Abstract
Description
上述したように、組織マトリックスの表面近傍のコラーゲンを部分的に変性させるのに利用可能な様々な方法がある。一実施例として、無細胞ブタ真皮の表面を、60Wで5000Hzの周波数および7.5cm/sの速度を使用してEPILOG(登録商標)Legend XL120 CO2レーザへの曝露後に、部分的に変性させた。その後、得られた組織500を、未処理の無細胞ブタ真皮(図5A)と比較してコラーゲン変性の程度(図5B)を評価するために、マッソンのトリクロームを用いて組織染色の処理を行った。図示のように、レーザ処理は、マトリックスの表面から有限の距離のところでコラーゲンの部分的な変性を可能にし、それにより、下方にあるマトリックスと密接に接触しかつそれに接続された変性表面510の形成を可能にする。
Claims (35)
- 組織製品の製造方法であって、
コラーゲン含有組織マトリックスを含む第1の材料を選択するステップと、
前記第1の材料の一部分を処理して、その部分の表面付近のコラーゲンを部分的に変性させるステップと、
コラーゲンを部分的に変性させるために処理された部分を、トランスグルタミナーゼ酵素を含む組成物と接触させるステップと、
トランスグルタミナーゼを含む組成物と接触した部分を、コラーゲン含有組織マトリックスを含む第2の材料と物理的に接触させるステップとを備えることを特徴とする方法。 - 請求項1に記載の方法において、
コラーゲン含有組織マトリックスを含む少なくとも1の追加の材料を選択するステップと、
少なくとも1の追加の材料の一部分を処理して、少なくとも1の追加の材料の一部分の表面付近のコラーゲンを部分的に変性させるステップと、
コラーゲンを部分的に変性させるために処理された少なくとも1の追加の材料の一部分を、トランスグルタミナーゼ酵素を含む組成物と接触させるステップと、
トランスグルタミナーゼを含む組成物と接触した少なくとも1の追加の材料の一部分を、第1の材料または第2の材料と物理的に接触させるステップとをさらに備えることを特徴とする方法。 - 請求項1または2に記載の方法において、
トランスグルタミナーゼ酵素を含む組成物が、水溶液を含むことを特徴とする方法。 - 請求項1乃至3の何れか一項に記載の方法において、
コラーゲン含有組織マトリックスを含む材料が、粒子状であることを特徴とする方法。 - 請求項1乃至3の何れか一項に記載の方法において、
コラーゲン含有組織マトリックスを含む材料が、シートの形態であることを特徴とする方法。 - 請求項1乃至5の何れか一項に記載の方法において、
コラーゲン含有組織マトリックスを含む材料が、無細胞組織マトリックスであることを特徴とする方法。 - 請求項6に記載の方法において、
コラーゲン含有組織マトリックスが、皮膚、筋膜、脂肪、心膜組織、硬膜、臍帯組織、胎盤組織、心臓弁組織、靭帯組織、腱組織、動脈組織、静脈組織、神経結合組織、膀胱組織、尿管組織、筋肉および腸組織から選択される組織に由来する組織から生成されることを特徴とする方法。 - 請求項6に記載の方法において、
コラーゲン含有組織マトリックスが、筋肉由来の組織から生成されることを特徴とする方法。 - 請求項6に記載の方法において、
コラーゲン含有組織マトリックスが、筋肉組織マトリックスを含むことを特徴とする方法。 - 請求項1に記載の方法において、
前記第1の材料および前記第2の材料の各々が無細胞組織マトリックスのシートを含み、前記第1の材料および前記第2の材料が互いに接触されて多層シートがもたらされることを特徴とする方法。 - 請求項10に記載の方法において、
前記多層シートが無細胞筋マトリックスを含むことを特徴とする方法。 - 請求項1乃至11の何れか一項に記載の方法に従って製造された組織製品。
- 治療方法であって、
コラーゲン含有組織マトリックスを含む材料を選択するステップと、
材料の一部分を処理して、その部分の表面近くのコラーゲンを部分的に変性させるステップと、
コラーゲンを部分的に変性させるために処理された部分を、トランスグルタミナーゼ酵素を含む組成物と接触させるステップと、
トランスグルタミナーゼを含む組成物と接触した部分を、組織部位と物理的に接触させるステップとを備えることを特徴とする方法。 - 請求項13に記載の方法において、
トランスグルタミナーゼ酵素を含む組成物が、水溶液を含むことを特徴とする方法。 - 請求項13または14に記載の方法において、
コラーゲン含有組織マトリックスを含む材料が、シートの形態であることを特徴とする方法。 - 請求項13乃至15の何れか一項に記載の方法において、
コラーゲン含有組織マトリックスを含む材料が、無細胞組織マトリックスを含むことを特徴とする方法。 - 請求項16に記載の方法において、
コラーゲン含有組織マトリックスが、皮膚、筋膜、脂肪、心膜組織、硬膜、臍帯組織、胎盤組織、心臓弁組織、靭帯組織、腱組織、動脈組織、静脈組織、神経結合組織、膀胱組織、尿管組織および腸組織から選択される組織に由来する組織から生成されることを特徴とする方法。 - 請求項16に記載の方法において、
コラーゲン含有組織マトリックスが、筋肉由来の組織から生成されることを特徴とする方法。 - 請求項16に記載の方法において、
コラーゲン含有組織マトリックスが、筋肉組織マトリックスを含むことを特徴とする方法。 - 治療方法であって、
コラーゲン含有組織マトリックスを含む材料を選択するステップであって、材料の表面に近接する材料の一部分が部分的に変性したコラーゲンを含む、ステップと、
部分的に変性したコラーゲンを含む部分を、トランスグルタミナーゼ酵素を含む組成物と接触させるステップと、
トランスグルタミナーゼを含む組成物と接触した部分を、組織部位と物理的に接触させるステップとを備えることを特徴とする方法。 - 請求項20に記載の方法において、
トランスグルタミナーゼ酵素を含む組成物が、水溶液を含むことを特徴とする方法。 - 請求項20または21に記載の方法において、
コラーゲン含有組織マトリックスを含む材料が、シートの形態であることを特徴とする方法。 - 請求項20乃至22の何れか一項に記載の方法において、
コラーゲン含有組織マトリックスを含む材料が、無細胞組織マトリックスを含むことを特徴とする方法。 - 請求項23に記載の方法において、
コラーゲン含有組織マトリックスが、皮膚、筋膜、脂肪、心膜組織、硬膜、臍帯組織、胎盤組織、心臓弁組織、靭帯組織、腱組織、動脈組織、静脈組織、神経結合組織、膀胱組織、尿管組織および腸組織から選択される組織に由来する組織から生成されることを特徴とする方法。 - 請求項23に記載の方法において、
コラーゲン含有組織マトリックスが、筋肉由来の組織から生成されることを特徴とする方法。 - 請求項23に記載の方法において、
コラーゲン含有組織マトリックスが、筋肉組織マトリックスを含むことを特徴とする方法。 - 請求項20乃至26の何れか一項に記載の方法において、
部分的に変性したコラーゲンを含む一部分を、トランスグルタミナーゼ酵素を含む組成物と接触させるステップ、並びに、トランスグルタミナーゼを含む組成物と接触した部分を、組織部位と物理的に接触させるステップが、
組織部位をトランスグルタミナーゼと接触させること、および、
部分的に変性したコラーゲンを含む材料の表面を、組織部位と接触させることによって行われることを特徴とする方法。 - 組織治療製品であって、
実質的に無傷の無細胞コラーゲン組織マトリックスを有する無細胞組織マトリックスと、部分的に変性したコラーゲンを含む表面領域とを備え、部分的に変性したコラーゲンが、実質的に無傷の無細胞コラーゲン組織マトリックスと同じ組織から形成されるコラーゲンマトリックスを含み、部分的に変性したコラーゲンが、無傷の組織マトリックスの天然コラーゲン骨格から形成されるとともに、無傷の組織マトリックスに結合されていることを特徴とする製品。 - 請求項28に記載の製品において、
コラーゲン含有組織マトリックスを含む材料が、粒子状であることを特徴とする製品。 - 請求項28または29に記載の製品において、
コラーゲン含有組織マトリックスを含む材料が、シートの形態であることを特徴とする製品。 - 請求項28乃至30の何れか一項に記載の製品において、
コラーゲン含有組織マトリックスが、皮膚、筋膜、脂肪、心膜組織、硬膜、臍帯組織、胎盤組織、心臓弁組織、靭帯組織、腱組織、動脈組織、静脈組織、神経結合組織、膀胱組織、尿管組織および腸組織から選択される組織に由来する組織から生成されていることを特徴とすることを特徴とする製品。 - 請求項28乃至30の何れか一項に記載の製品において、
コラーゲン含有組織マトリックスが、筋肉由来の組織から生成されることを特徴とする製品。 - 請求項28乃至30の何れか一項に記載の製品において、
コラーゲン含有組織マトリックスが、筋肉組織マトリックスを含むことを特徴とする製品。 - 組織治療製品であって、
コラーゲン含有無細胞組織マトリックスを含む第1の材料と、
コラーゲン含有無細胞組織マトリックスを含む少なくとも1の第2の材料とを含み、
第1の材料および第2の材料が、グルタミン残基のガンマ-カルボキシアミド基と第一級アミノ酸との間に形成された結合によって互いに接合されていることを特徴とする製品。 - 組織製品を製造する方法であって、
コラーゲン含有組織マトリックスを含む第1の材料を選択するステップと、
第1の材料の一部分を処理して、第1の材料の一部分の表面近くのコラーゲンを部分的に変性させ、それによってコラーゲン含有組織マトリックスに接続される部分的に変性されたコラーゲンの部分を形成するステップとを備えることを特徴とする方法。
