大な障害となる。EBVはまた、種々の血液癌及び非血液癌における腫瘍促進にも関与し
得る。また、EBVと鼻咽頭癌との間には強い相関性がある。したがって、EBV特異的
T細胞による予防的治療は、現在の療法に代わる優れた方法を提供する。

iTope(商標)及びTCED(商標)による分析を取り入れた。
型結合溝内の特異的結合ポケット(特にポケット位置;p1、p4、p6、p7及びp9
)と、ペプチドのアミノ酸側鎖との間の有利な相互作用を予測する。これらの対立遺伝子
は、世界的に見られる最も一般的なHLA-DR対立遺伝子を代表するものであり、何ら
かの特定の民族集団で最も優勢である対立遺伝子に起因する重み付けはしていない。20
個の対立遺伝子は「開状態」のp1構成を含み、14個は83位のグリシンがバリンで置
換されている「閉状態」の構成を含む。重要な結合残基の配置は、試験用タンパク質配列
にわたって1個のアミノ酸だけが重複する9量体ペプチドをin silico生成して
達成する。物理的なMHCクラスII結合実験と比較して、iTope(商標)を使用す
ると、MHCクラスII分子に結合するペプチドと結合しないペプチドとを高精度で良好
に識別できることがわかっている。in silicoのMHCクラスII結合アッセイ
に限界があれば、TCED(商標)を使用して緩和する。TCEDには、EpiScre
en(商標)のex vivoT細胞エピトープマッピングアッセイで事前にスクリーニ
ングされた配列に由来するペプチド(10,000超のペプチド)の大規模データベース
配列が含まれている。よってTCED(商標)を使用すると、無関係の抗体及びタンパク
質の配列に対して、あらゆる試験配列を検索し、実際のex vivoでの免疫原性との
相関性を見出すことができる。
対立遺伝子各々に対して試験したリンカー配列にわたって重複する9量体を用いて実施し
た。各9量体を、MHCクラスII分子との潜在的な「適合性」及び相互作用に基づいて
スコア付けした。ソフトウェアによって算出されるペプチドのスコアは0から1の間であ
る。高い平均結合スコア(iTope(商標)のスコア関数で0.55超)を得た非生殖
細胞系列ペプチドが強調表示され、50%以上のMHCクラスII結合ペプチド(すなわ
ち、34個の対立遺伝子のうち17個)が高い結合親和性(スコア0.6超)を有する場
合、このようなペプチドを「無差別な高親和性」MHCクラスII結合ペプチドと定義し
た(これは、CD4+T細胞エピトープを含むため高リスクであると考えられる)。50
%以上のMHCクラスII結合ペプチドがスコア0.55超を有する(ただし、0.6超
が過半数ではない)ペプチドは、「無差別な中親和性」MHCクラスII結合ペプチドと
定義した。配列の更なる分析はTCED(商標)を使用して実施した。先のEpiScr
een(商標)試験でT細胞応答を刺激した無関係なタンパク質から、ペプチド(T細胞
エピトープ)間の高配列相同性を同定するため、この配列を使用して、BLAST検索に
よりTCED(商標)を調べた。

ンカーについては、シグナル配列の切断部位から、リンカーを経て、HLA-A*02:

ついては、HLA-A*02:01のα3ドメイン末端の8個のアミノ酸から、リンカー
配列を経て、抗CD28 VHドメインの最初の8個のアミノ酸までの配列を分析した。

は中親和性(4位にp1アンカーを有する)である、2つの無差別なMHCクラスII結
合配列を含むことが見出された。C末端リンカーは、11位にp1アンカーを有する1つ
の無差別な中親和性のMHCクラスII結合ペプチドを含むことが見出された。
細胞エピトープデータベース(TCED(商標))のBLAST検索を実施し、先に特定
したエピトープとの相同性を判定した。TCED(商標)を使用して、EpiScree
n(商標)T細胞エピトープマッピングアッセイで試験した無関係の配列由来のペプチド
の大規模データベース(10,000超のペプチド)に対して、あらゆる試験配列を検索
した。リンカー配列のいずれにも、TCED(商標)で何の「ヒット」も含まないことが
わかった。
、リンカーへの配列変化を評価した。HLA-A*02:01のN末端が、pBFKsr
ベクターに設けられたシグナル配列またはその天然のシグナル配列のいずれかに直接融合
されるように、N末端リンカーを完全に除去できたことが確認された。これにより、融合
タンパク質のN末端がヒト生殖細胞系列配列のみを含み、T細胞エピトープのリスクを回
避できることが保証されたことになる。以下の推奨リンカー配列は、MHCクラスII結
合をバックグラウンド残存レベル(9量体のいずれかが結合した対立遺伝子が5未満)に
低減し、クローニングに適した制限部位をもたらすことが見出された(ただし、いずれの
配列もベクターの修飾を必要とする)。
GeneOptimizer(登録商標)を使用してコドンを最適化し、最適化した配
列を以下に示すようにクローニングして発現させた。

Biacore親和性データ及びその他の考察に基づいて、HC1::LC3及びHC
2::LC3の重鎖と軽鎖の組み合わせを、それぞれ第1及び第2のmAb候補として選
抜し、StableFast-NS0細胞株を作製した。
C3バイシストロニック発現ベクターの最終的なベクターマップを図14に示す。同じ手
法を用いて、pBFksr::HC2::LC3の構築を行った。
000万個の細胞(10x106)を45μgの線形(ΔPvuI)発現ベクターDNA
を用いてトランスフェクトした。増殖培地のバルク中で24時間、細胞を回復させた。回
復後、補充した無血清選択培地(コレステロール)中で細胞を洗浄し、選択培地で再懸濁
して、40x96ウェルプレートにウェル当たり200μLずつ分配した。実際の分配は
、HC1::LC3が1140細胞/ウェル、及びHC2::LC3が840細胞/ウェ
ルであった。プレートを37℃、5%CO2で1週間インキュベートし、フェノールレッ
ドを補充した選択培地を加えた。トランスフェクション後2週間で、多数のウェルが活発
に成長していくにつれて、培地の色が赤色から黄色へと変化した。
によりスクリーニングして、ヒトIgGの発現を検出した。HC2::LC3トランスフ
ェクションからは、合計612のウェルをスクリーニングした。IgGの濃度に基づいて
、それぞれ総量290及び101の細胞株を、HC1::LC3用とHC2::LC3用
の24ウェルプレートにスケールアップした。更なる分析のため、24時間の生産能アッ
セイを利用して最善な発現体を選別した。簡潔には、500μLの新鮮な培地中に5x1
05細胞となるように24ウェルプレートに播種した。24時間後、上清をELISA法
によりスクリーニングした。IgGの濃度に基づいて、それぞれ総量60及び24の細胞
株を、HC1::LC3用とHC2::LC3用の6ウェルプレートにスケールアップし
た。
潔には、1.5mLの新鮮な培地中に4x105細胞となるように6ウェルプレートに播
種した。3日後、細胞を計数して、上清をELISA法によりスクリーニングした。Ig
Gの濃度及び成長に基づいて、平均の比生産速度すなわちSPRをpg/細胞/日で算出
することができる。相対的SPRに基づいて、それぞれ総量20及び10の細胞株を、H
C1::LC3用とHC2::LC3用のT-75フラスコにスケールアップした。3日
間のSPRアッセイをT-75スケールで繰り返し、浮遊適応である最終細胞株を選別し
て、mAbを産生するためスケールアップした。
成長について再評価した。浮遊株をすべて堆積させた。mAbごとに最善に作用する細胞
株を小規模生産のため撹拌培養にスケール変更した。
3バイシストロニック発現ベクターを構築し、STABLEFAST-NS0細胞株を生
成した。ベクターマップを図15に示す。また、3つすべての融合タンパク質サブユニッ
ト(ヒトHLA-IgG4重鎖融合体、CD28軽鎖[LC3]、及びヒトβ2ミクログ
ロブリン)をコードするトリシストロニック発現ベクターのため、ヒトβ2ミクログロブ
リン遺伝子を含む発現カセット及びベクターも作製した。トリシストロニック構築物を図
16に示す。上清のELISA法及びウェスタンブロット分析により、一過性のHEK2
93培養に3つすべての遺伝子の発現が確認された。
CD28に対するヒト化モノクローナル抗体が、mAbをコーティングしたプレート上
で、新たに単離したPBMCの増殖を誘導する能力について試験した。図17に示すよう
に、ヒト化抗CD28は親と近似した機能を果たしており、スーパーアゴニストではない
。

以下の実施例は、分子量35KであるPLGA(LA:GA=1:1)から形成された
コアを有するナノ粒子の合成を示す。粒子のコロナは、PLGA-PEG-COOHまた
はPLGA-PEG-マレイミドと、PLGA-mPEG(メトキシPEG)の混合物か
らのPEGコポリマーによって形成する。COOH及びマレイミドの末端官能基により、
ポリペプチドのコンジュゲートが可能である。メトキシPEGは、PEG鎖上の末端官能
基に対して不活性である。PLGA部分は10~30K(例えば、約20K)の分子量を
有し、PEG部分の分子量は3K及び5Kである。この実施例では、ナノ粒子は、50%
のコアPLGA(35K)、及び25%のPLGA-PEG-COOHと25%のPLG
A-mPEGとの混合物により形成される。PLGA-PEG-COOH(またはマレイ
ミド)とPLGA-mPEGとの比率によって、粒子表面のリガンド密度の微調整が可能
である。例えば類似した分子量及び官能基密度を有する、PLA及びPLA-PEGなど
の他のポリマーを使用して類似した粒子を調製することができる。

定される微生物分解速度が2週間程度になるはずである。またタンパク質リガンドが外側
に配向されることで、同族のTCR及び共刺激受容体(例えばCD28、4-1BB)と
のT細胞の結合と比較して、必然的に最大限の生物学的活性をもたらす。更に、いくつか
の実施形態では、製剤化する際、ナノ粒子の疎水性コアに可溶性のペイロード(例えばI
L-2、抗TGF-b、IL-21または小分子薬)を充填することができる。

使用した。NI-19、NI-21、NI-22(Kbヒンジ二量体を含む)のバッチ、
ならびにNI-24及びNI-25(Dbヒンジ二量体を含む)の物理的特性を図32に
示す。NI-22は陰性対照である。
利用可能な一級アミンを介してコンジュゲートされるリガンドとともに、PLGA及び
PLGA-PEG-COOHのナノaAPCを調製した。PLGA 40(200nm)
を基とするPLGA粒子は、有意な安定性を示さなかった。PLGA-PEG-COOH
(40K/3K):PLGA-mPEG(17K/3K)を基とし、1:4のPEG:m
PEG比を有する粒子は、良好な安定性及び活性を示した。図20。

世代リガンドの部位特異的なチオールコンジュゲートによりナノaAPCを調製した。マ
ウス型とヒト化型両方のリガンドを使用して、ナノaAPCを調製した。ナノAPCは2
段階で調製した。まず、PLGA-mPEG及びPLGA-PEG-マレイミドからなる
粒子をナノ析出法によって調製する。例えば、PLGA-mPEGとPLGA-PEG-
マレイミドの混合物(PLGA-PEG-マレイミドの%w/wは1~55%の間で変化
する)をアセトニトリルに溶解して、最終濃度50mg/mLにする。この溶液を、シリ
ンジポンプを使用して5mL/分でPVA溶液(MnPVA=9kDa、0.5%w/v
)に撹拌しながら注入する。有機溶媒と水性溶媒の混合比は1:1~1:20の間で急変
させた。マイクロ流体装置、拘束衝突噴流装置及び多重注入口ボルテックスミキサー装置
を使用して、優れた均一性と狭い粒度分布を確保した粒子を調製することができる。粒子
は、タンジェンシャルフローフィルトレーション(TFF)またはAmicon遠心フィ
ルターを使用して精製する。次に粒子をコンジュゲート緩衝液(HEPES 50mM、
EDTA 10mM、pH=6.7)に再懸濁した。最後に、抗CD28及びKbSIY
/HLAリガンドの混合物を粒子にコンジュゲートすることにより、ナノaAPCを調製
する。リガンドの粒子とのコンジュゲートは、室温で一晩続行する。リガンド:粒子の質
量比は、1~500μg/mg粒子の範囲である。抗CD28:Kb/HLAの比は、0
~1の間で変化する。結合されなかったリガンドはSECまたはTFFを使用して除去す
る。90nmの平均粒径及び50~120nmの粒度分布を有する粒子を調製した。図3
2。
細胞の増殖を刺激する。更に、図23に示すように、PLGA-PEGベースのaAPC
は、凍結乾燥時に安定である。

- 免疫細胞活性化のためのポリペプチドリガンドを結合させたポリマーナノ粒子を含む人
工抗原提示細胞(aAPC)であって、該ナノ粒子が
PLGAまたはPLAポリマーコアと、
PEGポリマー部分がポリペプチドリガンドの末端との結合を有するPEGブラシから形
成される親水性シェルとを含む、前記aAPC。 - 前記親水性シェルが、同一のまたは異なる分子量を有するPEGブラシの混合物を更に
含む、請求項1に記載のaAPC。 - 前記ポリマーコアが、1:0~1:1の乳酸:グリコール酸比を基準とする、請求項1
または2に記載のaAPC。 - 前記ポリマーコアが、約1:1の乳酸:グリコール酸を有するPLGAである、請求項
3に記載のaAPC。 - 前記ポリマーコアがPLAである、請求項3に記載のaAPC。
- 前記コアポリマーが約10K~約50Kの分子量を有する、請求項1~5のいずれか1
項に記載のaAPC。 - 前記ポリマーナノ粒子が、PLGAもしくはPLAポリマーコア、ならびにPLGA-
PEG及び/またはPLA-PEGブロックコポリマーを含み、該PEG部分は親水性シ
