%を中和した。イバリズマブは広範なHIV分離株を強力に抑制することができるが、依然多くのHIVバリアントがイバリズマブの抑制活性から逃れることができる。HIVタイプ1エンベロープの可変領域5におけるアスパラギン結合グリコシル化部位の欠如がイバリズマブ耐性に関連することが最近報告された(Toma et al., J. Virology 85(8): 3872-2880, 2011; Pace et al., J. Acquir. Immune Defic. Syndr. Epub ahead of print: September 2012)。
本発明は、抗HIV抗体の活性を、該抗体の軽鎖および重鎖の可変領域におけるヒスチジン変異によって高めるための新規アプローチを提供する。
本発明の前記のおよび他の特徴、ならびにそれを得るおよび用いる方法は、以下の説明によってより明らかになり、よく理解されうる。
「抗原」とは、1つ以上のエピトープを含み、免疫学的反応を引き起こすことのできる分子をいう。「抗原分子」も、一般的な意味で、本書に開示されるグリカン修飾タンパク質の結合標的である分子をさして用いられる。
本書に開示される組換え抗体を含む医薬組成物を、該抗体を1つ以上の任意の薬学的に許容しうる担体と混合することによって調製することができる。薬学的に許容しうる担体は、溶媒、分散媒体、等張剤等を包含する。担体は、液体、半固体、例えばペースト、または固体の担体でありうる。担体の例は、水、塩類液もしくは他の緩衝剤(例えばリン酸、クエン酸緩衝液)、油、アルコール、タンパク質(例えば血清アルブミン、ゼラチン)、炭水化物(例えば、グルコース、スクロース、トレハロース、マンノース、マンニトール、ソルビトールまたはデキストリンを包含する、単糖、二糖および他の炭水化物)、ゲル、脂質、リポソーム、樹脂、多孔性マトリックス、結合剤、賦形剤、コーティング剤、安定剤、保存剤、アスコルビン酸およびメチオニンを包含する抗酸化剤、EDTAといったキレート剤;ナトリウムといった塩形成対イオン;TWEEN(商標)、PLURONICS(商標)またはポリエチレングリコール(PEG)といった非イオン性界面活性剤、またはその組み合わせを包含する。
さらなる側面において、本書に開示されるヒスチジン変異抗体は、場合により薬学的に許容しうる担体中に提供されて、対象におけるHIV感染の治療および予防ならびにHIV伝染の予防のために使用される。
pH依存的CD4結合活性を仲介することのできるイバリズマブCDR残基の特定
pH依存的抗原結合イバリズマブバリアントを特定するために、システマティックにイバリズマブVHおよびVL遺伝子において各CDR残基をヒスチジンに変異させ、各バリアントを発現させ、精製し、HRP標識野生型イバリズマブを用いる競合ELISAによって、各バリアントのpH7.4、6.0および5.0におけるCD4結合活性を野生型と比較して評価した。目的は、pH7.4におけるCD4結合活性の低下が最小限でありながら、pH7.4におけるCD4結合と比較してpH6.0におけるCD4結合(pH6.0/pH7.4)を最も大きく低下するイバリズマブのヒスチジンバリアントを特定することであった。pH5.0におけるpH依存的CD4結合活性のインビボ妥当性は明らかではないが、pH依存的CD4結合を仲介するヒスチジンバリアントの特定を、特に結合親和性に対する効果が小さいと期待された単一残基の寄与の評価に際して、より良く行うために、pH5.0の評価も含めた。
pH依存的CD4結合活性を仲介する最適なヒスチジン組み合わせの特定
重鎖の残基Y53a、D58、K96、D97、N98またはT100aならびにE61およびK64におけるヒスチジン変異の組み合わせがpH依存的CD4結合を改善しうるかを決定するために、pH依存的CD4結合活性が最も大きかった4つの軽鎖変異(L30、Y92、S93およびY94)を用いて、単一コンストラクトにおける軽鎖変異4つまでの組み合わせの効果を評価した。図3に示すように、個別に1.5のpH5.0/pH7.4差をもたらしたS93とY94の組み合わせは、3.0のpH5.0/pH7.4差をもたらした。S93/Y94二重変異体に、単独で1.7のpH5.0/pH7.4差をもたらしたY92を追加すると、pH5.0/pH7.4差は3.4に改善された。同様に、Y92H/S93H/Y94H三重バリアントに、単独で1.4のpH5.0/pH7.4差をもたらしたL30変異を追加すると、pH5.0/pH7.4差は5.7にさらに改善された。したがって、個別にpH依存的CD4結合活性を仲介するヒスチジン変異を単一分子中に組み合わせると、より大きいpH依存が仲介された。しかしながら、複数のヒスチジン変異の組み合わせは残念ながら、pH7.4におけるCD4結合活性に対しては悪影響を及ぼした。実際、組み合わせバリアントによるこの結合親和性の低下は、個々のバリアントの合計よりも実質的に大きかった。観察された組み合わせバリアントの結合親和性低下の線形回帰は、個々の変異の相乗(P=0.0008)対相加(P=0.06)効果に基づき期待された結合性低下に、より良くフィットした(図3参照)。したがって、pH7.4での十分なCD4結合親和性を維持するために、本発明者は、単一のVHヒスチジン変異と単一のVLヒスチジン変異の最適な組み合わせを特定することに焦点を絞った。
表面プラズモン共鳴によるpH依存的CD4結合活性の確認
BIACORE T3000光バイオセンサー(GE Healthcare, Piscataway, N.J.)を用いて結合親和性分析を行った。イバリズマブおよび全てのバリアントの固定を標準的な方法に従って行った。
薬物動態分析
アカゲザルにおける野生型イバリズマブ(Wt、青線)およびバリアントHC Y53a/LC Y94(His、緑線;およびHis FcRn、赤線)の薬物動態を図6Aに示す。図6Bには、各抗体を0.75mg/kgで投与されたアカゲザルにおける、野生型イバリズマブ(Wt、青線)およびバリアントHC Y53a/LC Y94(His、緑線;およびHis FcRn、赤線)によるCD4占拠時間を示す。各線は、各アカゲザルにおいて観察されたCD4占拠を示す。
ダウンモジュレーションの分析
図7に示すように、イバリズマブのヒスチジン変異バリアントは野生型イバリズマブよりも、CD4受容体ダウンモジュレーションを誘導する程度が小さかった。
