1.1 AAVコンストラクト
AAV2-ITR隣接ベクター骨格を有するプラスミドは、ヒトCMV-IE遺伝子、続いてニワトリベータアクチンプロモーターの切断型を含んでいた。これらは、全長コドン最適化ヒトppdyn配列(AA配列番号1)、あるいは変異体(AA配列番号2;配列番号3)又は切断変異体(ΔGFP)のいずれか又はEGFPの全長形式の発現を駆動する。遺伝子発現は、ウッドチャック肝炎ウイルス翻訳後エンハンサーエレメント(WPRE)、続いてウシ成長ホルモンに由来するpoly A+シグナル配列によって増強された。AAV2 ITRは、ssDNAゲノムを有するAAVベクターを得るために、その野生型立体配置であるか、あるいは、dsDNAゲノムを有する自己相補的AAVベクターが生じるように、1つのITRが切断された(図1)。
HEK 293細胞を、5%FCSを含むDMEMに25~33%の密集度で播種した。細胞を、リン酸カルシウムのコトランスフェクションによって24時間後にトランスフェクトした。AAVベクターは、本質的に他に記載されたように産生した(Mietzsch、Grasse et al.、2014)。簡潔には:AAV rep、cap、Ad5ヘルパー遺伝子のためのプラスミド、及び上述したppdyn(配列番号1)又はその変異体(配列番号2;配列番号3)を発現するrAAVのためのプラスミドを用いる、2プラスミド(pDG)コトランスフェクションプロトコル。培地を、FCS含有量が減少した(2%)培地によって12時間後に交換した。培養物を、細胞溶解のために3回の凍結溶解サイクルでトランスフェクションの72時間後に回収した。粗溶解物を、250U/mlのベンゾナーゼ(Merck)を用いて37℃で1時間消化して、インプット及びパッケージングされていないプラスミドDNAを分解し、8,000gで30分間遠心分離して、細胞残屑をペレット化した。
rAAVベクターを、血清型1又は2カプシドにパッケージングし、そして、ベンゾナーゼ処理した清澄凍結融解上清から、1ステップヘパリンセファロースクロマトグラフィーによって、又はAVBセファロースアフィニティークロマトグラフィーによって、AKTA精製機(GE Healthcare)で1mlの予め充填されたHiTrapカラムを用いて、以下のようにして精製した:凍結溶解上清を、1mMのMgCl2及び2.5mMのKClで補充した1・リン酸緩衝生理食塩水(PBS)(1・PBSMK)中で1:1希釈した後に、0.5ml/分でカラムにロードした。カラムを、1ml/分の速度で20mlの1・PBS-MKで洗浄した。AAVベクターを、1ml/分の速度で0.1Mの酢酸ナトリウム及び0.5MのNaCl pH2.5を用いて溶出し、1/10容量の1M Tris-HCl pH10を用いて直ちに中和する。精製したrAAV調製物を、Slide-A-Lyzer透析カセット(10,000 MWCO;Thermo Scientific)を用いて1xPBS-MKに対して透析した(Mietzsch、Grasse et al.、2014)。
高度に精製されたrAAVベクター調製物を、プロテイナーゼKを用いて2時間56℃で消化した。DNAを、フェノール及びクロロホルムで繰り返し抽出することによって精製し、エタノールで沈殿させた。カプシド放出AAVゲノムの連続希釈物を、定量的Light-Cycler PCRによって、Fast Start DNA Master SYBR Green kit(Roche)を用いて分析した。PCRプライマーは、ベクター骨格のウシ成長ホルモン遺伝子由来polyA部位に特異的であった(5’-CTAGAGCTCGCTGATCAGCC-3’及び5’-TGTCTTCCCAATCCTCCCCC-3’)。高度に精製されたAAV調製の力価を、AAV-DNA含有ゲノム粒子(gp)/mlとして測定した(Mietzsch、Grasse et al.、2014)。
動物
本研究では、C57BL/6N野生型及びpDynノックアウト(pDyn-KO)マウスを調査した。pDyn-KOマウスを、10世代にわたってC57BL/6Nバックグラウンド上に戻し交配した(Loacker et al.、2007)。飼育及び維持のために、マウスを食餌及び水に自由にアクセスできるようにして群に分けた。温度を23℃及び60%湿度に固定し、12時間の明暗サイクル(午前7時間から午後7時まで点灯)とした。動物に関わる全ての手順は、実験及び他の科学目的のために使用される脊椎動物の保護のための欧州条約ETS番号:123に従った、オーストリア動物実験倫理委員会、及びカナダ動物管理委員会によって、承認された。使用される動物の数を最小限にするためにあらゆる努力が成された。
カイニン酸注入及び電極移殖
雄マウス(12~14週)をケタミン(160 mg/kg、i.p.;Graeub Veterinary Products、Switzerland)で鎮静させ、次いで精密気化器(precise vaporizer)(Midmark、USA)を通してセボフルランで深く麻酔した。マウスに、以前に記載されたように(Loacker et al.、2007)、50nlの20mMのKA溶液を海馬に注入した(RC -1.80mm;ML+1.80mm;DV -1.60mm)。2つの電極(1つは皮質電極及び1つは深部電極)を、KA投与直後に埋め込んだ。エポキシラート被覆タングステン深部電極(直径250μm;FHC、USA)を、CA1領域に向けて海馬に配置した(RC -1.80mm;ML+1.80mm;DV -1.60mm)。表面電極は、運動皮質の上部の頭蓋骨内に配置した金めっきネジであり(RC+1.70mm;ML+1.6mm、ブレグマ(bregma)を基準点として)、異常なEEG活動の発生をモニタリングした。追加の表面電極を、基準及び参照として小脳上に配置した。電極を歯科用アクリレート(Heraeus Kulzer GmbH、Germany)で所定の位置に固定した。
rAAV注入
rAAV投与を必要とする実験のために、ガイドカニューレを移植し、これは海馬深部電極に取り付けられた。全ての動物は、麻酔処置として手術の20分前にメロキシカム(2mg/kg)を受けた。rAAV注入のために、動物を、注入の間(20分)セボフルランチャンバー内で穏やかに麻酔した。注入は、注入ポンプを有するガイドカニューレを介して、0,1μL/分の流量で行われ、2μLの総容量を注入した。
EEG記録及び分析
EEGは、無線記録装置(Neurologger、TSE、Germany)を用いて得られ、SciWorks Software(Datawave Technologies、USA)を用いて自動的に分析された。EEGを、70ms未満続く高振幅放電(>3×ベースライン)として定義されるてんかん様スパイクについてフィルタリングした。スパイクトレイン(Spike trains)を、1Hzよりも高い頻度でかつ少なくとも1秒間続く少なくとも3つのスパイクの発生として定義した(図2参照)。より低い頻度のスパイクは発作間スパイクとしてカウントした。長期海馬発作性放電(hpd)を、最小で10秒間続くスパイクトレインとして定義した(Zangrandi et al.、2016)。海馬深部及び運動皮質表面電極における高電圧EEG異常についての共出現によって、全般発作を視覚的に評価した(図3参照)。
空間記憶テスト(バーンズ迷路)
ナイーブ及び処置動物の学習及び記憶を評価するために、バーンズ迷路を使用した。バーンズ迷路は、その外周に20個の穴が開いた平らな丸テーブル(直径100cm)上で、60ルクスで行われた。それらのうち、1つのみがマウスを迷路から退避の暗い箱へと脱出させる。視覚的手がかりを90°の間隔でディスクの周りに置いた。初日は馴化日であり、標的の穴を開けて、マウスを5分間迷路を自由に探索させた。退避箱の位置は実験全体を通して一定に保った。マウスが穴を見つけると、箱は閉じられ、動物は2分間そこに維持されて、退避箱を安全な場所として関連付けさせた。動物が5分間の間に標的の穴を見つけられなかった場合、動物はやさしく穴へと導かれた。次の4日間に習得を行った。最大3分の試験を3回行い、マウスが穴を見つけるとすぐに、マウスを2分間内側に保持した。3分後にマウスが穴を見つけられなかった場合、マウスをやさしく穴へと導いた。動物によって成された主要なエラー及び穴を見つけるための潜時を計算し、学習基準として定義した。プローブ試験を6日目に行った。全ての穴を閉じ、マウスを5分間迷路を自由に探索させた。評価のためにボードを四分円に分割し、各四分円に費やされた時間を測定した(図5参照)。以前に開かれた退避穴を含んでいた四分円は、Q1と呼ばれ、これはQ2及びQ4に隣接し、Q3はQ1に対向している。
微量透析
微量透析を、微量透析実験の3週間前に上述のように一方の海馬にrAAV-pDyn注入を受けたpDyn-KO動物に対して行った。ベクター注入の時点で(RC -1.80mm;ML+1.80mm;DV -1.60mm)、動物に刺激電極(RC -4.20mm;ML+3.20mm;DV -4.90mm)、及びrAAV注入海馬の門を標的とするガイドカニューレを埋め込んだ。微量透析のために、MAB-2プローブ(SciPro、Sanborn、NY)を海馬内に配置し、人工CSF(140mM NaCl;3.0mMのKCl;1.25mMのCaCl2;1.0mMのMgCl2;1.2mMのNa2HPO4;0.3mMのNaH2PO4;3mMのグルコース、pH 7.2)によって0.4μL/分の流量で洗い流した。ACSFを、3x25分、続いて25分の低頻度刺激について集めた(300μA;10秒間隔で孤立した0.3msec方形波、ISO-STIM 01D、NPI、Tamm、Germany)。さらに25分のベースライン後、25分の高頻度刺激を行った(150μA;1秒間に20 msec離れた0.3msec方形波のトレイン;トレインは10秒によって分離された)。
ダイノルフィンB酵素免疫測定法(EIA)
微量透析実験の溶出液中の処理されたダイノルフィンBの含有量を、特定のDyn B(配列番号8)EIA(S-1429;Peninsula;San Carlos、CA)によって、製造業者のマニュアルに従って測定した。簡潔には、試料を抗血清と共に1時間インキュベートし、Bt-トレーサーと一晩のインキュベートに続く。2日目に、ストレプトアビジン-HRPを、EIA緩衝液を用いて5回洗浄した1時間後に添加した。さらに5回の洗浄後に、試料を5分間TMB溶液と反応させ、次に、プレートリーダーで450nmで分析した。Dyn B(配列番号8)含有量を、並行して行った較正試料に基づいて分析し、ng/mlとして表す(図4参照)。
統計分析
取得後に、pClamp 10.3(Molecular devices)を用いて電気生理学的記録を観察し分析した。Mac用のPrism 5(version 5.0f)を使用して、インビボ実験の統計分析を行い、数値を作成した。統計分析のために、Dunnett事後検定を用いた一元配置分散分析(one-way ANOVA)をインビボ実験に適用した。電気生理学のために、両側の対応のあるt検定を、得られた膜電位分析に適用し、一元配置分散分析を使用して、IPSCに対する薬物効果を比較した。0.05未満のp値を有意であるとみなした。データは、平均値±平均値の標準誤差(SEM)として表される。

発作閾値は、発作を起こしやすいことの尺度である。発作誘導剤又は刺激に対する抵抗性は、動物における読み出し(readout)として使用される。齧歯類の尾静脈へのGABAA受容体アンタゴニストであるペンチレンテトラゾールの注入は、発作閾値を測定するために認められた方法である。物質又は処置の抗けいれん作用は、適用時の発作閾値の増大によって実証され得る。pDyn欠損マウスは、野生型マウスよりも低い発作閾値を示す。これは、κオピオイド受容体アゴニストによって救うことができる。
Claims (20)
- プレプロダイノルフィン又はプレプロダイノルフィン変異体をコードするDNA配列を含む送達ベクターであって、前記送達ベクターが、プレプロダイノルフィン又はプレプロダイノルフィン変異体であるプレプロペプチドの発現を駆動し、ここで、前記プレプロペプチドが、シグナルペプチドを含み、かつ一方で、前記プレプロダイノルフィン又はプレプロダイノルフィン変異体が、以下の群から選択される配列:
a.配列番号7(配列番号1;ppDynのAA 207-223)であるDyn A、あるいは最初の13AA(N末端から最初の)からなるその変異体、又は最初の8AA(N末端から最初の)からなるその変異体、
b.配列番号8(配列番号1;ppDynのAA 226-238)である、Dyn B、
c.配列番号9(配列番号1;ppDynのAA 226-254)である、ロイモルフィン、
d.配列番号7に係るDyn Aの変異体であって、配列番号7のN末端から数えて最初の8AA(YGGFLRRI)内で少なくとも60%のアミノ酸配列同一性を有する、すなわち、配列番号7に含まれる配列YGGFLRRI内で少なくとも60%のアミノ酸配列同一性を有する、変異体、
e.配列番号8に係るDyn Bの変異体であって、配列番号8のN末端から数えて最初の8AA(YGGFLRRQ)内で少なくとも60%のアミノ酸配列同一性を有する、すなわち、配列番号8に含まれる配列YGGFLRRQ内で少なくとも60%のアミノ酸配列同一性を有する、変異体、
f.配列番号9に係るロイモルフィンの変異体であって、配列番号9のN末端から数えて最初の8AA(YGGFLRRQ)内で少なくとも60%のアミノ酸配列同一性を有する、すなわち、配列番号9に含まれる配列YGGFLRRQ内で少なくとも60%のアミノ酸配列同一性を有する、変異体、
の少なくとも1つを含む、送達ベクター。 - 前記変異体が、それぞれ、配列番号7のN末端から数えて最初の8AA(YGGFLRRI)、配列番号8のN末端から数えて最初の8AA(YGGFLRRQ)又は配列番号9のN末端から数えて最初の8AA(YGGFLRRQ)内で、少なくとも70%のアミノ酸配列同一性を有する、請求項1に記載の送達ベクター。
- 前記変異体が、それぞれ、配列番号7、配列番号8又は配列番号9のN末端から数えて最初の8AA内で、少なくとも80%のアミノ酸配列同一性を有する、請求項1又は請求項2に記載の送達ベクター。
- 請求項1から3のいずれか一項に記載の送達ベクターであって、前記送達ベクターが、プレプロダイノルフィン又はプレプロダイノルフィン変異体であるプレプロペプチドを駆動し、ここで、前記プレプロペプチドが、シグナルペプチドを含み、かつ一方で、前記送達ベクターが、プレプロダイノルフィン変異体をコードするDNA配列を含み、前記プレプロダイノルフィン変異体をコードするDNA配列が、以下の群から選択される変異体の配列:
a.配列番号10(YGZFLRRZRPKLKWDNQ)、
b.配列番号11(YGZFLRRZFKVVT)、
c.配列番号12(YGZFLRRZFKVVTRSQEDPNAYSGELFDA)、
の少なくとも1つを含み、
ここで、Zは、任意のアミノ酸を表し、かつ、a.;b.又はc.に係る配列内の少なくとも1つのZは、好ましくは、a.;b.又はc.に係る配列による前記ダイノルフィン断片の野生型配列と比較した場合に、別のアミノ酸によって置換されている、送達ベクター。 - 前記送達ベクターが、さらに、組み換えアデノ随伴ウイルス(AAV)ベクターゲノム又は組み換えレンチウイルスゲノムを含む、請求項1から4のいずれか一項に記載の送達ベクター。
- 組み換えアデノ随伴ウイルス(AAV)ベクターゲノムを含む、請求項1から5のいずれか一項に記載の送達ベクターであって、前記ベクターがヒト血清型ベクターであり、前記ヒト血清型ベクターが、血清型1、2、3、4、5、6、7、8、9、10、rh10、11、12、13、14、蛇行性(serpentine)AAV、祖先AAV、又はそれらに由来するAAVカプシド突然変異体を含む群から選択され、好ましくは血清型1又は2である、送達ベクター。
- 請求項1から6のいずれか一項に記載の送達ベクターを含む、組み換えウイルス粒子又はリポソーム。
- 前記送達ベクターが、さらに組み換えアデノ随伴ウイルス(rAAV)ベクターゲノムを含み、かつ前記rAAVベクターゲノムが、AAVカプシドにカプシド形成されているか、あるいは、前記送達ベクターが、さらに組み換えレンチウイルスベクターゲノムを含み、かつレンチウイルス粒子にパッケージングされている、請求項7に記載の組み換えウイルス粒子又はリポソーム。
- 中枢神経系の細胞に核酸を送達する方法であって、プレプロダイノルフィン又はプレプロダイノルフィン変異体をコードするDNA配列を前記細胞に導入するのに十分な条件下で、前記細胞と、請求項1から8のいずれか一項に記載の送達ベクター又は組み換えウイルス粒子又はリポソームとを接触させること、を含む、方法。
- 医薬としての使用のための、請求項1から8のいずれか一項に記載の送達ベクター又は組み換えウイルス粒子又はリポソーム。
- 対象の焦点性てんかん(focal epilepsy)、特に内側側頭葉てんかんの治療における使用のための、又は、焦点性てんかんを患う対象のてんかん発作の予防における使用のための、請求項1から8のいずれか一項に記載の送達ベクター又は組み換えウイルス粒子又はリポソームであって、前記送達ベクター又は組み換えウイルス粒子又はリポソームが、てんかん原性焦点(epileptogenic focus)におけるヒトκオピオイドレセプターの活性化を提供し、それによって発作を阻害する、送達ベクター又は組み換えウイルス粒子又はリポソーム。
- 対象の焦点性てんかん、特に内側側頭葉てんかんの治療における使用のための、又は、焦点性てんかんを患う対象のてんかん発作の予防における使用のための、請求項1から8のいずれか一項に記載の送達ベクター又は組み換えウイルス粒子又はリポソームであって、前記送達ベクター又は組み換えウイルス粒子又はリポソームが、てんかん原性焦点におけるヒトκオピオイドレセプターの活性化を提供し、それによって発作を阻害し、前記送達ベクター又は組み換えウイルス粒子又はリポソームが、てんかん原性焦点におけるヒトκオピオイドレセプターに対するアゴニスト作用を有するペプチドのオンデマンド放出(on-demand release)をもたらす、送達ベクター又は組み換えウイルス粒子又はリポソーム。
- 請求項10から12のいずれか一項に記載の、対象の焦点性てんかん、特に内側側頭葉てんかんの治療における使用のための、又は、焦点性てんかんを患う対象のてんかん発作の予防における使用のための、請求項1から8のいずれか一項に記載の送達ベクター又は組み換えウイルス粒子又はリポソームであって、前記ベクター又は組み換えウイルス粒子が、末梢投与あるいは頭蓋内又は脳内又は髄腔内(intrathecal)又は実質内投与に適している、送達ベクター又は組み換えウイルス粒子又はリポソーム。
- 請求項10から13のいずれか一項に記載の、対象の焦点性てんかん、特に内側側頭葉てんかんの治療における使用のための、又は、焦点性てんかんを患う対象のてんかん発作の予防における使用のための、請求項1から8のいずれか一項に記載の送達ベクター又は組み換えウイルス粒子又はリポソームであって、前記送達ベクター又は組み換えウイルス粒子又はリポソームが、脳内適用され、好ましくは焦点適用(applied focal)される、送達ベクター又は組み換えウイルス粒子又はリポソーム。
- 請求項1から8のいずれか一項に記載の送達ベクター又は組み換えウイルス粒子又はリポソーム、及び任意により薬学的に許容される担体を含有する、医薬リリースオンデマンド組成物(pharmaceutical release-on-demand composition)。
- 請求項1から8のいずれか一項に記載の送達ベクター又は組み換えウイルス又はリポソーム粒子により、好ましくはインビトロ又はエクスビボで、感染した細胞。
- 焦点性てんかん、特に内側側頭葉てんかんを有する対象を治療する方法、又は、焦点性てんかんを患う対象のてんかん発作を予防する方法であって:
対象に、請求項1から8のいずれか一項に定義された送達ベクター、組み換えウイルス粒子、又は医薬組成物を投与すること、を含み、好ましくは前記送達ベクター又は組み換えウイルス粒子又はリポソームは、プレプロペプチドをコードし、前記プレプロペプチドは、成熟及び放出後に、てんかん原性焦点におけるヒトκオピオイドレセプターの活性化を提供し、それによって発作を阻害し、ここで、好ましくは前記送達ベクター又は組み換えウイルス粒子又はリポソームは、脳内適用され、好ましくは焦点適用される、方法。 - 請求項1から6のいずれか一項に記載の送達ベクターのいずれかに由来するヒトκオピオイドレセプター(KOR)に対するアゴニスト作用を有するペプチドであって、好ましくは前記ペプチドが、配列番号10、配列番号11、配列番号12、配列番号13、配列番号14及び配列番号15を有するペプチドを含む群から選択される、ペプチド。
- ヒトκオピオイドレセプター(KOR)に対するアゴニスト作用を有するペプチドであって、前記ペプチドが、配列番号10、配列番号11、配列番号12、配列番号13、配列番号14及び配列番号15を有するペプチドを含む群から選択される、ペプチド。
- 請求項18又は19に記載のペプチドを含有する、医薬リリースオンデマンド組成物。
